Enzymatic transesterification for the synthesis of amino acid-sugar conjugates

  • Among the tested ten enzymes, Optimase M-440 showed the highest activity in transesterification of N-t-Boc-L-Phe-OTFE with D-glucose. Monosaccharides and their derivatives acted as good acyl acceptors in the Optimase M -440 catalyzed transesterification of N-t-Boc-L-Phe-OTFE. Optimase M-440 showed a preferable catalytic activity on the primary hydroxyl group of saccharides and a good regioselectivity. Optimase M-440 showed the highest activity in pyricline among the tested solvents. As acyl donors, trifluoroethyl esters of amino acids showed a high reactivity in transesterification. Optimase M-440 showed a broad substrate specificity towards amin 。 acid esters and saccharides.

Comparison of Chemical Compositions of Houttuynia cordata Thunb Cultivated from Different Local Area (재배지역이 다른 어성초의 부위별 화학성분 비교)

  • Proximate composition , volatile compounds, free amino acids, fatty acids and inorganic compounds in leaf, stem and root of Houttyunia cordata Thunb cultivated from two different area, Bosung and Sunchon , were analyzed. Each part of Houttuynia cordata Thunb from both local area showed moisture contents of 80-84% and crude ash contents of 2.1 ∼2.8%. Crude fat and protein contents were less than 3 % showing slightly higher contents in leaf than in both stem and root . Twenty six volatile compunds were identified from the parts of the plant, the volatile contents were high in the oder of leaf, root and stem. Major volatile compounds were mostly derivatives of decanoic acid ; decanoic acid, 20 tridecanoie, decanal and dodecanoic acid. Of free amino acids in leaf asparagine was the highest, while in stem hydroxyproline, proline and arginine were the major free amino acids. Linoleic acid was the highest in the stem and root, and linolenic acid was highest in leaf. the major minerals of all parts were K, Ca. Mg, P, Fe, Zn and Cu, showing highest with K.

Synthesis, Fastness and Spectral Properties of Some New Azo Pyrazole and Pyrazolotriazole Derivatives (Pyrazole과 Pyrazolotriazole 유도체의 합성 및 특성 연구)

  • Coupling of 5-amino-1,3-diaryl-pyrazoles 1a-c with diazonium salts of different aryl amines gave a series of novel 1,3-diaryl-5-amino-4-arylazopyrazoles 3a-l. Such compounds could be also obtained by reaction of 5-amino-1,3-diaryl-4-nitroso- 1H-pyrazoles 2a-c with different aryl amines in alkaline medium. Oxidation of azo derivatives 3a-l with cupric acetate, in dimethyl formamide and stream of air, gave 2,4,6-triaryl-2,4-dihydropyrazolo [4,3-d]-1,2,3-triazoles 4a-l. and the fluorescence properties of the cyclic triazoles were studied. Diazotization of 5-amino-1,3-diaryl-1H-pyrazoles 1a-c by sodium nitrite in ortho-phosphoric acid followed by coupling with some aryl amines gave o-aminoazo compounds 5a-f. Cyclisation of compounds 5a-f in pyridine and cupric acetate gave the corresponding triazoles 6a-f. The coupling of compounds 6a-f with different aryl diazonium salts gave compounds 7a-j. The synthesized dyes were applied to polyesters as disperse dyes and the fastness properties were evaluated.

Synthesis and pesticidal activity of ricinine derivatives (Ricinine 유도체(誘導體)의 합성(合成) 및 농약활성(農藥活性))

  • Chemical derivative synthesis of ricinine, an active compound of Ricinus communis which showed high mortality against brown planthopper (Nilaparvata lugens), was performed to improve its pesticidal activity and the toxicity of 12 synthetic derivatives against major insect pests and phytopathogenic fungi were examined. Carbamate derivatives of ricinine could be synthesized from the precursor of ricinine, chloronorricinine and norricinine, whereas the derivatives were not synthesized from chlororicinic acid and ricinic acid having ketone group of pyridine ring. In organophosphates, reaction with oxon type of phosphate gave better yield than thiono type. Among the organophosphate derivatives of ricinine, thiono type of derivative structure gave $96.3%{\sim}100%$ mortality of the brown planthopper and the two-spotted spider mite (Tetranychus urticae) at 500 ${\mu}g/ml$ level. On the other hand, carbamate derivatives did not show insecticidal activity. In the fungicidal activity of ricinine derivatives, the derivative having amino radical at the 2 position of ricinine gave 85 to 100% of mycelium growth inhibition effect against ten major plant pathogens at the 200 ${\mu}g/ml$ level. In particular, the control value of the derivative on the rice blast (Pyricularia grisea) and barley powdery mildew (Erysiphe graminis) at the 250 ${\mu}g/ml$ level in vivo under greenhouse conditions was 92% and 96%, respectively.

  • Pyrrolidone-anilide derivatives from L-glutamic acid and anilines were synthesized as follows: The products were identified by elementary analysis, IR, NMR and Mass spectra with (R)-2-pyrrolidone-5-carbox-anilide, (R)-2-pyrrolidone-5-cabox-p-chloroanilide, (R)-2-pyrrolidone-5-carbox-o-toluidide, (R)-2-pyrrolidone-5-carbox-m-toluidide and (R)-2-pyrrolidone-5-carbox-p-toluidide. The products were testes for their phytotoxicity on the germination and the seedling growth of radish and rice plants. Among them, (R)-2-pyrrolidone-5-carbox-anilide and (R)-2-pyrrolidone-5-carbox-p-chloroanilide derivatives were strongly inhibitory especially on the germination and the seedling growth of radish seeds. All the compounds also showed an inhibitory activity upon the germination of rice seeds. Additionally, the inhibiting rate of radish growth differs according to the isomeric position(ortho, meta and para) of the methyl group; (R)-2-pyrrolidone-5-carbox-m-toluidide derivative was more effective than both (R)-2-pyrrolidone-5-carbox-o-toluidide and (R)-2-pyrrolidone-5-carbox-p-toluidide derivatives.

Analysis on the Components of the Angelica dahurica Root (구릿대(Angelica dahurica) 뿌리의 성분 분석)

  • This study was carried out to analyze the components of the Angelica dahurica root for the studies of the physiological function. General components of A. dahurica root were 71.7% moisture, 10.6% carbohydrate, 9.3% crude protein, 6.6% crude ash and 1.9% crude fat. The content of reducing sugar was 1,850 mg/100 g. The total amount of free sugar was 80 mg/100 g, in which 19.3 mg/100 g fructose, 27.8 mg/100 g glucose, 28.4 mg/100 g sucrose and 4.5 mg/100 g maltose were present. In the results of mineral analysis, the content of K was the highest(2,145.03 mg/100 g) followed by 286.35 mg/100 g Mg and 145.23 mg/100 g Ca. The total amount of hydrolyzed amino acid was 71.68 mg/100 g, in which 20.98 mg/100 g essential amino acid and 50.70 mg/100 g of non-essential amino acid were present. Among them, proline(11.74 mg/100 g) was the highest. Total free amino acids were contained 17.04 mg/100 g, in which 6.67 mg/100 g of essential amino acids and 10.37 mg/100 g of non-essential amino acids were present. Among them alanine(5.96 mg/100 g) was the highest. Total content of amino acid derivatives was 3.37 mg/100 g.

Pathways Regulating the pbgP Operon and Colistin Resistance in Klebsiella pneumoniae Strains

  • In this study, we investigated colistin resistance mechanisms associated with the regulation of the pbgP operon in Klebsiella pneumoniae, using four isogenic pairs of colistin-susceptible strains and their colistin-resistant derivatives and two colistin-resistant clinical isolates. Amino acid sequence alterations of PhoPQ, PmrAB, and MgrB were investigated, and mRNA expression levels of phoQ, pmrB, pmrD, and pbgP were measured using quantitative real-time PCR. The phoQ and pmrB genes were deleted from two colistin-resistant derivatives, 134R and 063R. We found that phoQ, pmrD, and pbgP were significantly upregulated in all colistin-resistant derivatives. However, pmrB was significantly upregulated in only two colistin-resistant derivatives and one clinical strain. pmrB was not overexpressed in the other strains. The minimum inhibitory concentration of colistin was drastically lower in both phoQ- and pmrB-deleted mutants from a colistin-resistant derivative (134R) that was overexpressing phoQ and pmrB. However, colistin susceptibility was restored only in a phoQ-deleted mutant from a colistin-resistant derivative (063R) without overexpression of pmrB. In conclusion, two different regulations of the pbgP operon may associate with the development of colistinresisant K. pneumoniae.

Polyethyleneimine Derivative for Nucleic Acid Model

  • Water-soluble polyethyleneimine (PE) derivatives containing nucleic acid bases and hydrophilic amino acids such as homoserine (Hse) and serine were prepared by the activated ester method as nucleic acid models. From spectroscopic measurements, the polymers were found to interact with DNA accompanied by an induction of conformational change. Hypochromicity in UV spectra indicated that a stable polymer complex was formed between poly (A) with PEI­Hse-Ura by complementary hydrogen bonding with equimolar nucleic base units (adenine:uracil=1:1). The induced conformation of DNA by the interaction with the polymer containing uracil and homoserine (PEI-Hse-Ura) was concluded to be a super triple helical structure. The formation of the polymer complex, DNA: PEI-Hse-Ura, was found to be affected by the presence of metal ions such as $Ca^{2+}\;and\;Cu^{2+}$.

Effect of Thermal Pretreatment on the Functional Constituents of Waxy Corn (Zea mays L.)

  • Quantitative changes in functional constituents of waxy corn (Zea mays L.) by 5 different thermal pretreatments, including roasting, steaming, microwave, puffing, and extruding, were determined and compared with those of the raw waxy corn. There were no significant differences in fatty acid compositions among the corn treated with 5 thermal treatments. Levels of $\alpha$- and $\gamma$-tocopherols, soluble amino acids, and phytosterols decreased by thermal treatments, while those of p-coumaric and ferulic acids considerably increased by thermal treatments. In particular, the contents of tocopherols and phytosterols, and souble amino acid decreased significant in the steaming and puffing processes, respectively, whereas those of 2 free cinnamic acids increased significantly in the steaming and puffing processes. The extruding process showed smaller quantitative changes in tocopherols, phytosterols, and hydroxycinnamic acid derivatives compared to other heat pretreatments. These results suggest that the extruding process have a positive effect on valuable phytochemicals in waxy corn.

  • Liquid chromatographic enantiomer separation of N-tert-butoxycarbonyl (BOC) $\alpha$-amino acids and their ethyl esters was performed on covalently immobilized chiral column (Chiralpak IC) derived from polysaccharide derivative. The solvent versatility of the covalently immobilized Chiralpak IC in enantiomer separation of N-BOC $\alpha$-amino acid ethyl ester derivatives was shown and the chromatographic parameters of their enantioselectivities and resolution factors were greatly influenced by the nature of the mobile phase. Also the effect on the dissolving solvent for these analytes on the enantiomer separation using the same mobile phase and the examples of preparative enantiomer separation on analytical column were shown.
