Anti-obesity and LDL-cholesterol lowering effects of silkworm hemolymph in C57BL/6N mice fed high fat diet (고지방식이 비만 유도 mouse에서 누에체액의 항비만 및 LDL-cholesterol 저하효과)

  • Purpose: Obesity, a worldwide epidemic, is associated with insulin resistance, hyperlipidemia, hypertension, cardiovascular disease, and certain cancers. Many strategies, including natural alternative anti-obesity agents, are used widely to prevent obesity. This study examined the effects of silkworm hemolymph on the weight control of C57BL/6N mice fed with a high-fat diet. Methods: The mice were divided into five groups: normal group (N), high-fat diet group (HFC), high-fat diet and silkworm hemolymph (at dose of 1 mL/kg BW (HFS-1), 5 mL/kg BW (HFS-5) and 10 mL/kg (HFS-10) for 12 weeks. Results: After 12 weeks treatment, the administration of silkworm hemolymph decreased the final body weight significantly along with a decrease in the weights of epididymal fat and total fat. The plasma LDL-cholesterol concentration was significantly lower in the HFS-1, HFS-5, and HFS-10 groups than in the HFC group. In addition, the leptin level of the HFS groups was significantly lower than those of the HFC group without a change in the plasma insulin concentration. Conclusion: These findings suggest that the silkworm hemolymph may have the potential to prevent obesity.

A Study on the Effect of Electroacupuncture at Hyunjong (GB39) on Anti-Oxidation (현종에 시술한 저주파 전침자극의 항산화 효과에 대한 실험적 연구)

  • Objectives & Methods : The purpose of this study is to investigate the anti-oxidative effects of electroacupuncture at Hyunjong (GB39) in rats. The author performed several experimental items including change of body weight, relative liver weight, level of albumin, total bilirubin, LDL-Cholesterol, LDH, GOT and GPT in blood serum, level of SOD, glutathione, catalase, NO and MDA in liver, histological change of liver. Results : 1. In the GB39-EA group, body weights were decreased in the early stages, but increased later on gradually. 2. In the GB39-EA group, the level of LDL-cholesterol was significantly decreased in comparison with that of the holder group, sham-EA group and GB39-NR group. 3. In the GB39-EAgroup, liver SOD, glutathione, catalase activity were increased, and NO density was decreased in comparison with those of the holder group, sham-EA group and GB39-NR group. MDA activity was significantly decreased in comparison with that of the holder group. 4. In the GB39-EA group, the density of liver tissue was more similarly maintained to the normal group in comparison with that of the holder group and sham-EA group. 5. The results of the GB39-EA group showed similar tendency with those of the GB39-EA group, but the effects were lower than those of the GB39-EA group. Conclusions : These results suppose that electroacupuncture at GB39 has an anti oxidative effect in human.

Investigation of Novel Pharmacological Action of Arctii Fructus and its Compound

  • Arctii Fructus (AF), which contains arctigenin (ARC) as a major constituent, is traditionally used as an anti-inflammatory medicine to treat inflammatory sore throat. Although several studies have proven its anti-inflammatory effects, there have been no reports on its use in inflammation related disorders such as obesity, cancer metastasis, and allergic responses. This study investigated the anti-obesity effect and anti-metastasis effect of AF and ARC. AF and ARC inhibited weight gain by reducing the mass of white adipose tissue in high fat diet (HFD)-induced obese mice. Serum cholesterol levels were also improved by AF and ARC. In in vitro experiments, AF and ARC decreased differentiation of white adipocytes. Furthermore, AF induced differentiation of brown adipocytes, which are able to consume surplus energy through non-shivering thermogenesis. Also, AF and ARC inhibited colon cancer and lung metastasis of colon cancer. They suppressed not only colorectal cancer cell progression by inhibiting cell growth, but also prohibited lung metastasis by regulating epithelial-mesenchymal transition (EMT), migration, and the invasion. These effects were confirmed in an experimental metastasis mouse model. In addition, AF and ARC inhibited mast cell mediated allergic responses. Collectively, our study suggests that AF and ARC might show inhibitory effects on inflammation related diseases, including obesity, cancer, cancer metastasis, and allergic responses.

Effects of Mucilage from Yam (Dioscorea batatas DECENE) on Blood Glucose and Lipid Composition in Alloxan-induced Diabetic Mice (마(Dioscorea batatas DECENE) 점질물이 Alloxan 유발 당뇨 마우스의 혈당 및 지질 성분에 미치는 영향)

  • Effects of Mucilage fraction from Yam (Dioscorea batatas DECENE), which has long been used as a wild vegetable and folk medicine, on blood glucose and lipid composition in alloxan-induced diabetic mice were investigated. Diabetes mellitus was induced in male ICR mice by the injection of alloxan into the tail vein at a dose of 75 mg/kgBW. Alloxan-induced diabetic mice were administered the yam mucilage fraction1 orally ; y500F1 group (500 mg/kgBW/day fraction1) and y750F1 group (750 mg/kgBW/day fraction1); and the normal and alloxan-control group were orally administered with saline for 10 days. The body weight gain and food intake were monitored every day. The concentrations of glucose, triglyceride, total cholesterol, and LDL-cholesterol of serum and liver levels of glucose, triglyceride, and cholesterol were determined. Also weight of liver, heart, spleen and kidney were measured. The fraction 1 of yam mucilage lowered body weight gain significantly (p<0.05) and decreased serum glucose levels in alloxan-induced diatetic mice compared to that of alloxan-control group. In alloxan induced diabetic mice serum triglyceride level was lowered and liver HDL-cholesterol level increased significantly (p<0.05). In conclusion, it was assumed that yam mucilage fraction 1 has anti-hyperglycemic and anti-obesitic effects by reducing body weight gain and decreasing serum glucose and triglyceride level.

Effects of the Combined Extracts of Grape Pomace and Omija Fruit on Hyperglycemia and Adiposity in Type 2 Diabetic Mice

  • Grape products have been known to exert greater antioxidant and anti-obesity than anti-hyperglycemic effects in animals and humans. Omija is used as an ingredient in traditional medicine, and it is known to have an anti-hyperglycemic effect. We investigated whether the combined extracts of grape pomace and omija fruit (GE+OE) could reduce fat accumulation in adipose and hepatic tissues and provide beneficial effects against hyperglycemia and insulin resistance in type 2 diabetic mice. C57BL/KsJ-db/db mice were fed either a normal control diet or GE+OE (0.5% grape pomace extract and 0.05% omija fruit extract, w/w) for 7 weeks. GE+OE decreased plasma leptin and resistin levels while increasing adiponectin levels and reducing the total white adipose tissue weight. Furthermore, GE+OE lowered plasma free fatty acid (FFA), triglyceride, and total-cholesterol levels as well as hepatic FFA and cholesterol levels. Hepatic fatty acid synthase and glucose 6-phosphate dehydrogenase activities were decreased in the GE+OE group, whereas hepatic ${\beta}$-oxidation activity was increased. Furthermore, GE+OE supplementation not only reduced hyperglycemia and pancreatic ${\beta}$-cell failure but also lowered blood glycosylated hemoglobin and plasma insulin levels. The homeostasis model assessment of insulin resistance levels was also decreased and the decrease seems to be mediated by the lowered activities of hepatic glucose-6-phosphatase and phosphoenolpyruvate carboxykinases. The present data suggest that GE+OE may have the potential to reduce hyperglycemia, insulin resistance, and obesity in patients with type 2 diabetes.

Anti-Obesity Effect of a New Dietary Supplement Consisting of Hydroxycitrate, Carnitine and Red Pepper (3D-Relax Diet) in High-Fat Fed Rats

  • Anti-obesity effect of a new dietary supplement (3D-relax) in high-fat fed rats. The aim of this study was to assess the effects of 3D-relax; a proprietary formulation containing hydroxycitrate (233 mg/g), carnitine (150 mg/g) and red pepper (150 mg/g); on body weight, body fat, and serum lipids levels in rats fed a high-fat diet. Male SD 7-wk-old rats (n=8) were fed a high fat diet [52% total dietary energy (E%) from fat, 15.4 E% protein, 32.6E% carbohydrate] with or without 3D-relax administration (1 g/kg body weight/day) for 3 weeks. Administration of 3D-relax significantly reduced the increase in body weight compared to the group fed high fat without 3D-relax. Food efficiency ratio (FER) tended to be decreased with administration of 3D-relax, but was not significant. The perirenal and epididymal fat pad weights of vats administered 3D-relax were significantly lower than those of the high fat group that did not ingest 3D-relax during the 3 weeks. The oral administration of 3D-relax significantly increased HDL-cholesterol level and lowered total cholesterol level compared to those of high fat alone group. These results suggest that 3D-relax reduced body weight and fat gains, and those effects are presumably linked to its inhibitory effects on lipogenesis.

Anti-obesity and Anti-hyperlipidemic Effects of Taeyeumjowee-tang and its modified prescription (태음조위탕(太陰調胃湯)과 그 가감방(加減方)의 항비만 및 항고지혈증 효과에 관한 연구)

  • Objective : The purpose of this study was to investigate the anti-obesity and anti-hyperlipidemic Effects of Taeyeumjowee-tang and its modified prescription on the animal model of obesity and hyperlipidemia induced high-fat diet. Method : 1) The extracts of Taeyeumjowee-tang (TJT) and its modified prescription, Taeyeumjoweetang gagam-bang (TJGB) were evaluated for its inhibitory effects on obesity. 2) The body weight and feed weight were determined in the pre-treated and post-treated mice and the lipid profiles in the serum were analyzed in order to evaluate the anti-hyperlipidemia action of the extracts. 3) The effect of each extract was investigated for the influences on monoamine oxidase activity and HMG-CoA reductase activity. Results 1. TJT and TJGB extracts dose-dependently reduced the body weight and feed intake in normal mice. The effect of TJGB extract was better than that of TJT extract. 2. TJGB extract diminished the body weight increase and reduced the feed intake in the pre-treatment or post-treatment of the extract 3. TJGB extract decreased the amount of total cholesterol slightly and triglyceride potently after the pre-treatment or post-treatment, but HDL cholesterol exhibited no remarkable change compared with control. 4. TJGB extract weakly potentiated the monoamine oxidase activity, but its effect was better than that of TJT extract. 5. TJGB extract weakly inhibited the HMG-CoA reductase activity, but its effect was better than that of TJT extract. Conclusion : Taeyeumjowee-tang and its modified prescription can clinically be useful as anti-obesity drug and also for the improvement of hyperlipidemia.

Effects of Dietary Lentinus Edodes and Kinds of Dietary Lipids on the Lipids Metabolism in Rats Fed High Cholesterol Diets (식이내 표고함량과 지방의 종류가 고콜레스테롤식이를 섭취한 흰쥐의 지방대사에 미치는 영향)

  • This study was designed to evaluate the anti hypercholesterolemic effect in either soybean oil or lard containing hypercholesterol diet in rats. Rats were fed, ad libitum, for 5 weeks with diets containing 0, 5, 10% of L.edodes in either soybean oil or lard with 1% exogenous cholesterol. Forty-two male sprague-dawley rats weighing 71.6$\pm$8.1g were divided into 6 groups. The results of this study were summairzed as follows: 1. Weight gain, total feed intake, feed intake efficiency ratio, total calorie intake and calorie efficiency ratio during five weeks showed significant difference among groups. The lowest value was found in soybean oil + L.edodes-free group and the highest value was found in lard + L.edodes-free group. The weight of organs were significantly higher in lard groups than soybean oil groups. 2. The contents of serum triglyceride showed significant difference kinds of lipids. It tended to be lower soybean oil groups than lard groups. It was difference with dietary L.edodes levels in soybean oil groups. The contents of total serum cholesterol showed significant difference by kinds of lipids, it was lower soybean oil groups than lard groups. But it was not differ with dietary L.edodes levels in soybean oil groups. 3. The contents of total liver cholesterol showed no difference kinds of lipids. And it was lower in soybean oil groups than lard groups. This result showed that the soybean oil decreased the contents of total liver cholesterol.

Effects of Chicken Treated with Hwangki-Beni Koji Sauces on Body Weight, Serum and Hepatic Lipid Profiles of Rats Fed High Fat and High Cholesterol Diets (황기홍국소스를 처리한 계육이 고지방 및 고콜레스테롤 식이 흰쥐의 체중과 혈청 및 간 조직의 지질 함량에 미치는 영향)

  • Dietary effects of chicken treated with Hwangki-Beni Koji sauces (HBS) on serum and hepatic lipid profiles in rats fed high a fat and high cholesterol diet were investigated. Experimental plots (5 rats per each group) were divided into normal control diet group (NC), high fat and high cholesterol diet group (HFC), HFC plus 15% base sauce treated chicken supplemented diet group (HFC-BS), HFC plus 15% HBS-treated chicken supplemented diet group (HFC-HBS), and fed for 5 weeks. Feed intakes in HFC-HBS group were higher than those of HFC and HFC-BS groups, whereas body weight gain and feed efficiency ratio of the HFC-HBS group were lower than those of the HFC and HFC-BS groups. Serum alanine aminotransferase and aspartate aminotransferase activities of HFC-HBS group were significantly lower than those of HFC and HFC-BS groups. Level of serum triglyceride, total cholesterol and LDL-cholesterol in HFC-HBS group were 34.06%, 6.28% and 31.00% lower than those of HFC-BS group, respectively, whereas HDL-cholesterol level and atherogenic index in HFC-HBS group were 1.72-fold higher and 2.52-fold lower, respectively, than those of HFC-BS group. Level of hepatic total lipid, triglyceride and total cholesterol in HFC-HBS group were 9.53%, 5.25% and 22.20% lower than those of HFC-BS group, respectively. In the morphological results of liver, the hepatocytes of central vein lesion and intact hepatic cell plate are preserved well from steatosis in HFC-HBS group, whereas the hepatocytes in HFC and HFC-BS groups are distended by accumulation of multiple fat droplets, and sinusoids are occluded. In the above results, we expected that chicken treated with Hwangki-Beni Koji sauce may have functionalities of anti-obesity, anti-cholesterol and also, lowering anti-atherogenic index in high fat and high cholesterol diets.

Anti-cholesterol Effects and Molecular Mechanism Study of Mixture of Atractylodes Macrocephala and Amomum Villosum Extracts (백출과 양춘사 추출 혼합물의 항콜레스테롤 효과 및 기전 연구)

  • Atractylodes macrocephala (AM) and Amomum villosum (AV) are the most common herbs in Korean Medicine to treat digestive diseases. In this study, we investigated the cholesterol lowering effects of mixtures of AM and AV extracts on high cholesterol diet (HCD) induced dyslipidemia mouse model. We classified animals into six different groups; Group 1: Normal diet, Group 2: HCD, Group 3: AV extracts : AM extracts (1:1) (200 mg/kg) + HCD, Group 4: AV extracts : AM extracts (1:2) (200 mg/kg) + HCD, Group 5: AV extracts : AM extracts (1:3) (200 mg/kg) + HCD, Group 6: Simvastatin 40 mg/kg + HCD. After 4 weeks of oral administration of respective drugs, we checked body, liver and epididymal fatweights along with liver and serum triacylglyceride (TG) concentration, total and low density lipoprotein (LDL) cholesterol in serum. Moreover, 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase (HMGCR), LDL receptor (LDLR), and sterol regulatory element-binding protein 2 (SREBP2) were detected by RT PCR or western blot analysis. The overall results showed that mixtures of AM and AV extracts inhibited HCD-induced increases of total cholesterol and LDL cholesterol in serum. Those effects seem to be caused by AM and AV extracts through inhibition of HMGCR expression. And thus blood cholesterol is induced into the liver by increasing LDLR expression, which is regulated by SREBP2 transcrption factor. The cholesterol lowering effects and mechanism of mixtures of AM and AV extracts was similar to the statin. We have identified the potential mixtures of AM and AV extracts as a new treatment for dyslipidemia.
