Effect of Magnolia Obovata Water-extracts on the Atherosclerosis Treated with High-cholesterol Diet in Rat (고(高)콜레스테롤 식이로 유발된 흰쥐 동맥경화에 Magnolia obovata가 미치는 영향)

  • Magnolia obovata exhibit several beneficial effects including anti-oxidant effects. Magnolia obovata is used as a therapeutic agent to stop hemorrhages and a tonic to promoted health in Korean and Chinese medicine. The pharmacokinetic profiles of the main Magnolia obovata is still not accurately investigated. In present study, I examined the effects of water extracts of Magnolia obovata on atherosclerosis induced high cholesterol diet in rats in serum and abdominal aorta. A total of 3-week old 9 male rats of Sprague-Dawley were divided into 3 groups and fed with the basal diet (normal group), high cholesterol diet (atherosclerosis induced group) for 8 weeks, high cholesterol diet supplemented with water extracts of Magnolia obovata (Magnolia obovata group) for 2 weeks. And rats were sacrificed, serum lipid level, abodominal aortic anti-oxidant activities and lipid peroxide were measured. These results indicated that serum total cholesterol, LDL-cholesterol, triglycerides concentration significantly lower in Magnolia obovata group than high cholesterol diet group. But HDL-cholesterol concentration had significantly higher in Magnolia obovata group than high cholesterol diet group. In conclusion, Magnolia obovata controls of lipid level of hyperlipidemia and hypercholesterolemia, thus suppresses the proliferation of intima in endothelial cells, which is risk factors of atherosclerosis induced high cholesterol diet in rats. I consider as Magnolia obovata is worth to using as atherosclerosis treatment.

Anti-obesity and Cholesterol-lowering Effects of Germinated Brown Rice in Rats Fed with High Fat and Cholesterol Diets (발아현미의 섭취에 의한 흰쥐의 비만 억제 및 콜레스테롤 저하 효과)

  • To investigate the anti-obesity and cholesterol-lowering effects of germinated brown rice (GBR), male Sprague-Dawley rats were randomly divided into the following 4 groups and fed with high fat and cholesterol diets for 5 weeks; control group fed with experimental diet, rice group fed with diet containing 50% rice, brown rice group fed with diet containing 50% brown rice and GBR group fed with diet containing 50% GBR. All groups showed no significant difference in body weight, but the GBR group showed the lowest value in body weight, $471.2{\pm}17.8\;g$. Body weight gain and FER of GBR group, $310.6{\pm}14.7\;g$ and $41.8{\pm}2.0%$, respectively, were also lower than those of other groups. Blood triglyceride and cholesterol levels of the GBR group were $54.0{\pm}23.4\;mg/dL$ and $64.8{\pm}14.7\;mg/dL$, respectively, which were significantly lower than those of rice groups and were the lowest values among the experimental groups. The weights of epididymal and kidney fat of GBR group also showed the lowest values compared to other groups. The liver total lipid and total cholesterol of the GBR group, $216.3{\pm}35.7\;mg/g$ liver and $16.5{\pm}0.7\;mg/g$ liver, respectively, were lower than those of other groups, and total lipid, total cholesterol and triglyceride of GBR group showed the same results. These results suggest that GBR diet reduces body weight and fat gain, and has cholesterol-lowering effect.

Effects of Ixeris dentata ext. on Lowering Lipid and Anti-oxidation (씀바귀의 지질강하 및 항산화효과)

  • To investigate effects of Ixeris dentata EtOH ext. on lowering lipid levels and anti-oxidation activity, hyperlipidemic rats were treated with Ixeris dentata EtOH ext. and lipid levels and anti-oxdation activity were measured. The Ixeris dentata EtOH ext. groups showed low concentration of plasma FFA, plasma triglyceride, plasma total cholesterol, and plasma LDL-cholesterol compared to control group. However, concentration of plasma HDLcholesterol was not significantly different among all the treatment groups. The Ixeris dentata EtOH ext. groups showed lower level of liver total cholesterol, liver triglyceride, plasma TBARS, and liver TBARS than those of control group. The Ixeris dentata EtOH ext. groups also showed higher level of GSH-Px activity, SOD activity, and CAT activity than those of control group. Moreover, the Ext. showed lower level of TNF-${\alpha}$, Apo-B, Apo-E, and leptin expression than those of control group. The results in this study shows that the Ixeris dentata EtOH ext. have positive effect in lowering lipid level, and anti-oxidative activity.

The Influence of Long-term Aloe Supplement on Anti-oxidative Defenses and Cholesterol Content in Brain and Kidney of Aged Rats

  • The present study was investigated the anti-oxidative effects of aloe vera ingestion on brain and kidney in aged rats by monitoring several oxidative-related parameters. Male specific pathogen-free Fischer 344 rats were randomly divided into four groups of five rat each: Group A was fed test chow without aloe supplementation; Group B was fed a diet containing a 1% freeze-dried aloe filet; Group C was fed a diet containing a 1% charcoal-processed, freeze-dried aloe filet; and Group D was fed a diet containing a charcoal-processed, freeze-dried, whole leaf aloe in drinking water. Analyses of tissues were done at 4 months and 16 months of age. Results showed that a long-term intake of aloe, regardless of the preparation used, enhanced antioxidant defenses against lipid peroxidation, as indicated by reduced phosphatidylcholine hydroperoxide levels in both brain and kidney. The additional benefit of aloe intake on the anti-oxidative action was evidenced by enhanced superoxide dismutase and catalase activity in all aloe-ingested groups. Another beneficial effect of aloe shown in this study, although not an anti-oxidative parameter, was its cholesterol-lowering effect as detected in brain and kidney with significant decreases at age16 months of aloe-fed rats. Based on these findings, we conclude that a long-term dietary aloe supplementation modulated the anti-oxidative defense systems and cholesterol level.

Effects of Hangbisan, an Oriental Medicine, on Body Weight Gain in Diet-Induced Obese (DID) rats

  • This study was designed to determine possible weight loss effects of Hangbisan, an oriental medicine, on Sprague-Dawley (SD) rats. SD rats that were fed a high-fat diet for 6 weeks to induce obesity and subsequently fed with a basic diet containing 10%(w/w) Hangbisan or 10%(w/w) cellulose for 8 weeks. The Hangbisan fed rats demonstrated a significantly (p<0.05) reduced weight gain compared to the cellulose fed rats as well as a reduced level of plasma cholesterol and triglycerides and an increased level of (HDL)-cholesterol. These results suggest that dietary Hangbisan has an anti-obesity effect in the high-fat diet-induced obesity (DIG) rat and therefore a potential use as an anti-obesity agent in the application of oriental medicine compounds.

Effect of Artemisia Capillaris Thunberg EtOH Ext. on Lowering Lipid, Anti-oxidation and Concentration of Plasma Inflammatory Mediators Using Rats Fed on High-oxidized Fat (인진호(茵蔯蒿) 추출물이 과산화지질 투여한 쥐의 지질강하, 항산화효과 및 염증매개물질의 생산에 미치는 영향)

  • Objectives : The present study investigated effects of Artemisia Capillaris Thunberg ethanol extract(EtOH ext). on lowering lipid, anti-oxidation and concentration of plasma inflammatory mediators using rat fed on high oxidized fat. Methods : We divided fat sprague-dawley rats fed on high oxidized into 4 groups. They were normal group, feed with 100 mg/kg Artemisia Capillaris Thunberg group, feed with 200 mg/kg Artemisia Capillaris Thunberg group and feed with 300 mg/kg Artemisia capilaris Thunberg group. They were administered for 4 weeks. We measured concentration of plasma free fatty acid(FFA), plasma triglyceride, plasma total cholesterol, and plasma low density lipoprotein-cholesterol(LDL-cholesterol), plasma high density lipoprotein-cholesterol(HDL-cholesterol), concentration of liver total cholesterol and liver triglyceride (TG), concentration of plasma thiobarbituric acid reactive substance(TBARS) and liver thiobarbituric acid reactive substance(TBARS), glutathione peroxidase (GSH-Px) activity, superoxide dismutase(SOD) activity and catalase(CAT) activity, plasma nitric oxide(NO), ceruloplasmin and ${\alpha}-glycoprotein$. Results : 1. The Artemisia Capillaris Thunberg EtOH ext. groups showed low concentration of plasma FFA, plasma triglyceride, plasma total cholesterol and plasma LDL-cholesterol compared to control group. However, concentration of plasma HDL-cholesterol was increased in the Artemisia Capillaris Thunberg EtOH ext. groups. 2. Concentration of liver total cholesterol and liver TG showed a significantly decrement in all Artemisia Capillaris Thunberg EtOH ext. groups than that of control group. 3. The Artemisia Capillaris Thunberg EtOH ext. groups showed lower values in concentration of plasma TBARS and liver TBARS than that of control group. The values of GSH-Px activity, SOD activity and CAT activity were increased in the Artemisia Capillaris Thunberg EtOH ext. groups. 4. The values of plasma NO, ceruloplasmin and ${\alpha}-glycoprotein$ were decreased in Artemisia Capillaris Thunberg EtOH ext. groups. Conclusions : Based on the results in this study, the Artemisia Capillaris Thunberg EtOH ext. showed a positive effect in lowering lipid, anti-oxidation and decrement of plasma inflammatory mediators.

Anti-hyperlipidemic Effects of Bacillus strain-fermented Cheonggukjang Products in Mice

  • To evaluate the hypolipidemic effects of Cheonggukjang (CGJ), which is frequently used in Korea similar to Natto in Japan and Douchi in China like a dairy product, boiled soybeans were fermented with two Bacillus strains, B. subtilis and B. licheniforms, isolated from rice straw and their antihyperlipidemic effects of their products were investigated. Treatment with the CGJs significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels and increased HDL cholesterol levels in Triton WR-1339-induced hyperlipidemic mice. The treatment of non-fermented soybeans alone also reduced blood TG and TC levels, but not significantly. Feeding the CGJs significantly lowered high blood TG and TC levels as well as body and epididymal mass weights in hyperlipidemic mice induced by the long-term feeding of a high-fat diet that increased blood HDL cholesterol levels. The B. subtilis-fermented CGJ products more potently reduced TG and TC levels, although the differences between the starters were not significant. These finding suggest that CGJ products may be effective as hypolipidemic foods by the synergistic interaction of soy and Bacillus strains.

  • It has been well known that mushroom polysaccharide extracts have superoxide and hydroxyl radical scavenging activities. So we studied the efficacy and anti-oxidative effect of mushroom(Auricularia auricula U.) in rats treated with water and ethanol extracts of Auricularia auricula U after inducing dryness of skin by the application of detergent. The results of the research for of the efficacy and anti-oxidative effect Auricularia auricula U. are as follows thats. Weight gain and feed intake in animals treated with water extracts and ethanol extracts are significantly higher(p<0.05) than animal treated with detergent. Animals treated with water extracts of Auricularia auricula U. compared to animals treated with ethanol extracts have the higher HDL-cholesterol levels in the blood and the lower LDL-cholesterol levels in the blood. SOD-like activity of the ethanol extracts of Auricularia auricula U. was $27.39{\pm}1.61%$ at 2000ppm.

BS21, a combination of Phyllostachys pubescens and Scutellaria baicalensis extracts, reduces adiposity and hyperuricemia in high-fat diet-induced obese mice (죽엽황금복합추출물 BS21의 고지방식이 유도 비만 동물모델에서의 지방생성 및 고요산혈증 개선 효과)

  • Objectives : Phyllostachys pubescens and Scutellaria baicalensis are considered to be effective in promoting blood circulation in traditional medicine. In this study, we examined whether a mixture of P. pubescens leaves and S. baicalensis root (BS21) had any anti-obesity, anti-hyperlipidemia, or anti-hyperuricemia effects and the possible mechanisms of action. Methods : We examined the effects of BS21 in high-fat diet (HFD)-induced obese mice. Mice were fed HFD with BS21 (75, 150, or 300 mg/kg) or Garcinia cambogia extracts (245 mg/kg) as a positive control for 8 weeks. At the end of 8 weeks, body weight, liver and adipose weight, adipocyte size, plasma lipid profiles, adipokine and uric acid levels, and adipose tissue expression levels in obesity and uric acid production-related genes were examined. Results : BS21 decreased body weight gain, white adipose tissue, liver weight, adipocyte size, and liver triglyceride accumulation. It also reduced levels of plasma glucose, triglycerides, non-esterified fatty acids, total cholesterol, low-density lipoprotein cholesterol, alanine transaminase, leptin, and uric acid. In contrast, BS21 increased adiponectin levels. Furthermore, BS21 decreased the expression levels of adipogenesis-related genes, such as peroxisome proliferator-activated receptor γ, sterol regulatory element binding protein-1c, and fatty acid synthase, as well as xanthine oxidoreductase, which is involved in uric acid production. Conclusions : These results suggest that BS21 may exert anti-obesity, anti-hyperlipidemia, and anti-hyperuricemia effects in HFD-induced obese mice by regulating the expression of xanthine oxidoreductase and adipogenesis-related genes.

The Effect of Cichorium intybus Extract on Hepatotoxicity in Rats

  • Cichorium intybus (Compositae) has been used for fevers, dyspepsia, headache and jaundice, as a demulcent. Also, it has relaxation effects and relief effects against coffee and teas, and is widely used as food. We investigated anti-lipid peroxidative effects and liver protective activity on $CCl_4$ induced lipid peroxidation and hepatotoxicity in rats. MeOH Ex. enhanced the inhibition of anti-lipid peroxidative effects in liver lipid. In chemical parameters obtained from serum analysis, MeOH Ex. revealed significant decrease on hepatotoxicity. The results were as follows; 1. The inhibitory effects of lipid peroxidation were shown in accordance with the increase of samples' concentration level. 2. In chemical parameters obtained from serum analysis, the activities of GOT, GPT, AlP were restored to near the normal level. The contents of cholesterol and BUN showed inhibitory effects with valence. 3. The weights of liver and spleen were not able to restore to the normal level. But on a general level, they were reduced more than the control group.
