Enhancement of L-Threonine Production by Controlling Sequential Carbon-Nitrogen Ratios during Fermentation

  • Controlling the residual glucose concentration is important for improving productivity in $\text\tiny{L}$-threonine fermentation. In this study, we developed a procedure to automatically control the feeding quantity of glucose solution as a function of ammonia-water consumption rate. The feeding ratio ($R_{C/N}$) of glucose and ammonia water was predetermined via a stoichiometric approach, on the basis of glucose-ammonia water consumption rates. In a 5-L fermenter, 102 g/l $\text\tiny{L}$-threonine was obtained using our glucose-ammonia water combined feeding strategy, which was then successfully applied in a 500-L fermenter (89 g/l). Therefore, we conclude that an automatic combination feeding strategy is suitable for improving $\text\tiny{L}$-threonine production.

Effect of Solvents as Subcritical and Supercritical Fluid on Decomposition and Extraction of Used Automotive Tire (아임계와 초임계유체로써 폐타이어 분해와 추출에 미치는 용매의 영향)

  • Side wall samples from a used automotive tire were subjected to subcritical and supercritical decomposition and extraction with three solvents, water, 28% ammonia solution and ammonia. For 6mm cube samples the rate of supercritical extraction with water followed a first-order kinetics with an activation energy of 140 kJ/mol. Solvent power of 28% ammonia so lotion at supercritical condition was found to be higher than supercritical water at initial extraction as pressure decreased. These phenomena were considered to be an effect of ammonia involved in water.

  • Removal of nitrogen compound from waste water is essential and often accomplished by biological process. To prevent washout and to develop an efficient bioreactor, immobilization of sutibal microorganisms could be sensible approach. Strains and permeabilized cell encapsulated in cellulose nitrate microcapsules and immobilized on polystyrene films were prepared by the method described in the previous study. In the wastewater treatment system, nitrification of ammonia component is generally known as rate controlling step. To enhance the rate of nitrification, firstly nitrifying strains Nitrosomonas europaea(IFO14298), are permeabilized chemically, and immobilized on polystyrene films and secondly oxidation rates of strain system and permeabilized strain system are compared in the same condition. with 30 minute permeabilized cells, it took about 25 hours to oxidize 70% of ammonia in the solution, while it took about 40 hours to treat same amount of ammonia with untreated cells. All the immobilization procedures did not harm to the enzyme activity and no mass transfer resistance through the capsule well was shown. In the durability test of immobilized system, the system showed considerable activity for the repeated operation for 90 days. With these results, the system developed in this study showed the possibility to be used in the actual waste water treatment system.

  • Selective Catalytic Reduction (SCR) technology is well-known to be effective for the reduction of NOx emission. So car manufacturers has adopted Ures-SCR system to be satisfied with emission regulation. This paper discusses the effective of $NH_3/NOx$ ratio and SCR catalyst temperature in the $NH_3$-SCR reactor on DeNOx performance. So it is shown the characteristic of NOx conversion and ammonia slip using the $NH_3$ instead of Urea-Solution. From the result of this study, it is found to optimize $NH_3/NOx$ ratio to have the best case of high NOx conversion and low ammonia slip at variable SCR catalyst temperatures. Lastly, it is also found the characteristics of NOx conversion and ammonia slip with compared with Urea.

  • In this study, a water electrolysis was studied to investigate the effect of ammonia on current density and H2 gas production. A H type cell with three electrodes was used and KOH solution was used as electrolyte. The conventional platinum foil was used for working electrode, whereas nickel foam was used for counter electrode. CV experiment was performed to see the activity of ammonia oxidation reaction. In addition, CP experiment was done to examine the dependence of Faraday efficiency of hydrogen on current density and NH3 concentration. The CV result shows the 0.5M NH3 concentration required for highest current density and reliable operation. The CP result shows the increased current density leads to higher H2 generation. The higher H2 production and subsequent energy efficiency was observed for 0.5M NH3 using a Pt/13%Rh coil for a cathode as compared to those in water electrolysis.

Electromagnetic Interference Shielding Efficiency Characteristics of Ammonia-treated Graphene Oxide (암모니아수 처리된 그래핀 옥사이드의 전자파 차폐효율 특성)

  • In this study, nitrogen doped graphene oxide (GO) was prepared using liquid phase ammonia treatment to improve its electrical properties. Also, the aminated GO was manufactured into a film format and the electromagnetic interference (EMI) shielding efficiency was measured to evaluate its electrical properties. The XPS result showed that the increase of liquid phase ammonia treatment concentration led to the increased nitrogen functional group on the GO surface. The measurement of EMI shielding efficiency reveals that EMI shielding efficiency of the liquid phase ammonia treated GO was better than that of non-treated GO. When GO was treated using the ammonia solution of 21% concentration, the EMI shielding efficiency increased by -5 dB at higher than 2950 MHz. These results were maybe due to the fact that nitrogen functional groups on GO help to improve the absorbance of electromagnetic waves via facile electron transfer.

  • This study was investigated on the recycle feasibility of the waste AC(Ammonium Carbonate) solution produ-ced in a commercial AUC(Ammonium Uranyl Carbonate) conversion plant. AUC particles were produced with the AC solution which was prepared with AC solid-agent instead of ammonia and carbon-dioxide gases. As the results particles of monoclinic shapes has been obtained regardless of the pH change if the carbonate concentration is sufficient in the mother liquore. Also a lot of twinned or aggregated particles were formed in case of the increase of pH in the reaction system but not affected in the change of temperature. Consequen-tly the characteristics of the particles which converted for AUC were produced withAC solution to UO2, particles specific surface area shape sintered density and others were similar to that of the particles which were produced with gases only when the pellets are fabricated in the nuclear fuel manufacturing process So the waste AC solution which is produced in the commercial AUC conversion plant is possible to recycle.

An Assessment on the Behavior of Nitrogenous Materials during the First High-rate Phase in Composting Process (퇴비화 공정의 1차 발효단계에서 질소성 물질의 거동 평가)

  • Composting of N-rich wastes such as food waste and wastewater sludges can be associated loss of with substantial gaseous N, which means loss of an essential plant nutrient but may also lead to environmental pollution. We investigated the behavior of nitrogenous materials during the first high-rate phase in composting of food waste. Air dried food waste was mixed with shredded waste paper or wood chip and reacted in a bench scale composting reactor. Samples were analyzed for pH, ammonia, oxidized nitrogen and organic nitrogen. The volatilized ammonia nitrogen was also analyzed using sulfuric acid as an absorbent solution. Initial progress of composting reaction greatly influenced the ammonification of organic nitrogen. A well-balanced composting reaction with an addition of active compost as an inoculum resulted in the promoted mineralization of organic nitrogen and volatilization of ammonia. The prolongation of initial low pH period delayed the production of ammonia. It was also found that nitrogen loss was highly dependent on the air flow supplied. With an increase in input air flow, the loss of nitrogen as an ammonia also increased, resulted in substantial reduction of ammonia content in compost. The conversion ratio of initial nitrogen into ammonia was in the range of 28 to 38% and about 77~94% of the ammonia produced was escaped as a gas. Material balance on the nitrogenous materials was demonstrated to provide an information of importance on the behavior of nitrogen in composting reaction.

Electrochemical Decomposition Characteristics of Ammonia by the Catalytic Oxide Electrodes (촉매성 산화물 전극에 의한 암모니아의 전기 화학적 분해 특성)

  • In order to know the electrochemical decomposition characteristics of ammonia to nitrogen, this work has studied several experimental variables on the electrolytic ammonia decomposition. The effects of pH and chloride ion at $IrO_2$, $RuO_2$, and Pt anodes on the electrolytic decomposition of ammonia were compared, and the existence of membrane equipped in the cell and the changes of the current density, the initial ammonia concentration and so on were investigated on the decomposition. The performances of the electrode were totally in order of $RuO_2{\approx}IrO_2>Pt$ in the both of acid and alkali conditions, and the ammonia decomposition was the highest at a current density of $80mA/cm^2$, over which it decreased, because the adsorption of ammonia on the electrode surface was hindered due to the evolution of oxygen. The ammonia decomposition increased with the concentration of chloride ion in the solution. However, the increase became much dull over 10 g/l of chloride ion. The $RuO_2$ electrode among the tested electrodes generated the most OH radicals which could oxidized the ammonium ion at pH 7.

  • Fundamental ideas of the free energy gradient method are briefly reviewed with three applications: the stable structures of glycine and ammonia-water molecule pair in aqueous solution and the transition state (TS) structure of a Menshutkin reaction $NH_3 + CH_3Cl → CH_3NH_3^+ + Cl^-$ in aqueous solution, which is the first example of full TS optimization of all internal degrees of freedom.
