Variation of Univariate Flow Karyotypes and Chromosomal DNA Contents in Maize (Zea mays L.)

  • Analyses of now karyotypes using different maize (Zea mays L.) inbred lines have been performed. The accumulation and isolation of high quality and quantity metaphase chromosomes from root tips can be achieved from many kinds of maize lines. The chromosome suspensions were prepared by a simple slicing method from synchronized maize root tips and analyzed with a now cytometry. The variations of experimental now karyotypes were detected among inbred lines in terms of the positions and/or the numbers of chromosome peaks. The 2C DNA amount among 8 inbred lines ranged from 5.09 to 5.52 pg. The variability of DNA content in maize chromosome 1 was 9.1 % ranging from 0.685 to 0.747 pg. The selection of appropriate maize lines is critical for sorting specific single chromosome types. At least five different chromosome types can be discriminated and sorted from five maize lines.

Inhibitory Effect of Mannose on Auxin-Induced Ethylene Production in Corn (Zea mays L.) Coleoptiles (옥수수 자엽초에서 오옥신 유발 에틸렌 생성에 대한 Mannose의 억제작용)

  • Effect of mannose on auxin-induced ethylene production in corn (Zea mays L.) coleoptiles was studied. Auxin induced ethylene production decreased in proportion to mannose concentrations. The inhibitory effect of mannose appeared after 2 h of incubation. Ethylene production was significantly depressed by mannose at high concentration (10-5M-10-4M) of indole acetic acid (IAA), but not at low concentrations (10-8M-10-6M). The inhibition of auxin-induced ethylene production by mannose was specific, since other sugars such as galactose, glucose, sucrose and mannitol did not have an inhibitory effect. In an effort to elucidate mechanisms of mannose the effect on the auxin induced ethylene production, effect of the sugar on ACC synthase activity and ACC induced ethylene production was studied. Mannose failed to inhibit ACC mediated ethylene production, but decreased both the ACC content and ACC synthase activity in the tissue. These results suggest that the inhibitory effect of mannose on auxin induced ethylene production results from suppression of auxin induction of ACC synthase.

New Host Plants of Charops striatus (Uchida) (Hymenoptera: Ichneumonidae: Campopleginae) from South Korea

  • The genus Charops is one of the small genera of the subfamily Campopleginae with 30 species reporded worldwide. All of the known host insects of Charops striatus belong to the lepidopteran family Zygaenidae, perceived as harmful. C. striatus, one of the endoparasitoids from Elcysma westwoodi, is the most damaging lepidopteran pest of Prunus yedoensis. In South Korea, host plants of C. striatus were reported by Choi & Lee in 2008. In this paper, two new host plants, Crepidiastrum denticulatum and Zea mays, of Charops striatus are reporded for the first time from South Korea. Digital images and redescription of this species are provided.
