Identification and Molecular Characterization of PERV Gamma1 Long Terminal Repeats

  • Porcine endogenous retroviruses (PERVs) gamma1 in the pig genome have the potential to act as harmful factors in xenotransplantation (pig-to-human). Long terminal repeats (LTRs) are known to be strong promoter elements that could control the transcription activity of PERV elements and the adjacent functional genes. To investigate the transcribed PERV gamma1 LTR elements in pig tissues, bioinformatic and experimental approaches were conducted. Using RT-PCR amplification and sequencing approaches, 69 different transcribed LTR elements were identified. And 69 LTR elements could be divided into six groups (15 subgroups) by internal variation including tandem repeated sequences, insertion and deletion (INDEL). Remarkably, all internal variations were indentified in U3 region of LTR elements. Taken together, the identification and characterization of various PERV LTR transcripts allow us to extend our knowledge of PERV and its transcriptional study.

Arg243, Invariably Critical for the Transcriptional Activation of Yeast Gcn4p

  • The arginine residue at position 243 (Arg 243) of the yeast transcription factor, Gcn4p, is invariably conserved among bZIP transcription factors. Using site-directed oligonucleotide saturation mutagenesis involving two-step polymerase chain reaction (PCR) amplification, random mutations were successfully introduced at the codon of 243 in the basic domain of Gcn4p. This mutant library was transformed ito Gcn4p defective yeast strain and selected for the transcriptionally active colonies. All colonies which were transcriptionally active had arginines in the codon 243. In this study, the strand preference by Taq polymerase during mutagenesis was also tested. Oligonucleotides were specially designed to test whether or not the polymerase was preferred using the strand as a template. A population of randomly mutated products were cloned into an appropriate vector and characterized by DNA sequencing analysis. Saturation mutagenesis which was performed efficiently by this method revealed a strong bias in terms of strand preference of Taq polymerase by an approximate ratio of 3 to 1 in this study.

Modeling of Public Risk Perception and Risk Communication Research: In A Social-Cognitive Direction

  • This study begins with a review of commonly discussed dimensions of public risk perception that may influence public opinion toward risks. Factors that have been revealed by the literature to have substantial impact on risk perception, such as demographic background, trust, and media environment, are also discussed. Meanwhile, we evaluate two well-known research models in the realm of risk analysis: 1) the psychometric paradigm, and 2) the social amplification of risk framework (SARF). Based on a literature review, this study suggests that, besides the psychological and social approach, models of risk perception and risk communication research should shift to a more comprehensive one by considering the interrelations between laypeople and the environment. This study proposes a research model from the perspective of social cognitive theory (SCT) as a potential framework for future studies: 1) in the societal environment, individuals' risk perception and information seeking behavior, which is determined by risk perception will be influenced by trust in regulators and interpersonal trust; 2) in the media environment, individuals' risk perception and information seeking behavior will be influenced by individuals' perceived information characteristics. Knowledge about risk accumulated through information seeking will change risk perception in a longitudinal process.

  • Broadcast nature of wireless medium and path-loss reduction create a favourable condition for collaborative communications (CC) among single-antenna users to gain the powerful benefits of multi-antenna system without the demand for physical arrays. This paper proposes a CC strategy adapting to the propagation environment changes by optimizing the transmit signal amplification factors to simplify the structure of maximum likelihood (ML) detector and to obtain the minimum error probability as well. The closed-form BER expression was also derived and compared to the simulation results to evaluate the performance of the suggested solution. A variety of numerical results revealed the cooperation significantly outperforms non-cooperative counterpart under flat Rayleigh fading channel plus AWGN (Additive White Gaussian Noise).

  • Breast cancer is the most common cancer worldwide among women and the second most common cancer. Approximately 15-23% of breast cancers over-express human epidermal growth factor receptor2 (HER2), a 185-kDa transmembrane tyrosine kinase, which is mainly found at the cell surface of tumor cells. HER2-positive breast cancer, featuring amplification of HER2/neu and negative expression of ER and PR, has the three following characteristics: rapid tumor growth, lower survival rate, and better response to adjuvant therapies. Clinically, it is notable for its role in a pathogenesis that is associated with increased disease recurrence and acts as a worse prognosis. At the same time, it represents a good target for anti-cancer immunotherapy despite the prevalence of drug resistance. New treatments are a major topic of research, and a brighter future can be expected. This review discusses the role of HER2 in breast cancer, therapeutic modalities available and prognostic factors.

  • An acoustic finite element model of a bridge is developed to evaluate the noise generated by the traffic-induced vibration of the bridge. The dynamic response of a multi-girder bridge, modeled by a 3-dimensional frame element model, is analyzed with a 3-axle(8DOF) truck model and a 5-axle(l3DOF) semi-trailer. The flat plate element is used to analyze the acoustic pressure due to the fluid-structure interactions between the vibrating surface and contiguous acoustic fluid medium. The radiation fields of noise with a specified distribution of vibrating velocity and pressure on the structural surface are also computed using the Kirchhoff-Helmholtz integral. In an attempt to illustrate the influence of the structural vibration noise of a bridge to total noise level around the bridge, the random function is used to generate the vehicle noise source including the engine noise and the rolling noise interacting between the road and tire. Among the diverse parameters affecting the dynamic response of bridge, the vehicle velocity, the vehicle weight, the spatial distribution of the road surface roughness, the stiffness degradation of the bridge and the variation of the air temperature changing the air density are found to be the main factors that increase the level of vibration noise. Consequently, The amplification rate of noise increases with the traveling speed and the vehicle weight.

Modification of Site Classification System for Amplification Factors considering Geotechnical Conditions in Korea (국내 지반 특성에 따른 합리적 증폭 계수의 결정을 위한 지반 분류 체계 개선 방안 고찰)

  • For the site characterization at two representative inland areas, Gyeongju and Hongsung, in Korea, in-situ seismic tests containing boring investigations and resonant column tests were performed and site-specific ground response analyses were conducted using equivalent linear as well as nonlinear scheme. The soil deposits in Korea were shallower and stiffer than those in the western US, from which the site classification system and site coefficients in Korea were derived. Most sites were categorized as site classes C and D based on the mean shear wave velocity to 30 m, Vs30 ranging between 250 and 650 m/s. Based on the acceleration response spectra determined from the site-specific analyses, the site coefficients specified in the Korean seismic design guide underestimate the ground motion in the short-period band and overestimate the ground motion in mid-period band. These differences can be explained by the differences in the bedrock depth and the soil stiffness profile between Korea and western US. The site coefficients were re-evaluated and the preliminary site classification system was introduced accounting for the local geologic conditions on the Korean peninsula.

  • Numerical analysis of PSC box bridge bearings for high speed KTX train vehicles has been carried out as a virtual simulation for Ubiquitous Technology. Improved numerical models of bridge, vehicle and interaction between bridge and train are considered, where bending and torsional modes are provided, whereas the exist UIC code is applied by the simplified HL loading. Dynamic and static analysed results are compared to get Dynamic Amplification Factors (D. A. F.) for maximum deflections and bending stresses up to running speed of 500 km/h. Equation from the regression analysis for the D. A. F. is presented. Sliding distance of the bearings for various KTX running speeds is compared with maximum and accumulated distances by the dynamic behaviors of PSC box bridge. Dynamic and static simulated sliding distances of the bearings according to the KTX running speed are proved as a major parameter in spite of the specifications of AASHTO and EN1337-2 focused on the distance by temperature variations.

Assessment of genetic diversity and distance of three Cicuta virosa populations in South Korea

  • Cicuta virosa L. (Apiaceae) is a perennial emergent plant designated as an endangered species in South Korea. According to the former records, only four natural habitats remain in South Korea. A former study suggested that three of four populations (Pyeongchang: PC, Hoengseong: HS, Gunsan: GS) would be classified as different ecotypes based on their different morphological characteristics and life cycle under different environmental conditions. To evaluate this suggestion, we estimated genetic diversity in each population and distance among three populations by random amplification of polymorphic DNA. Seven random primers generated a total of 61 different banding positions, 36 (59%) of them were polymorphic. Nei's gene diversity and the Shannon diversity index increased in the order of PC < HS < GS, which is the same order of population size. In the two-dimensional (2D) plot of first two principal components in principal component analysis with the presence of 61 loci, individuals could be grouped as three populations easily (proportion of variance = 0.6125). Nei's genetic distance for the three populations showed the same tendency with the geographical distance within three populations. And it is also similar to the result of discriminant analysis with the morphological or life-cycle factors from the previous study. From the results, we concluded that three different populations of C. virosa should be classified as ecotypes based on not only morphology and phenology but genetic differences in terms of diversity and distance as well.

Research on the Efficiency Improvement of the Cymbal-type Piezoelectric Energy Harvester (심벌형 압전 에너지 하베스터 에너지 수율 향상 연구)

  • The pollution problem of fossil energy sources has caused the development of green energy harvesting systems. Piezoelectric energy harvesting technology has been developed under those external environmental factors. A piezoelectric energy harvester can be defined as a device which transforms mechanical vibration or impact energy into electrical energy. Most researches have focused on bender structures. However, these have a limitation on energy efficiency because of the small effective electromechanical coupling factor, around 10%. Therefore, we should look for a new design for energy harvesting. A cymbal energy harvester can be a good candidate for the high-power energy harvester because it uses a high amplification mechanism using endcaps while keeping a higher electromechanical coupling factor. In this research, we focused on energy efficiency improvements of the cymbal energy harvester by changing the polarization direction, because the electromechanical coupling factor of the k33 mode and the k15 mode is larger than that of the k31 mode. Theoretically, we checked the cymbal harvester with radial polarization and it could obtain 6 times larger energy than that with the k31 direction polarization. Furthermore, we verified the theoretical expectation using the finite element method program. Consequently, we could expect a more efficient cymbal harvester with the radial polarization by comparing two polarization directions.
