Screening for down syndrome using trophoblast retrieval and isolation of the cervix: preliminary study

  • Purpose: Trisomy 21, the cause of Down syndrome (DS) with various medical problems, is the most common aneuploidy during the fetal period. For diagnosis, a non-invasive screening test using maternal blood, which cannot be confirmed and invasive confirmation test with a risk of miscarriage, may be performed. The trophoblast retrieval and isolation of the cervix (TRIC) have been proposed by some researchers as an alternative to overcome the limitations of current tests. We experimented using TRIC to identify the possibility of trisomy 21 for the first time in Asia. Materials and Methods: Three cases of DS were analyzed confirmed by invasive tests (chorionic villus sampling, amniocentesis). All samples of trophoblasts immediately were immersed in phosphate-buffered saline and processed with formalin for fixation. The trophoblasts were isolated using an anti-human leukocyte antigen-G antibody coupled to magnetic nanoparticles. β-human chorionic gonadotropin (hCG)-expressing cells were considered as trophoblast cells, and the detection rate calculated. DS was confirmed by fluorescence in situ hybridization (FISH). Results: The mean trophoblast detection rate using β-hCG was 78.1%, and the detection rate using FISH was 22.2%. In all cases, the trisomy of chromosome 21 was identified. Conclusion: Trophoblast can be obtained from the five weeks of gestation and has a high detection rate, so it is noted that it can replace the current prenatal genetic test. To realize the clinical application as a prenatal genetic test, we will need additional efforts to identify trisomy 21 as well as other chromosomal abnormalities in future large-scale studies.

Preference of Subterranean Termites among Community Timber Species in Bogor, Indonesia

  • Many methods have been explored to increase the palatability of pine (Pinus merkusii), the most common wood used for termite baiting. However, because of the undersupply of pine in Indonesia, it is crucial to vary the wood species for termite baiting and look for potential alternatives. Furthermore, various studies have shown that baiting time influences the intensity and pattern of termite attacks. Therefore, the present research aimed to study the preferences of subterranean termites and find the ideal baiting time among community wood species from Bogor, West Java, as a baiting alternative to pine. The woods tested were Acacia mangium (acacia), Falcataria moluccana (sengon), Anthocephalus cadamba (jabon), Maesopsis eminii (manii), Swietenia mahagoni (mahogany), Hevea brasiliensis (rubberwood), and P. merkusii (pine). Field tests were carried out based on the American Society for Testing and Materials D 1758-06 at the Arboretum, Faculty of Forestry and Environment, IPB University, with a baiting time of one to six months. The results led to the identification of four species of termites, namely Microtermes sp., Macrotermes sp., Shedorhinotermes sp., and Capritermes sp.. The frequency of termite attacks on the test site reached 93.1%. Rubberwood was the most potential wood bait for subterranean termites, indicated by the highest average weight loss value (65.8%) with a shorter optimal baiting time (up to one month) than that of other tested woods.

다회용 마스크 유형에 따른 성능 비교연구 (A Comparison of Performance Evaluations of Reusable Masks)

  • Objectives: This study was designed to evaluate the filtration efficiency, inhalation resistance, and liquid resistance of commercial reusable masks in comparison to the performance of KF-AD masks (reference masks). Methods: Thirty-six masks were purchased from an online market. Filtration efficiency and inhalation resistance were determined in the same manner as in MFDS certification testing, respectively using TIL/IL & Filter Test 1300S (SFP Services, UK) and Breathing Resistance Analyzer DA2001 (Lambda Ray, South Korea). Results: Most of the filtration efficiencies found for the 19 masks were far lower than the test standard for KF-certified masks. Four masks met KF94 and three masks even met KF80. Most inhalation resistances were also much lower than the standard, with many almost one-half of the standard. In addition, all the masks except KF-AD masks did not pass the liquid resistance test. Conclusions: Although most of the filtration efficiency performance results found in this study for the reusable masks, which emerged as an alternative, were lower than the standard for health masks, multi-use masks can be used with a replacement filter inserted.

  • 3Y-TZP (3 mol% yttria-stabilized tetragonal zirconia polycrystals) ceramics have excellent mechanical properties including high fracture toughness, good abrasion resistance as well as chemical and biological stability. As a result, they are widely used in mechanical and medical components such as bearings, grinding balls, and hip implants. In addition, they provide excellent light transmittance, biocompatibility, and can match tooth color when used as a dental implant. Recently, given the materials' resemblance to human teeth, these ceramics have emerged as an alternative to titanium implants. Since the introduction of CAD/CAM in the manufacture of ceramic implants, they've been increasingly used for prosthetic restoration where aesthetics and strength are required. In this study, to improve the surface roughness of zirconia implants, we modified the 3Y-TZP surface with a biocomposite of hydroxyapatite and forsterite using room temperature spray coating methods, and investigated the mixed effect of the two powders on the evolution of surface microstructure, i.e., coating thickness and roughness, and biological interaction during the in vitro test in SBF solution. We compared improvement in bioactivity by observing dissolution and re-precipitation on the specimen surface. From the results of in vitro testing in SBF solution, we confirmed improvement in the bioactivity of the 3Y-TZP substrate after surface modification with a biocomposite of hydroxyapatite and forsterite. Surface dissolution of the coating layer and the precipitation of new hydroxyapatite particles was observed on the modified surface, indicating the improvement in bioactivity of the zirconia substrate.

Assessment of lactic acid bacteria isolated from the chicken digestive tract for potential use as poultry probiotics

  • Objective: The use of probiotics as an alternative to antibiotics in animal feed has received considerable attention in recent decades. Lactic acid bacteria (LAB) have remarkable functional properties promoting host health and are major microorganisms for probiotic purposes. The aim of this study was to characterize LAB strains of the chicken digestive tract and to determine their functional properties for further use as potential probiotics in poultry. Methods: A total of 2,000 colonies were isolated from the ileum and cecal contents of the chickens based on their phenotypic profiles and followed by a preliminary detection for acid and bile tolerance. The selected 200 LAB isolates with exhibited well-tolerance in acid and bile conditions were then identified by sequencing the 16S rDNA gene, followed by acid and bile tolerance, antimicrobial activity, adhesion to epithelial cells and additional characteristics on the removal of cholesterol. Then, the two probiotic strains (L. ingluviei and L. salivarious) which showed the greatest advantage in vitro testing were selected to assess their efficacy in broiler chickens. Results: It was found that 200 LAB isolates that complied with all measurement criteria belonged to five strains, including L. acidophilus (63 colonies), L. ingluviei (2 colonies), L. reuteri (58 colonies), L. salivarius (72 colonies), and L. saerimneri (5 colonies). We found that the L. ingluviei and L. salivarius can increase the population of LAB and Bifidobacterium spp. while reducing Enterobacteria spp. and Escherichia coli in the cecal content of chickens. Additionally, increased concentrations of valeric acid and short chain fatty acids were also observed. Conclusion: This study indicates that all five Lactobacillus strains isolated from gut contents of chickens are safe and possess probiotic properties, especially L. ingluviei and L. salivarius. Future studies should evaluate the potential for growth improvement in broilers.

Effect of phytic acid as an endodontic chelator on resin adhesion to sodium hypochlorite-treated dentin

  • Objectives: Phytic acid (IP6), a naturally occurring agent, has been previously reported as a potential alternative to ethylenediaminetetraacetic acid (EDTA). However, its effect on adhesion to sodium hypochlorite (NaOCl)-treated dentin and its interactions with NaOCl have not been previously reported. Thus, in this study, the effects of IP6 on resin adhesion to NaOCl-treated dentin and the failure mode were investigated and the interactions between the used agents were analyzed. Materials and Methods: Micro-tensile bond strength (µTBS) testing was performed until failure on dentin treated with either distilled water (control), 5% NaOCl, or 5% NaOCl followed with chelators: 17% EDTA for 1 minute or 1% IP6 for 30 seconds or 1 minute. The failed specimens were assessed under a scanning electron microscope. The reaction of NaOCl with EDTA or IP6 was analyzed in terms of temperature, pH, effervescence, and chlorine odor, and the effects of the resulting mixtures on the color of a stained paper were recorded. Results: The µTBS values of the control and NaOCl with chelator groups were not significantly different, but were all significantly higher than that of the group treated with NaOCl only. In the failure analysis, a distinctive feature was the presence of resin tags in samples conditioned with IP6 after treatment with NaOCl. The reaction of 1% IP6 with 5% NaOCl was less aggressive than the reaction of the latter with 17% EDTA. Conclusions: IP6 reversed the adverse effects of NaOCl on resin-dentin adhesion without the chlorine-depleting effect of EDTA.

The Reliability and Validity of a Portable Hand-held Spirometer for the Measurement of Various Lung Functions in Healthy Adults

  • Objective: This study aims to assess the reliability and validity of the new hand-held spirometer as a potential substitute for traditional pulmonary function testing (PFT) devices. Design: Cross-sectional study. Methods: In this study, thirty healthy adults underwent spirometry using both the new hand-held spirometer and the MIR spirometer, which is a standard PFT device. Parameters including peak expiratory flow (PEF), forced expiratory volume in one second (FEV1), and forced vital capacity (FVC) were measured and analyzed for validity and reliability. Inter-rater reliability and validity were evaluated through 95% limits of agreement (LOA) and intraclass correlation coefficients (ICC). Statistical analyses, including the Bland-Altman plots and the ICC, were utilized to assess agreement between the two devices. Results: The new hand-held spirometer exhibited a good agreement with intra-class coefficient (ICC [2,1]) ranging 0.762 to 0.956 and 95% LOA of -1.94 to 1.80 when compared with MIR. The test-retest reliability of the hand-held spirometer analyzed using - ICC [2,1] demonstrated a good level of consistency (ICC [2,1] =0.849-0.934). Conclusions: In conclusion, the study aimed to assess the potential of the new hand-held spirometer as a viable alternative to traditional PFT devices, with a specific focus on its reliability and validity in spirometric measurements. The new hand-held spirometer exhibited good test-retest reliability across all measured variables, suggesting its potential as a valid and reliable tool for simultaneous PFT measurements.

워드클라우드 분석을 통한 제작공정 교육용 확장 현실 콘텐츠 사용성 평가 (Usability Evaluation of XR Content for Production Training Through Word Cloud Analysis)

  • 본 연구는 제작공정 훈련을 위한 확장 현실 콘텐츠의 사용성을 탐색하고, 사용자 경험을 분석하는 목적으로 수행되었다. 이를 위해 공정교육용 확장 현실사용성 평가 보고서에 제시된 사용자 하드웨어, 사용자인터페이스, 콘텐츠 만족도에 대한 인터뷰 내용을 키워드 추출 및 워드 클라우드 시각화를 위한 파이선 패키지를 사용하여 분석하였다. 분석 결과, 하드웨어의 착용감은 만족스러웠지만, 중량과 열 문제는 개선이 필요하다는 피드백이 나타났다. 사용자인터페이스 측면에서는 가독성에는 긍정적 응답이 많았으나, 핸드트래킹 기반 상호작용 방식의 인식률이 낮아 개선이 요구되었다. 또한, 사용자는 확장 현실 기술을 의료 및 교육 분야에 적용할 가능성을 인식하고 긍정적인 미래 기대감을 표현하였다. 본 연구는 산업용 확장 현실 콘텐츠의 사용성 및 품질 개선을 위한 기초 자료를 제공하며, 향후 실질적인 산업 현장 적용을 위한 발전 방향을 수립하는 데 활용될 수 있다.

치과용 진공 융착 다이아몬드 드레서의 연삭성 연구 (Evaluation of the grinding performance of vacuum-brazed diamond dressers in dentistry)

  • Purpose: This study evaluated the potential of a vacuum-brazed dental diamond dresser as an alternative to commercially available electroplated diamond dressers and assess its suitability for clinical use. Methods: Commercially available electroplated diamond dressers were selected as the control group (n=5), whereas vacuum-brazed diamond dressers with 80 grit and 100 grit particles were fabricated as the experimental groups (n=5 each). Prepared specimens were evaluated for surface roughness, wear resistance, and abrasiveness against a wear-inducing element (ZrO2 ball). The results were reported as means and standard deviations. Statistical analyses were performed using one-way ANOVA followed by Tukey's HSD test using SPSS Statistics ver. 23.0 (IBM), with the significance level set at 0.05. Results: Surface roughness was highest in the 80 grit group (23.99±3.62 ㎛) followed by the 100 grit group (20.38±1.79 ㎛) and the control group (17.59±0.56 ㎛). Wear resistance was greatest in the control group (0.0060±0.0009 g) followed by the 80 grit group (0.0018±0.0003 g) and the 100 grit group (0.0013±0.0001 g). Abrasiveness against the ZrO2 ball was highest in the 80 grit group (0.120±0.007 g) followed by the 100 grit group (0.101±0.007 g) and the control group (0.086±0.004 g). Conclusion: This study offers key insights for enhancing the performance of diamond dressers through vacuum-brazing and is expected to aid in the development of more efficient and durable diamond dressers for future applications.

산성광산배수슬러지의 토양 중금속 안정화 적용 가능성 (The Applicability of the Acid Mine Drainage Sludge in the Heavy Metal Stabilization in Soils)

  • BACKGROUND: Recent studies using various industrial wastes for heavy metal stabilization in soil were conducted in order to find out new alternative amendments. The acid mine drainage sludge(AMDS) contains lots of metal oxides(hydroxides) that may be useful for heavy metal stabilization not only waste water treatment but also soil remediation. The aim of this study was to investigate the applicability of acid mine drainage sludge for heavy metals stabilization in soils METHODS AND RESULTS: Alkali soil contaminated with heavy metals was collected from the agricultural soils affected by the abandoned mine sites nearby. Three different amounts(1%, 3%, 5%) of AMDS were applied into control soil and contaminated soil. For determining the changes in the extractable heavy metals, $CaCl_2$ and Mehlich-3 were applied as chemical assessments for metal stabilization. For biological assessments, lettuce(Lactuca sativa L.) and chinese cabbage(Brassica rapa var. glabra) were cultivated and accumulation of heavy metals on each plant were determined. It was revealed that AMDS reduced heavy metal mobility and bioavailability in soil, which resulted in the decreases in the accumulation of As, Cd, Cu, Pb, and Zn in each plant. CONCLUSION: Though the high level of heavy metal concentrations in AMDS, any considerable increase in the heavy metal availability was not observed with control and contaminated soil. In conclusion, these results indicated that AMDS could be applied to heavy metal contaminated soil as an alternative amendments for reducing heavy metal mobility and bioavailability.
