굴에 기생하는 Amoeba에 대하여 (AMOEBA INFECTION IN OYSTER (CRASSOSTREA GIGAS))

  • 거제만의 수하식 양식굴에서 난소의 이상발육과 팽융된 부분이 관찰되었다. 1978년 $9\~10$월과 1979년 10월에 조사한 결과는 다음과 같다. 1) 7개지점중 화도와 송구에서만 $6.3\~13.3\%$ 검출 되었다. 2) amoeba에 기생된 굴의 무게는 $6.2\~7.0g$이다. 3) 기생된 굴은 수심에 관계없이 상${\cdot}$${\cdot}$하층에서 다 검출되었다. 4) 난세포에서만 검출되며 amoeba의 크기는 $3\~23\mu$이다. 5) 난세포내에는 $1\~3$개체의 amoeba가 가장 많이 검출되었다. 6) amoeba는 PAS 염색 e양성이며 Eosin에 잘 염색된다. 7) amoeba의 형태와 성상이 Swayer가 보고한 Acanthamoeba sp.와 유사했다. 8) 순수배양하여 형태변화를 봐 고정하고저 한다.

광에 대한 Amoeba 와 Planaria 의 습성화에 따른 단백질 합성유형의 변화 (Change of Protein-synthetic Patterns with Habituations of Amoeba and Planaria by the Light Stimuli)

  • When the light stimulus was presented continuously, habituations were formed, after 13 and 14 hrs in amoeba and planaria, repectively. As amoeba more habituated, the light-escaping velocity of streaming was decreased with a negative-exponential function. in two-dimensional electrophoretic patterns of proteins of amoeba habituated, a newly synthesized protein (pI 7.0, MW 20 kD) was detected. Therefore, it is conjectured that similarity or commonality may exist between amoeba and planaria habituations to light, and in these behavioral modifications, specific protein(s) may be involved in molecular level.

  • 이 연구의 주된 목적은 공간 클러스터의 범역을 설정하는 GIS-기반 방법론을 개발하는 것이다. 주요 과제는 지리적 경계 분석과 LISA-기반 클러스터 탐지에 대한 기존 방법론을 비교 검토함으로써 진일보한 방법론을 고안하고, 그것을 실행하는 GIS-기반 프로그램을 개발하는 것이다. 주요 연구 결과는 다음과 같다. 첫째, 기존 방법론을 검토한 결과, LISA를 이용한 AMOEBA 기법이 가장 타당한 것으로 판단되었다. 둘째, 수정 AMOEBA 기법의 알고리즘을 확립했으며 실행 소프트웨어를 상용 GIS 프로그램의 확장 기능형태로 개발하였다. 셋째, 수정 AMOEBA 기법을 실험 데이터와 실 데이터에 적용한 결과 제안된 기법의 유용성이 확인되었다.

Amoeba proteus에 있어서 박테리아 감염에 의한 변이주 특이성 단백질의 손실 (Loss of a Strain-Specific Protein by Bacterial Infection in Amoeba proteus)

  • Amoeba proteux xD strain에서 추출한 공생박테리아를 tD strain에 감염시키고, 숙주의 박테리아 감염으로 인한 변이주 특이성 단백질의 손실을 2차원 전기영동에 의해 탐지하였다. 유도 식작용에 의한 실험감염 50일 만에 숙주인 아메바는 tD strain 특이성 단백질을 손실하였며, 이는 $27^\\circC$ 배양에 의해 감염 박테리아 및 xD strain 특이성 단백질을 제거한 후에도 재합성되지 못하였다. 이 시기면 숙주인 아메바는 박테리아에 완전히 의존한 것으로 판명되었다. 이상의 결과 및 Lorch Jeon (1981, Science 221:549)의 결과를 볼 때 감염된 숙주핵이 감염되지 않은 아메바 원형질과 양립하지 못하는 것이나 숙주의 박테리아에 대한 의존 유발은 박테리아 감염에 의해 세포 특이성 유전인자의 비가역적인 불활성화 또는 솔실로 인한 것이 분명하다.

First Record of Potentially Pathogenic Amoeba Vermamoeba vermiformis (Lobosea: Gymnamoebia) Isolated from a Freshwater of Dokdo Island in the East Sea, Korea

  • Vermamoeba vermiformis is a very important free-living amoeba for human health in association with Legionnaires' disease and keratitis. This interesting amoeba was firstly isolated from a freshwater of Dokdo (island), which was historically used for drinking water. Trophozoites and cyst forms of V. vermiformis strain MG1 are very similar to previous reported species. Trophozoites of V. vermiformis strain MG1 showed cylindrical shape with prominent anterior hyaline region. The average ratio of length and width was about 6.5. Typically, cysts of the strain MG1 showed a spherical or slightly ovoidal shape with smooth wall, and lacked cyst pores. Some cysts had crenulate-walled ectocyst, which was separated from endocyst wall. Further, 18S rRNA gene sequence of V. vermiformis strain MG1 showed very high similarity to other V. vermiformis species (99.4%-99.9% identity). Molecular phylogenetic analysis based on 18S rRNA gene sequences clearly confirmed that the isolate was one strain of V. vermiformis with maximum bootstrap value (maximum likelihood: 100%) and Bayesian posterior probability of 1. Thus, the freshwater of Dokdo in Korea could harbor potentially pathogenic amoeba that may cause diseases in humans.

상수원수 및 정수처리공정별 가시아메바 분포특성에 관한 연구 (Distribution of Acanthamoeba spp. in Raw Water and Water Treatment Process)

  • The free-living amoeba and Acanthamoeba sp. are widely distributed in fresh water, soil, air and dust in the world. We studied distribution of amoeba from low Nakdong River(Mulgum and Maeri) and removal efficiency in water treatment process of Busan metropolitan city. During this investigation, water quality showed pH $7.4{\sim}9.6({\pm}1.1)$, water temperature $2.0{\sim}29.0({\pm}17)^{\circ}C$, turbidity $4.8{\sim}27.4({\pm}11.0)$ NTU, chlorophyll-a $10.3{\sim}109.0({\pm}44.3)\;mg/m^{3}$, BOD $1.7\sim4.9({\pm}2.6)$ mg/L, COD $3.1\sim-6.9({\pm}5.0)$ mg/L and total coliform $17\sim920({\pm}200.5)$ MPN/100 mL. The free-living amoeba were detected highly than Acanthamoeba sp., 11 out of 22 in raw water samples were positive (50%) for Acanthamoeba sp. from February 2005 to December 2005. The seasonal characteristics of tree-living amoeba and Acanthamoeba sp. in raw water were mainly distributed through the spring to the early fall. When tree-living amoeba and Acanthamoeba sp. were passed through the water treatment of pilot-plant, approximately 80% was sure to be removed through pre-ozonation, sedimentation, send filtration. 100% was removed after post-ozonation process. All of the isolated amoebas from Nakdong River were Acanthamoeba sp. AC311 18S ribosomal RNA gene with 98% nucleotide sequence homology.

공간통계기법을 이용한 토지피복변화의 핫스팟 탐지 (Hotspot Detection for Land Cover Changes Using Spatial Statistical Methods)

  • 최근 인구집중으로 인한 도시개발 및 사회기반 시설의 확충 등의 인위적 현상과 지구온난화 및 가뭄, 홍수, 산불 등 자연적 현상 등 다양한 요인으로 인해 토지피복 변화가 발생하고 있다. 이러한 토지피복변화의 원인과 영향, 그리고 탐지방법에 대한 다양한 연구가 진행되고 있는데, 기존 연구들은 대부분 주어진 임계치를 기준으로 '변화' 또는 '무변화'의 양분된 결과를 도출한다. 따라서 본 연구에서는 확률에 기초하여 공간자기상관과 핫스팟 탐지를 결합한 변화탐지 방법을 제시하고자 한다. 또한 기존의 핫스팟 탐지 알고리즘인 AMOEBA(A Multidirectional Optimal Ecotope-Based Algorithm)에서 나타나는 과다탐지의 경향을 개선하여 핵심적인 토지피복 변화의 핫스팟을 탐지하는 AMOEBA-CH(core hotspot)를 개발 및 적용하였다. 본 연구에서는 토지피복 변화의 확률과 공간자기상관에 기초한 픽셀 간의 공간적 상호작용을 고려하였으며, AMOEBA-CH의 인접-우세 모형을 통해 토지피복 변화의 핵심적인 핫스팟을 탐지하고자 한다. 본 연구에서 제시한 방법의 가용성을 테스트하기 위해 남한지역의 2000년과 2008년의 NDVI자료를 이용하여 토지피복 변화의 핫스팟 탐지를 수행하였다.

광자극에 대한 Amoeba 의 반응양상과 phosphatase 의 특성 및 활성도 변화 (The Characterization and Activity Changes of Phosphatases in Amoeba sp. to the Light Stimuli and Its Response Pattern)

  • Amoeba sp. was cultured under the light and the dark conditions, and the activity of phosphatases was investigated. There was a linear correlation between the early reaction time and the activity of phosphatases when phosphatases were incubated at 30℃. Then the activity of acid phosphatase was about 2 times higher than that of alkaline phosphatase. The activity of phosphatase was optimal at pH 5.0 in acidic part and at pH 8.0 in alkaline part, respectively. The optimal temperature of phosphatases was near the 40℃. The isozyme patterns of cytoplasmic acid phosphatase were compared with those of membraneous one. Both the isozyme patterns were shown to bo polymorphic on the polyacyamide gel, but different band patterns were observed in the isozymes of the cytoplasmic and the membraneous acid phosphatases. The number of Amoeba sp. under the light stimulus for 48 hours decreased negative exponentially from the illumination. The activity of acid and alkaline phosphatases under the illumination of light incresed 1.7 and 1.5 times higher, respectively, than the activity of those under the dark condition. This result apperars to be related to the mechanism of the autophosphorylation.

단항체를 이용한 아메바(Amoeba proteus) 의 2종 핵막 단백질에 대한 면역형광현미경적 및 생화학적 특성 조사 (Immunofluorescence Microscopy and Biochemical Characterization of Two Nuclear Envelope Proteins of Amoeba proteus by Using a Monoclonal Antibody)

  • Distribution of the antigens during the cell cycle of amoebae was followed by immunof-luorescence microscopy using a monoclonal antibody against the nucleus as a probe. While the cells were in the interphase, the antigen was localized on the nucleus membrane. But it was dispersed all over the cytoplasm during mitosis and cytokinesis. The molecular weights of the immunoreacted antigens were 210 KD and 190 KD as determined by SDS PAGE and western blotting of the purified nuclei. The antigens were not soluble in non-ionic detergent, but were released from the nucleus by incubation with 0.05 M sodium carbonate, pH 10.6 or with 8 M urea at serial chemical extraction. Thus the antigens appeared to be peripheral proteins of the nurBeus envelope. The isoelectic point of both antigens was 7.64 as determined by 2 D PAGE and transfer blotting. Considering the peiipherd association with the nucleus membrane and the dispersed distribution during mitosis, the antigens could be lamin like proteins. Hourever, it appears also possible that they are the component molecules of the unusually structured aurous lamina of amoeba nucleus since they have the large molecular weight and the basic pl.

  • In this paper, there analyzed the UV behavior and detection images due to corona discharge of insulators at 22.9kV. As the results, the shapes of UV detection images are jellyfish, sunflower, and amoeba type. Generally, the jellyfish and sunflower type appeared in air discharge. In surface discharge, Amoeba is generated. Also, there are classified 8 patterns such as sunflower A or B, jellyfish A or B, amoeba A, B, C, D. This method of the facilities assessment has an immediate effect in the field.
