Allozyme Variations in Local Populations of Parafossrulus manchouricus (Gastropoda: Prosobranchia) in Korea (한국산 왜우렁이 집단의 지역적 Allozyme 변이에 관한 연구(I))

  • The most medically important snail species of Korea is Parafossarulus manchouricus, a member of the freshwater prosobranch family Bithyniidae. The human parasite that this snail transmits is Clonorchis sinensis, the "Chinese live fluke". On the other hand, this snail has physiological characteristics that reduce the turbidity of freshwater by its filiter feeding activity. However, a few basic studies have been carried out so far. The present studies were attempted to know 1) the possibility of culturing the snails and 2) allozyme variations among 5 local populations of this bithyniid snails. As the results of the studies, P. manchouricus was able to be cultivated in the lavoratory and showed considerable allozyme variations especially in the Chongpyung and Paldaing populations out of those collected from 5 localities in Korea. It is quite suggestive that the endemicity of clonorchiasis might be related to the allozyme variability.

Detection of Sugar Process Contamination Using Dextran Binding hages Produced by Batch Fermentation of Escherichia coli (대장균의 회분식 발효에 의해 생산된 덱스트란 결합 파아지를 활용한 설탕 제조공정 오염 검출)

  • Sequential passes through $Sephadex^{TM}$ columns were used to select phages that displays ligands for dextran ($\alpha$-1,6 linked linear chains) from a phage antibody library. Those phages that bound to the $Sephadex^{TM}$ in each iteration were replicated in E. coli. A phage preparation isolated on the third round selection produced 5.4 nephelos turbidity units (NTU) in a dextran specific immunonephelometric assay, a 2.2 fold higher value than the phage preparation from the first round selection. This phage gave $72\;{\pm}\;10$ normalized intensity (N.I.) in a dip-stick assay against high molecular size dextran (T2000, $2\;{\times}\;10^6) and significantly lower color ($30\;{\pm}\;6$ N.I.) against low molecular size dextran (T10, $10^4$). The presence of an Fab insert in each of these phages was confirmed using a $\beta-galactosidase linked assay and polymerase chain reaction.

Geochemical Equilibria and Kinetics of the Formation of Brown-Colored Suspended/Precipitated Matter in Groundwater: Suggestion to Proper Pumping and Turbidity Treatment Methods (지하수내 갈색 부유/침전 물질의 생성 반응에 관한 평형 및 반응속도론적 연구: 적정 양수 기법 및 탁도 제거 방안에 대한 제안)

  • The formation of brown-colored precipitates is one of the serious problems frequently encountered in the development and supply of groundwater in Korea, because by it the water exceeds the drinking water standard in terms of color. taste. turbidity and dissolved iron concentration and of often results in scaling problem within the water supplying system. In groundwaters from the Pajoo area, brown precipitates are typically formed in a few hours after pumping-out. In this paper we examine the process of the brown precipitates' formation using the equilibrium thermodynamic and kinetic approaches, in order to understand the origin and geochemical pathway of the generation of turbidity in groundwater. The results of this study are used to suggest not only the proper pumping technique to minimize the formation of precipitates but also the optimal design of water treatment methods to improve the water quality. The bed-rock groundwater in the Pajoo area belongs to the Ca-$HCO_3$type that was evolved through water/rock (gneiss) interaction. Based on SEM-EDS and XRD analyses, the precipitates are identified as an amorphous, Fe-bearing oxides or hydroxides. By the use of multi-step filtration with pore sizes of 6, 4, 1, 0.45 and 0.2 $\mu\textrm{m}$, the precipitates mostly fall in the colloidal size (1 to 0.45 $\mu\textrm{m}$) but are concentrated (about 81%) in the range of 1 to 6 $\mu\textrm{m}$in teams of mass (weight) distribution. Large amounts of dissolved iron were possibly originated from dissolution of clinochlore in cataclasite which contains high amounts of Fe (up to 3 wt.%). The calculation of saturation index (using a computer code PHREEQC), as well as the examination of pH-Eh stability relations, also indicate that the final precipitates are Fe-oxy-hydroxide that is formed by the change of water chemistry (mainly, oxidation) due to the exposure to oxygen during the pumping-out of Fe(II)-bearing, reduced groundwater. After pumping-out, the groundwater shows the progressive decreases of pH, DO and alkalinity with elapsed time. However, turbidity increases and then decreases with time. The decrease of dissolved Fe concentration as a function of elapsed time after pumping-out is expressed as a regression equation Fe(II)=10.l exp(-0.0009t). The oxidation reaction due to the influx of free oxygen during the pumping and storage of groundwater results in the formation of brown precipitates, which is dependent on time, $Po_2$and pH. In order to obtain drinkable water quality, therefore, the precipitates should be removed by filtering after the stepwise storage and aeration in tanks with sufficient volume for sufficient time. Particle size distribution data also suggest that step-wise filtration would be cost-effective. To minimize the scaling within wells, the continued (if possible) pumping within the optimum pumping rate is recommended because this technique will be most effective for minimizing the mixing between deep Fe(II)-rich water and shallow $O_2$-rich water. The simultaneous pumping of shallow $O_2$-rich water in different wells is also recommended.

Detecting red tides in turbid waters

  • As an example of many possible applications of OSMI data, we present a method to detect red tides. In Korean waters, red tides usually occur in the South Sea where the turbidity is usually high due to strong tidal mixing in the shallow sea. The conventional case 1 chlorophyll algorithm cannot be applied since it cannot distinguish chlorophyll from SS (suspended sediments). In October 1998, a red tide outbreak occurred off the coast of Kunsan. We analyzed the SeaWiFS data of the outbreak. The standard SeaWiFS chlorophyll algorithm OC2 was poor in identifying the red tides. However, comparison of spectra of normalized water-leaving radiance indicates that red tide pixels can be distinguished from sediment-laden pixels. Channel 443 and 555 were effective in showing the spectral characteristics. We suggest K490 algorithm as an example in summarizing the information of the spectra and thereby in distinguishing the red tide pixels. Further development is desirable.

  • 본 논문에서는 대형 탱커선의 의무 장비인 유배출 감시 장비 (Oil Discharge Monitoring Equipment)를 위한 유분 농도계의 설계에 대하여 연구하였다. ODME는 국제해사기구 (IMO)에서 일정 ton 이상의 대형 선박에 의무적으로 설치하도록 규정하고 있으며 이는 날로 심각해지는 해양 오염을 방지하는데 목적이 있다. ODME의 구성 요소는 중앙 제어 장치, 유분 농도계, 유량계, 선속계, Sampling System, 배출 제어 장치로 이루어져 있으며 이중 가장 중요한 것은 유분 농도계이다. 이는 선박에서 배출되는 물에 포함된 유 성분을 ppm 단위로 계측하는 것이다. 이를 위한 탁도 측정 방법에는 여러 가지가 있지만, 본 연구에서는 선박에서 사용이 용이한 광산란 방식과 방폭성을 가지는 광섬유를 이용하여 탁도를 측정하는 유분 농도계를 설계하였으며 아울러 이에 대한 탁도 계산 알고리즘에 대하여 고찰하였다. 측정의 정밀도는 0-999ppm 범위에서 $\pm$5%의 오차 이내로 측정이 가능함을 보였다. 이는 IMO의 규격 ($\pm$10%의 오차)에 만족하는 수치이다.

Membrane Filtration Technology for Drinking Water Treatment & Night Soil Treatment

  • 1. The flux for hydrophilic CA membrane is higher than that for hydrophobic PES membrane at any operating conditions. The difference in bpth fluxes becomes greater as the water recovery is lower. 2. Backwash pressure should be more than twice as high as filtration pressure in order to maintain the higher flux. Backwash frequency is independent of the flux when the UF is operated under the same water recovery. 3. The relatively lower crossflow velocity of around 0.1 m/s would be appropriate because of the lower energy consumption per treated water. 4. The membrane fouling occurring at high turbidity and high concentration of organic compounds in raw water can reduce the flux and increase the removal of the organic compounds. 5. It is confirmed by the pilot plant testing that the UF by using the CA membrane module was well applicable to the drinking water treatment.

Comparative study of thermal gelation properties and molecular forces of actomyosin extracted from normal and pale, soft and exudative-like chicken breast meat

  • Objective: The objectives of this study were to investigate the thermal gelation properties and molecular forces of actomyosin extracted from two classes of chicken breast meat qualities (normal and pale, soft and exudative [PSE]-like) during heating process to further improve the understanding of the variations of functional properties between normal and PSE-like chicken breast meat. Methods: Actomyosin was extracted from normal and PSE-like chicken breast meat and the gel strength, water-holding capacity (WHC), protein loss, particle size and distribution, dynamic rheology and protein thermal stability were determined, then turbidity, active sulfhydryl group contents, hydrophobicity and molecular forces during thermal-induced gelling formation were comparatively studied. Results: Sodium dodecyl sulphate-polyacrylamide gel electrophoresis showed that protein profiles of actomyosin extracted from normal and PSE-like meat were not significantly different (p>0.05). Compared with normal actomyosin, PSE-like actomyosin had lower gel strength, WHC, particle size, less protein content involved in thermal gelation forming (p<0.05), and reduced onset temperature ($T_o$), thermal transition temperature ($T_d$), storage modulus (G') and loss modulus (G"). The turbidity, reactive sulfhydryl group of PSE-like actomyosin were higher when heated from $40^{\circ}C$ to $60^{\circ}C$. Further heating to $80^{\circ}C$ had lower transition from reactive sulfhydryl group into a disulfide bond and surface hydrophobicity. Molecular forces showed that hydrophobic interaction was the main force for heat-induced gel formation while both ionic and hydrogen bonds were different significantly between normal and PSE-like actomyosin (p<0.05). Conclusion: These changes in chemical groups and inter-molecular bonds affected protein-protein interaction and protein-water interaction and contributed to the inferior thermal gelation properties of PSE-like meat.

Interpretation of tube agglutination test for bovine brucellosis with turbidimetric readings and international unit

  • The tube agglutination test has been used for bovine brucellosis diagnosis in Korea since middle 1950s. The reported high specificity was its value in eradication program. However, the reading of reaction mostly depends on personal experience, thus here we report a way to improve accuracy and uniformity of reading. The tube agglutination was conducted according to the protocol provided by Korean Ministry of Agriculture and Forestry. The intensity of reaction was measured by spectrophotometer. The relationship between turbidity and percentage clearing was generally in direct proportion and linear. The correspondent percent transmittance at 75, 50, and 25% clearing were 91, 82, and 73, respectively. Then, the degree of percentage of clearing at given international unit was measured. With about 1.5 unit of serum, the maximum percentage clearing was observed. The international unit showing 25, 50, and 75 percentage clearing were 0.61, 0.83 and 1.35, respectively. Based on the information obtained using international standard serum, the calculation of international unit of test serum was available. According to the protocol for bovine brucellosis diagnosis which provided by Korean Ministry of Agriculture and Forestry, the available range of detectable international unit was between 15 and 538. And the corresponding international units for suspicious case ranged between 42 and 127. Of the 35 sera from B abortus infected cattle, about half of them had more than 538 international units. Collectively, the reading of turbidity using spectrophotometer and application to international unit improved accuracy and uniformity of reading.

Distribution and Species Prediction of Epilithic Diatom in the Geum River Basin, South Korea (금강권역 주요 하천의 돌 부착돌말류 분포 및 출현예측)

  • In order to understand the relationship between the distribution of epilithic diatoms and the habitual environments, land-use, water qualities, and epilithic diatoms were studied at 141 sampling sites in the midwestern stream of Korean peninsula (Geum river, Mangyeong river, Dongjin river, and Sapgyo river). The total 183 diatom taxa was appeared in the study, while the dominant species were found to be Nitzschia palea (10.9%) and Achnanthes convergens (8.4%). Based on the abundance of epilithic diatoms, a cluster analysis results indicate that the sampling sites divided the sampling sites into 4 groups (G) at the 25% level. In term of geographic and aquatic environments, G-I and -II accounted for the upper and mid streams of the Geum river, and had large forest areas and good in water quality. G-III accounted for farmland and urban, and high concentration nutrient levels (TN and TP) and electric conductivity. G-IV accounted for mostly farmland, and high levels in turbidity, BOD, nutrient and electric conductivity. CCA results showed that the saproxenous taxa Meridion circulare was the indicator species of G-I, which strongly influenced by altitude and forests. In G-II, the indifferent taxa Navicula cryptocephala was influenced by Chl-a, AFDM, and DO. In G-III and -IV, the indifferent taxa Fragilaria elliptica and saprophilous taxa Aulacoseira ambigua were influenced by electric conductivity, turbidity, and nutrient counts. Meanwhile, random forest results showed that the predicting factor of indicator species appearance in G-I, -II, and -III was found to be electric conductivity whereas in G-IV it was found to be turbidity. Collectively, the distribution of diatoms in the midwestern of Korean peninsula was found to depend more on the land-use and its subsequent water qualities than the inherent characteristics of the aquatic environment.
