Biological Control on Rhizoctonia Blight of Turfgrasses in Golf Courses (골프장 잔디의 Rhizoctonia 마름병에 대한 생물학적 방제)

  • This study was undertaken to find a new formulation of soil amendment, and selection of antogonists and to effectively control brown and large patch of turfgrasses caused by Rhizoctoniz solani AG1-1 and AG 2-2. Fourteen inorgainc chemicals (1%, w/w) were added individually in vitro, and some chemicals showed suppressiveness to R. solani. Alum suppressed effectively mycelial growth of R. solani in the range of 17 to 77% as compared with control. The four chemicals such as Al2(SO4)3, alum, CaO, and NH4NO3 were finally selected. Out of three organic compounds, composted pine bark (CPB) showed prominent suppressive effect as compared with milled alfalfa and pine leaves. After inoculation of R. solani isolates AG-1 and AG2-2 on the turf seedlings, water soaked lesions and blight symptoms were developed on the whole seedlings. According to inhibition zone method, mycelial growth of the fungus were greatly suppressed by culture filterates of the antagonists, Gliocladium virens (Gl1-) and Pseudomonas sp. (P713). CPB soil amendment mixed with antagonists (1% w/w) controlled not only brown and large patch of turfgrasses, but also promote the good growth of the seedlings. In addition, the controlling effect was maintained more than 30 days. Especially, the controlling effect of two antagonists was similar to Cㅖㅠ soil amendment with the antagonists and also stimulated a favorable growth of the seedlings. Therefore, its is expected that continuous control of Rhizoctonia blight of turfgrasses can be obtained in field by subsequent applications of the antagonists.

Coloring Effect and Functionality of Rayon Based Cool Fabrics Treated by Various Mordant and Gallnut Extract (레이온계 여름용 직물에서 매염제 종류에 따른 오배자 염색의 색채발현 및 기능성에 관한 연구)

  • Gallnuts are known to exert various pharmaceutical properties that include anti-inflammatory, antimicrobial, antioxidant, and detoxifying effects. Gallnut extract is thought to be a particularly safe antimicrobial agent for textile application due to its natural origins. Hence, textiles were dyed with gallnut to develop multi-functional clothing material with no harmful effects. In this study, three kinds of cool touch woven fabrics, each comprised of 100% viscose rayon filament, viscose rayon filament/high absorption & quick dry (polyester) filament, and viscose rayon $filament/tencel^{(R)}$ spun yarn, were prepared and used as substrates for gallnut dyeing to especially develop functional cool touch textiles for summer clothing material. On the other hand, gallnut dyeing was conducted with mordanting with iron, lime or alum, which induced different colors on the dyed textiles. Dyed textiles were then investigated by SEM, FTIR, antibacterial test, and antioxidant test. Subsequently, gallnut dyed textiles indicated excellent antibacterial ability and deodorization activity regardless of mordant species. However, only gallnut dyed textiles mordanted with iron and alum showed significant antioxidant ability. In addition, the dyeing processes impair the cool touch feeling of the textiles by changing the micro structure of the textile surface.

Effect on Immune Cells of Proteoglycan Originating from Rhanella aquatilis (Rhanella aquatilis 유래 당단백질의 면역세포에 미치는 영향)

  • So as to evaluate its use an immune stimulator in humans, the toxicity and action against immune cells by an anti-yeast substance (AYS), a bacterial proteoglycan, were investigated. The AYS did not possess hemolytic activity with human red blood cells (hRBC). Nor did it exhibit cytotoxicity against human peripheral blood mononuclear cells (hPBMC). In addition, the AYS did not induce hPBMC proliferation, but it did agglutinate hPBMCs in vitro. Moreover, hPBMC induced inflammatory cytokines such as IL-6, IL-5, IFN-${\gamma}$ and TNF-${\alpha}$ with the AYS during culture. Compared with alum, the AYS as an adjuvant has an increased antibody production rate against bovine serum albumin (BSA) in mice.

Development of Livestock Manure Additives for Ammonia Reduction in High School Field Education (고등학교 현장 교육에서의 암모니아 저감용 축분첨가제 개발)

  • This study focused on high school laboratory research and the main purpose was to develop alternative additives for livestock waste and ammonia volatilization methods with high school students as participants and to provide information to business owners based on the results. Compared to the control groups, The bentonite and illite treatment groups had similar ammonia volatilization, pH, EC, and total nitrogen content. In particular, the alum and aluminum chloride mixed treatment group showed low pH and ammonia volatilization, and high EC and total nitrogen content for poultry litter. As a result, when focusing on high school laboratory research, the alum and aluminum chloride mixed agent treatment fulfilled its role as an alternative additive for ammonia reduction. In addition, this approach can be suggested as a method to solve difficulties in adapting to the field through a practical cooperative relationship with livestock farms.

Influence of barley grain treated with alkaline compounds or organic extracts on ex vivo site and extent of digestion of starch

  • Objective: Two ex vivo experiments were conducted to verify the effect of barley grain (Nusrat cultivar) treated with alkaline compounds (AC) including alum, ammonium, and sodium hydroxide or cation-exchanged organic extracts (OE) prepared from alfalfa hay, sugar beet pulp and Ulva Fasciata, on extent and digestion of starch. Methods: In the first study, the in vitro first order disappearance kinetic parameters of dry matter (DM), crude protein (CP) and starch were estimated using a non-linear model ($D_{(t)}=D_{(i)}{\cdot}e^{(-k_d{\cdot}time)}+I$, where: $D_{(t)}$ = potentially digestible residues at any time, $D_{(i)}$ = potentially digestible fraction at any time, $k_d$ = fractional rate constant of digestion (/h), I = indigestible fraction at any time). In the second experiment, the ruminal and post-ruminal disappearance of DM, CP, and starch were determined using in situ mobile nylon bag. Results: Barley grains treated with alum and alfalfa extract had a higher constant rate of starch digestion (0.11 and 0.09/h) than others. Barley grain treated with OE had a higher constant rate of CP digestion and that of treated with AC had a higher constant rate of starch digestion (0.08 and 0.11/h) compared with those of the other treatments. The indigestible fraction of starch treated with alum and sugar beet pulp extract was higher than that of the control group (0.24 and 0.25 vs 0.21). Barley grain treated with AC and OE had significant CP disappearance in the rumen, post-rumen and total tract, and also starch disappearance for post-rumen and total tract compared with the untreated (p<0.001). Conclusion: This study demonstrated that AC and OE might have positive effects on the starch degradation of the barley grain. In addition, treating barley grain with alum and sugar beet pulp extract could change the site and extend digestion of protein and starch.

Removal of Cs and Sr in Water Using Chemical and Natural Coagulants (화학 및 천연 응집제를 이용한 수중 Cs, Sr 제거)

  • This study investigated removal of radionuclides (Cs, Sr) in surface water by coagulation and precipitation. Jar tests were conducted with various chemical and natural coagulants to remove the stabilized radionuclides ($^{133}Cs$, $^{88}Sr$). Chemical coagulants included aluminum sulfate, poly aluminum chloride, and poly aluminum hydroxide chloride silicate (PACS); natural coagulants (minerals) included illite and zeolite. Chemical coagulant alone could achieve removals of Cs and Sr less than 10%; The removals increased up to 23.1% for Cs and 17.8% for Sr with addition of poly-dimethyldiallylammonium chloride (polyDADMAC) along with the chemical coagulants. Compared with chemical coagulants, natural coagulants (minerals) could achieve higher removals of Cs and Sr. Addition of zeolite along with PACS and polyDADMAC increased the removal rates up to 36.9% for Cs and 17.1% for Sr.

Effect of Aluminum Sulfate Addition to Litter and Dietary Protein Levels of Broilers on Litter Nitrogen Content (육계에 급여되는 사료중의 단백질 수준과 깔짚 내의 Aluminum Sulfate 첨가가 깔짚 내의 질소 함량에 미치는 영향)

  • The two experiments in this study compared litter nitrogen (N) contents after broiler chicks were raised for 42 days. Experiment I compared litter treated with aluminum sulfate (alum) [Al$_2$(SO$_4$)$_3$ㆍ14$H_2O$] (T$_1$) with nontreated litter (T$_2$) when the broiler chicks were fed the same levels of dietary protein (23% for 0∼3 weeks, 21% for 4∼6 weeks) Experiment 2 compared the alum treated litters of broiler chicks fed high protein diets (T$_3$) having 20.4% protein for 0∼3 weeks and 19.3% for 4∼6 weeks, with lower protein diets (T$_4$) having protein levels of 18.0% for 0∼3 weeks and 17.0% for 4 6 weeks. Each treatment had four replicate cages. As shown in Table 1, T$_1$ had a significantly (P<0.0l) lower pH and significantly (P<0.05) higher total N (TN), NH$_4$-N and inorganic N (IN) than T$_2$. T$_1$ and T$_2$ had similar moisture, organic carbon (OC), NO$_3$-N and organic N (ON). Alum treatment increased available N (AN) significantly (P<0.05) from 13.75$\pm$0.0l mg/g to 14.90$\pm$0.01 mg/g and predicted available N (PAN) significantly (P<0.05) from 15.00$\pm$0.0l to 16.50$\pm$0.02. The C : N ratios were 18.84$\pm$0.40 (T$_1$) and 19.46$\pm$0.10 (T$_2$) while the C : ON ratios were 28.49$\pm$1.15 (T$_1$) and 28.34$\pm$0.20 (T$_2$) although C : N ratios or C : ON ratios did not show any difference between T$_1$ and T$_2$. In Table 2, T$_3$ had significantly (P<0.05) higher moisture, TN, NH$_4$-N, ON and IN than T$_4$, while the pH, OC and NO$_3$-N were similar in both groups. The AN of T$_3$ increased significantly (P<0.05) from 10.99$\pm$0.0l mg/g to 12.98$\pm$0.03 mg/g, while the PAN increased significantly (P<0.05) from 12.39$\pm$0.10 mg/g (T$_4$) to 14.68$\pm$ 0.30 mg/g (T$_3$). The C : N ratios increased significantly (P<0.0l) from 20.07$\pm$0.20 (T$_3$) to 24.40$\pm$0.10 (T$_3$). The C : ON ratios also increased significantly (P<0.0l) from 28.99$\pm$1.15 (T$_3$) to 35.51$\pm$0.20 (T$_4$). These current research results show increased AN contents and PAN contents in alum treated litter or with increased CP levels regardless of alum treatment. However, none of the litters in this study could initially increase mineralization.

Effect of Metal Salt Coagulant on Membrane Fouling During Coagulation-UF Membrane Process (응집-UF 막 공정의 적용시 금속염 응집제가 막오염에 미치는 영향)

  • The objectives of this research are to investigate the mechanism of coagulation affecting UF, find out the effect of metal salt coagulant on membrane fouling. Either rapid mixing + UF or slow mixing + UF process caused much less flux decline. For PACl coagulant, the rate of flux decline was reduced for both hydrophilic and hydrophobic membrane than alum due to higher formation of flocs. In addition, the rate of flux decline for the hydrophobic membrane was significantly greater than for the hydrophilic membrane, regardless of pretreatment conditions. In general, Coagulation pretreatment significantly reduced the fouling of the hydrophilic membrane, but did little decrease the flux reduction of the hydrophobic membrane. When an Al(III) salt is added to water, monomers, polymers, or solid precipitates may form. Different Al(III) coagulants (alum and PACl) show to have different Al species distribution over a rapid mixing condition. During the rapid mixing period, for alum, formation of dissolved Al(III) (monomer and polymer) increases, but for PACl, precipitates of $Al(OH)_{3(s)}$ increases rapidly. This experimental results pointed out that precipitates of $Al(OH)_{3(s)}$ rather than dissolved Al(III) formation is major factor affecting flux decline for the membrane.

Increased Immunogenicity and Protective Efficacy of a P. aeruginosa Vaccine in Mice Using an Alum and De-O-Acylated Lipooligosaccharide Adjuvant System

  • Pseudomonas aeruginosa (P. aeruginosa) is an opportunistic pathogen that commonly causes fatal infections in cystic fibrosis and burn patients as well as in patients who are hospitalized or have impaired immune systems. P. aeruginosa infections are difficult to treat owing to the high resistance of the pathogen to conventional antibiotics. Despite several efforts, no effective prophylactic vaccines against P. aeruginosa are currently available. In this study, we investigated the activity of the CIA06 adjuvant system, which is composed of alum and de-O-acylated lipooligosaccharide, on a P. aeruginosa outer membrane protein (OMP) antigen vaccine in mice. The results indicated that CIA06 significantly increased the antigen-specific IgG titers and opsonophagocytic activity of immune sera against P. aeruginosa. In addition, the antibodies induced by the CIA06-adjuvanted vaccine exhibited higher cross-reactivity with heterologous P. aeruginosa strains. Finally, mice immunized with the CIA06-adjuvanted vaccine were effectively protected from lethal P. aeruginosa challenge. Based on these data, we suggest that the CIA06 adjuvant system might be used to promote the immunogenicity and protective efficacy of the P. aeruginosa OMP vaccine.

X-RAY PHOTOELECTRON SPECTOSCOPIC ANALYSIS OF ALUMINUM COMPOUND ADSORBED ON PULP FIBER SURFACES

  • aluminum sulfate (alum) as a representative retention aid in papermaking processes was added to pulp suspensions, and the aluminum components adsorbed on the pulp were investigated quantitatively by two types of X-ray elementary analyses with regard to simultaneous changes of their surface charges. X-ray photoelectron spectroscopy (XPS) and X-ray fluorescence analysis (XFA) were applied to determine the aluminum components retained in pulp pads up to ca. 10 nm and 100${\mu}$m depth, respectively. In other words, XPS was utilized to analyze the outermost surface layers of the samples, and XFA was available for measurement of their extensive regions. A particle charge detector (PCD) was used to monitor streaming potentials at various pHs of the pulp mixtures under moderate sharing conditions. At pH 4.5 of pulp suspensions containing alum, surface charges of pulp fibers varied from negative to slight negative (approximately neutral) according to adsorption of aluminum components onto the pulp fibers. Subsequently, when a dilute NaOH solution in limited amounts was added to pulp mixtures, both streaming potentials and surface aluminum content of the pulp fibers increased distinctly although little total aluminum retention increased. Further addition of alkali solutions brought drastic decreases of the surface charges and surface aluminum content, while total aluminum content, on the contrary, increased gradually under neutral conditions. These results indicate that residual aluminum ions remained in pulp suspensions are predominantly adsorbed on surfaces of pulp fibers by adequate alkali additions and they must sufficiently cationize the fiber surfaces with increases of somewhat cationic aluminum complexes formed on the surfaces. On the other hand, aluminum components formed in higher pH ranges have nearly no contribution to improvement of charge properties of the pulp fiber surfaces, even though aluminum retention in pulp pads increases. XPS and XFA analyses combined with streaming potential measurement using a PCD suggest close relationships between aluminum content on the pulp fiber surfaces and their charge properties.
