On-Off Type Mechanofluorochromism of a Novel Fluorescent Amorphous Molecular Material, N-{4-[Bis(4-methylphenyl)amino]benzylidene}aniline

  • The title compound (BMBZA) was designed and synthesized as a novel fluorescent amorphous molecular material. BMBZA was found to exhibit solvatofluorochromism and to readily form an amorphous glass by cooling the melt on standing. In addition, the morphological change from crystalline state to amorphous one could be induced by mechanical grinding. Although fluorescence was scarcely observed for the crystalline sample of BMBZA, the grinding the sample was found to enhance the fluorescence emission, that is, BMBZA exhibited on-off type mechanofluorochromism.

  • 유류오염 토양에서 난분해성 물질인 PAH (polycyclic aromatic hydrocarbon)들을 잘 분해하는 균주 중 SOD (superoxide dismutase) 활성이 높은 균주인 Sphingomonas sp. KS 301의 SOD특성을 알아보기 위하여 Ammonium sulfate 침전, DEAE-Sepharose 크로마토그래피, Superose-12 겔 여과 크로마토그래피, Uno-Q1 이온교환 크로마토그래피를 이용하여 SOD 단백질을 정제하였다. Sphingomonas sp. KS 301은 DEAE-Sepharose 크로마토그래피로 분석한 결과, 기존의 알려진 세균들과는 달리 서로 다른 5가지의 SOD 활성을 가지고 있는 것으로 나타났으며 본 연구에서는 그중 SOD III를 부분 정제하였다. 정제한 SOD III는 Mn type 및 Fe type Escherichia coli SOD와 비교했을 때 비활성도(specific activity)가 5배로 높게 나타났다. SOD III의 분자량은 SDS-PAGE에서는 23 kDa으로 측정되었으며 Superose-12겔 여과 크로마토그래피 후 native 상태의 분자량은 71 kDa으로 정제한 SOD는 3개의 소단위체로 구성되어 있는 것으로 보여진다. 정제한SOD III의 최적 pH는 7.0 이었고 $20^{\circ}C$에서 최적의 활성을 보였다. 또한 SOD의 종류를 알 수 있는 억제물질 $NaN_{3},\;H_{2}O_{2},\;KCN$를 이용한 억제효과를 살펴보았더니 $NaN_{3}$에만 억제되어 Mn type의 SOD임을 알 수 있었다. 또한 이 효소의 아미노 말단의 아미노산 서열은 Psudomonase ovalis 및 Vibrio cholerae의 SOD와 가장 유사하였다.

MLSB 항생제 내성인자인 ErmSF의 N-terminal 38개 아미노산 제거가 항생제 내성 효소활성에 미치는 영향 (Effect of Truncation of 38 Amino Acids in N-terminal Region of ErmSF, a MLSB Antibiotic Resistance Factor Protein, on Enzymatic Activity)

  • ErmSF는 macrolide 항생물질인 tylosin을 생성하는 Streptomyces fradiae가 보유한 4개의 항생제 내성인자 단백질 중 하나로 23S rRNA의 $A_{2058}$에 dimethylation 시킴으로써 항생제가 부착되는 것을 막음으로써 그 내성을 일으킨다. ErmSF는 다른 Erm 단백질과는 달리 긴 N-terminal end region을 가지고 있어서 그 역할을 알아보기 위해 1-38번째의 아미노산을 제거한 결손변이 단백질을 고안하고 대장균에서 발현하여 그 활성을 in vivo와 in vitro에서 관찰하였다. 결손변이 단백질을 발현하는 대장균은 결손에 의한 활성저하에 기인하여 야생형 단백질을 발현하는 대장균에 비하여 항생제에 대한 내성이 손상된 것을 관찰하였다. 세포 외 in vitro에서의 활성은 야생형 ErmSF에 비하여 약 20%가 손상된 것으로 나타났다. 이렇게 관찰된 활성의 저하는 결손 변이에 의한 활성화 부위에서 일어난 결손에 의한 것이라기 보다는 기질의 부착 또는 생성물의 효소에서의 이탈 과정이 손상되어서 나타나는 것으로 사료된다.

The Differences in Chemical Composition, Physical Quality Traits and Nutritional Values of Horse Meat as Affected by Various Retail Cut Types

  • The effects of retail cut type on chemical, quality and nutritional characteristics of horse meat were studied. Jeju female breed horses (n = 9) at 32-mo-old were slaughtered and the carcasses at 24 h post-mortem were fabricated into 10 retail cuts including: tender-loin, loin, strip-loin, shoulder-chuck-roll, shoulder-clod, top-round, outside-round, brisket, short-plate-brisket, and shank. The results revealed that all of parameters (chemical, meat quality and nutritional composition) examined significantly (p<0.05) differed between the cuts. The chemical composition range (minimum to maximum) of cuts was found as such: moisture 65.06% to 71.69%; protein 19.07% to 21.28%; collagen 1.40% to 2.45%; fat 2.56% to 12.14% and cholesterol 55.76 to 79.50 mg/100 g. Shoulder-chuck-roll had the highest pH and water-holding capacity, while top-round had the highest cooking loss. Shear force ranged between the cuts from $2.80kg/cm^2$ to $4.98kg/cm^2$. The Cu, Fe, and Zn contents ranged between the cuts from 1.52 mg/kg to 2.75 mg/kg, 21.25 mg/kg to 30.85 mg/kg, and 16.51 mg/kg to 40.42 mg/kg, respectively. Additionally, most of the cuts studied showed favorable polyunsaturated fatty acid/saturated fatty acid, n-3/n-6 and essential amino acid/non-essential amino acid ratios.

뼈 연화 고등어(Scomber japonicus) 가공품의 식품성분 특성 (Food Component Characteristics of Bone-softened Mackerel Scomber japonicus Product)

  • The present study investigated the food component characteristics of a bone-softened mackerel Scomber japonicus product (FC-M). The proximate composition of the FC-M was 57.4% moisture, 25.2% crude protein, 14.1% crude lipid, and 2.7% ash. The energy in the FC-M was 241.5 kcal/100 g, which corresponds to 8.9-16.1% of the daily energy intake recommended for Koreans ${\geq}6$ years of age. Regardless of type of taste, there were no significant differences in taste intensity when the FC-M was compared to a control (canned mackerel) but the odor intensity was significantly higher. Although there were no significant differences in hardness, the Hunter color values of redness and yellowness were significantly lower than those of the control. The sensory score for the FC-M was significantly superior to that of the control. The total amino acid content in the FC-M was 24.12 g/100 g and the major amino acids were aspartic acid, glutamic acid, leucine, and histidine. The major fatty acids in the FC-M were 16:0, 18:1n-9, 20:5n-3, and 22:6n-3 and the FC-M was a good source of minerals.

Poliovirus Sabin 1 as a Live Vaccine Vector: Expression of HIV-1 p24 Core Protein

  • The poliovirus Sabin 1 strain has features that make it a particularly attractive live recombinant mucosal vaccine vehicle. Sabin 1 cDNA was manipulated to have multiple cloning sites and a viral specific 3C-protease cutting site at the N-terminal end of the polyprotein. The gene for the N-terminal 169 amino acids of the HIV-1 p24 was cloned into the multiple cloning site of the manipulated Sabin cDNA. A recombinant progeny virus was produced from HeLa cells when it was transfected with the RNA synthesized from the p24-Sabin chimeric cDNA. The recombinant progeny virus expresses substantial amounts of the HIV-1 p24 protein, which was clearly detected in the infected cell lysates and culture supernatants in Western blot experiments with rabbit anti-p24 serum and AIDS patients' sera. Differing from the Mahoney strain, the recombinant Sabin 1 poliovirus maintained the foreign gene stably during the subsequent passages. Replication capacity was about 1 to 1.5 log lower than that of the wild-type Sabin 1. Other physicochemical stability characteristics of the recombinant virus were similar to that of the wild-type Sabin 1. These results suggest that the manipulated Sabin 1 poliovirus can be used as a live viral vaccine vector for the development of mucosal vaccines.

Influence of Diet Induced Changes in Rumen Microbial Characteristics on Gas Production Kinetics of Straw Substrates In vitro

  • The effect of diets varying in level and source of nitrogen (N) and fermentable organic matter on dynamic characteristics of microbial populations in rumen liquor and their impact on substrate fermentation in vitro was studied. The diets tested were straw alone, straw+concentrate mixture and straw+urea molasses mineral block (UMMB) lick. The same diets were taken as substrates and tested on each inoculum collected from the diets. Diet had no effect on the amino acid (AA) composition of either bacteria or protozoa. Differences among the diets in intake, source of N and OM affected bacterial and protozoal characteristics in the rumen. Upper asymptote of gas production (Y$\alpha$) had a higher correlation with bacterial pool size and production rate than with protozoal pool size and production rate. Among the parameters of the gas production model, Y$\alpha$ and lag time in total gas has showed significant (p<0.01) correlation with bacterial characteristics. Though the rate constant of gas production significantly differed (p<0.01) between diet and type of straw, it was least influenced by the microbial characteristics. The regression coefficient of diet and type of straw for Y$\alpha$ indicated that the effect of diet on Y$\alpha$ was threefold higher than that of the straw. As microbial characteristics showed higher correlation with Y$\alpha$, and diet had more influence on the microbial characteristics, gas production on a straw diet could be used effectively to understand the microbial characteristics.

  • Crystal structure of two inclusion compounds Cd(pn)Ni(CN)4·0.25G (G=toluene and p-xylene, pn=1,2-diaminopropane) have been determined from single-crystal X-ray diffraction data; toluene clathrate: monoclinic P21/n, a=13.838(6), b=26.893(5), c= 7.543(5) Å, γ=90.92(3)°, Z=4, R=0.0616; p-xylene clathrate: monoclinic P21/n, a=13.895(2), b=26.900(3), c=7.613(1) Å, γ=91.06(1)°, Z=4, R=0.0486. The host structures determined for toluene- and pxylene-guest clathrates are substantially identical to the U-type structures observed for the straight chain aliphatic-guest clathrates. However, the alternating arrangement of occupied channels with the guest molecules and vacant channels appears in the host structure. The non-centrosymmetric toluene molecules are distributed about the inversion center to give an image like p-xylene molecule. The guests, toluene and p-xylene, prefer the U-type channel, favoring the interaction between the π-electrons of the aromatic ring and the pn-amino groups to hold the aromatic ring vertical to the cyanometallate meshes.

  • Citrin deficiency는 염색체 7q21.3의 SLC25A13 유전자 돌연변이에 의해 발생하는 상염색체 열성질환으로 neonatal-onset type II citrullinemia (CTLN2) 및 adult-onset CTLN2로 분류된다. 생후 16일된 여아가 신생아 대사 검사에서 citrullinemia (CTLN)가 의심되었고 혈중 아미노산 분석에서 CTLN 및 고암모니아혈증을 보였고, SLC25A13 유전자 분석 결과 c.221C>T in exon4 and c.1645C in exon16 (p.[Ser74Phe]+[Gln549X]) 의 새로운 돌연변이가 발견되어 neonatal-onset CTLN2로 진단되었다. 저자들은 신생아 대사 검사에서 citrullinemia를 보이는 영아들에서 확진을 위한 유전자 검사 및 혈중 아미노산 분석이 필요하다고 생각한다.

세미(洗米)에 의한 미곡(米穀)의 영양손실(營養損失)에 관한 연구(硏究) (Effect of Washing on the Loss of Nutrients in Rice)

  • 세미(洗米)로 인한 미곡(米穀)의 주요(主要) 영양성분 손실을 품종별(品種別) 도정도별(搗精度別)로 분석한 결과는 다음과 같았다. 1) 세미(洗米)에 의한 각영양소 평균손실율의 범위는 총고형분 $1{\sim}2%$, 단백질 $5{\sim}7%$, 칼슘 $18{\sim}26%$, 철분 $19{\sim}47%$, $vitamin\;B_1\;22{\sim}40%$, $B_2\;11{\sim}24%$ 그리고 niacin이 $36{\sim}45%$이었다. 2) 일반미인 아끼바레(Japonica type)보다 통일미(Indica type)에서 vitamin 성분을 제외한 총고형분, 단백질, 칼슘 및 철분의 손실율은 더 높았다. 3) 백미(白米)의 도정도가 높을수록 총고형분, 가용성무질소물 및 vitamin의 손실율은 높아지는 경향이었다. 그러나 단백질, 칼슘 및 철분들의 손실율은 도정도에 따른 일정한 경향은 없었으나, 손실되는 절대량은 도정도가 낮은 5분도미에서 더 많았다. 4) 세미(洗米)중 구성 amino acid의 손실율은 Iysine이 가장 높았고, 다음 histidine, glycine 및 alanine등이었다.
