답토양(畓土壤)의 부식(腐植)과 질소형태(窒素形態)에 관(關)한 연구(硏究) (Studies on the Characteristics of Humic Substances and Fractionations of Nitrogen in Paddy Soils)

  • 제주도(濟州道) 및 경기도(京畿道)를 제외(除外)한 전국농토배양답(全國農土培養畓)에서 비교적(比較的) 작물생산력이 높은 보통답토양(普通畓土壤) 7점(點)과 작물생산력이 낮은 사질답토양(砂質畓土壤) 7점(點)을 채취(採取)하여 부식(腐植)의 성상(性狀)과 질소(窒素)의 형태(型態)를 조사(調査), 비교(比較)한 결과(結果)는 다음과 같다. 1. 공시토양(供試土壤)의 Humus(Humic acid+Fulvic acid)의 양(量)은 665mg/100g 2680mg/100g의 수준(水準)이었으며, Humic acid는 보통답(普通畓) 평균치(平均値)가 1436mg/100g, 사질답(砂質畓) 평균치(平均値)는 970mg/100g이었다. 2. 웅전법(熊田法)에 의(依)한 부식산(腐植酸)이 분류형(分類型)은 B형(型), P형(型), RP형(型)에 분포(分布)되어 있었다. 3. 공시토양(供試土壤)의 무기태질소함량(無機態窒素含量)은 전질소함량(全窒素含量)의 1.04-2.86%였으며 사질답(砂質畓) 평균치(平均値)와 보통답(普通畓) 평균치(平均値)는 각각(各各) 1.72%, 1.75%였다. 4. 산가수분해(酸加水分解)에 의(依)한 유기태질소분별정량(有機態窒素分別定量) 결과(結果) 보통답(普通畓), 사질답토양(砂質畓土壤) 공(共)이 Amino산태질소(酸態窒素)가 가장 많았으며 사질답토양(砂質畓土壤)에서는 Amino산태질소(酸態窒素)가 31.52%, Humin태질소(態窒素) 20.63% Amino당태질소(糖態窒素)는 18.70%였다.

SPF 닭에서 재조합 H9N3 조류 인플루엔자 백신의 효능과 안전성 평가

  • To reduce the economic impact and control Low pathogenic avian influenza (LPAI), vaccination with inactivated vaccine has been considered in this country. We tried to develop inactivated vaccine with reassorted H9N3 AI virus which has different type of neuraminidase compare to those of field AI virus. Before reassorted vaccine was produced, we confirm the virus as master seed by limiting dilution, RT-PCR and sequencing method. Also, we evaluate the biological characteristics of the virus to find out the possibility of prevention against field infection of AI virus. Finally, we evaluate the safety and efficacy of the vaccine made of reassorted AI virus in the specific pathogen free (SPF) chickens. After limiting dilution, we choose RV7CE4 as a vaccine candidate and compare the gene sequence of this vaccine strain to those of AI05GA which is parents strain. Compared to amino acid sequences of specific gene of AI05GA and RV7CE4, exhibited a high degree of amino acid sequence homology. In the safety and efficacy test, there were no specific clinical signs or mortality. Reassorted H9N3 viruses were reisolated in cloaca swab on 5 days post inoculation. In the vaccine study, once or twice vaccination was performed and challenged with H9N2 field virus (01310). Vaccine has no adverse effect on birds and formed good immune capability which reduce viral shedding in the birds infected with 01310. Based on the above result, we developed reassorted H9N3 vaccine which will efficiently prevent the low pathogenic AIV (H9N2) infection in the poultry farms.

Synthesis and Antitumor Evaluation of cis-(1,2-Diaminoethane) dichloroplatinum (II) Complexes Linked to 5- and 6-Methyleneuracil and -uridine Analogues

  • The search for platinum (II)-based compounds with improved therapeutic properties was prompted to design and synthesize a new family of water-soluble, third generation cis-diaminedichloroplatinum (II) complexes linked to uracil and uridine. Six heretofore unreported uracil and uridine-platinum (II) complexes are; [N-(uracil-5-yl-methyl)ethane-1,2-di-amine]dichloroplatinum (II) (3a), [N-(uracil-6-yl-methyl)ethane-1,2-diaminel dichloroplatinum (II) (3b), t[N-($2^1$, $3^1$,$5^1$-tri-O-acetyl)uridine-5-yl-methyl] ethane-1,2-diamineldichloroplatinum (II) (6a), {[N-($2^1$,$3^1$, $5^1$-tri-O-acetyl) uridine-6-yl-methyl]ethane-1,2-diamine)dichloroplatinum (II) (6b),[N-(uridine- 5-yl-methyl)ethane-1,2-diamine]dichloroplatinum (II) (7a), [N-(uridine-6-yl- methyl)ethane-1,2-diamine]dichloroplatinum (II) (7b). These analogues were prepared from the key starting materials, 5-chloromethyluracil (1a) and 6-chloromethyluracil (1b) which were reacted with ethylenediamine to afford the respective 5-[(2-aminoethyl)aminol methyluracil (2a) and 6-[(2-aminoethyl)amino]methyluracil (2b). The cis-platin complexes 3a and 3b were obtained through the reaction of the respective 2a and 2b with potassium tetrachloroplatinate (II). The heterocyclic nucleic acid bases 1a and 1b were efficiently introduced on the .betha.-D-ribose ring via a Vorbruggen-type nucleoside coupling procedure with hexamethyldisilazane, trimethylchlorosilane and stannic chloride under anhydrous acetonitrile to yield the stereospecific .betha.-anomeric 5-chloromethyl- $2^1$,$3^1$,$5^1$-tri-O-acetyluridine (4a) and 6-chloromethyl-$2^1$,$3^1$,$5^1$-tri-O-acetyluridine (4b), respectively. The nucleosides 4a and 4b were coupled with ethylenediamine to provide the respective 5-[(amino-ethyl)aminolmethyl-$2^1$,$3^1$,$5^1$-tri-O-acetyluridine (5a) and 6-[(aminoethyl)amino] methyl-$2^1$,$3^1$,$5^1$-tri-O-acetyluridine (5b). The diamino-uridines 5a and 5b were reacted with potassium tetrachloroplatinate (II) to give the novel nucleoside complexes, 6a and 6b, respectively which were deacetylated into the free nucleosides, 7a and 7b by the treatment with CH$_{3}$ONa. The cytotoxic activities were evaluated against three cell lines (FM-3A, P-388 and J-82) and none of the synthesized compounds showed any significant activity.

채취시기별 방사무늬김(Porphyra yezoensis)의 식이섬유, 아미노산 및 지방산 함량 변화 (Seasonal Variation in the Dietary Fiber, Amino Acid and Fatty Acid Contents of Porphyra yezoensis)

  • Porphyra yezoensis is potentially an excellent source of dietary fiber, amino acids, and eicosapentaenoic acid (EPA) because this red seaweed is available in large quantities and is rich in polysaccharides, proteins, and n-3 fatty acids. This study determined the insoluble dietary fiber (IDF), soluble dietary fiber (SDF), neutral detergent fiber (NDF), acid detergent fiber (ADF), amino acid, and fatty acid contents of P. yezoensis harvested monthly from November 2011 to March 2012. The total dietary fiber (TDF) and IDF contents ranged from 27.2-34.9% and 18.5-26.9%, respectively, and were greater in March than November. The SDF content ranged from 4.9-8.4% and did not differ significantly during growth. Galactose and 3,6-anhydro galactose were the major sugars in IDF and SDF. The higher levels of galactose and 3,6-anhydro galactose in IDF might be due to associated porphyran-type polysaccharides. Mannose and xylose were also major sugars in IDF. The total amino acid contents decreased gradually from November to March. The total amino acid composition of Porphyra was dominated by alanine, glutamic acid, arginine, and aspartic acid. No significant changes in the fatty acid profile were observed throughout the study period. The dominant fatty acid during all seasons was EPA, which comprised as much as 50% of the total fatty acid content.

시판 전통식 된장의 품질평가 (Quality Assessment of Commercial Doenjang Prepared by Traditional Method)

  • To standardize quality characteristics of commercial doenjang prepared by traditional method, general components, organic acids, amino acids and fatty acid composition were investigated. Moisture, crude protein, crude lipid, titratable acidity (expressed as milliliter of 0.1 N NaOH), acid value and salinity were 54.7%, 13.8%, 8.0%, 14.4mL, 45.7mg/g and 11.8%, respectively. The content of amino type nitrogen was 345.3mg% (w/w) in the range of 207.6 to 443.5mg% being 1.1~2.2-fold for all samples. Average value of Hunter color for L(lightness), a(redness) and b(yellowness) were 37.4, +9.7 and +21.3, respectively. Browning index value(expressed as optical density at 425 nm) of water-soluble compound(2.58) was above 7.1 times higher than that of the water-insoluble compound(0.38). Total contents of free and total amino acid were 3.81 and 9.72%, respectively, and then the former(1.1~3.7 times) was more different in all samples tested than that of the latter(1.1~2.0 times). Among amino acids, glutamic acid was abundant and its content of free and total amino acids was 0.57 and 1.24%, respectively. Of organic acids, lactic acid was the most abundant being 0.34%(average) and ranged from 0.15 to 0.86%. Linoleic acid(52.17%, as area%) was the highest content of the total fatty acids. Unsaturated and polyunsaturated fatty acids were 83.89 and 61.189%, respectively. Polyunsaturated fatty acid was markedly different among all samples and ranged from 51.52 to 64.91%.

  • ABA type block copolymers composed of poly($\gamma-benzyl$ L-glutamate) (PBLG) as the A component and poly (ethylene glycol) as the B component were obtained by polymerization of $\gamma-benzyl$ L-gletamate N -carboxyanhydride, initiated by amino groups at both ends of poly(ethylene glycol) . From circular dichroism measurements in ethylene dichloride solution as well as from infrared spectTa measurements in solid state, it was found that the polypep- tide block exists in the a-helical conformation, as in PBLG homopolymer. $Poly-N^5$ (3-hydroxypropyl glutamine) (PHPG)/poly(ethylene glycol)block copolymer hydrogel was obtained by the treatment of PBLG/PBG block copolymer with the mixture of 3-ammine-1-propanol and diamlnooctane. The water content of PHPG/PEG block copolymer hydrogel was about 80wt% when the concentration of crosslinking agent was below 5 mole % per polymer.

  • PSS hetero 돼지를 이용하여 PSS와 관련된 유전자를 cloning 하여 유전자 구조를 분석하고 세포내의 유전자의 존재를 확인하여 PSS 돼지의 유전양식을 밝히고자 실시되었고, 그 결과를 요약하면 다음과 같다. 615번 amino acid가 N과 n에서 각각 arginine과 cysteine으로 존재함을 확인할 수 있었다. 그리고 6번 염색체(PSS 관련유전자)로부터 우성유전자 N과 열성유전자 n이 각각 유전자 좌위(locus)에 대립 유전자(alleles)로 존재함을 확인할 수 있었으며, 이러한 alleles로 존재함으로써 각각의 유전자가 나뉘어져 유전됨을 알 수 있었다.

실리카 나노입자 표면에 결합된 아미노기와 Glycidyl Methacrylate의 반응에 관한 분광학적 연구 (Spectroscopic Studies on the Reaction between Amino Groups on Silica Nanoparticle Surface and Glycidyl Methacrylate)

  • 본 연구에서는 실리카 나노입자를 dipodal 형태의 bis[3-(trimethoxysilyl)propyl]amine(BTMA) 실란 커플링제로 실리카 표면을 개질한 후, glycidyl methacrylate(GMA)로 표면 처리를 하여 실리카에 결합된 BTMA의 N-H기와 GMA의 epoxide기의 개환 반응에 의하여 실리카 표면에 중합용 methacrylate기를 도입하는 연구를 수행하였다. 반응시간, 반응온도 및 투입하는 GMA의 농도 변화가 BTMA의 N-H기와 GMA의 epoxide기 사이의 반응에 미치는 영향을 Fourier transform infrared spectroscopy(FTIR), elemental analysis(EA) 및 고체상태 $^{13}C$ cross-polarization magic angle spinning(CP/MAS), nuclear magnetic resonance spectroscopy(NMR)법을 사용하여 분석하였다. BTMA로 개질된 실리카를 GMA로 처리하면 실리카 입자에 결합되어 있는 BTMA의 N-H기와 GMA의 epoxide기가 열리면서 상호 반응이 일어났으며, 실험한 조건에서는 반응시간, 반응온도 및 투입하는 GMA 농도가 증가할수록 실리카 표면에 도입되는 methacrylate기가 증가함을 확인하였다.

ErmSF에서 특이적으로 발견되는 N-terminal end region에 존재하는 1-25번째 아미노산을 함유하는 peptide segment의 효소 활성에서의 역할 (Functional Role of Peptide Segment Containing 1-25 Amino Acids in N-terminal End Region of ErmSF)

  • ERM protein 은 23S rRNA의 $A_{2058}$에 dimethylation시킴으로써 $MLS_B$계 항생제의 부착을 저해하여 항생제의 활성을 억제하는 내생인자 단백질이다. ERM 단백질의 하나인 ErmSF의 N-말단부위(N-termimal end region, NTER)에 존재하는 1-25번째 아미노산을 함유하는 펩타이드의 활성에서의 역할을 알아보기 위해 이률 제거한 변이 단백질을 표현하는 유전자를 클로닝하고 대장균에서 수용성 단백질로 12.65 mg/L culture의 수율로 대량생산하였다. 이렇게 대량생산된 단백질의 활성을 in vivo와 in vitro에서 확인하였다. 그 결과 in vitro에서 야생형(wild type)의 단백질에 비해 15%의 활성이 감소한 것을 확인하였고 이는 제거된 펩타이드가 기질과 상호작용하여 효소의 활성에 영향을 미친다는 것을 시사하고 있다. 이렇게 감소된 효소의 활성은 생체 내(in vivo) 활성에도 적용되어 처음에는 변이 단백질을 함유하는 세포가 항생제의 작용에 의하여 성장억제를 받지만 시간의 경과와 함께 내성을 회복하여 밤샘 배양하였을 경우는 야생형 단백질을 함유한 세포와 동일한 내성 즉 항생제에 의한 성장억제지역(inhibition zone)을 전혀 나타내지 않는 것으로 밝혀졌다.

버섯첨가된장의 맛성분 조성 (The Taste Components Composition in Various Mushrooms-Added Korean Soybean Paste (Doenjang))

  • The purpose of this study is to evaluate the sensory characteristics and the taste components of Korean soybean paste (doenjang) with mushrooms such as Lentinus edodes, Ganoderma lucidum and Phellinus Lentinus. edodes doenjang turned out to have a good taste, odor and color, but Ganoderma lucidum and Phellinus liteus doenjang were worse than control doenjang in the taste, odor and color as well as in sensory evaluation. The contents of amino type nitrogen were higher in Lentinus edodes and Phellinus liteus doenjang than in control doenjang. Of organic acids, succinic acid was the most abundant and Ganoderma lucidum doenjang had a little higher acetic, butyric and propionic acid. Fructose and glucose were detected as free sugar of mushroom doenjang, of which glucose was considerably contained in Ganoderma lucidum doenjang, Total contents of free amino acid were 2.247 ∼ 2.833mg/100g in doenjangs added mushrooms. Glutamic acid and alanine were dominant in mushroom doenjangs.
