Effect of oral antioxidants on the progression of canine senile cataracts: a retrospective study

  • Background: Cataracts are the leading cause of impaired vision or blindness in dogs. There are many antioxidants that can prevent cataract progression, but whether they are clinically effective in dogs has not been established. Objectives: To analyze the delaying or preventing effect of oral antioxidants on canine senile cataracts through retrospective analysis. Methods: Medical records of dogs from January 1, 2015 to July 10, 2020 were reviewed. Dogs that were 8 yr of age or older with senile cataracts were included in this study. The dogs were divided into two treatment groups (dogs administered with Ocu-GLO supplement and dogs administered with Meni-One Eye R/C supplement) and a control group (dogs that were not administered any supplement). Dogs with incipient and immature cataracts were included in this study. Altogether, 112 dogs (156 eyes) with incipient cataracts and 60 dogs (77 eyes) with immature cataracts were included. The period of time that cataracts progressed from incipient to immature, and from immature to mature was recorded for each dog. Results: There was no significant delaying effect on the progression of incipient cataracts. However, both Ocu-GLO (hazard ratio = 0.265, p = 0.026) and Meni-One (hazard ratio = 0.246, p = 0.005) significantly delayed the progression of immature cataracts compared to the control group. Conclusions: Although there was no significant delaying effect of oral antioxidants on incipient cataract progression, antioxidants could be used to delay the progression of senile immature cataract.

Ginsenoside compound K reduces the progression of Huntington's disease via the inhibition of oxidative stress and overactivation of the ATM/AMPK pathway

  • Background: Huntington's disease (HD) is a neurodegenerative disorder caused by the expansion of trinucleotide CAG repeat in the Huntingtin (Htt) gene. The major pathogenic pathways underlying HD involve the impairment of cellular energy homeostasis and DNA damage in the brain. The protein kinase ataxia-telangiectasia mutated (ATM) is an important regulator of the DNA damage response. ATM is involved in the phosphorylation of AMP-activated protein kinase (AMPK), suggesting that AMPK plays a critical role in response to DNA damage. Herein, we demonstrated that expression of polyQ-expanded mutant Htt (mHtt) enhanced the phosphorylation of ATM. Ginsenoside is the main and most effective component of Panax ginseng. However, the protective effect of a ginsenoside (compound K, CK) in HD remains unclear and warrants further investigation. Methods: This study used the R6/2 transgenic mouse model of HD and performed behavioral tests, survival rate, histological analyses, and immunoblot assays. Results: The systematic administration of CK into R6/2 mice suppressed the activation of ATM/AMPK and reduced neuronal toxicity and mHTT aggregation. Most importantly, CK increased neuronal density and lifespan and improved motor dysfunction in R6/2 mice. Conversely, CK enhanced the expression of Bcl2 protected striatal cells from the toxicity induced by the overactivation of mHtt and AMPK. Conclusions: Thus, the oral administration of CK reduced the disease progression and markedly enhanced lifespan in the transgenic mouse model (R6/2) of HD.

Antibiofilm Activity and Binding Specificity of Polyclonal DNA Aptamers on Staphylococcus aureus and Escherichia coli

  • Aptamers are short, chemically synthesized, single-stranded DNA or RNA oligonucleotides that fold into unique three-dimensional structures. In this study, we aim to determine the antibiofilm activity and binding specificity of the six polyclonal DNA aptamers (S15K3, S15K4, S15K6, S15K13, S15K15, and S15K20) on Staphylococcus aureus BPA-12 and Escherichia coli EPEC 4. Aptamer S15K6 showed the highest percentage of antibiofilm activity against S. aureus BPA-12 (37.4%) as shown by the lowest OD570 value of 0.313. Aptamer S15K20 showed the highest percentage of antibiofilm activity against E. coli EPEC 4 (15.4%) as shown by the lowest OD570 value of 0.515. Aptamers S15K13 and S15K20 showed antibiofilm activities against both S. aureus BPA-12 and E. coli EPEC4, and thus potentially have broad reactivity. Furthermore, based on the binding capacity and Kd values from our previous study, the binding specificity assay of selected polyclonal DNA aptamers (S15K3 and S15K15) against S. aureus BPA-12, E. coli EPEC 4, S. aureus BPA-6, S. agalactiae, E. coli MHA-6, and Listeria monocytogenes were performed using qPCR. Aptamers S15K3 and S15K15 showed specific binding to S. aureus BPA-12, E. coli EPEC 4, S. aureus BPA-6, and S. agalactiae, but could not bind to E. coli MHA-6 and L. monocytogenes. Therefore, this study showed that the polyclonal DNA aptamers have antibiofilm activity and were able to bind to S. aureus BPA-12 and E. coli EPEC 4 bacteria.

돼지정자의 수정능력과 Reactive Oxygen Species의 관계분석 I. Xanthine과 Xanthine Oxidase에 의한 정자의 전배양 (Analysis of Relationship Between Spermatozoa Ability and Reactive Oxygen Species in Porcine: I. Sperm Preincubation by Xanthine and Xanthine Oxidase)

  • 본 연구는 xanthine과 xanthine oxidase가 첨가된 배양액내에서 전배양된 돼지동결정액의 수정능력에 있어서 catalase의 영향을 검토하였다. 체외수정을 위한 기본 배양액내에서 0 또는 30분간 전배양한 정자는 catalase를 첨가 (40 및 15%)한 경우 보다는 무첨가 (66 및 38%)시 유의적으로 높은 정자침입율을 나타냈지만 (p<0.05), 배양액내에 xanthine을 첨가해 정자를 0, 30 및 60분간 전배양했을 때에는 catalase 무첨가 (33, 41 및 19%) 보다는 첨가시 (68, 70 및 49%) 유의적으로 높은 정자침입율이 인정되었다 (p<0.05). 그러나 xanthine oxidase를 첨가하여 정자의 전배양을 행하지 않은 경우는 catalase의 첨가 (13%) 보다는 무첨가 (51%)시 유의적으로 높은 정자침입율을 나타냈지만 (p<0.01), 전배양(30 및 60분)후에는 catalase의 존재유무와 정자전배양시간에 관계없이 매우 낮은 정자침입율 $(10\sim21%)$을 나타냈다. xanthine과 xanthine oxidase를 동시에 첨가하여 0, 30 및 60분간 정자를 전배양한 경우 catalase의 무첨가 (14, 4 및 8%)보다 첨가 (75, 55 및 52%)시 유의적으로 높은 정자침입율을 나타냈다 (p<0.001). 한편, 다정자침입율은 xanthine, xanthine+xanthine oxidase의 첨가시 정자전배양기간이 길어짐에 따라 감소하였으며, catalase의 첨가보다는 무첨가시 낮은 다정자침입율을 나타냈다. 본 연구의 결과로부터 xanthine과 xanthine oxidase를 동시에 첨가시 catalase의 존재는 정자의 전배양후에도 다정자침입을 억제하면서 수정능력의 유지를 위해 매우 효과적인 것으로 추측되었다.

한우 송아지의 설사 분변에서 Escherichia coli(K99), Clostridium perfringens와 Cryptosporidium parvum의 검출 (Detection of Escherichia coli(K99), Clostridium perfringens and Cryptosporidium parvum in Diarrhetic Feces of Korean Native Calves)

  • The prevalence of E. coli(K99), Clostridium perfringens and Cryptosporidium parvum on acute diarrhea in suckling Korean native calves was evaluated in the field by a veterinary practice. In diagnosis, fecal samples were directly collected from calves that had diarrhea between 2 and 98 days of age. 40 samples were analyzed in October, 2006 and December, 2006. Clostridium perfringens and Cryptosporidium parvum were detected in 15(37.5%) and 4(10.0%) of the samples from diarrhetic calves, respectively. However, E. coli(K99) was not detected in the samples from diarrhetic calves. There was no significant difference(p>0.05) between October(5, 25.0%) and December(10, 50.0%) in incidence of detected Clostridium perfringens from diarrhetic calves. On the other hand, significant differences (p<0.05) in the detection rate of Clostridium perfringens were found between the within 1 month age and all other age groups. In the detection of Cryptosporidium parvum, there was no significant difference(p<0.05) between October (2, 10.0%) and December(2, 10.0%) in the incidence of detected Cryptosporidium parvum from diarrhetic calves. These results suggest that causative agents of calf diarrhea occurred frequently with Clostridium perfringens infection than E. coli(K99) and Cryptosporidium parvum.

  • The purpose of this study is to determine the changes of ultrasonographic appearance of stomach, pylorus and duodenum in relation to food intake and administration of contrast medium in dogs. Ultrasonograms were obtained with 5.0MHz sector transducer. After placing the dogs in dorsal recumbency, scanning was performed at the 8th intercostal space. The mean inner diameter of stomach immediately after food intake, 2, 4 and 6 hours was 47.9, 51.5, 46.8 and 40.6mm, respectively. The diameter of pylorus immediately after food intake, 2, 4 and 6 hours was 7.4, 9.7, 8.9 and 6.0mm, respectively(p<0.01). The diameter of duodenum immediately after food intake 2, 4 and 6 hours was 12.5, 7.9, 11.9 and 11.8mm, respectively(p<0.01). Before administration of contrast medium, the mean inner diameter of stomach, pylorus and duodenum was 38.6, 7.2 and 9.5mm, respectively. After administration of contrast medium, the diameters of stomach, pylorus and duodenum was 42.8, 7.7 and 9.9mm, respectively. It may be concluded that the ultrasonographic values determined in this study can be used as references for the diagnosis of gastrointestinal disease in dogs.

Development of Modified Selective Media to Differentiate Cryptococcus Species Complex and its Serotypes using Natural Materials

  • The formation of brown colonies due to phenol oxidase activity on classic agar media containing natural material extracts of Helianthus annuus or on medium containing L-3,4-dihydroxyphenylalanine has been used to identify Cryptococcus species complex. In this study, various natural materials were used to develop a modified medium and to identify five major serotypes of Cryptococcus species complex. Serotypes A, D, and A/D were pigmented on medium using Perilla frutescens var. japonica Hara (PerJ agar) after a three-day incubation. Serotypes B and C were pigmented on PerJ agar after four- and five-day incubations, respectively. Growth time and pigmentation of the five serotypes occurred more rapidly on PerJ agar than on the other media. In addition, colony morphology, size, and pigmentation were specific by serotype. In conclusion, PerJ agar should be used in clinic settings to identify Cryptococcus species complex and its serotypes rapidly.

Histopathologic and electron microscopic findings of canine malignant melanoma from the lower limb and digit

  • A 16-year-old female mixed dog was submitted for examination at the pathology division of national veterinary research and quarantine service (NVRQS). Grossly, white or grayish spherical, multinodular, firm to friable masses were present in the tibiofibula to the pharenge area of the right limb, and dysphagia, breathing difficulties and tachypnea were shown. Various-sized white or grayish black masses were scattered in lungs and diaphragm and one mass was observed in the trachea and in the jejunum, respectively. Histopathologically, the neoplastic cells were composed of polygonal or spindle shaped cells with various sized round to oval nuclei and abundant cytoplasm. These cells formed lobules or nests separated by fine connective tissue and contained little amount of melanin pigments. Melanin pigments were stained dark gray or black with Fontana-Masson method. Melanosomes were also ultrastructurally demonstrable by electron microscopy. Based on above results, the present case was diagnosed as canine malignant melanoma originating from the lower limb and digit.

백년초 추출물(Opuntia ficus-indica)을 이용한 개의 만성 외이도염 치료 효과 (The Efficacy of Opuntia ficus-indica for the Treatment of Chronic Otitis Externa in Dogs)

  • To determine the antimicrobial and anti-inflammatory efficacy of Opuntia ficus-indica for chronic otitis, we evaluated the effects of topical applications of the methanol extracts with 20 cases of dogs which had chronic malignant otitis by pathogens with antibiotic resistance. The dogs had revealed recurrent symptoms of malignant otitis and were not treated by conventional therapeutic agents. However, in this study, the clinical cure rates of Opuntia ficus-indica was 75% and the average alleviation period was $1.21{\pm}0.42$ week, and the mean recovery period was $1.06{\pm}1.06$ week after the initiation of treatment. As the results of this study, topical Opuntia ficus-indica extracts was found to be highly effective for the treatment of chronic malignant otitis with clinical cure rates of 75% within 1.06 weeks of therapy. Further evaluation of Opuntia ficus-indica extracts will allow us to establish and to optimize the therapeutic strategy for the malignant otitis in veterinary practice, and the potential usefulness of this complementary treatment on recurrent infectious pathology.

개 파보바이러스성 장염에서 산-염기 장애의 진단 (Diagnosis of Acid-Base Disorders in Canine Parvoviral Enteritis)

  • Abstract: This study was aimed to diagnose acid-base disorders of dogs with canine parvoviral enteritis (CPE) and data to establish a rational fluid therapy regimen for patients with CPE. A total of 43 dogs which had clinical signs of CPE and had detected canine parvovirus by polymerase chain reaction, were bled anaerobically from jugular vein at the time of admission. Blood chemical test, determination of electrolytes and blood gas analysis were conducted, and calculated values were obtained from each measured items. The values of blood chemical and electrolytes of dogs with CPE were various depending on the degree of clinical signs, and these tests were not specific to diagnose for CPE. Hypochloremia (20.9%), hyperchloremia (11.6%), hypokalemia (7.0%), hyperkalemia (11.6%), hyponatremia (9.3%) and hypernatremia (18.6%) were diagnosed as abnormalities of electrolytes from 43 dogs with CPE. The 29 out of 43 dogs (67.4%) were metabolic acidosis and 3 dogs (7.0%) were metabolic alkalosis. The acid-base status of 11 dogs out of 43 dogs (25.6%) was normal.
