Mechanism and Regulation of Amino Acid Transport in Mammary Gland - Review -

  • Several amino acid transport systems in mammary gland have been characterized during the last few years. These systems may be divided into two broad categories based on whether they are sodium-dependent or $Na^{+}$-independent, and each of these categories is subdivided into 3 groups depending on whether the systems prefer zwitterionic, cationic or anionic substrates. The zwitterion preferring transport processes in mammary gland are $Na^{+}$-dependent system A and $Na^{+}$-independent systems L and T. System $y^{+}$ is a $Na^{+}$-independent transporter of cationic amino acids and $X_{AG^{-}}$ is a $Na^{+}$-dependent system for anionic amino acids. A ($Na^{+}+Cl^{-}$)-dependent system, selective for $\beta$-amino acids has been reported in rat mammary tissue. In addition, there is yet another class of transporters that have still broader specificity. The $Na^{+}$-dependent systems $BCl^{-}$-dependent and $BCl^{-}$-independent and $Na^{+}$-independent system $y^{+}L$ have been reported to mediate the transport of zwitterionic as well as cationic amino acids. Each system has been characterized with respect to its substrate specificity, affinity, kinetics and ion-dependence. Transport of amino acids by mammary tissue is regulated by i) the intracellular substrate concentration, ii) lactogenic hormones and iii) milk stasis. Four of the above transport systems (i.e. A, L, $y^{+}$ and $BCl^{-}$-independent) are up-regulated by lactogenic hormones (insulin, cortisol and prolactin) in mammary gland.

동물성 아미노산 시용이 토양이화학성과 노지고추 생육 및 품질에 미치는 영향 (Effect of "Animal Amino Acid's Bestamin" on the Physicochemical Properties of Soil, the Growth and Fruit Quality of Hot Pepper (Capsicum annuum L.))

  • This study was carried out to evaluate the effect of applying levels of Bestamin, animal amino acid, on growth and quality of hot pepper and physicochemical properties of soil. Treatment was given with 200, 400, 800, 1600kg per 10a to control of Bestamin, difference of physicochemical properties was lower than Bestamin, and $P_2O_5$ seems to be reduced, also $K^+$, $Ca^{++}$ were obviously low compared to the control. The content of $NO_{3-}N$ was low compared to control at the Bestamin treated plot. Plant height was longest at 800 treatment and main stem length, main stem weigh and the number of leaves were significantly different with 400 and 800 treatment. No difference was found among the fruit weight, length, diameter in first harvest, but there was significantly different at control of 2nd, 3rd harvest and more increased than 400 and 800 treatment. Fruit weight per plant was the heaviest at 400 and 800 treatment, and the number of fruit was no difference at red pepper but increased with 400 and 800 at green pepper, and yield per 10a was significantly increased to 4503.6kg and 4582.5kg, respectively. Nitrogen in mesophyll accumulation content was obviously reduced at Bestamin treatment compared to control, and amino acid was reduced with control.

Additivity of digestible energy and nutrient concentrations in hatchery byproducts fed to nursery pigs

  • Objective: The objective was to test additivity of digestible energy and nutrient concentrations in the hatchery byproduct mixture fed to nursery pigs. Methods: In the previous studies, energy, phosphorus, calcium, and amino acid digestibility of infertile eggs, unhatched eggs, culled chicks, and a mixture of 3 hatchery byproduct ingredients was determined in nursery pigs (initial body weight = 9.4 to 14.2 kg). An additivity test was conducted using these determined values. Results: No difference was observed between determined and predicted metabolizable energy values in the mixture (3,998 and 3,990 kcal/kg as-is basis, respectively). Measured standardized total tract digestible phosphorus in the mixture was less than the predicted value (4.5 vs 5.3 g/kg as-is basis, respectively; p<0.05). Measured standardized total tract digestible calcium in the mixture was greater compared with the predicted value (40.0 vs 31.7 g/kg as-is basis, respectively; p<0.05). Measured standardized ileal digestible tryptophan in the mixture was greater than the predicted value (3.7 vs 3.1 g/kg as-is basis, respectively; p<0.05) whereas other amino acid values were additive. Conclusion: Energy and most of amino acid concentrations in hatchery byproducts are additive in the mixture fed to nursery pigs.

Isolation and Characterization of the C-type Lysozyme Gene from the Common Cutworm Spodoptera litura

  • We have isolated and characterized a new insect chicken type (c-type) lysozyme gene from the common cutworm, Spodoptera litura. The full-length cDNA of Spodoptera lysozyme is cloned by rapid amplification of cDNA ends PCR (RACE-PCR). The isolated cDNA consists of 1039 bp including the coding region for a 142-amino acid residue polypeptide, which included a signal peptide of 21-amino acid residue and a mature protein of 121-amino acid residue. The predicted molecular weight of mature lysozyme and its theoretical isoelectric point from amino acid composition is 13964.8 Da and 9.05, respectively. The deduced amino acid sequence of Spodoptera lysozyme gene shows the highest similarity (96.7%) to Spodoptera exigua lysozyme among other lepidopteran species. Amino acid sequence comparison with other the c-type lysozymes, Spodoptera lysozyme has the completely conserved $Glu^{32}$ and $Asp^{50}$ of the active site and eight Cys residues are completely conserved in the same position as that of other lepidopteran lysozymes.

Optimizing hormonal and amino acid combinations for enhanced cell proliferation and cell cycle progression in bovine mammary epithelial cells

  • Objective: The number of bovine mammary epithelial cells (BMECs) is closely associated with the quantity of milk production in dairy cows; however, the optimal levels and the combined effects of hormones and essential amino acids (EAAs) on cell proliferation are not completely understood. Thus, the purpose of this study was to determine the optimal combination of individual hormones and EAAs for cell proliferation and related signaling pathways in BMECs. Methods: Immortalized BMECs (MAC-T) were treated with six hormones (insulin, cortisol, progesterone, estrone, 17β-estradiol, and epidermal growth factor) and ten EAAs (arginine, histidine, leucine, isoleucine, threonine, tryptophan, lysine, methionine, phenylalanine, and valine) for 24 h. Results: Cells were cultured in a medium containing 10% fetal bovine serum (FBS) as FBS supplemented at a concentration of 10% to 50% showed a comparable increase in cell proliferation rate. The optimized combination of four hormones (insulin, cortisol, progesterone, and 17β-estradiol) and 20% of a mixture of ten EAAs led to the highest cell proliferation rate, which led to a significant increase in cell cycle progression at the S and G2/M phases, in the protein levels of proliferating cell nuclear antigen and cyclin B1, cell nucleus staining, and in cell numbers. Conclusion: The optimal combination of hormones and EAAs increased BMEC proliferation by enhancing cell cycle progression in the S and G/2M phases. Our findings indicate that optimizing hormone and amino acid levels has the potential to enhance milk production, both in cell culture settings by promoting increased cell numbers, and in dairy cows by regulating feed intake.

Energy and Standardized Ileal Amino Acid Digestibilities of Chinese Distillers Dried Grains, Produced from Different Regions and Grains Fed to Growing Pigs

  • Two experiments were conducted to determine the digestibility of crude protein (CP), amino acids and energy in three Chinese corn distillers dried grains with solubles (DDGS), one rice DDGS, one American corn DDGS and one American high protein distillers dried grains (HP-DDG). In Exp. 1, the apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of CP and amino acids in the six samples were determined using cannulated barrows (initial BW: $43.3{\pm}1.7$ kg). In Exp. 2, the digestible energy (DE) and metabolizable energy (ME) content of these six samples were determined using crossbred barrows (initial BW: $46.0{\pm}2.5$ kg). The results of the two experiments indicated that Chinese corn DDGS is generally similar to American DDGS in chemical composition, digestibility of amino acids, DE and ME. However, Chinese DDGS had a lower Lys concentration (0.50% vs. 0.74%) and SID Lys (52.3% vs. 57.0%, p<0.01). The DE and ME values in Chinese corn DDGS were 3,427 and 3,306 kcal/kg, respectively. Rice DDGS had a similar DE and ME (3,363 and 3,228 kcal/kg) but higher Lys concentration (0.64% vs. 0.50%) to corn DDGS, while the SID of Lys was quite low (61.8%, p<0.01). HP-DDG had high value of SID of Lys, DE and ME (79.8%, 3,899 and 3,746 kcal/kg). In conclusion, except for a lower Lys concentrations and availability, the chemical composition, digestibility of amino acids, DE and ME values in Chinese corn DDGS are similar to American corn DDGS. Additionally, the rice DDGS had lower Lys content and digestible Lys values than that in corn DDGS. Thirdly, HP-DDG has higher levels of digestible amino acids and energy than DDGS.

Ileal Amino Acid Digestibility of Broken Rice Fed to Postweaned Piglets with or without Multicarbohydrase and Phytase Supplementation

  • Most of amino acid (AA) digestibility values for feed ingredients are obtained using pigs cannulated in the distal ileum. The ileal-cannulated pig model uses pigs older than six weeks due to difficulties related to implanting the T-cannula in distal ileum of younger pigs and complications during the post-surgical recovery. However, to properly formulate the diet of weaned pigs, the nutritive value of feed ingredients should be determined with younger pigs. Thus, 25 weaned pigs were used to determine the apparent total tract digestibility (ATTD) of nutrients, energy, and apparent ileal digestibility (AID) and standardized ileal digestibility (SID) ileal AA digestibility of broken rice (BR), with or without multicarbohydrase (MC) and phytase (Phy) supplementation. Piglets were weaned at 23 d of age and individually housed in digestibility cages until 45 d of age. The trial consisted of 7 d of adaptation to the experimental diets and 3 d of excreta (feces and urine) collection. Ileal digesta was collected at slaughter (about 6 weeks of age). A completely randomized experimental design was used to determine the effects of MC and Phy. Reference diets (RD, 5% casein) was replaced by 30% of BR with or without MC, Phy, or MC+Phy. The RD was used to quantify endogenous AA losses. BR with Phy supplied had increased the ATTD of dry matter (p<0.05) and SID of histidine (p = 0.05), arginine, leucine, lysine, valine, alanine, and proline (p<0.05). BR with MC had been increased digestible energy and protein and SID for histidine (p<0.05). There was no interaction between Phy and MC on the BR nutrient digestibilities. Standardized amino acid digestibilities of BR, without enzymes, were lower than those values reported in the literature. The MC and Phy improved the digestibility of some nutrients and energy of BR in post-weaned piglet diets.

버크셔의 성별이 돈육의 관능 평가, 지방산 및 아미노산 조성에 미치는 영향 (Effects of Gender on Sensory Scores, Fatty Acid and Amino Acid of Berkshire Pork)

  • 본 연구는 버크셔종의 성별에 따른 육질 특성을 파악하고자 실시되었으며, 60 kg의 Berkshire 40두를 2 돈방에 암퇘지와 거세돼지 각각 20두씩 배치하여 출하 시(약 110 kg)까지 비육한 후 도축하여 실험에 공시하였다. 신선육 관능검사에서 암퇘지보다 거세 돼지가 육색, 마블링 점수가 높아 전체적인 기호도가 높게 나타났으며, 가열육 관능검사에서도 암퇘지보다 거세 돼지가 육색, 향 및 기호성에서 높은 점수를 나타내었다(p<0.05). 지방산 조성 중 linoleic acid와 arachidonic acid는 암퇘지보다 거세 돼지가 낮아(p<0.05) 필수 지방산과 다가 불포화 지방산도 낮은 경향을 보였으나 전체적으로 성별에 따른 지방산 조성에는 유의적인 차이를 보이지 않았다.(p>0.05). 아미노산 조성은 암퇘지보다 거세 돼지가 감미계인 alanine, 방향족인 tyrosine, 필수 아미노산인 arginine은 높게 나타났고, proline과 감미계이자 필수 아미노산인 threonine, 필수아미노산인 histidine은 낮게 나타나(p<0.05) 전체적으로 암퇘지보다 거세 돼지의 풍미가 낮은 경향이었으나 glutamic acid, 감미계, 황함유, 방향족 및 필수 아미노산 각각의 총 함량은 성 간에 유의적인 차이를 보이지 않았다(p>0.05).

사육일령이 육계의 가슴 및 다리살의 아미노산·지방산 변화에 미치는 영향 (Effect of Raising Periods on Amino Acids and Fatty Acids Properties of Chicken Meat)

  • 본 연구는 1일령 초생추(Ross)를 부화장에서 암 수 분리한 후 구입하여, 수병아리만 평당 66수를 수용해 왕겨 깔짚을 사용한 평사에서 사육을 시작하여 사육일령이 30, 36, 42일령 일 때 임의로 선발하여 도계한 다음 닭고기의 가슴 및 다리육의 아미노산, 유리아미노산, 지방산 변화를 구명하기 위하여 수행하였다. 육계의 사육일령별 닭고기의 육질변화에서 아미노산 중 glutamic acid의 변화는 가슴살에서 사육일령이 경과할수록 감소하는 경향을 나타내었고, 다리육에서도 가슴육과 유사한 경향을 나타내었다. 총 유리아미노산 변화에서는 가슴살에서 246.3~370.6 mg%를 나타내었고, 다리육은 321.9~470.16 mg%를 나타내어 가슴살보다 다리육에서 전반적으로 더 높은 값을 나타내었다. 유리아미노산에서 glutamic acid도 다른 유리아미노산과 비슷하게 사육 기간이 경과할수록 감소하는 경향을 나타내었다. 지방산 변화는 가슴살에서 필수지방산 중 가장 많은 부분을 차지하는 linoleic acid 함량은 사육일령이 경과할수록 증가하는 경향을 나타내었으며, 뇌세포의 주성분인 DHA의 성분도 가슴육의 경우 30일령보다 42일령에서 1.9배 정도 증가하였다. 이러한 경향은 다리육에서도 유사하게 나타내었다. 가슴육 및 다리육의 총 불포화지방산 비율도 육계의 사육일령이 경과할수록 증가하는 경향을 나타내었다.

다양한 유기산제들의 급여가 이유자돈의 성장과 영양소 소화율에 미치는 효과 (Comparison of the Efficacy of Different Organic Acids on Growth Performance and Nutrient Digestibility in Weaned Pigs)

  • 본 연구는 다양한 유기산제들의 급여가 이유자돈의 성장과 영양소 소화율에 미치는 영향을 비교하기 위하여 2번의 사양실험을 수행하였다. 실험 1과 실험 2에서는 각각 이유자돈 180두 (4처리, 3반복, 15두/반복)를 공시하였다. 실험 1은 기초사료에 formic acid, ammonium-formate, lactic acid 그리고 acid mixture를 각각 0.5%씩 첨가하여 총 5주간 사양실험을 실시하였으며, 실험 2는 formic acid, ammonium-formate, lactic acid 그리고 acid mixture를 각각 0.3%씩 첨가하여 총 6주간 사양실험을 실시하였다. Acid mixture는 formic acid와 lactic acid가 50:50의 비율로 혼합된 제제를 사용하였다. 외관상 회장아미노산 소화율을 측정하기 위하여 각 처리별 3두, 총 12두를 공시하여 T-cannula를 회장 말단부에 부착시킨 후 적응기간을 거쳐 회장내용물을 채취하였다. 사양성적은 유기산 종류에 따른 개선 효과는 없는 것으로 나타났으나 (실험 1), acid mixture 급여구에서 증체량이 lactic acid 급여구보다 높은 것으로 나타났다 (실험 2). 외관상 회장 아미노산 소화율은 lactic acid 급여구보다 acid mixture 급여구에서 개선되는 것으로 나타났다 (실험 1과 2). 실험 1과 2의 결과를 종합하였을 때 acid mixture (formic acid + lactic acid) 급여는 이유자돈의 사양성적과 외관상 회장 아미노산 소화율을 개선시키는 것으로 판단된다.
