Anti-atopic Effect of Hot Water and Supercritical Carbon Dioxide Fluid Extract of Persimmon (Diospyros kaki) Peels (감껍질 열수 및 초임계 유체 추출물의 항아토피 효과)

  • This study aimed to investigate the anti-atopic effect of hot water (PPWE) and supercritical-carbon dioxide fluid extract of persimmon peels (SPPE) on atopic dermatitis (AD)-like skin lesions in hairless mice. Histological analyses demonstrated that SPPE treatment more strongly inhibited the dermal infiltration of inflammatory cells in AD-like skin lesions than that by PPWE. Compared to PPWE, SPPE significantly decreased the dermatitis clinical score and the epidermal thickness and potently suppressed serum IgE and interleukin (IL)-4 production in hairless mice with AD. Furthermore, compared to PPWE, SPPE potently inhibited the production of nitric oxide, prostaglandin $E_2$, and proinflammatory cytokines such as IL-6 and IL-$1{\beta}$ in lipopolysaccharide-stimulated RAW264.7 macrophages. These results suggested that SPPE exhibited anti-atopic dermatitis activity via the regulation of inflammatory responses.

Anti-inflammatory and Antioxidant Effects of Zizyphus jujuba Miller var. inermis extracts (대추 추출물의 항산화 및 아토피 피부염 관련 항염증 효과)

  • In the present study, the effect of Z. jujuba Miller var. inermis Rehder extracts on the secretion of atopic dermatitis (AD)-related cytokines and hyaluronidase activity was investigated. We prepared four fractions, butylene glycol (JB), ethanol (JE), and water (JW), with Z. jujuba Miller var. inermis extracts. JW significantly reduced the secretion of interleukin-8 and JE reduced the secretion of tumor necrosis factor-alpha in human keratinocyte HaCaT cells. Also, hyaluronidase activity was measured by enzyme assay and the fractions inhibited the activity in a dose-dependent manner. In addition, the human dermal fibroblast, HDF-n cells were treated with the extracts and antioxidant activities were measured. The results showed that the extracts increased the free radical scavenging activity and the superoxide dismutase activity. Taken together, Z. jujuba Miller var. inermis extracts reduced the secretion of AD-related cytokines and inhibited the hyaluronidase. In addition, the extracts showed antioxidant activity.

  • The efficacies of anti-atopic preparations were investigated in hairless mice suffering from 2,4-dinitro-chlorobenzene (DNCB)-induced atopic dermatitis-like lesion. Solid lipid nanoparticle (SLN) containing ceramide and astaxanthin was prepared by a melt-homogenization method using Aminsoft-CT 12 (Cocoyl glutamate) as an emulsifier. And then, the SLNs were coated with silk fibroin (SF) by taking advantage of an electrostatic interaction between the surface of SLNs and SF. SLNs were included in lotion (FL) and cream (FC) types of preparations. Anti-atopic efficacies of the preparations were investigated in terms of appearance of skin surface, spleen index, serum IgE level, and serum cytokine level. SLN-containing preparations suppressed IgE production and IL-4 expression, but promoted $IFN-{\gamma}$ expression.

Effects of Lespedeza Caneata (LC) Extracts on Atopic Dermatitis in DNCB-Induced Mice (DNCB로 유도된 생쥐에서 아토피 피부염에 대한 야관문추출물의 효과)

  • The purpose of this study was to examine the effects of Lespedeza Caneata (LC) extracts on atopic dermatitis. For the experiment, mice were divided into a normal group, an AD group, and AD-LC groups (100 mg/Kg, 300 mg/Kg, and 500 mg/Kg). For the AD and AD-LC groups, 1% DNCB $200{\mu}{\ell}$ was applied at a hair-removed site for 2 weeks, and then 0.1% DNCB $150{\mu}{\ell}$ was applied every 2 days for 4 weeks. Skin thickness is a symptom of atopic dermatitis, and in this experiment, the AD group had the thickest skins, and the AD-LC group (500 mg/Kg) had the same skin thickness as the normal group. Mast cells show inflammatory reaction, and in this experiment, the AD group had the largest number of mast cells. Collagen fibers are usually observed on normal skins, and in this experiment, the AD-LC group (500 mg/Kg) had a uniform level of those fibers. Based on the study results, it turned out that LC extracts have the anti-atopic efficacy improving keratinization, erosion, and uredo.

Inhibitory Effects of Black-red Ginseng Extracts on Allergic Inflammation In Vitro and In Vivo (In Vitro 및 In Vivo 알러지성 염증반응에 대한 흑홍삼의 억제효과)

  • Black-red ginseng (BRGP) exhibits more potent biological activities than white and red ginseng. However, the effect of BRGP on allergic inflammation has not been studied extensively. In this study, we attempted to evaluate the effects of BRGP on allergic inflammation in vitro and in vivo. The effects of BRGP on pro-inflammatory mediators in phorbol myristate acetate (PMA) and A23187-stimulated human mast cells (HMC-1) and on trimellitic anhydride (TMA)-induced atopic dermatitis-like skin lesions in BALB/c mice were evaluated. BRGP suppressed the expression of $TNF-{\alpha}$, IL-6 and IL-8 from HMC-1 stimulated with PMA and A23187. Furthermore, the oral administration of BRGP markedly significantly reduced apparent severity of atopic dermatitis-like skin lesions, ear swelling, lymph node weight gains and serum IgE levels induced by TMA in mice. BRGP treatment also ameliorated epidermal hyperplasia and infiltration of inflammatory cells including mast cells. Taken together, BRGP possesses significant anti-atopic efficacy, suggesting that it could be used as a potential therapeutic agent for allergic inflammatory diseases, including atopic dermatitis.

Anti-dermatitis Effects of KamiCheongsimyeonjatang on GATA-3 Regulation in NC/Nga Mouse (NC/Nga 생쥐에서 가미청심연자탕(加味淸心蓮子湯)의 GATA-3 조절에 의한 항아토피 피부염 효과)

  • Objectives : The purpose of this study is to investigate effectiveness of KamiCheongsimyeonjatang(KCSYJT) medicines to suppress atopic dermatitis in mouse model experimentally. Methods : First, in vitro, we isolated B cells from 18 weeks of atopicdermatitis-like skin NC/Nga mouse. Then we analyzed FACS(Fluorescence Activated Cell Sorter) by intracellular staining of IFN-$\gamma$, GATA-3+ analyzed cytokines by using real-time PCR. Secondly, in vivo, after administration of KCSYJT to atopic dermatitis NC/Nga mouse at 12 weeks of age, we analyzed serum IgE and the change of activated cell in PBMCs(Peripheral Blood Mononuclear Cells). Results : In vitro, KCSYJT medicines supressed IL-1$\beta$, IL-6, TNF-$\alpha$, and TGF-$\beta$ mRNA and increased IL-10 mRNA in B cells. Also, KCSYJT medicines decreased the levels of GATA-3$^+$CD4$^+$ and increased the levels of IFN-$\gamma^+$CD4$^+$T Cell. In vivo, serum IgE levels dreased in KCSYJT group than control group and In PBMCs, the activated cell percentage of granulocytes, CD3+, CD3+/CD4+, B220+/CD23+, and CCR3+ decreased and CD19+, CD3+/CD8+ increased in KCSYJT group than control group. Conclusions : This study demonstrates immunological activity of KCSYJT on atopic dermatitis-like model mice.

Anti-inflammatory Effects of Effective Microorganism Fermentation Substance on Atopic Dermatitis-like NC/Nga Mouse Model (아토피 피부염 유사 NC/Nga 마우스 모델에서 유용 미생물 발효물질의 항염증효과)

  • Atopic dermatitis (AD) is a chronically relapsing pruritic inflammatory skin disease. To find new anti-inflammatory products for skin inflammatory disease such as AD and contact dermatitis, we produced the effective microorganism fermentation substance (EM-S) by fermentation of medicinal plants with effective microorganisms including photosynthetic bacteria, lactic acid bacteria and yeast, screened the effects of EM-S on NC/Nga model mice. Murine AD-like skin lesions were made by painting Dermatophagoides farinae (Df) extract. Topically applied EM-S significantly reduced clinical severity score, ear thickness and histological grade in AD-like NC/Nga mouse model by Df antigen sensitization. In addition, the serum IgE and Th2 chemokine levels (TARC/CCL17, MDC/CCL22 and CTACK/CCL27) were significantly reduced by EM-S. Futhermore, skin tissue expressions of Th2 chemokines were significantly reduced by EM-S. These results demonstrate that topical application of EM-S may be improve the AD-like skin lesion by suppressing IgE and Th2 chemokines.

Effect of SHT on the anti-oxidant activity and atopic dermatitis related inflammatory cytokines (석자해기탕(石紫解肌湯)이 아토피피부염 유관 인자인 산화적 손상과 염증 인자에 미치는 영향)

  • In order to investigate the possibility of SHT as therapeutic for the treatment of atopic dermatitis (AD), cytotoxicity, anti-oxidant activity, modulatory and suppression activities of SHT were tested. 90% or higher cell viability was observed in all tested groups from 25 to 200 ug/ml using Raw 264.7 cells. SHT showed dose-dependent DPPH scavenging activity, with more than 80% scavenging activities at 400 and 800 ug/ml concentrations. SHT showed dose-dependent suppression activity of ROS production, especially at 200 ug/ml of 57.4%. SHT decreased NO production activity dose dependently, expecially at 200 ug/ml of 28.8%. IL-$1{\beta}$, IL-6, MCP-1, TNF-${\alpha}$ production rate were decreased by 45.7%, 15.5%, 8.9%, 16.5% respectively when Raw 264.7 cells were treated with LPS and with SHT of 200 ug/ml. However, only IL-6 and TNF-${\alpha}$ showed significant changes. The results above indicate that SHT significantly reduces the effect of oxidative and inflammatory cytokines. The use of SHT in dermatitis can be widely suggested.

Hwangnyeonhaedok-tang Extracts Ameliorates Atopic Dermatitis via Epidermal Lipid Barrier Regeneration in NC/Nga Mouse (황련해독탕의 피부지방장벽개선을 통한 Th2 분화조절이 아토피피부염 완화에 미치는 효과)

  • Objectives Hwangnyeonhaedok-tang is a Korean herbal medical treatment that removes toxic heat, fever and inflammation. The purpose of this study was to investigate the effect of Hwangnyeonhaedok-tang treatment on the relief of atopic dermatitis (AD) through regeneration of skin lipid barrier. Methods Male NC/Nga mice (20 g, 6 week age) were used. Each 10 mice were allocated to the control group (Ctrl), the AD-induced with no treatment group (AE), and the group which induced AD after administering Hwangnyeonhaedok-tang extract (HT). To induce AD-like skin lesions, sodium dodecyl sulfate (SDS) (Sigma-Aldrich, USA) was rubbed on the back of each mouse to remove the lipid lamella of the stratum corneum, and Dermatophagoides (D.) farinae crude extract was applied. HT group was orally administered Hwangnyeonhaedok-tang after induction of AD. IL-4 IL-13, $p-I{\kappa}B$, iNOS, Sudan Black B (SB), loricrin, and filaggrin were observed to confirm the effect. Results In HT group, AD skin score was decreased by 46%. The cytokine IL-4 and IL-13, which can identify Th2 differentiation, was reduced by 73% and 58% each. Anti-inflammatory effects were observed in $p-I{\kappa}B$ and iNOS by 69% and 54%, respectively. Finally, SB showed that the regeneration of the lipid layer and the increase of the regeneration power of loricrin and filaggrin were increased by 437% and 464%, respectively. Conclusions From the study result, we observed that Hwangnyeonhaedok-tang treatment alleviates AD by decreasing skin score, reducing Th2 differentiation, inducing anti-inflammatory, and increasing skin lipid barrier regeneration. Thus, Hwangnyeonhaedok-tang treatment would be considered as an effective AD relieving treatment.

Properties of Fucoidan as Raw Materials of Water-holding Cream and Cosmetics (후코이단의 보습제 및 화장품재료로서 특성)

  • Properties of fucoidan used for functional cosmetics agents were studied. Fucoidan was extracted from Undaria pinnatifida sporophylls. To test possibility of fucoidan as a cosmetics material, water-holding property measurement, DPPH free radical scavenging assay and MTT assay were done. Water-holding property of fucoidan was higher than that of hyaruronic acid, which is known as the one of the best water-holding material. The water-holding strength of fucoidan slightly increase as molecular weight of fucoidan decrease. Fucoidan showed high stability from MTT assay and good anti-oxidation property from DPPH assay. To evaluate the effect of water-holding property and anti-alergy property of fucoidan on the atopic dermatitis(AD), 46 AD patients were treated with fucoidan cream. After 6 weeks treatment, Investigation Global Assessment(IGA) scores decreased from 3.04 to 2.15, that is fucoidan cream had a 39.8% benefit effect on atopic dermatitis.
