Screening for Antioxidative Activity in Soybean Local Cultivars in Korea

  • Sixty local soybean cultivars were evaluated on the antioxidative activity by superoxide dismutase (SOD), 1, 1-diphenyl-2-picrylhydrazyl (DPPH), thiobarbituric acid(TBA), and chemiluminescence using the FI-CL system. Soybean were collected throughout the country, and were grown over two years (1997 and 1998) for measuring antioxidative activity in soybean seeds. There were differences in antioxidative activity depen-ding on the method of measurement and variation of the crop year. Soybeans from Kwangyang-shi-1 (76.78%) in 1997 and Kangjin-gun-3(79.14%) in 1998 showed the highest SOD activity, whereas those from Hwasoon-gun (80.43%) in 1997 and Kangjin-gun-2 (49.82 %) in 1998 exhibited the highest DPPH activity. Soybeans from Chongup-gun-2 (75.77%) in 1997 and from Yochon-shi-5 (69.17%) in 1998 exhibited the highest TBA activity, and those from Jinahn-gun (48.99%) in 1997 and Kohung-gun (49.73%) in 1998 exhibited the highest activity using the chemiluminescence method. These results suggest that it may be possible to develop soybean varieties with higher antioxidative activity.

Antioxidative Activities and Quantitative Determination of Gallotannins from Barks of Acer ginnala Maxim (신나무 수피로부터 Gallotannin 화합물의 항산화 활성 및 함량분석)

  • Activity guided isolation of 80% acetone extract from the barks of Acer ginnala Maxim. yielded five gallotannins [6-galloyl-1,5-anhydroglucitol (ginnalin B) (1), acertannin (3,6-digalloyl-1,5-anhydroglucitol) (2), methyl gallate (3), acertannin (2,6-digalloyl-1,5-anhydroglucitol) (4) and gallic acid (5)]. All of these isolated compounds from Acer ginnala(1-5) were firstly isolated from Acer ginnala Maxim. And contents of compounds from barks of Acer ginnala (Comp. 1: 0.73$\pm$0.002%, Comp. 2: 0.48$\pm$0.001%, Comp. 3: 0.66$\pm$0.002%, Comp. 4: 1.05$\pm$0.002% and Comp. 5: 0.29$\pm$0.001%) were evaluated by HPLC analysis. And, in order to evaluate anti-oxidative activities on Comp. 1-5 isolated from Acer ginnala, DPPH radical scavenging activity was measured in vitro. All of these isolated compounds from Acer ginnala exhibited potent DPPH radical scavenging activities.

Repressive effects of red bean, Phaseolus angularis, extracts on obesity of mouse induced with high-fat diet via downregulation of adipocyte differentiation and modulating lipid metabolism

  • Obesity is generally caused by quantitative changes in adipocyte differentiation and fat metabolism. Only a few studies have been determined the effect of red beans extract on obesity and plasma cholesterol concentration. We have been studied the functional activities of red-bean extracts including anti-oxidative effect against DNA and cell damages. Histological study including micro CT analysis showed that the accumulation of fat in hepatocytes and intestines was significantly decreased in red bean extract treated group. In addition, plasma cholesterol and triglyceride levels were decreased in blood samples. In addition, it was confirmed that the red bean extract inhibited the expression of $PPAR{\gamma}$, Fabp4 and RETN genes, which regulate total adipocyte differentiation and lipid metabolism. Red bean extract inhibits the expressions of transcription factors associated with adipocyte differentiation in a dose-dependent manner, thereby inhibiting fat accumulation and decreasing blood lipid levels in obese mice induced by high fat diet.

Phenolics of Propolis and in vitro Protective Effects against Oxidative Stress Induced Cytotoxicity

  • Anti-neurodegenerative activities of aqueous extract from propolis were investigated. The aqueous extracts showed strong antioxidant activities in malondialdehyde (MDA) assay, and it effectively inhibited lipid peroxidation. In addition, the aqueous extract presented protective effects against $H_2O_2-induced$ neurotoxicity in a dose-dependent manner. Our study verified that the aqueous extract has strong antioxidant activity and neuronal cell protective effect which is correlated with its total phenolics (290.75 mg GAE/g) including p-coumaric acid, vanillic acid and gallic acid. These phenolics of propolis may reduce the risk of neurodegenerative disorders, and can be utilized as effective and safe resources of functional food.

Antioxidant Activities of Methanol Extracts from Root Parts of Korean Salad Plants

  • Phenolics level, total flavonoid content, and antioxidant were determined from the methanol extracts of the eight medicinal plants using roots. Total phenolics were found as the highest levels in the methanol extracts of Arctium lappa, and followed by Youngia sonchifolia and Cirsium japonicum. Total amount of the each phenol compounds were detected in C. japonicum extracts ($319.2\;mg\;kg^{-1}$) as the greatest component, and followed by A. lappa ($96.3\;mg\;kg^{-1}$) and Y. sonchifolia ($22.9\;mg\;kg^{-1}$). Total flavonoid content showed the highest amount in methanol extracts from A. lappa (68.1 mg $100\;g^{-1}$) and followed by Y. sonchifolia (11.2 mg $100\;g^{-1}$) Lycoris radiate extracts (87.2%) had the highest nitrite scavenging activity and followed by A. lappa (81.5%) and Y. sonchifolia (77.5%). Methanol extracts of A. lappa at 25 mg $100\;g^{-1}$ exhibited the highest DPPH radical scavenging activity by 90.8%, even though less activity than synthetic antioxidants Vitamin C or butylated hydroxytoluene (BHT). Level of polyphenols was highly correlated with anti oxidative activity ($r^2$ = 0.85). The results suggest that several medicinal plants selected had the potent biological activities, and that their activities were differential depending on plant species.

Antioxidant Activity of Ethyl acetate Fraction of Oat in Caenorhabditis elegans (귀리 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과)

  • Oat, the seeds of Avena sativa L. (Gramineae), is an important dietary staple for people in many countries. Previous studies reported that A. sativa had various pharmacological effects such as anti-inflammatory, antitumor, neurotonic, and antispasmodic activities. In this study, Caenorhabditis elegans model system was used to investigate the antioxidant activity of methanol extract of oat. The ethyl acetate soluble fraction of the oat methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using GFP-expressing transgenic worm. As a result, the ethyl acetate fraction increased SOD and catalase activities, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity compared to the control.

Effect of Bojungikgi-tang on cytochrome P450 and LKB1-AMPK anti-oxidant signaling pathway (보중익기탕이 cytochrome P450 및 LKB1-AMPK 항산화 신호에 미치는 영향)

  • Objectives : We investigated the effects of Bojungikgi-tang (BJIGT) on P450 cytochrome enzyme and oxidative stress in the cells. Methods : We enrolled the HepG2 hepatocyte cell line to assess MTT assay, flow cytometer, and immunoblotting analysis. Expression of CYP450 was confirmed by immunoblotting analysis in the Huh7 cell line. Results : We determined that BJIKT markdely changed the expression of the CYP2C19, CYP2D6, and CYP2E1. Moreover, BJIKT inhibited the cell toxicity induced by arachidonic acid + iron treatment, as assessed by FACS analysis. BJIKT induced AMPK activation, which increased the phophorylation of ACC. Conclusions : This study verified the effects of BJIKT, on P450, ROS production, mitochondrial damage and AMPK signaling pathway, which might give us the scientific information about the traditional herbal prescription.

Changes in Antioxidant Activity and Total Phenolic Content of Water Spinach (Ipomoea aquatic Forsk.) under In Vitro Biomimicking System

  • The purpose of current study was to examine bioaccessibility of antioxidant activity and total phenolic content in each part of water spinach (Ipomoea aquatic Forsk.). In vitro biomimicking system simulated human digestive fluid was employed in order to measure bioavailable anti-oxidative effect and phenolic content. Antioxidant activity and total phenolic content was measured by using the DPPH method and the Folin-Ciocalteu assay, respectively. Stem of water spinach had a higher DPPH free radical scavenging effect (5.43 mg/mL for $IC_{50}$) than leaf (5.95 mg/mL for $IC_{50}$), while leaf had a greater level of total phenolic content (287.45 ${\mu}g$ GAE/mL) than stem (216.45 ${\mu}g$ GAE/mL). Bioaccessible antioxidant capacity and digestive stability of total phenolic content showed a similar pattern to what found in raw materials. Our result also indicated that total phenolic content was not found to be a major marker for prediction of antioxidant activity. It is plausible that other constituents such as vitamin E and C in water spinach could be contributors for antioxidant activities.

Developmental toxicity of dimethachlor during zebrafish embryogenesis mediated by apoptosis and oxidative stress

  • Dimethachlor is a synthetic herbicide, belonging to the chloroacetanilide group, that inhibits the undesirable growth of weeds via the suppression of very long-chain fatty acid synthesis. Although dimethachlor has been shown to run off from agricultural fields into aquatic ecosystems, the toxicity of dimethachlor on aquatic invertebrates and vertebrates is unknown. In our study, we assessed the toxicity of dimethachlor on developing zebrafish embryos by analyzing viability, hatching ability, and phenotypic changes. Embryonic viability decreased from 48 h post-fertilization (hpf) at the highest concentration of dimethachlor. Decreased hatching ratio, shortened body length, and pathological changes in the eye, heart, and yolk sac were observed at sub-lethal concentrations. Additionally, dimethachlor increased the number of apoptotic cells and level of reactive oxygen species 120 hpf. Our results indicate that dimethachlor may act as an anti-developmental toxicant when accumulated in an aquatic environment.

Quality Characteristics of Sponge Cakes made with Air Potato (Dioscorea bulbifera) Powder (열매마(Dioscorea bulbifera) 분말을 첨가한 스펀지케이크의 품질특성)

  • The purpose of this study was to investigate the possibility of using air potato (Dioscorea bulbifera) powder to make sponge cakes. The sponge cake batter was made by adding 0, 10, 20, 30, and 40% of air potato powder, and the resultant anti-oxidative properties and quality characteristics were analyzed. The study showed that the height, batter yield, and loss rate of sponge cakes decreased as increasing amounts of air potato powder were added, but the weight, viscosity, moisture content, and specific gravity increased. An evaluation of the color showed that the L and b values were highest in the control group but the a value was highest in the 40% group. There was no significant difference between samples in terms of cohesiveness, although the study showed a significant increase in the hardness, chewiness, and gumminess as the quantity of air potato powder in the sponge cakes increased. The total polyphenol content and DPPH radical scavenging activity increased noticeably as more air potato powder was added to the sponge cakes. The results thus showed that the study groups with the addition of air potato powder showed higher antioxidant activity than the control group.
