Searching and review on the Three Rs information in Korea: time for quality assessment and continued education

  • Scientists planning research that involves the use of animals are required by international and/or national law to examine the possibilities for the implementation of Replacement, Reduction and/or Refinement (the Three Rs principles of Russell and Burch) in experiments for research, testing, and education. There are two Korean laws legislating humane use of animals and ethical review prior to animal experiments. This report reviews current practice of the literature search by the researchers and protocol review by the Institutional Animal Care and Use Committees on the Replacement, Reduction and Refinement alternatives in Korea. The promotion and protection of the laboratory animals are one of the core competencies of investigators exploring the ethical conduct of research and good science.

광감작성 시험에서의 동물대체 시험법 (Invirto alternatives to photosensitization Test)

  • 동물시험을 최소화 하고, 동물시험에 사용되는 노력과 비용을 줄이기 위해 동일한 동물에서 광독성과 광알러지성을 함께 보는 새로운 방법을 제안하고, in vitro alternatives로서 photostability와 spectrophotometric carbonyl assay의 유용성을 검토하였다. Modified photosensitization model에서는 Ichikawa 방법과는 달리 동물수, 휴식 기간, 첩포 횟수를 단순히하고 폐쇄 첩포를 추가하였다. 방법의 유용성을 보기 위해 Benzocaine, 6-methyl coumarine을 Ichikawa 방법 및 본 시험 방법으로 결과를 비교한 결과 macroscopic, histologic examination에서 비슷한 결과를 얻었다. Photostability 시험에서는 pbotoreactive agent 들이 모두 20%이상의 흡광도 감소를 보여 pbotosensitization potential을 측정하기 위한 전 단계 실험으로서의 가능성을 보여주었고, spectrophotometric carbonyl assay에서는 salmonella를 이용한 광독성 실험에서는 나타나지 않았던 저농도의 Amthracene, Hematoporphyrin에서 조차도 phtotoxic prtential을 보여주어 in vitro alternatives로서의 가능성을 보여주었다.

Integration of the PubAnnotation ecosystem in the development of a web-based search tool for alternative methods

  • Finding publications that propose alternative methods to animal experiments is an important but time-consuming task since researchers need to perform various queries to literature databases and screen many articles to assess two important aspects: the relevance of the article to the research question, and whether the article's proposed approach qualifies to being an alternative method. We are currently developing a Web application to support finding alternative methods to animal experiments. The current (under development) version of the application utilizes external tools and resources for document processing, and relies on the PubAnnotation ecosystem for annotation querying, annotation storage, dictionary-based tagging of cell lines, and annotation visualization. Currently, our two PubAnnotation repositories for discourse elements contain annotations for more than 110k PubMed documents. Further, we created an annotator for cell lines that contain more than 196k terms from Cellosaurus. Finally, we are experimenting with TextAE for annotation visualization and for user feedback.

  • Objectives Korea's Act on the Registration and Evaluation of Chemicals (K-REACH) was enacted for the protection of human health and the environment in 2015. Considering that about 2000 new substances are introduced annually across the globe, the extent of animal testing requirement could be overwhelming unless regulators and companies work proactively to institute and enforce global best practices to replace, reduce or refine animal use. In this review, the way to reduce the animal use for K-REACH is discussed. Methods Background of the enforcement of the K-REACH and its details was reviewed along with the papers and regulatory documents regarding the limitation of animal experiments and its alternatives in order to discuss the regulatory adoption of alternative tests. Results Depending on the tonnage of the chemical used, the data required ranges from acute and other short-term studies for a single exposure route to testing via multiple exposure routes and costly, longer-term studies such as a full two-generation reproducibility toxicity. The European Registration, Evaluation, Authorization and Restriction of Chemicals regulation provides for mandatory sharing of vertebrate test data to avoid unnecessary duplication of animal use and test costs, and obligation to revise data requirements and test guidelines "as soon as possible" after relevant, validated replacement, reduction or refinement (3R) methods become available. Furthermore, the Organization for Economic Cooperation and Development actively accepts alternative animal tests and 3R to chemical toxicity tests. Conclusions Alternative tests which are more ethical and efficient than animal experiments should be widely used to assess the toxicity of chemicals for K-REACH registration. The relevant regulatory agencies will have to make efforts to actively adopt and uptake new alternative tests and 3R to K-REACH.

A comparison of metabolomic changes in type-1 diabetic C57BL/6N mice originating from different sources

  • Animal models have been used to elucidate the pathophysiology of varying diseases and to provide insight into potential targets for therapeutic intervention. Although alternatives to animal testing have been proposed to help overcome potential drawbacks related to animal experiments and avoid ethical issues, their use remains vital for the testing of new drug candidates and to identify the most effective strategies for therapeutic intervention. Particularly, the study of metabolic diseases requires the use of animal models to monitor whole-body physiology. In line with this, the National Institute of Food and Drug Safety Evaluation (NIFDS) in Korea has established their own animal strains to help evaluate both efficacy and safety during new drug development. The objective of this study was to characterize the response of C57BL/6NKorl mice from the NIFDS compared with that of other mice originating from the USA and Japan in a chemical-induced diabetic condition. Multiple low-dose treatments with streptozotocin were used to generate a type-1 diabetic animal model which is closely linked to the known clinical pathology of this disease. There were no significantly different responses observed between the varying streptozotocin-induced type-1 diabetic models tested in this study. When comparing control and diabetic mice, increases in liver weight and disturbances in serum amino acids levels of diabetic mice were most remarkable. Although the relationship between type-1 diabetes and BCAA has not been elucidated in this study, the results, which reveal a characteristic increase in diabetic mice of all origins are considered worthy of further study.

Advancing Risk Assessment through the Application of Systems Toxicology

  • Risk assessment is the process of quantifying the probability of a harmful effect to individuals or populations from human activities. Mechanistic approaches to risk assessment have been generally referred to as systems toxicology. Systems toxicology makes use of advanced analytical and computational tools to integrate classical toxicology and quantitative analysis of large networks of molecular and functional changes occurring across multiple levels of biological organization. Three presentations including two case studies involving both in vitro and in vivo approaches described the current state of systems toxicology and the potential for its future application in chemical risk assessment.

  • Cytotoxicity assays using artificial skins have been proposed as in vitro alternatives to minimize animal ocular and dermal irritation testing. Accordingly, the responses of artificial skins to the well-characterized chemical irritants toluene, glutaraldehyde, and sodium lauryl sulfate (SLS), and the nonirritant polyethylene glycol were studied. The evaluation of the irritating and non-irritating test chemicals was also compared with the responses observed in human dermal fibroblasts and human epidermal keratinocytes grown in a monolayer culture. The responses monitored included an MTT mitochondrial functionality assay. In order to better understand the local mechanisms involved in skin damage and repair, the production of several mitogenic proinflammatory mediators, interleukin-l$\alpha$, 12-HETE, and 15-HETE, was also investigated. Dose-dependent increases in the levels of かIn and the HETEs were observed in the underlying medium of the skin systems exposed to the two skin irritants, glutaraldehyde and SLS. The results of the present study show that both human artificial skins can be used as efficient in vitro testing models for the evaluation of skin toxicity and for screening contact skin irritancy.

OECD TG데이터를 이용한 그래프 기반 딥러닝 모델 분자 특성 예측 (Toxicity prediction of chemicals using OECD test guideline data with graph-based deep learning models)

  • 본 연구에서는 OECD test guideline 데이터를 이용하여 graph기반 딥러닝 모델들의 성능을 비교하고자 한다. OECD TG는 화학물질들이 인체와 환경에 미칠 잠재적 영향에 대해 시험하는 방법이며, 많은 실험이 동물실험을 통해 독성을 확인한다. 동물실험은 많은 시간과 비용이 들며, 윤리적 이슈가 있어 대안을 찾거나 최소화하는 방법들이 연구되고 있다. 딥러닝은 화학물질을 활용하는 다양한 분야에서 사용되고 있으며, 독성예측 분야에도 사용되고 있으며, 특히 graph 기반 모델에 대한 연구가 활발하다. 우리의 목표는 OECD TG 데이터에 대한 graph기반 딥러닝 모델들의 성능을 비교하여 가장 성능이 좋은 모델을 찾는 것이다. 우리는 OECD에서 운영하는 웹사이트 eChemportal.org에서 OECD TG를 따른 결과를 수집하였으며, 전처리 과정을 통해 학습이 불가능하거나 부적절한 화학물질은 제거하였다. 수집된 OECD TG데이터와 화학물질 특성 예측 성능의 벤치마크 데이터셋인 MoleculeNet 데이터를 활용하여 5개의 graph기반 모델들의 독성 예측 성능을 비교하였다.

아크릴계 내화도료 연소가스의 유해성 평가와 배기온도에 대한 고찰 (Review of Hazard Test of Combustion Gas and Exhaust Temperature of Acrylic Fire Protection Paint)

  • 국내 건축물에 사용되는 내화도료는 품질시험을 통하여 내화구조로 인정받아야 사용할 수 있다. 품질시험은 내화시험 및 부가시험이며 내화도료의 부가시험 항목은 가스유해성과 부착강도이다. 이중 가스유해성의 경우 관련 KS 등에 따라 동물실험을 하며 매건 16마리의 살아있는 쥐가 희생된다. 이에 실험자는 윤리적 부담을 느끼게 되며 실험기관은 관련 법률에 따른 실험동물운영위원회 운영 등 행정적 부담을 지게 된다. 이에 연소 가스의 성분 분석을 통한 유해성 측정 등 상기 동물실험을 대체하려는 연구와 노력이 지속되고 있으나 현재까지는 동물실험이 유지되고 있다. 연소가스의 유해성 평가 시 시험체 가열에 따라 발생하는 배기온도가 측정되는데 특정 타입 내화도료에 대한 배기온도를 분석하여 유의한 초기 반응 양상을 파악할 수 있다면 동물실험 대체에 기여할 수 있을 것이라 사료된다. 이에 본 논문을 통하여 가스유해성 시험 시 쥐의 행동정지시간과 배기온도와의 관계를 파악하여 현 동물실험의 개선점을 찾아보고자 하였다.

3차원 배양 시스템을 이용한 돼지 유선 상피 세포 주 특성과 설정 (Establishment and characterization of porcine mammary gland epithelial cell line using three dimensional culture system)

  • 조직특이적 프로모터 및 벡터를 연구하고 검증하기 위해서는 조직 및 종의 특이성을 유지하는 세포 시스템을 개발하는 것이 바람직하다. 이러한 시스템은 형질전환동물 모델에 대한 효과적인 대안이다. 우리는 베타 카제인 (CSN2)의 세포 형태와 mRNA 수준에 기초하여 일차 배양으로부터 돼지 유선 상피 세포 주 (PMECs)를 확립하였다. 선택된 PMECs는 cytokeratin 항체에 의해 염색되었으며, 유선 상피 세포에 존재한다고 생각되어지는 유즙 단백질 유전자 (CSN2, 락토페린 및 유청 단백질)를 발현하는 것으로 나타났다. 또한, 3D 배양에서 PMECs932-7의 acini 구조를 확인하기 위해 살아있는 세포를 핵산에 결합하는 SYTO-13으로 염색하였다. 우리는 마트리겔 (matrigel)에 있는 PMECs의 acini가 말초 세포의 응집에 의해 형성되고 공간의 lumen을 특징으로 한다는 것이 관찰하였다. 우리는 PMECs의 matrigel 사용법과 세포 밀도를 포함한 세포 배양 조건의 영향을 시험함으로서 시스템을 시연했다. 이러한 결과는 PMCEs의 유선 상피 세포는 유전적 또는 구조적 특징을 갖고 있음을 시사하고 있다.
