Potential antifilarial activity of the fruit, leaf and stem extract of Melia azedarach Linn. on cattle filarial parasite Setaria cervi in vitro

  • The effect of aqueous and alcoholic extracts of the fruit, leaf and stem of Melia azedarach Linn. (Meliaceae) on the spontaneous movements of both the whole worm and the nerve-muscle (n.m.) preparation of Setaria (S.) cervi and on the survival of microfilariae in vitro was studied. Alcoholic extracts of fruit, leaf and stem caused inhibition of the spontaneous movements of the whole worm and the n.m. preparation of S. cervi, while only aqueous extract of fruit caused inhibition of the spontaneous movements of the whole worm and the n.m. preparation of S. cervi. The initial stimulatory effect was not observed by the aqueous and alcoholic extracts of fruit on n.m. preparation. The concentrations required to inhibit the movements of the whole worm and n.m. preparation for alcoholic extracts of fruit, leaf and stem were 250, $40\;{\mu}g/ml$; 280, $40\;{\mu}g/ml$ and 270, $25\;{\mu}g/ml$ respectively, whereas an aqueous extract of fruit caused inhibition of whole worm and n.m. preparation at $200\;{\mu}g/ml$ and $40\;{\mu}g/ml$ respectively. Alcoholic extracts of the fruit, leaf and stem and aqueous extract of the fruit of M. azedarach caused concentration related inhibition on the survival of microfilariae (m.f.) of S. cervi. The $LC_{50}$ and $LC_{90}$ as observed after 6 h were found to be 5, 15, 10, 20 ng/ml and 10, 25, 20 and 35 ng/ml, respectively. This work was conducted in view of the exploration of potential antifilarial herbal drug.

  • 본 연구는 음주가정폭력행위자에 개입하는 중독관리통합지원센터 실무자의 실천경험이 어떠했는지를 총체적으로 이해하기 위해 15명의 실무자를 대상으로 포커스집단인터뷰, 개별인터뷰를수행한 녹취자료와 수퍼비전 및 자문, 워크샵 등을 통해 수집한 제반자료에 대하여 질적사례연구 방법을 활용하여 분석하였다. 연구결과, 음주가정폭력행위자에 개입하는 실무자의 실천경험 영역에서는 상위범주로서 '음주가정폭력행위자에 대한 버거운 대응', '선 실천을 통한 역량강화', '난제해결을 위해 계속되는 고민들', '음주가폭 개입으로 인한 원하지 않은 변화'로 나타났다. 음주가정폭력 실천현장의 현실과 대안 영역에서는 '현장의 필요와 현실간의 괴리', '역량강화를 위한 구체적 필요'의 상위범주로 구분되었다. 이를 근거로 음주가정폭력행위자 개입에 필요한 효과적인 전략으로서, 중독관리통합지원센터라는 현장의 특수성을 반영한 실천적, 정책적, 이론적 함의를 제시하였다.

Assessment of the Human Risk by an Intake of Ethyl Carbamate Present in Major Korean Fermented Foods

  • Levels of ethyl carbamate, a potential carcinogen produced naturally during fermentation, in major Korean fermented foods and alcoholic beverages were determined by GC/MS/SIM, and their average daily intake and excess cancer risk in Korean people were estimated. In GC/MS/SIM analysis n.d.-4.26, 1.40-58.90, n.d.-3.76, n.d.-1.87, and 0.40-10.07 $\mu$g/kg of ethyl carbamate were detected in kimchi, soy sauces, fermented pastes, fermented dairy products, and alcoholic beverages, respectively. The average daily intake of ethyl carbamate and excess cancer risk through major Korean fermented foods and alcoholic beverage consumption were 6.0 ng/kg bw/day and $3.0\times10^{-7}$, respectively for the average Korean person aged 3-64 years, and were mainly contributed by Chinese cabbage kimchi, soy sauces, and Soju.

  • Objectives: The purpose of this study is to report the clinical effectiveness of Korean medicine, especially Palmijihwang-hwan and Oryung-san, on an alcoholic liver disease patient with fatigue and dizziness. Methods: A patient was treated with Korean medicine, including herbal decoction, acupuncture, moxibustion, and cupping. Then, the patient was asked about his condition every day and underwent liver function tests three times. Results: After 13 days of treatment, fatigue decreased by 80%, and dizziness disappeared on the second day of hospitalization. It also recovered from indigestion and abdominal distension. Conclusions: According to this study, Korean medicine, including Palmijihwang-hwan and Oryung-san, is an effective treatment for symptoms associated with alcoholic liver disease.

  • 저자들은 뇌하수체저하증을 동반한 조기에 발병된 소아 비만 환자에서 비알코올성 지방간질환의 진행으로 인한 간경화를 경험하였기에 이를 보고하는 바이며, 향후 뇌하수체저하증이 동반된 비만 환자에서 비알코올성 지방간질환의 발생 및 진행에 대한 주의 깊은 관찰이 필요할 것으로 사료된다.

급성적인 알콜성 지방간 생성에서 Cytochrome P-450 2E1의 역할에 관한 연구 (Lack of Evidence for Involvement of Cytochrome P-450 2E1 in Acutely Induced Alcoholic Fatty Liver)

  • The role of cytochrome P-450 2E1 (P450 2E1) in the early phase of alcoholic fatty liver was examined. Female rats were pretreated with either allyl sulfide (200 mg/kg, po), disulfiram (500 mg/kg, po), YH 439 (250 mg/kg, po) or pyrazine (200 mg/kg/day$\times$2 days, ip). Marked changes in carbon tetrachloride-induced hepatotoxicity and caboxyhemoglobin (COHb) elevation due to dichloromethane administration were observed in rats treated with one of the P450 2E1 modulators. A single dose of ethanol (6 g/kg, po) increased the hepatic triglyceride contents approximately 2 fold, which was inhibited completely by YH 439 pretreatment. However, the other P450 2E1 modulators failed to alter the ethanol-induced hepatic triglyceride accumulation. In vitro hepatic microsomal enzyme activity was determined in 4 week old premature and 12 week old adult rats. Aminopyrine-N demethylation was not different, but p-nitrophenol hydroxylation and p-nitroanisole O-demethylation were significantly higher in premature rats. However, no difference in the triglyceride accumulation induced by an intraperitoneal dose of ethanol (3 g/kg) was noted between premature and adult rats. The results suggest that the P450 2E1 activity dose not play an important role in the induction of acute alcoholic fatty liver.

알코올성 간질환에 대한 한의학 연구 경향 분석 : 국내논문을 중심으로 (A Review of Domestic Research on Traditional Korean Medicine for Alcoholic Liver Disease)

  • Objectives: This study aimed to investigate the trend in the research on alcoholic liver disease (ALD) using Korean traditional medicine. Methods: This review was conducted using six electronic databases (NDSL, KMBASE, Koreantk, KISS, KISTI, and KoreaMed) with no restrictions in year. The search terms were "alcoholic liver disease", "alcoholic fatty liver", "alcoholic hepatitis", "alcoholic cirrhosis", "herbal medicine", "acupuncture" and "traditional medicine". The searched studies were analyzed according to the type of research. Results: After screening, 37 studies were selected among 552. The types of studies were as follows: 8 in vivo studies, 17 case reports, 7 case series, 1 assessment scale study, 1 randomized controlled trial, and 3 research reviews. The in vivo studies reported the efficacy and its mechanism in the animal phase of single or complex herbal medicine. In the clinical research, interventions such as herbal medicine and acupuncture were most commonly used for ALD treatment. Conclusions: We analyzed the trends in ALD treatment using Korean traditional medicine through this review. The results showed that Korean traditional medicine could be an effective method for ALD treatment. Conducting related in-depth studies, such as well-designed randomized controlled trial based on the results of experimental research, is necessary.

6-O-Galloylsalidroside, an Active Ingredient from Acer tegmentosum, Ameliorates Alcoholic Steatosis and Liver Injury in a Mouse Model of Chronic Ethanol Consumption

  • We have previously reported that Acer tegmentosum extract, which is traditionally used in Korea to reduce alcohol-related liver injury, suppresses liver inflammation caused by excessive alcohol consumption and might improve metabolism. The active ingredient, 6-O-galloylsalidroside (GAL), was isolated from A. tegmentosum, and we hypothesized that GAL could provide desirable pharmacological benefits by ameliorating physiological conditions caused by alcohol abuse. Therefore, this study focused on whether GAL could ameliorate alcoholic fat accumulation and repair liver injury in mice. During chronic alcohol consumption plus binge feeding in mice, GAL was administered orally once per day for 11 days. Intrahepatic lipid accumulation was measured in vivo using a noninvasive method, 1H magnetic resonance imaging, and confirmed by staining with hematoxylin and eosin and Oil Red O. The serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured using a Konelab system, and the triglyceride content was measured in liver homogenates using an enzymatic peroxide assay. The results suggested that GAL alleviated alcohol-induced steatosis,e as indicated by decreased hepatic and serum triglyceride levels in ethanol-fed mice. GAL treatment also correlated with a decrease in the Cd36 mRNA expression, thus potentially inhibiting the development of alcoholic steatosis via the hepatic de novo lipogenesis pathway. Furthermore, treatment with GAL inhibited the expression of cytochrome P450 2E1 and attenuated hepatocellular damage, as reflected by a reduction in ALT and AST levels. These findings suggest that GAL extracted from A. tegmentosum has the potential to serve as a bioactive agent for the treatment of alcoholic fatty liver and liver damage.

보리수열매주의 알코올성 지방간 형성 억제 효과 (Cherry Silverberry (Elaeagnus multiflora) Wine Mitigates the Development of Alcoholic Fatty Liver in Rats)

  • 본 연구에서는 보리수 열매를 원료로 하여 만든 보리수열 매주의 항산화 활성을 조사하고, 흰쥐에 보리수열매주의 급여가 알코올 지방간 완화에 미치는 영향을 조사하였다. 알코올의 섭취로 지방간을 유도하기 위하여 Lieber-DeCarli 액체식이를 공급하였으며, 액체 표준식이만 공급받는 동물군을 대조군(control), 대조군과의 열량 차이를 에탄올로 보충한 액체식이를 공급받는 동물군을 에탄올군(ethanol), 에탄올 대신 보리수열매주를 혼합한 액체식이를 공급받는 동물군을 보리수열매주군(cherry silverberry wine, CSW)으로 6마리씩 3그룹으로 나누어 6주간 사육하였다. 실험 시작 전, 3주째, 그리고 6주째에 혈액을 채취하였고 간은 6주 혈액 채취직후 적출하였다. 보리수주의 총 페놀함량은 37.24 mg/100 mL, SOD 유사활성은 22.26%, DPPH 라디칼 소거능은 13.69 %로 나타났다. 혈액의 주요 지방간 표지자들의 농도는 대조군에 비해 에탄올군에서는 유의적으로 증가하였으나 보리수열매주군에서는 유의적으로 감소하였다. 대조군에 비해 에탄올군의 간은 지방구가 축적되어 비대해진 것을 확인할 수 있었으며, 보리수열매주군의 간은 지방구의 크기나 수가 대조군 수준으로 감소한 것으로 나타났다. 이와 유사하게 간조직의 총지방량 농도도 보리수열매주군에서 유의적으로 감소하였다. 본 실험을 통해 보리수열매주가 알코올 섭취로 증가된 혈중 지질 수준과 간기능 지표 수준을 개선시키는 효과를 나타냈으며 간조직에 지방간 형성이 유의적으로 억제하는 것을 간조직 분석을 통해 확인하였다. 이는 보리수열매에 함유되어 있는 다양한 피토케미칼 성분의 항산화 효과로 인해 알코올에 의한 간 손상을 보호하는데 기여할 수 있음을 나타낸 결과로 사료된다.

Alcohol oxidase 효소센서를 이용한 알코올 음료 중의 에탄올 정량 (Determination of Ethanol in Alcoholic Beverages by Alcohol Oxidase Sensor)

  • 알코올 함량을 보다 신속하고 정확하게 측정하고자, 알코올 센서를 사용하였다. Alcohol oxidase를 glutaraldehyde로 nylon net에 고정화시킨 다음 산소전극에 연결하여 기질과의 반응에서 소모되는 용존산소 소비량의 변화를 용존산소측정기로 측정하여 알코올을 정량하였다. 알코올 센서의 최적조건에서 시판되는 각종 주류를 측정해 본 결과, 각각 맥주는 $4{\sim}5%$, 저알코올성 음료는 0.71%, 포도주는 10.06%, 청주는 16.12%, 소주는 25.71%, 탁주는 6.18%로 정량되었으며, 이는 가스 크로마토그래피로 측정한 값과 좋은 상관관계를 가졌다.
