Amino Acid Concentrations in the Blastocoelic Fluid of In Vitro-Produced Bovine Blastocysts (체외생산된 소 배반포강 내의 아미노산 농도)

  • Concentrations of free amino acids in the BF of IVP bovine BL and HBL were examined in this study. The embryos derived from IVF oocytes were cultured in a SOFM containing BSA, EAA and NEAA. BF was aspirated from BL (180 h of age after insemination) and HBL (216 h of age after insemination), and introduced into drops of SOFM (30$\mu$l/drop) containing PVA through micromanipulation. The medium containing BF was then subjected to measurement of 20 amino acids by an automatic amino acid analyzer. The concentrations of isoleucine, leucine and methionine were higher (p〈0.05) in the BF from HBL than from BL, and no difference was found in aspartate or glutamate concentrations between BL and HBL, while threonine, alanine (p〈0.01) and the rest of the amino acids (p〈0.001) were significantly higher in the BF from HBL than from BL. Cystine was not found in either BL or HBL. A high concentration of glutamine was found in the BF from both BL and HBL, although it was not added to the culture medium. These results indicate that bovine BF contains several EAA (methionine in BL and isoleucine, leucine and methionine in HBL) and NEAA (alanine, glutamate, glycine, proline, serine and aspartate in BL, and glutamate and aspartate in HBL), and there is significant differences in the amino acid concentration in the BF between BL and HBL derived by WP.

Lysinoalanine in Protein Recovered from Frozen Belanger's Croaker, Johnius grypotus, Using Alkaline Processing (알칼리 처리하여 회수한 냉동깡치 어육 단백질의 Lysinoalanine 함량)

  • The formation of lysinoalanine (LAL) in protein recovered from the belanger's croaker, Johnius grypotus, using a pH shifting process was measured by amino acid analysis. The LAL peak was detected at 49.24 min, between phenylalanine and histidine peaks in the amino acid analyzer. LAL was not detected in the fish muscle or in protein recovered using the alkaline pH shifting process. LAL was not formed in protein recovered after storage for up to 9 hrs at pH 11, but was detected in the soluble protein fraction at pH 11, followed heating at $90\;^{\circ}C$. The myosin heavy chain decreased with storage time at pH 11. The results suggest that the alkaline shifting process for recovering fish muscle protein is safe, and that no LAL forms.

Analysis of Theanine Contents in Commercial Green Tea (시판 녹차로부터 Theanine 함량의 분석)

  • The theanine contents of commercial green teas manufactured by the different harves times(1st tea, 2nd tea and 3rd tea) and processes(steamed and parched teas) were determined by amino acid analyzer. There were remarkable differences in the theanine contents among teas of the different harvest times. The theanine content in 1st steamed green tea was 2235 mg% and one in 1st parched green tea was 2106 mg%. The theanine contents in 2nd, 3rd tea were gradually decreased. There were slight differences in the theanine contents among teas of the different process.

  • To separate and quantitate antitumor protein-bound polysaccharides of Coriolus versicolor, the constituents were obtained from the submerged culture of the mycelia(C) and from the extract of the carpophores of the wild fungus(N). The polysaccharides were degraded by methanolysis. The neutral monosaccharides were separated and quantitated by HPLC using microbondapak carbohydrate analysis column, refractive index detection and water-acetonitrile acetic acid elution. The analyses of these constituents by HPLC showed that the natural constituent(N) consisted of glucose, xylose and mannose, the average amount being 96.44, 2.16 and 1.73%, respectively. The fermentation constituent(C) consisted of mannose, glucose, xylose and galactose, the average amount being 61.30, 14.00, 12.95, and 11.75%, respectively. The analyses of these constituents by an amino acid analyzer showed that both C and N contained 17 amino acids.

  • Taurine is a type of sulfur-containing amino acid having a sulfate functional group, that is biosynthesized from cysteine. It is mainly distributed in high concentrations in animal tissues and is known to have various effects such as osmotic pressure control, calcium control, anti-inflammatory, antioxidant, and hepatocellular protection. Also, taurine deficiency causes a variety of symptoms, including visual impairment. In particular, in the case of cats, taurine is not biosynthesized and must be supplied through food, so it is classified as an essential amino acid. In this study, an analysis method using mass spectrometry was developed instead of the commonly used derivatization method to quickly, environmentally, and precisely analyze taurine in various animal feeds. The developed analytical method showed good linearity (R2 > 0.99), accuracy (81.97-105.78%), and precision (0.07-12.37%). In addition, the developed method was further verified through quantitative comparison with the derivatization method. This developed method was used in the determination of taurine in 20 animal feed samples obtained from South Korea. The levels of taurine found ranged from 81.53 to 6,743.53 mg/kg. The developed analysis method will be used for the detection and quantification of taurine in domestic feed.

  • A potential biocontrol bacterium, YB-70 was isolated from a rhizosphere in suppressive soil and identified as a strain of Bacillus subtilis. In several biochemical and in vitro antibiosis tests on Fusarium solani with the culture filterates from B. subtilis YB-70, we found that antifungal mechanism of B. subtilis YB-70 was mediated by antibiotic substances produced from the bacterium. These antifungal substances were appeared to be hear-resistant, micromolecular, and ethy alcohol soluble. Antifungal agents produced by B. subtilis YB-70 showed strong inhibified against root-rotting fungi F. solani in in vivo pot test. An antifungal substance. YBS-1s, was purified from the culture broth of B. subtilis YB-70 by isoelectronic precipitation, silica gel column chromatography and Sephadex LH-20 column chromatography analysis by Fab-MASS, $^{1}$H-NMR, $^{13}$C-NMR, DEPT, and amino acid analyzer revealed that the YBS-1A was a peptide antibiotics of iturin class containing seven amino acids from five different groups, and the other(YBS-1B) was an analogue of iturin group composed of 11 amino acids with larher molecular weight of about 1, 500 dalton, which was lager than that of iturin A.

Varietal Analysis and Quantification of Amino Acid in Mulberry Fruits (뽕나무 계통별 오디의 아미노산 함량 분석)

  • To reveal the possibility as a source of functional food and to increase the usability of mulberry fruits, amino acid analysis with mulberry fruits was carried out with L-8800 Amino acid auto analyzer. We analyzed 16 amino acid components including 6 essential amino acids. According to the results, the mean content of total amino acid was 4,640.8 mg/100 g DW. 'Sabangso(II)' showed the highest total amino acid content (6,919.3 mg/100 g DW) among the fifty accessions tested, whereas 'Gukkwang' showed the lowest content (3,389.1 mg/100 g DW). The distribution rate of amino acid components decreased in the order of Glu ($20.1\%$) > Gly ($12.7\%$) > Asp ($12.5\%$) > Thr ($8.2\%$) > Ala ($7.9\%$) > Arg ($7.5\%$) > Pro ($6.1\%$) > Ser ($6.1\%$) > Val ($5.7\%$) > Leu ($5.3\%$) > Ile ($2.7\%$) > His ($2.5\%$) > Met ($0.9\%$) > Tyr ($0.8\%$) > Lys ($0.7\%$) > Cys ($0.3\%$). 'Sabangso(II)' showed the highest content in 11 amino acid components, which were Glu (1,293.2 mg/100 g), Ser (390.2 mg/100 g), Thr (564.4 mg/100 g), His (175.3 mg/100 g), Arg (590.4 mg/100 g), Lys (53.5 mg/100 g), Gly (589.4 mg/100 g), Ala (499.3 mg/100 g), Leu (481.9 mg/100 g), Ile (240.9 mg/100 g), and Val (407.6 mg/100 g). Tyr and Cys were the highest in 'Jangsosang' (70.8 mg/100 g, 19.3 mg/100 g, respectively), and Asp, Pro, and Met were the highest in 'Jasan(I)', 'Hwansipchosaeng' and 'Palcheongsipyung(I)' (1,098.8 mg/100 g, 364.4 mg/100 g, 68.8 mg/100 g, respectively).

  • Screening was performed to isolate biopolymer-producing microorganisms from natural sources. The bacteriological characteristics of this strain and physicochemical properties of the biopolymer produced were investigated. The bacterial strain was identified as a Bacillus coagulans. Crude biopolymer treated with ethanol and actone was purified to fraction I and II by ion exchange chromatography and gel chromatography (Shephadex G-100 and Shepharose CL-2B). Analysis of chemical composition and various color reaction revealed that the polymer is composed of amino acids. It was confirmed that fraction II is a homopolymer of glutamic acid and fraction I is a homopolymer of lysine by analysis data of amino acid analyzer, GC and IR.

Purification and Characterization of an Antioxidant Protein from Fertilized Eggs

  • Free radicals may attack cells or tissue, leading to chronic diseases, and antioxidant consumption is potentially useful for removing free radicals. Egg proteins may be used as potential sources of antioxidant considering their ability of scavenging free radicals to apply for food or cosmetics industry. In this study, we obtained a natural antioxidant protein from fertilized eggs, which was a dietary supplement in some Asian countries. Meanwhile, antioxidant activities of these proteins were evaluated using different oxidation systems. With increasing incubation time, the antioxidant activity of these proteins increased during 15 d of incubation. The samples on day 15 were performed for isolation of antioxidant protein. The protein, named P4-1 (MW, 45 kDa), was isolated and purified by consecutive chromatographic methods. P4-1 contained 17 amino acids, which was determined by liquid chromatography-mass spectrometry and Amino Acid Analyzer. Moreover, the amino acid sequence was highly similar to that of ovalbumin. Differential scanning calorimetry showed that the denaturation temperature of P4-1 was $57.16^{\circ}C$. Furthermore, P4-1 suggested high oxygen radical-absorbance activity in ${\cdot}OH$ assays, and its antioxidant activity was stable at $30-50^{\circ}C$ in acidic and neutral pH. Thus, these results revealed that P4-1 may be a potential resource as a natural antioxidant.

  • Objectives : The aim of this study is to investigate that Sunghyangjeongki-San which has been frequently medicated in the early stage of stroke can protect against ischemic damage in rat brain Methods : Extracellular levels of amino acids(glutamate, aspartate, GABA, glycine, taurine, tyrosine, alanine), organic acids(pyruvate, lactate), and cerebral infarction volume were measured at the striatum of rats subjected to permanant focal cerebral ischemia induced by 2 hours of middle cerebral artery occiusion(MCAO). Rats were orally administered with Sunghyangjeongki-San at 30mins before MCAO and the microdialysate was collected by intracerebral microdialysis three times before MCAO and six times after MCAO at 20mins interval and analyzed by HPLC. After a microdialysis study, the brain was sliced and stained with cresyl violet buffer for the measurement of cerebral infarcted area and volume by image analyzer system Results : The concentrations of glutamate, aspartate, and tyrosine known as excitatory neurotransmitters were significantly decreased in Sunghyangjeongki-San group compared with control group, The concentrations of GABA, glycine, taurine and alanine known as inhibitory neurotransmitters were significantly increased in Sunghyangjeongki-San group compared with control group. The concentrations of pyruvate and lactate showed little significant change in Sunghyangjeongki-San group compared with control group. The measurement of cerebral infarcted area and volume by image analyzer system were significantly decreased in Sunghyangjeongki-San group compared with control group. Conclusions : Sunghyangjeongki-San can affect on protecting against cerebral ischemia by regulating extracellular levels of excitatory and inhibitory amino acid neurotransmitters and improve the conditions of the patients in the early stage of stroke.
