Liquid Crystal-based Imaging of Biomolecular Interactions at Roller Printed Protein Surfaces

  • In this study, the orientational behavior of thermotropic liquid crystals (LC) supported on a film of protein receptors was examined. Avidin was roller printed and covalently immobilized onto the surface of gold using NHS/EDC chemistry. The orientation of nematic 4-cyano-4'-pentylbiphenyl (5CB) was found to be parallel to the plane of the printed avidin surface before incubation with a solution of biotin. However, protein-receptor complexation induced a random orientation of 5CB, where protein-receptor complexes disturbed the nanoscale topography of the printed protein surface. Atomic force microscopy and ellipsometry was used to confirm printing and the specific interaction of proteins. These results demonstrate that the combination of LC and roller printing can be used to detect specific interactions between biomolecules by manipulating the orientational behavior of LC to the printed protein surfaces.

Cultural Characteristics for the Enhanced Mycelial Growth of Ramaria botrytis

  • The culture conditions for the enhanced mycelial growth of Ramaria botrytis was investigated. The optimal temperature and pH for the mycelial growth were $24^{\circ}C$ and 5.0, respectively. It was shown that starch was best of several carbon sources in Czapek-Dox medium as a minimal medium for the enhanced mycelial growth. Organic nitrogen sources were better than inorganic ones for mycelial growth. The appropriate vitamin and mineral salt were biotin and FeCl3, respectively. When this strain was cultured with $FeCl_3$ for 30 days, 19.23 g/l of dry mycelium of R. botrytis was obtained.

  • A case of melanotic neuroectodermal tumor of infancy(MNTI) in 5 month old girl is presented with review of the literature. The review of literature indicated that there have been 189 reported cases of this lesion and there were 9 cases of malignant tumor for a rate of 4.8%. This rate of malignancy is very high in view of the fact that the tumor has been described as benign. And immunohistochemical study using avidin-biotin conjugate method demonstrated that this tumor had been originated from neural crest.

  • LSPR(Localized Surface Plasmon Resonance) sensor measures the refractive index change on the sensor surface. The detection of biological reaction with the unknown refractive index needs to be converted into the signal sensitivity for the refractive index change for comparison with other measurements. To find the signal sensitivity, the three steps of signal processing are proposed, which are signal modeling, signal calibration and signal normalization of LSPR sensor. The detected signal of biotin-streptavidin interaction has been converted into unit of [RU](Resonance Unit) using the proposed method. The converted signal directly can be compared with the other sensors including commercialized one.

  • Mitochondrial DNA (mtDNA) mutation was investigated in a hepatoma patient using a polymerase chain reaction (PCR) and an in situ hybridization technique. Biotin-labeled probes for the subunit m of cytochrome c oxidase revealed differences in the in situ hybridization. A PCR assay using biopsied and microdissected tissues showed that common deletion (4,977 bp) was more pronounced in the cancer region than in the normal parts of the same patient. These results suggest that mtDNA deletion might be associated with tumorigenesis in hepatoma.

  • The design of DNA nanostructures is of fundamental importance, the intrinsic value of DNA as a building-block material lies in its ability to organize other bio-molecules with nanometer-scale spacing. Here, we report the fabrication of DNA scaffolds with nano-pores (<10 nm size) that formed easily without the use of additives (i.e., avidin, biotin, polyamine, or inorganic materials) into large-scale structures by assembling DNA molecules at near room temperature ($30^{\circ}C$) and low pH (~5.5). Protein immobilization results also confirmed that a fibronectin (FN) proteins/large scale DNA scaffolds/aminopropylytriethoxysilane (APS)/SiO2/Si substrate with high sensitivity formed in a well-defined manner. The DNA scaffolds can be applied for use with DNA-based biochips, biophysics, and cell biology.

  • For the purpose of developing more effective chelating agents, we have synthesized a diethylene triamine pentaacetic acid(DTPA) analogue by using an amino acid. S-(N-Boc-aminophenyl)-Cys(t-Bu4-DTPA) methylester was prepared in 6 steps with total yield of 47.9%. For the sake of introducing a biomolecule to the DTPA derivative, a selective hydrolysis was performed with 3 M HCl/Ethylacetate = 1 : 3 ($25{^{\circ}C}$, 30 min, vigorous stirring). $^{166}Ho$-Cys-DTPA and $^{166}Ho$-Biotin-Cys-DTPA were prepared by mixing $^{166}Ho$ with DTPA derivatives at room temp in a HCl solution (pH = 5) and the radiochemical stabilities (> 99%) were maintained for over 6 hrs in vitro.

Biosensor Based on Distributed Bragg Reflector Photonic Crystals for the Detection of Protein A

  • The functionalized photonic crystals of porous silicon biosensor was prepared for the application as a label-free biosensor based on distributed Bragg reflector interferometer. Prepared distributed Bragg reflector of porous silicon biosensor displayed sharp reflection in the optical reflective spectra. The mean of construction of molecular architectures on distributed Bragg reflector of porous silicon surfaces was investigated for the step-by-step binding interaction with amines, biotin, avidin, and biotinylated protein A. The subsequent introduction of avidin, and biotinylated protein A resulted in the reflectivity shifted to longer wavelengths, indicative of a change in refractive indices induced by binding of biomolecules.

  • Many photosynthetic bacteria capable of hydrogen production were isolated from samples of mud flats of paddy field collected in Kim Hae and Dae Jeo. A strain 230 was selected for the high capability of hydrogen evolution. As the results of examination in physiological, morghological and cultural characteristics, the strain 230 was identified as Rhodopseudomonas sphaeroides.

  • It is desirable to improve the tumor targeting and blood clearance pharmacokinetics of radiolabeled monoclonal antibodies. To achieve this goal, several avidin-biotin (Bt) binding systems have been developed to decouple large molecular weight antibodies from small radiolabels, thereby achieving high tumor-to-background radioactivity ratios. We inserted a readily catabolizable linker, triglycine (TG), between 3-[/sup125/I]iodobenzoate and dendrimer (G3). We also neutralized the positive charges of G3 by acylation with tetrafluorophenyl glycolate, thereby blocking proximal tubular reabsorption of G3 mediated by charge attraction. (omitted)
