The Temporal and Spatial Expression of the Cytokeratin in Keratinocytes during Cutaneous Wound Healing on the Amphibian(Bombina orientalis) (양서류 피부 상처회복과정 중 각질화세포 cytokeratin의 분포)

  • The system of wound healing is very complex biological processing that includes inflammatory, reepithelialization, and matrix construction. For identification of the transitional pathway of the keratinocytes, we have employed immunohistochemical analysis using cytokeratin antibody after wounding. Epithelium in skin of the frog(Bombina orientalis) was examined with transmission electron microscopy. Cytokeratin was expressed in normal basal and gland cavity cells. At 3-hour basal layer cells were strong positive, however cells of the upper layer were negative reaction. Day1 and 2 after post-wounding, regenerating epithelial cell layer was positive reaction, especially basal layer cells were strong positive. At day 10 after wounding, the degree of positive reaction to basal cells of regenerating epithelial tissue was equal to day 7 wound tissue. At day of 19th, basal and spinous layer cells were strong positive reaction. Regenerating epithelial cells were positive but some basal cells were strong positive at day 27. From this result, we identified that the migration of the keratinocytes in amphibian skin wounds is initiated from basal layer fells and the keratinocytes migrate into basal and middle of the wound area.

  • Amphibians play a pivotal role in the ecosystem as a mediator between aquatic and terrestrial environment. Currently they are directly exposed to a variety of chemicals in the aquatic environment throughout their life cycle. Azole fungicides have been widely used in medical applications and agricultural activities. The direct exposure of azole fungicides causes an alarming situation for various ecosystem. Recently, teratogenesis and endocrine disruption by azole fungicides have been reported in amphibians. In an effort to provide the current information for amphibian toxicity of azole fungicides and to make the guidelines for safe usage of azole-based materials, the effects of azole fungicides including imidazole, triazole, thiazole, oxazole, and pyrazoleon on early development, differentiation and reproduction of amphibians were reviewed.

Differences in Habitat Structure and Herpetofauna Populations Caused by Thinning (간벌에 의한 서식지 구조와 양서·파충류 개체군의 특성 차이)

  • This study was conducted to clarify the differences in habitat structure and amphibian and reptile (herpetofauna) populations between before and after thinning in the coniferous and deciduous forests from June to September 2014 and 2015 at the Mt. Maewha of Hongcheon, Gangwon province, Korea. Forest structure was dramatically changed after thinning. Coverage of understory and mid-story were decreased while volume and number of downed trees were increased after thinning. We used line transect sampling method on a total of 4 transect lines, 2 lines in each forest type. 119 individuals of 5 amphibian species and 5 individuals of 3 reptile species were recorded. Mean number of amphibian species were decreased after thiining in the coniferous forest. There was no observed reptiles after thinning in the coniferous and deciduous forests. Detailed monitoring with different area, frequency and intensity might be needed to clarify more precise differences characterisitics of herpetofauna by thinning.

A Study on Riparian Habitats for Amphibians Using Habitat Suitability Model (서식지적합성 모형을 이용한 수변지역 양서류 서식지 분석)

  • The objective of this study was to analyze characteristics of distribution of amphibian species and the affecting ecological factors. For the study, habitat environment factors were determined and applied to a habitat suitability model for the data collected from the Seom River in Hoengseong County and Wonju City, Gangwon Province, Korea between March 2013 to October 2013. The analyzed amphibian species were Rana nigromaculata, Hyla japonica, Rana dybowski, and Rana rugosa Temminck and Schlegel, and a logistic regression model was used with the pseudo-absence data. The result of the model analysis suggests that the major factors for Rana nigromaculata are distance to vegetation and rock and that for Hyla japonica is waterway. Rana dybowski and Rana rugosa Temminck and Schlegel have similar habitat characteristics, but the latter is shown to be dominant due to its wider habitat preference. According to the species richness model, the analyzed amphibian species are shown to have tendency to move between valleys or streams. This study quantitatively analyzed habitat environment characteristics using species distribution model, however, there is a limitation in terms of analysis on food factor and connectivity with other species. Combined with additional density or habitat analysis on birds or fish, this study can lead to more comprehensive analysis on biological environment factors.

Developmental Toxicity of Alkylphenols in Amphibians: A Review (알킬페놀류 화합물의 양서류 발생독성: 종설)

  • Aquatic contamination by organic pollutants has been a suspected reason for rapid decrease of amphibian populations whose embryonic and larval stages are in an aquatic environment. Amphibian embryos can be a useful model to study the ecoctoxicologial impacts of aquatic pollutants. The obtained toxicological data are useful references for the management of aquatic pollutants in public health because amphibia share many developmental events with terrestrial vertebrates including humans. Safety guidelines for the toxicological effects of aquatic contaminants of chemicals identified as hazardous should be addressed at multiple endpoints. Alkylphenols have been widely-used in agricultural, industrial, and household activities; they contaminate and can persist in aquatic environments. Exposure to alkylphenols results in endocrine disruption in aquatic animals. In this review, we summarize the developmental toxicities of alkylphenols in amphibian embryos and larva according to the exposure route, chemical concentration, duration of exposure, and affected developmental stage together with mechanisms of toxicity and typical patterns of developmental abnormality. The merits of amphibian embryos as a toxicity test model for mid- to long-term exposure to aquatic pollutants are discussed proposed.

Characteristics of Amphibian and Reptile Populations in a Coniferous Plantation and a Deciduous Forest (침엽수림과 활엽수림에 서식하는 양서류와 파충류 개체군 특성)

  • This study was conducted to clarify the characteristics of amphibian and reptile (herpetofauna) populations in a coniferous plantation and a deciduous forest from July to September 2013 in national forest, Hongcheon, Gangwon province, Korea. Coverage of understory, mid-story and overstory were more developed in a deciduous forest. We used line transect sampling method on a total of 4 transect lines, 2 lines in each forest type. Fifty two individuals of 4 amphibian species and 11 individuals of 3 reptile species were recorded. Mean observed number of herpetofauna species, individuals and species diversity index (H') were not significantly different between coniferous plantation and deciduous forest. This result related to ecological characteristics of herpetofauna. Habitats of herpetofauna were influenced on not only vertical structure of forests, but also climatic variables, thermoregulation and prey availability. The long-term monitoring and research on habitat preference and seasonal ecological traits of herpetofauna would be needed for conservation and management of herpetofauna in forest areas.

The Ultrastructure of the Mucus Secreting Cells in the Amphibian Skin (양서류 피부 점액분비세포의 미세구조)

  • The authors observed the ultrastructure of the mucous glandular epithelial cells in the amphibian skin by mean of electron microscope. The specimens from the experimental animals were fixed in 2.5% glutaraldehyde-oaraformaldehyde fixative in phosphate buffer at pH 7.2 prior to fixation in 1% osmium tetroxide, dehydrated in graded ethanol and acetone, embedded in Epon 812 mixture, and sectioned with Sorvall MT-2 ultramicrotome. The ultrasections were contrasted with uranyl acetate and lead citrate and observed with a JEOL-100B electron microscope. The results were as follows: 1. The cutaneous mucous glands in amphibia consisted of the glandular epithelial and the myoepithelial cells. 2. Several different cells in ultrastructure were observed in the mucous glandular epithelium of the adult amphibian skin. a. The dark and the light cells were observed in Hynobius leechi. b. The mitochondria-rich and the round secretory granule-containing cells were observed in Bombina orientalis. c. The round secretory granule-containing and the foam-like granule mass-containing cells were observed in Kaloula tornieri. d. The cutaneous mucous gland of Rana nigromaculata were divided into two types: A and B-type glands. In the A-type mucous gland, the mitochondria-rich and the round secretory granule-containing cells and in the B-type mucous gland, the mitochondria-rich, the secretory granule-containing and the ER-rich cells were observed. 3. Based upon the above findings, the authors infer that the mucous granular epithelium of the amphibian skin consists of the mitochondria-rich undifferentiated, the secetory granule-containing and mature, and the ER-rich evacuated cells.
