Effects of High Pressure Treatments and Added Binders on the Physico-Chemical Properties of Restructured Pork Meat

  • In general, the application of HP contributes to improvement of functionalities of meat protein. In the current study, HP allowed the lower thermal processing of restructured meat product. Low thermal processing allowed fresh-like meat color which is one of problems in hot-set restructuring, while they showed slight discolorization induced by HP. In addition, HP processing combined with thermal processing could be achieved the palatable binding strength in restructured meat product. The addition of non-meat protein had an effect in binding strength. However, they showed no effect on water binding properties at 200 MPa, especially in milk proteins such as casein and whey protein. This is probably due to protein aggregation or to increase in surface hydrophobicity under HP. This result indicates that the application of HP on meat restructuring is more significant than the addition of binders. Therefore, the application of HP has apotential benefit in restructured meat product, and further investigations are needed.

Biohydrogenation Pathways for Linoleic and Linolenic Acids by Orpinomyces Rumen Fungus

  • The objective of this study was to identify biohydrogenation pathways for linoleic, linolenic, oleic and stearic acids by Orpinomyces species of rumen fungus during in vitro culture. Biohydrogenation of linoleic acid produced conjugated linoleic acid (cis-9, trans-11 C18:2), which was then converted to vaccenic acid (trans-11 C18:1) as the end product of biohydrogenation. Biohydrogenation of linolenic acid produced cis-9, trans-11, cis-15 C18:3 and trans-11, cis-15 C18:2 as intermediates and vaccenic acid as the end product of biohydrogenation. Oleic acid and stearic acid were not converted to any other fatty acid. It is concluded that pathways for biohydrogenation of linoleic and linolenic acids by Orpinomyces are the same as those for group A rumen bacteria.

RESTRICTION BY MOTHERWORT (Leonurus sibiricus L) OF LACTATION SUPPRESSED BY PREGNANCY-DEPENDENT MAMMARY TUMORS IN GR/A MICE

  • As a possible step to evaluate in the mammary gland the role of motherwort (Leonurus sibiricus L), a representative medicinal plant used traditionally for the therapy of gynecologic diseases, the effects of the agent on lactation which was suppressed by preganacy-dependent mammary tumors (PDMT) were studied in GR/A mice. Beginning the day of placing with males at 45-50 days of age, female mice were given 60% methanol-extract of the aerial part of motherwort as drinking water at the concentration of 0.5% throughout the experiment. Mice developing PDMT during pregnancy [PDMT(+)] and given motherwort were similar to mice developing no PDMT [PDMT(_)] with or without motherwort treatment and were significantly higher than PDMT(+) mice given tap water in litter growth and mammary RNA/DMA ratio on day 12 of the 2nd lactation. Mammary DNA and RNA contents were also elevated by motherwort in PDMT(+) mice. The results suggest that motherwort can ameliorate lactation suppressed by PDMT through its stimulation of both growth and function of the mammary glands.

Effects of Fruit By-product Extracts on Blood Characteristics, Antioxidant Activity, and Immune Response in Pigs

  • This experiment was conducted to determine the effects of extracts from fruit by-products on the blood characteristics, antioxidant activities, and immune response to Escherichia coli lipopolysaccharide (LPS) in growing pigs. A total of 96 pigs [(Landrace ${\times}$ Yorkshire) ${\times}$ Duroc] with an initial BW of $27.94{\pm}0.92kg$ were used in a 6-week feeding trial. The pigs were randomly placed into one of four treatment groups with six replications (four pigs per replication) per treatment according to their initial BW. Treatments were: 1) CON (basal diet), 2) PRO (CON + 0.5% procyanidin), 3) HES (CON + 0.5% hesperetin), 4) TAN (CON + 0.5% tannin). At the end of the sixth week, five pigs (total 20 pigs, $BW=27.94{\pm}0.92kg$) were selected from each treatment and injected with LPS ($100{\mu}g/kg$ of BW). Blood samples were collected 3 h after LPS injection to assess anti-oxidative and inflammatory responses. After the LPS challenge, the concentration of serum cholesterol decreased with fruit by-product treatment compared with CON (p<0.05). The administration of TAN increased the concentration of blood total protein compared with the CON group 3 h after LPS challenge (p<0.05). The albumin concentration was also higher with PRO treatment compared to HES treatment (p<0.05). The concentration of IgM was increased by fruit by-product supplementation at 0 and 3 h (p<0.05). In addition, IgG concentration was higher in PRO, HES, and TAN treatments compared to CON treatment at 0 h, and IgG concentrations were also higher in the HES group compared to the CON group at 3 h (p<0.05). The concentration of IgA also increased with fruit by-product treatments at 3 h (p<0.05). In conclusion, dietary supplementation with fruit by-products may moderate the immune response after a LPS challenge in growing pigs.

인삼부산물 급여에 따른 비육돈의 생산성 및 육질 특성에 미치는 영향 (Effects of Dietary Ginseng By-product on Growth Performance and Pork Quality Parameters in Finishing Pigs)

  • 본 시험은 인삼부산물 급여 유무에 따른 돼지 생산성, 혈액, 도체 및 육질 특성에 관하여 조사하였다. 시험 동물은 2원 교잡종(LandraceYorkshire) 육성-비육돈 24두를 공시하였으며 개시 체중은 65.81±2.02kg이였다. 기초 사료를 대조구(CON)로 설정하였고, 처리구(GBP)은 인삼 부산물을 기초사료의 lupin 2.5%로 대체하였다. 시험구는 완전임의 배치법으로 암·수 동일한 비율로 각 처리구당 4마리씩 3반복 실시하였다. 생산성에서 일당사료섭취량은 GBP가 CON 보다 유의적으로 낮았다(P<0.0001). 혈장의 생화학적 조성에서는 total protein, blood urea nitrogen, glucose, albumin, calcium 그리고 inorganic phosphate는 각각 GBP가 CON 보다 유의적으로 높은 함량을 나타내었다. 도체 및 육질 특성은 GBP 및 CON간에 유의적인 차이가 나타나지 않았다. 돈육의 총 ginsenoside 함량은 GBP가 CON 보다 유의적으로 높은 함량을 나타내었고 TBRAs는 저장 6일과 13일에 GBP가 CON 보다 유의적으로 낮았다. 이상의 시험 결과를 종합하면, 인삼부산물 급여는 혈장의 화학적 조성의 변화를 가져왔고, 돈육의 ginsenoside의 함량을 증가시키고 지방산패도를 감소시켰는데 이 사실은 인삼부산물이 돼지의 사료첨가제로 이용될 수 있는 가능성을 나타낸다.

Potential to mitigate ammonia emission from slurry by increasing dietary fermentable fiber through inclusion of tropical byproducts in practical diets for growing pigs

  • Objective: Research was conducted to test the effect of including fiber-rich feedstuffs in practical pig diets on nutrient digestibility, nitrogen balance and ammonia emissions from slurry. Methods: Three Vietnamese fiber sources were screened, namely cassava leaf meal (CL), cassava root residue (CR), and tofu by-product (TF). Accordingly, a control diet (Con) with 10% of dietary non-starch polysaccharides (NSP) and three test diets including one of the three fiber-rich feedstuffs to reach 15% of NSP were formulated. All formulated diets had the same level of crude protein (CP), in vitro ileal protein digestible and metabolisable energy, whereas the in vitro hindgut volatile fatty acid (VFA) production of the test diets was 12% to 20% higher than the control diet. Forty growing barrows with initial body weight at $28.6{\pm}1.93kg$ ($mean{\pm}standard$ deviation) were allocated to the four treatments. When pigs reached about 50 kg of body weight, four pigs from each treatment were used for a nitrogen balance trial and ammonia emission assessment, the remaining six pigs continued the second period of the feeding trial. Results: The TF treatment increased fecal VFA by 33% as compared with the control treatment (p = 0.07), suggesting stimulation of the hindgut fermentation. However, urinary N was not significantly reduced or shifted to fecal N, nor was slurry pH decreased. Accordingly, ammonia emissions were not mitigated. CR and CL treatments failed to enhance in vivo hindgut fermentation, as assessed by fecal VFA and purine bases. On the contrary, the reduction of CP digestibility in the CL treatment enhanced ammonia emissions from slurry. Conclusion: Dietary inclusion of cassava and tofu byproducts through an increase of dietary NSP from 10% to 15% might stimulate fecal VFA excretion but this does not guarantee a reduction in ammonia emissions from slurry, while its interaction with protein digestibility even might enhance enhanced ammonia emission.

Effects of Replacing Fish Meal With Ensiled Shrimp By-Product on the Performance and Carcass Characteristics of Growing Pigs

  • A feeding trial was conducted to evaluate the effects of replacing fish meal (FM) with ensiled shrimp by-product (ESB) in a cassava root meal and rice bran-based diet on the performance and carcass characteristics of growing pigs. Thirty six crossbred (Large White$\times$Mong Cai) pigs, with an average initial BW of 19.4 kg, were randomly allocated to one of three different dietary treatments in which the crude protein of the FM was replaced with 0, 50 or 100% ESB. The animals were fed restrictedly and at the end of the experiment at a BW of about 90 kg, 6 representative animals in each treatment group were slaughtered for carcass quality evaluation. Animal growth performance and daily feed intake were significantly reduced (p<0.05) by the inclusion of shrimp by-products in the diets, whereas feed conversion ratios and carcass measurements were not significantly affected (p>0.05). Daily weight gains of the pigs fed the 100% FM diet and 50% ESB diet were significantly (p<0.05) higher than those of pigs fed the 100% ESB diet. In conclusion, from an economical as well as performance point of view, ESB can replace 50% of the crude protein of FM in cassava root meal and rice bran-based diets for growing pigs with a low genetic growth potential.

Perception of the HACCP system operators on livestock product manufacturers

  • The purpose of this study was to investigate crucial factors on HACCP system implementation in domestic livestock product plants, and to offer job satisfaction and the career prospect of HACCP system operators. The survey was carried out by selecting 150 HACCP system operators who implemented HACCP system. The respondents claimed that the most important contents in HACCP system operation were to assemble HACCP team (21.8%), and the second was to monitoring (20.0%). Documentation and recording (16.9%) and verification (11.1%) were followed. The respondents answered the major factor in sanitation management was cleaning/washing/disinfection (18.9%) and inspection (18.4%). The results showed that there were significant differences in the prospect of occupation in HACCP system operator by the gender (p < 0.015), age, livestock product facilities, service period, and position (p < 0.001). The respondents from HACCP system operator were satisfied with their job (73%) and also showed optimistic prospect of occupation (82%).

Study on radiolabeling method of chitosan to improve the radiolabeling yield of the final product

  • Chitosan is a polysaccharide derived from chitin by deacetylation. Chitosan is non-toxic, biodegradable, and biocompatible, so that it can be used in wide variety of medical applications such as wound healing and antimicrobial biomaterials. It also used as dermal fillers due to its ability to inject with liquid formulations. For investigation on in vivo distribution of these chitosans, Bolton-Hunter-conjugated chitosan (Chitosan-BH) was synthesized by the reaction between the primary amino group of chitosan and N-hydroxysuccinimide ester group of Bolton-Hunter reagent. Then Chitosan-BH was radiolabeled with 125I (Chitosan-BH-125I) using a Chloramine-T method. The effects of each radiolabeling step on the radiolabeling yield of the final product were tested. The results showed that purification step had significant effects on the radiolabeling yield of the final product. Finally, SPECT/CT images were obtained to evaluate in vivo uptake of the radiolabeled chitosan (Chitosan-BH-125I) in several organs. The highest uptake was found in the site of injection at 21 days post-injection. The results of this study suggest that chitosan is expected to be useful for biomaterials of dermal fillers.

Antioxidative and Probiotic Properties of Lactobacillus gasseri NLRI-312 Isolated from Korean Infant Feces

  • We selected a Lactobacillus spp. from Korean healthy infant feces based upon their antioxidant activity. This strain was identified as Lactobacillus gasseri by 16S rDNA sequencing, and named Lactobacillus gasseri NLRI-312. In the present study, we investigate the protective effect of this strain on the $H_2O_2$ induced damage to cellular membrane lipid and DNA in Jurkat cells. To estimate the extent of cellular lipid peroxidation inhibition, MDA (malondialdehyde) was measured, and DNA damage was tested by the comet assay. We also examined probiotic properties including tolerance to acid and bile, antibiotic resistance. From the results obtained, the supplementation of Jurkat cells with NLRI-312 decreased in DNA damage, while no effect was shown on MDA decrease. In probiotic properties, this strain was resistance to both acid and bile, showed considerably higher survival when incubated in pH 2 or 1% bile salts (w/v). We concluded that the NLRI-312 could be used as potential probiotic bacteria, with the effect of reducing DNA damage induced by $H_2O_2$.
