Effects of LED Light Conditions on Growth and Analysis of Functional Components in Buckwheat Sprout (LED 광 조건에 따른 메밀 새싹의 생육 및 기능성 물질 분석)

  • Buckwheat sprouts are a vegetable; a functional food should provide health benefit and enhance performance as high nutritionally important substances. Buckwheat noodles are the major buckwheat food in Japan, Korea and China. In addition, Buckwheat as preventive medicine has undergone a great advancement in the last decade. Comparison of the functional properties distribution and utilization in tatary buckwheat is required of understanding the metabolites. The study was conducted to identify the sorts of phenolic compounds and metabolites in tatary buckwheat seedling at 4, 7, and 10 days seedling under the different combinations of light-emitting diode (LED) such as blue, red, mix (red, blue, and white), dark, and natural lights in stem and leaves. After breaking the dormancy, buckwheat seeds were grown in culture room under lights for 14 hrs and the dark condition for 10 hrs, at $25^{\circ}C$ for 10 days. Length of buckwheat was gradually increased under all of the conditions. Using HPLC, rutin was highest at 7 days under mix and natural light in stem and leaf, respectively. Quercetin was highest at 4 and 7 days under natural light in both. Chlorogenic acid was highest at 7 days under mix and natural in stem and leaf, respectively. Taken Together, this study indicates that phenolic compounds and metabolites present in those plants could be helpful for the human health and nutritional additive.

Comparative Study on Antioxidant Capacities and Polyphenolic Contents of Commercially Available Cocoa-containing Products (유통되는 코코아함유 가공품의 항산화능과 폴리페놀 함량에 관한 연구)

  • UV-VIS spectrophotometric and high-performance liquid chromatographic (HPLC-DAD) methods were applied in order to identify and quantify the composition of polyphenols in commercial cocoa-containing products. Total polyphenolic contents of cocoa mix, choco-syrup, milk chocolate, and dark chocolate were evaluated using Folin-Ciocalteu's phenol reagent according to a UV-VIS spectrometric method. Antioxidant capacities of cocoa extracts by methanol were evaluated by 2,2'-azino-bis(3-ethyl benzothiazoline-6-sulfonic acid) (ABTS) assay and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. The obtained results show that the polyphenolic contents and antioxidant capacities of cocoa products and chocolates depend on solid cocoa content. Among the tested cocoa products and chocolates, the most abundant phenolic compound was epicatechin.

Evaluation of the Anti-thrombosis Activities of the Aerial Parts of Sageretia thea (상동나무 지상부의 항혈전 활성)

  • Recently, thrombotic diseases have become rapidly more prevalent due to Westernized lifestyles and high-fat diets. In this study, the anti-thrombosis activities of the aerial parts of Sageretia thea were evaluated using ethanol extracts of the leaf (ST-L), branch (ST-B), and fruit (ST-F), and their anti-coagulation, platelet aggregation inhibition, and hemolytic toxicity were assessed. In comparison to the ST-F extract, the ST-B exhibited 6.7 times more polyphenol content, and the ST-L had 2.7 times more total flavonoid content. The ST-L and ST-B extracts showed stronger inhibitions of thrombin, prothrombin, and blood coagulation factors than aspirin, berry extracts, or commercial oriental herbs. Furthermore, ST-L and ST-B showed superior platelet aggregation-inhibitory activities than aspirin. The ST-F extract demonstrated only minor anti-thrombosis effects, and none of the extracts showed hemolysis against red blood cells up to 1 mg/ml. Phenolic acid and flavonoid analysis of the ST-L and ST-B extracts showed abundant rutin, isoquercitrin, and astragalin as the major active compounds. Further research on the anti-thrombotic activity of isoquercitrin, a rare flavonoid from quercetin, is necessary. This is the first report of isoquercitrin in Sageretia thea, and our results suggest that ST-L and ST-B extracts could therefore developed as anti-thrombosis agents.

Preparation of Cheonggukjang added onion (Allium cepa L.) and its antioxidative activity (양파(Allium cepa L.) 첨가 청국장의 제조 및 항산화능 평가)

  • Yellow and black soybean Cheonggukjangs (YBSC) prepared with an addition of onion (Allium cepa L.) in different contents (0, 5, 10, 20, and 30%, w/w) were manufactured, and the sensory evaluation was carried out. The optimum addition ratio of onion was determined to be 20%. Total phenolic and flavonoid contents of YBSC prepared with an addition of onion were higher than those of the YBSC prepared without an onion. In addition, YBSC prepared with an addition of onion showed higher 1,1-diphenyl-2-picrylhydrazyl radical-scavenging activities than those prepared without an onion. Furthermore, rat blood plasma 1 hour after oral administration of YBSC prepared with an onion was more effective in suppressing the accumulation of cholesteryl ester hydroperoxide than those prepared without onion and control. These results indicated that the consumption of the YBSC prepared with onion may contribute to the antioxidant defense in vivo.

Non-thermal treatment of Prunus mume fruit and quality characteristics of the dehydrated product (청매실의 비가열 전처리 및 건조매실의 특성)

  • To maintain the microbiological safety of Prunus mume fruit before it is processed, it was treated with a combination of 0.5% citric acid and 0.1% Tween 20, and stored at $4{\pm}1^{\circ}C$ for seven days. The combined treatment reduced total aerobic bacteria, yeast, and mold populations in the fruit by 2.20 and 1.70 log CFU/g, respectively, compared to those in the control. Organic acid contents and the Hunter $L^*$, $a^*$, and $b^*$ values were not affected by the treatment during the storage. In addition, the dried Prunus mume fruit prepared with 40% red algae extract (RAE) or maltodextrin (MD) treatment and hot-air drying were compared with respect to the fruit's physicochemical properties such as color, total phenolic and flavonoid content, and microstructure. The hot-air dried samples had undesirable color changes and inferior textures. The RAE-treated samples had a higher total phenolic content (225.15 mg gallic acid equivalent (GAE)/100 g) and total flavonoid content (49.25 mg quercetin equivalent (QE)/100 g) than the other treatments. The treatment of Prunus mume fruit with RAE can provide better-dried products than can MD treatment or hot-air drying. These results suggest that the combined treatment with citric acid and Tween 20 can be effective in preserving the microbiological safety of Prunus mume fruit, and its dehydration using RAE is an efficient drying method.

Polyphenol Contents and Antioxidant Activities of Lentil Extracts from Different Cultivars (품종별 렌틸 추출물의 폴리페놀화합물 함량 및 항산화 활성)

  • Lentils (Lens culinaris) have been gaining increasing attention recently as a top five superfood, as they are high in protein and other essential nutrients, including folate, iron, potassium, and various antioxidants. In the present study, phenolic extracts from four different lentil cultivars (green, red, French, and beluga) were evaluated for their total phenolic contents and in vitro antioxidant activities. Total polyphenol and flavonoid contents of four different lentil extracts were 27.30~30.30 mg tannic acid equivalents (TAE)/g and 13.14~16.29 mg quercetin equivalents (QUE)/g, respectively. Beluga and red lentil extracts showed higher polyphenol contents than others (P<0.05), whereas there was no significant difference in flavonoid contents among the four lentil cultivars. $RC_{50}$ values of the lentil extracts for DPPH radical, ABTS radical, and $H_2O_2$ were $57.42{\sim}64.49{\mu}g/mL$, $66.11{\sim}75.69{\mu}g/mL$, and $59.72{\sim}72.86{\mu}g/mL$, respectively. Among the four lentil extracts, beluga lentil extract showed the most potent scavenging effect in all three reactive oxygen species (ROS) scavenging assays, and thus beluga extract was further tested for its inhibitory effect on early peroxidation of linoleic acid. The results showed that beluga lentil extract significantly inhibited linoleic acid peroxidation in a dose-dependent manner (concentration required for 50% reduction=$222.76{\m}g/mL$). In addition, beluga lentil extract showed a significant protective effect against alcohol-induced cytotoxicity in AML-12 cells (normal mouse hepatocyte cell line). Taken together, these results suggest that lentil extracts represent potential sources of natural antioxidants, and further studies will be necessary to determine their protective effects against oxidative stress in vivo.

Anti-skin Aging Potential of Alcoholic Extract of Phragmites communis Rhizome

  • Chronological aging and photoaging affect appearance, causing wrinkles, pigmentation, texture changes, and loss of elasticity in the skin. Phragmites communis is a tall perennial herb used for its high nutritional value and for medicinal purposes, such as relief from fever and vomiting and facilitation of diuresis. In this study, we investigated the effects of ethanol extract of P. communis rhizome (PCE) on skin aging. The total flavonoid and total phenolic content in PCE were 2.92 ± 0.007 ㎍ of quercetin equivalents (QE) and 231.8 ± 0.001 ㎍ of gallic acid equivalents (GAE) per 100 mg of dried extract (n = 3). The half-maximal inhibitory concentration (IC50) values of PCE for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) and hydrogen peroxide scavenging activities were 0.96 and 0.97 mg/mL, respectively. PCE showed inhibitory effects on tyrosinase when L-tyrosine (IC50 = 1.25 mg/mL) and L-3,4-dihydroxyphenylalanine (IC50 = 0.92 mg/mL) were used as substrates. PCE treatment up to 200 ㎍/mL for 24 h did not cause any significant cytotoxicity in B16F10 melanocytes, human dermal fibroblasts (HDFs), and HaCaT keratinocytes. In B16F10 melanocytes, PCE (25 and 50 ㎍ /mL) inhibited melanin production and cellular tyrosinase activity after challenge with α-melanocyte-stimulating hormone (α-MSH; p < 0.05). In HDFs, PCE suppressed the mRNA expression of matrix metalloproteinase-1 (MMP-1) and reduced the activity of elastase (p < 0.05). In addition, ultraviolet B (UVB)-mediated downregulation of hyaluronic acid synthase-2 gene expression in HaCaT keratinocytes was also effectively suppressed by PCE treatment. Overall, our results showed that PCE has potential anti-skin aging activity associated with the suppression of hyperpigmentation, wrinkle formation, and reduction in dryness. PCE is a promising candidate for the development of an anti-skin aging cosmetic ingredient.

Effect of Defective Onion Extract on the Onion Productivity by Organic Farming (양파 파치 추출물이 유기농 양파성에 미치는 영향)

  • This study evaluated the close crop recycling method by using the extract from defective onions. The mixture of the diluted extract (1/500) and nutrients was applied seven times on the onion leaves without spilling it onto the field. The yield and quality of the onions produced by this method (recycle) were compared to those grown organically (control) and conventionally (normal). The yield from the recycling field was increased significantly by 9% compared to that of the control field, while it was decreased by 11% compared to the normal field (p < 0.05). This lower yield was explained by the differences of the mulching vinyl and the fertilizers between the treatments which effect the onion growth during the winter. The content of quercetin in the onions from the recycling filed was increased significantly by 34% and 44% compared to those of the control and normal field, respectively (p < 0.05). It seemed that minerals and biologically active substances in the defective onion extract were effective in increasing the onion growth. In terms of soil microbial biomass, arbuscular mycorrhizal fungi was increased significantly in the recycle field by 40.1% and 30.6% compared to those of the control and normal fields, respectively (p < 0.05). On the other hand, microbial stress (cy19:0/18:1w7c) was decreased in the recycle field by 21.0% and 14.1% compared to those of the control and normal fields, respectively.

Antioxidant Contents and Antioxidant Activities of Hot-Water Extracts of Aronia (Aronia melancocarpa) with Different Drying Methods (건조방법에 따른 아로니아(Aronia melancocarpa) 열수 추출물의 항산화 성분 함량 및 항산화 활성)

  • This study determined the antioxidant levels and activities of hot water aronia extracts by different drying methods such as sun drying, sun drying after steam treatment, freeze-drying, and oven drying. The total polyphenol content, calculated as gallic acid equivalent, was the highest in the freeze-dried sample (910 mg), followed by sun-dried after steam treatment (779 mg), sun-dried (769 mg), and oven-dried (757 mg) samples. Similar patterns were observed for the total flavonoid and anthocyanin contents. Freeze-dried aronia samples contained the highest polyphenol, flavonoid, and anthocyanin contents as compared to the samples dried by other methods. All antioxidant activities were found to increase in a dose-dependent manner. For the hot water-extracted freeze-dried aronia powder (200 mg/mL), the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radicalscavenging activities were 65.5% and 61.7% and the hydroxyl and superoxide anion radical-scavenging activities were 50.5% and 52.1%, respectively. These results suggest that comparatively, freeze-drying is a better method for preserving the bioactive components and the antioxidant activities of aronia.

Determination of Antioxidant Activities and Bioactive Compounds from Rosa rugosa Extract (해당화 추출물의 주요물질 분석에 따른 폴리페놀 함량 및 항산화 활성 탐색)

  • The purpose of this study was to evaluate the antioxidant properties of Rosa rugosa extract and to identify which of its components are responsible for these properties. Reactive oxygen species play an important role in diseases such as cancer, arteriosclerosis, and heart disease as a consequence of increased metabolic rates, gene mutations, and relative hypoxia. Therefore, the antioxidant effect of R. rugosa extract was confirmed by HPLC, HPLC-MS/MS, the total polyphenol content, the total flavonoid content, and the radical scavenging activity. HPLC and HPLC-MS/MS analyses were conducted to identify and quantify the main components of the R. rugosa extract. Gallic acid and epigallocatechin gallate were identified as the main components, with 17.4 and 4.35 mg/g dry matter (DM), respectively. The antioxidant activity of R. rugosa extract was evaluated based on its total polyphenol content, total flavonoid content, and radical scavenging activity, which were 72.3 mg gallic acid equivalent/g DM, 11.2 mg quercetin equivalent/g DM, and 87.9%, respectively. The radical scavenging activities of the main components, gallic acid and epigallocatechin gallate, were 80.5% and 89.7%, respectively. Therefore, R. rugosa has a high polyphenol content and antioxidant activity, and it can be used as a natural antioxidant in food, cosmetics, and pharmaceuticals.
