Inotodiol Inhabits Proliferation and Induces Apoptosis through Modulating Expression of cyclinE, p27, bcl-2, and bax in Human Cervical Cancer HeLa Cells

  • Inonotus obliquus is a medicinal mushroom that has been used as an effective agent to treat various diseases such as diabetes, tuberculosis and cancer. Inotodiol, an included triterpenoid shows significant anti-tumor effect. However, the mechanisms have not been well documented. In this study, we aimed to explore the effect of inotodiol on proliferation and apoptosis in human cervical cancer HeLa cells and investigated the underlying molecular mechanisms. HeLa cells were treated with different concentrations of inotodiol. The MTT assay was used to evaluate cell proliferating ability, flow cytometry (FCM) was employed for cell cycle analysis and cell apoptosis, while expression of cyclinE, p27, bcl-2 and bax was detected by immunocytochemistry. Proliferation of HeLa cells was inhibited by inotodiolin a dose-dependent manner at 24h (r=0.9999, p<0.01). A sub-$G_1$ peak (apoptotic cells) of HeLa cells was detected after treatment and the apoptosis rate with the concentration and longer incubation time (r=1.0, p<0.01), while the percentage of cells in S phase and $G_2$/M phase decreased significantly. Immunocytochemistry assay showed that the expression of cyclin E and bcl-2 in the treated cells significantly decreased, while the expression of p27 and bax obviously increased, compared with the control group (p<0.05). The results of our research indicate that inotodiol isolated from Inonotus obliquus inhibited the proliferation of HeLa cells and induced apoptosis in vitro. The mechanisms may be related to promoting apoptosis through increasing the expression of bax and cutting bcl-2 and affecting the cell cycle by down-regulation the expression of cyclin E and up-regulation of p27. The results further indicate the potential value of inotodiol for treatment of human cervical cancer.

Neuroprotective Effect of Citri Pericarpium On Transient Global Ischemia in Gerbils

  • The current study was carried out to evaluate neuroprotective effects of Citri Pericarpium after transient global ischemia in gerbils. Male Mongolian gerbils weighing 60-80g were anesthetized with 2% isoflurane mixed with 30% oxygen and 70 % nitrogen. Bilateral common carotid arteries were occluded for 5 minute with microaneurysm dips. On 3 or 7 days after ischemic surgery, the gerbils were sacrificed. The brain were removed, embedded in paraffin and sectioned at 8㎛-thickness. Gerbils that received ischemic insult for 5 min showed extensive neuronal damage in the hippocampal CA1 region, and the number of viable neuronal cell was 51.0±2.5/mm, 32.2% of normal group at 7 days after ischemic surgery. In animals that underwent the extract of Citri Pericarpium treatment, the number of viable neuronal cell were significantly better preserved at 110.58±3.58/mm, 72.0% of normal group than those of ischemic group (P<0.01). In the immunohistochemistry of Bax and Bcl-2, the Citri Pericarpium treated group down-regulated the expression of Bax protein at 72hr after transient global ischemia. In contrast, Bcl-2 protein level was not changed. The appearance in TUNEL assay is similar to the pattern of Bax protein. The water extract of Citri Pericarpium significantly reduced the number of TUNEL-positive CA1 pyramidal neurons at 72hr. The results suggest that Citri Pericarpium has potential neuroprotective effects in the transient global ischemia and the increase in the ratio of Bcl-2 to Bax may contribute to the anti-apoptotic effect of Citri Pericarpium.

자궁경부암세포(子宮經部癌細胞)(HeLa cell)에서 유향(乳香) 세포고사(細胞枯死) 기전(機轉) 연구(硏究) (Olibanum-induced Apoptosis Signaling in Human Cervical Carcinoma HeLa Cells)

  • Purpose : To address the ability of Olibanum to induce cell death, we investigated the effect of olibanum on cell apoptosis. Twenty-four hours later, apoptosis occurred following olibanum exposure in a dose-dependent manner. Methods : We culture HeLa cell which is human metrocarcinoma cell in D-MEM included 10% fetal bovine serum(Hyclone Laboratories) below $37^{\circ}C$, 5% CO2. Then we observed apoptosis of log phage cell which is changed cultivation liquid 24 Hours periodically. Results : The treatment of BAPTA-AM regulated olibanum-induced apoptosis in HeLa human cervical carcinoma cells. The 24 hr-earlier -thapsigargin-pretreated cell showed the resistance against olibanum-induced apoptosis and the Ru360-mitochondrial uniporter-inhibited olibanum-induced apoptosis, too. It means that olibanum leads to the accumulation of calcium and the resultant apoptosis in HeLa cells. Immunoblotting data also shows that the expression of GRP78, ER stress marker protein, was induced by the olibanum. Bcl-2, anti-apototic protein, was decreased and that the expression of Bax, pro-apoptotic protein, was increased by the addition of olibanum. Interestingly, the olibanum increased the activity of caspase-8 as well as calpain cysteine pretense in HeLa cervical carcinoma cells. Calpain inhibitor-calpastatin as well as caspase-8C/A expression abrogated olibanum-induced apoptosis in the carcinoma cells. The inhibition of caspase-8 regulated olibanum-induced calpain activation but the inhibition of calpain did not have any effect on the caspase-8 activation in HeLa human cervical carcinoma cells. Conclusion : We conclude that olibanum induces the accumulation of calcium and the resultant apoptosis in which caspase-8 and calpain are involved.

The Protective Effect of Epigallocatechin-3 Gallate on Ischemia/Reperfusion Injury in Isolated Rat Hearts: An ex vivo Approach

  • The aim of this study was to evaluate the preventive role of epigallocatechin-3 gallate (EGCG, a derivative of green tea) in ischemia/reperfusion (I/R) injury of isolated rat hearts. It has been suggested that EGCG has beneficial health effects, including prevention of cancer and heart disease, and it is also a potent antioxidant. Rat hearts were subjected to 20 min of normoxia, 20 min of zero-flow ischemia and then 50 min of reperfusion. EGCG was perfused 10 min before ischemia and during the whole reperfusion period. EGCG significantly increased left ventricular developed pressure (LVDP) and increased maximum positive and negative dP/dt (+/-dP/dtmax). EGCG also significantly increased the coronary flow (CF) at baseline before ischemia and at the onset of the reperfusion period. Moreover, EGCG decreased left ventricular end diastolic pressure (LVEDP). This study showed that lipid peroxydation was inhibited and Mn-SOD and catalase expressions were increased in the presence of EGCG. In addition, EGCG increased levels of Bcl-2, Mn-superoxide dismutase (SOD), and catalase expression and decreased levels of Bax and increased the ratio of Bcl-2/Bax in isolated rat hearts. Cleaved caspase-3 was decreased after EGCG treatment. EGCG markedly decreased the infarct size while attenuating the increase in lactate dehydrogenase (LDH) levels in the effluent. In summary, we suggest that EGCG has a protective effect on I/R-associated hemodynamic alteration and injury by acting as an antioxidant and anti-apoptotic agent in one.

카드뮴으로 인한 마우스 간 손상에 대한 진피의 보호효과 (Protective effect of Citri Unshius Pericarpium against cadmium-induced liver damage in mice)

  • Objective : Citri Unshius Pericarpium (Citrus unshiu peel) has been used in Korean medicine to treat indigestion, vomiting, coughing and phlegm. This study investigated the hepatoprotective effect of ethanol extract of Citrus unshiu peel (CEE) in cadmium (CdCl2)-treated mouse model. Methods : CEE was dissolved in water and administered orally to mice once a day for 7 consecutive days. The mice were then exposed to a single intraperitoneal (i.p.) injection of cadmium (4 mg/kg body weight) to induce acute hepatotoxicity. At the end of the experiment, blood and liver tissue samples were collected, analyzed for alanine aminotransferase (ALT), aspartate aminotransferase (AST), and histopathological evaluation. Liver damage was assessed as the percentage of degenerative areas of the hepatic parenchyma, the number of degenerative hepatocytes, and the number of infiltrated inflammatory cells. Results : In cadmium-treated rats, pretreatment with CEE significantly reduced the serum ALT and AST levels associated with liver damage. Histopathologically, CEE prevented degenerative changes on the hepatic tissues including confluent necrosis, congestions and infiltration of inflammatory cells. CEE also reduced the elevation of oxidative stress markers (nitrotyrosine and 4-hydroxynonenal) and apoptosis markers (cleaved caspase-3 and cleaved PARP) positive cells. PARP protein expression in liver tissue was also restored by CEE. Conclusion : This study showed that CEE exerted antioxidant and anti-apoptotic effects against cadmium-induced liver injury. Thus, it can be concluded that CEE can be used to prevent liver damage caused by cadmium.

인체 대장암 세포주 SW480에서 docosahexaenoic acid에 의한 침윤억제 기전 (Mechanism of Anti-Invasive Action of Docosahexaenoic Acid in SW480 Human Colon Cancer Cell)

  • 대장암은 미국 등 서양 국가뿐만 아니라 국내에서도 2번째로 많이 발병이 되는 암으로 알려져 있다. 역학조사에 의하면 ${\omega}3$-PUFAs를 많이 섭취한 인종에서 대장암 발생빈도가 감소하고 최근 ${\omega}3$-PUFAs는 수종의 암에 대해 항암작용을 나타낸다고 한다. 이에 본 연구에서는 대장암에서 DHA 등 ${\omega}3$-PUFA의 항침윤 기전을 규명하여 다음과 같은 결과를 얻었다. DHA및 EPA는 대장암 세포주 SW480의 증식을 농도 의존적으로 억제하였으나 AA는 거의 영향이 없었으며 TUNEL assay로 apoptotic cell death가 확인 되었다. DHA는 $\beta$-catenin 단백 및 TCF/LEF luciferase 활성을 농도 의존적으로 억제 하였다. SW480 세포의 침윤능은 DHA의 농도에 의존적으로 억제되었다. DHA처리 후 MMP-9 및 MMP-2 mRNA양이 감소되었을 뿐만 아니라 그 promoter의 reporter 활성도 억제되었다. NF-kB 및 p-IkB 단백질양도 DHA의 처리농도에 의존적으로 감소하였으며 NF-kB promoter의 활성도 억제되었다. 이상의 결과로 ${\omega}3$-PUFA는 대장암에서 NF-kB 신호전달 차단에 의한 MMP-2 및 MMP-9 발현을 억제하여 침윤을 억제하여 항암작용을 나타낼 수 있음을 시사하며, 따라서 ${\omega}3$-PUFA는 대장암의 예방 및 치료에 유용하게 사용될 수 있으리라 생각된다.

개에서 DNCB에 의해 유발된 알레르기성 접촉피부염에 대한 폴리감마글루탐산의 항염증 효과 (Anti-Inflammatory Effects of Poly-${\gamma}$-Glutamic Acid on DNCB-Induced Allergic Contact Dermatitis in Dogs)

  • 알레르기성 접촉피부염은 T세포와 대식세포가 관여하는 세포매개성 면역반응으로 항원에 노출된 뒤 수 일 후에 증상이 나타나는 지연형 반응이다. 그 과정은 감작기와 유발기로 나뉘는데 감작기에는, 표피장벽을 통해 유입된 항원이 표피기저층에 있는 항원전달세포에 의해 처리된 후 림프절로 이동되어 T세포에 의해 인식되고 그 T세포는 항원특이 T세포로 활성화된다. 유발기는 동일 항원이 재감작될 때 항원특이 T세포의 반응을 활성화시키고 다양한 cytokine 분비를 통해 염증세포를 항원 유입부위로 이동시킨다. 본 연구에서는 DNCB로 알레르기성 접촉피부염을 유발한 개의 모델에 폴리감마글루탐산의 항염증 효과를 평가하였다. 폴리감마글루탐산은 12일간 적용하였고 실험기간 동안 이틀 간격으로 피부 생리학적 지표를 측정하였으며 적용 후 cytokine 측정과 조직병리학적 검사를 실시하였다. DNCB 적용후 피부 생리학적 지표의 변화로 표피경유수분손실, 피부 수화도, 피부 두께 그리고 홍반지수는 증가하였고 피부 산도는 감소하였다(p < 0.05). 조직병리학적 검사결과 염증세포 침윤과 부종성 변화에 의한 상피두께 증가 및 진피 결합조직의 감소가 특징적으로 나타났다. 또한 진피에서 pro-inflammatory cytokine인 TNF-${\alpha}$와 IFN-${\gamma}$ 수치 및 상피에서 apoptotic change의 지표인 caspase-3와 PARP 면역반응세포의 수치가 유의적으로 증가하였다(p < 0.01). 하지만 폴리감마글루탐산 적용으로 피부 생리학적 지표(p < 0.05) 및 조직병리학적 변화가(p < 0.01) 기본 수치로 회복되었다. 따라서 본 연구를 통해 개에서 DNCB에 의한 알레르기성 접촉피부염 유발 및 폴리감마글루탐산의 우수한 항염증 효과를 확인하였고 그 결과 폴리감마글루탐산은 향후 피부염에 대한 치료제로 사용할 수 있을 것으로 기대된다.

Gaucher병에서 resveratrol의 세포주기 조절자 p21을 통한 세포보호 효과 연구 (Resveratrol Upregulates p21, Cell Cycle Regulator, in Gaucher Disease Cells)

  • 고셔병은 세포내의 글루코세레브로시데이즈의 결핍으로 인하여 리소좀 내의 글루코세레브로사이드가 분해되지 못하고 축적되는 질환으로 알려져 있으며, 유형의 종류에 따라 신경퇴행성 질환으로 나타나는 것으로 보고되어 있으나 아직까지 정확한 기전이 밝혀져 있지 않다. 본 논문에서는 항산화 효과 및 신경보호 효과가 있는 것으로 알려진 레스베라트롤을 고셔병 환자의 fibroblast 세포에 투여하여 세포 생존율 변화 여부 및 세포주기 조절에 관하여 분자 생물학적 기전을 알아보고자 하였다. 고셔병 세포의 p21의 mRNA 발현 수준과 단백질 발현 양상을 확인한 결과 mRNA 상의 정량적 차이는 관찰되지 않았으나 단백질 발현수준은 레스베라트롤의 농도가 높아짐에 따라 증가 되는 것을 확인하였다. 또한 세포사멸의 표지 인자 단백질로 알려진 PARP의 변화양상을 확인한 결과 레스베라트롤의 농도가 높아짐에 따라 감소하는 것을 확인 할 수 있었다. 이를 통해 폴리페놀계 천연물인 레스베라트롤이 고셔병에서 세포 손상을 치유하며, 궁극적으로 세포사멸을 억제하는 효과를 가져올 것으로 생각할 수 있으며, 본 질환에서 병증을 완화 시킬 수 있을 것으로 사료된다.

A1E Induces Apoptosis via Targeting HPV E6/E7 Oncogenes and Intrinsic Pathways in Cervical Cancer Cells

  • A1E is an extract from traditional Asian medicinal plants that has therapeutic activities against cancers, metabolic disease, and other intractable conditions. However, its mechanism of action on cervical cancer has not been studied. In order to ascertain if A1E would have pronounced anti-cervical cancer effect, cervical cancer cells were incubated with A1E and apoptosis was detected by nuclear morphological changes, annexin V-FITC/PI staining, cell cycle analysis, western blotting, Reverse-transcription polymerase chain reaction, and measurement of mitochondrial membrane potential. Expression of human papiloma virus E6 and E7 oncogenes was down-regulated in A1E-treated cervical cancer cells, while p53 and retinoblastoma protein levels were enhanced. A1E also perturbed cell cycle progression at sub-G1 and altered cell cycle regulatory factors in SiHa cervical cancer cells. A1E activated apoptotic intrinsic pathway markers such as caspase-9, caspase-3 and poly ADP-ribose polymerase, and down-regulated expression of Bcl-2 and Bcl-xl. A1E induced mitochondrial membrane potential collapse and cytochrome c release, and inhibited phosphatidylinositol 3-kinase (PI3K)/Akt, key factors involved in cell survival signaling. Taken all these results, A1E induced apoptosis via activation of the intrinsic pathway and inhibition of the PI3K/Akt survival-signaling pathway in SiHa cervical cancer cells. In conclusion, A1E exerts anti-proliferative action growth inhibition on cervical cancer cells through apoptosis which demonstrates its anti-cervical cancer properties.

Construction of Mammalian Cell Expression Vector for pAcGFP-bFLIP(L) Fusion Protein and Its Expression in Follicular Granulosa Cells

  • FLICE inhibitory protein (FLIP) is one of the important anti-apoptotic proteins in the Fas/FasL apoptotic path which has death effect domains, mimicking the pro-domain of procaspase-8. To reveal the intracellular signal transduction molecules involved in the process of follicular development in the bovine ovary, we cloned the c-FLIP(L) gene in bovine ovary tissue with the reverse transcription polymerase chain reaction (RT-PCR), deleted the termination codon in its cDNA, and directionally cloned the amplified c-FLIP(L) gene into eukaryotic expression vector pAcGFP-Nl, including AcGFP, and successfully constructed the fusion protein recombinant plasmid. After identifying by restrictive enzyme BglII/EcoRI and sequencing, pAcGFP-bFLIP(L) was then transfected into follicular granulosa cells, mediated by Lipofectamine 2000, the expression of AcGFP observed and the transcription and expression of c-FLIP(L) detected by RT-PCR and Western blot. The results showed that the cattle c-FLIP(L) was successfully cloned; the pAcGFPbFLIP(L) fusion protein recombinant plasmid was successfuly constructed by introducing a BglII/EcoRI cloning site at the two ends of the c-FLIP(L) open reading frame and inserting a Kozak sequence before the start codon. AcGFP expression was detected as early as 24 h after transfection. The percentage of AcGFP positive cells reached about 65% after 24 h. A 1,483 bp transcription was amplified by RT-PCR, and a 83 kD target protein was detected by Western blot. Construction of the pAcGFP-bFLIP(L) recombinant plasmid should be helpful for further understanding the mechanism of regulation of c-FLIP(L) on bovine oocyte formation and development.
