Inhibitory Activity on the Diabetes Related Enzymes of Tetragonia tetragonioides (번행초 추출물의 당뇨관련 효소에 관한 저해 활성)

  • In this study, we examined the anti-diabetic activity in vitro by the crude extracts of Tetragonia tetragonioides which has been known to superior plants for the traditional prevention and treatment of stomach-related diseases. $\alpha$-Amylase and $\alpha$-glucosidase, the principal enzymes involved in the metabolism of carbohydrates, and aldose reductase, the key enzyme of the polyol pathway, have been shown to play the important roles in the complications associated with diabetes. A hexane (HX) fraction of T. tetragonioides were shown to inhibit more than 50% of salivary and pancreatin $\alpha$-amylase activity at concentration of 2.882 mg/mL and 2.043 mg/mL, respectively. In addition, the HX and ethylacetate (EA) fraction showed the highest inhibitory activity on yeast $\alpha$-glucosidase at values of $IC_{50}$ of 0.723 mg/mL and 1.356 mg/mL respectively. The HX, dichloromethane (DCM) and EA fraction showed more higher inhibitory activity on yeast $\alpha$-glucosidase than commercial agent such as 1-deoxynorjirimycin and acarbose. Also, the aldose reductase from human muscle cell had been inhibited strongly by the DCM fraction and HX fraction at 51.95% and 47.22% at a concentration of 1 mg/mL, respectively. Our study, for the first time, revealed the anti-diabetic potential of T. tetragonioides and this study could be used to develop medicinal preparations or nutraceutical and functional foods for diabetes and related symptoms.

Inhibitory Effects of Retinoic Acid and Melanization of B16 Melanoma Cell by Epimedium koreanum Nakai and $\alpha$ -MSH (음양곽과 $\alpha$ -MSH에 의한 B16 Melanoma 세포의 멜라닌화와 Retinoic Acid의 억제 효과)

  • Melanization of B16 melanoma cells was comparatively studied by the aqueous extract of Epimedium koreanum Nakai(EK) and $\alpha$-MSH. In addition, inhibitory effects of RA was investigated. B16 melanoma cells(about 1${\times}10_5$) have been shown an increase in tyrosinase activity and melanin contents in proportion to concentration of $\alpha$-MSH when treated with $\alpha$-MSH and incubated for 72 hrs. They indicated a 350% increase in tyrosinase activity and a 290% increase in melanin contents at 8 ng/mL. In case of EK, they have been shown a 200% increase in tyrosinase activity and a 180% increase in melanin contents at 100 ${\mu}g$/mL. In addition of RA to the above condition, they have been shown an inhibition from 350% to 210% in tyrosinase activity and from 290% to 250% in melanin contents in $\alpha$-MSH, and inhibition from 200% to 100% in tyrosinase activity and from 180% to 120% in melanin contents in EK. From the above results, it is suggested that EK promotes melanization of B16 melanoma cells through cAMP pathway, whereas RA inhibits it.

Quantitative Structure Activity Relationship between Diazabicyclo-[4.2.0]octanes Derivatives and Nicotinic Acetylcholine Receptor Agonists

  • Three dimensional quantitative structure activity relationship between diazabicyclo[4.2.0]octanes and nicotinic acetylcholine receptor($h{\alpha}4{\beta}2$ and $h{\alpha}3{\beta}4$) agonists was studied using comparative molecular field analysis(CoMFA) and comparative molecular similarity indices analysis(CoMSIA). From 11 CoMFA and CoMSIA models, CoMSIA with steric and electrostatic fields gave the best predictive models($q^2=0.926$ and 0.945, ${r^2}_{ncv}=0.983$ and 0.988). This study can be used to develop potent $h{\alpha}4{\beta}2$ receptor agonists with low activity on $h{\alpha}3{\beta}4$ subtype.

Effects of Red Light on $\alpha$-Amylase Isozmyes of the Germinated Barley (Hordeum distichum L.) (맥아의 $\alpha$-Amylase Isozyme에 미치는 Red Light의 영향)

  • This study carried out to changes $\alpha$-amylase activity and isozymes in barley during germination in the dark and red light The specific activity of u-amylase was increased during the germination periods in the dark, giving 355.0 and 523.7 units/mg protein at 3 and 5 day, and the activity was increased by the red light up to 48 and 15% at 3 and 5 days of germination, respectively. The ratio of $\alpha$-amylase I and II was approximately 95:5 at both 3 and 5 days of germination in the dark while the different ratio was found by the red light i. e. 60 : 40 and 90 : 10 at 3 and 5 days of germination, respectively.

  • 4-Aryl-2-hydroxytetronic acids which can be construded as a "lipophilic vitamin C analogue", are known to have antilipidemic and antiaggregatory activities in stead of antiscorb utic activity of vitamin C. A new compound 4-(p-chlorobiphenyl)-2-hydroxytetronic acid (8), which is structurally closely connected with the most active 4-(p-chlorophenyl)-2-hydroxytetronic acid, was synthesized from alpha-(p-chlorobiphenyl)-alpha-hydroxyacetic acid (1) through 7 steps as a potentially bioactive compound.

Effects of Clove Extracts on the Autoxidation and Thermal Oxidation of Soybean Oil (대두유의 자동산화 및 가열산화에 미치는 정향추출물의 항산화 효과)

  • This study was conducted to investigate the antioxidant activities of clove extracts in water, methanol and ether. The clove extracts, BHA and ${\alpha}$-tocopherol were added to each oil at a level of 200 ppm. The activities of the substrate oils and controls were tested under autoxidation and thermal oxidation conditions. The degree of the effects of the antioxidant activities under autoxidation condition were in the following order; ether extract 〉 methanol extract 〉BHA 〉 ${\alpha}$-tocopherol 〉 water extract = control group. The induction periods of the control, water, methanol and ether extracts, and BHA and ${\alpha}$-tocopherol were 9.5, 9.6, 10.7 11.8, 10.4 and 9.7 days, respectively. Under thermal oxidation condition, the methanol extract showed stronger antioxidant activity than those of the water and ether extracts. The antioxidant activity of the methanol extract was attributed to ${\alpha}$-tocopherol and BHA.

  • 1. $\alpha$-amylase from B. circulans F-2 was purified with specific activity 55.0 u/mg. protein (about 23 times of the original specific activity) and the yield of 25.5%, by means of corn starch absorption, salting out with ammonium sulfate (80% saturation), gel filtration on Bio-Gel P-100 and DE-32 column chromatography. 2 The purified enzyme showed two closely migrated protin bands on polyacrylamide disc gel electrophoresis, both of which have amylase activity judging from the activity staining of the gel. On SDS-polyacrylamide disc gel electrophoresis, however, the purified enzyme showed a single band suggesting that those two bands are the charge isomers of an amlyase having the slightly different charge. 3. Plot of log mobility of two bands versus polyacrylamide gel concentration according to Hedrick and Smith gave the parallel lines indicating them to be charge isomers. 4. To confirm the action pattern of two enzyme protein bands, each band was separated and was eluted from the gel and eluates were incubated with soluble starch. Oligosaccharide pattern produced by each eluate was examined by paper chromatography. The eluates of two bands showed the same action pattern. 5. The maltohexaose was the only hydrolysis product of soluble starch in the early stage of hydrolysis.

Antidiabetic Activity and Enzymatic Activity of Commercial Doenjang Certified for Traditional Foods (전통식품 품질인증 일부 시판 된장의 효소활성 및 항당뇨 활성)

  • We investigated the anti-diabetic activity and enzymatic activity of 24 commercial doenjang samples certified for traditional foods. Twenty four doenjang samples showed the wide ranges in enzymatic activities (protease activities 0-50.45 unit/g, ${\alpha}$-amylase activities 0-675.9 unit/g, ${\beta}$-amylase 13.6-308.6 unit/g), and there were no difference in enzymatic activity by the producing region. To evaluate the potential anti-diabetic activity of 24 doenjang samples, we examined the effect of doenjang methanol extract (DME) on 2-[n-(7-nitrobenz-2-oxa-1, 3-diazol-4-yl) amyno]-2-deoxy-d-glucose (2-NBDG) uptake. Ten samples among 24 samples significantly stimulated the uptake of 2-NBDG. When the cells were treated with DME at 400 ug/mL, No. 17 and 23 specially stimulated 2-NBDG uptake by 1.23-fold and 1.25-fold, respectively, compared with untreated control cell. And there were no cytotoxicity in the C2C12 cells treated with DME at concentration of 500 ug/mL. Among 24 samples, No. 6, 7, 12, 21 and 24 showed the ${\alpha}$-glucosidase inhibitor activity at concentration of 10 mg/mL; however, they were less effective than acarbose which is a commercial ${\alpha}$-glucosidase inhibitor.

Studies on Antioxidant and Antidiabetic Effects of Fermented Cnidium officinale Makino (발효(醱酵) 천궁(川芎)의 항산화 및 항당뇨 활성(活性)에 관한 연구(硏究))

  • Objectives : We investigated the antioxidant and Antidiabetic effects of Cnidii Rhizoma Fermentata by Aspergillus oryzae and Aspergillus Kawauchi for 3days. Methods : In this study we compared Cnidii Rhizoma Fermentata with Aspergillus oryzae and Aspergillus Kawauchi that examined using reducing sugar, DPPH radical scavenging activity, ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibitory activity. Also determined changes of pH and sugar content during fermentation for 3days. Results : The values for DPPH radical scavenging activity of Cnidii Rhizoma fermented by Aspergillus oryzae (86.6%) was higher than that of Aspergillus Kawauchi (77.9%). In ${\alpha}$-amylase inhibitory activity, fermented by Aspergillus Kawauchi had the highest inhibitory activity among other groups. But in ${\alpha}$-glucosidase inhibitory activity, fermented by Aspergillus oryzae had the highest inhibitory activity among other groups. While all groups of the sugar content increased During 3days fermentation, the pH was decreased. Conclusions : Based on these results, It was suggested that Cnidii Rhizoma Fermentata can be a useful and cost-effective resource for fuctional food and medicine.

Antioxidant and α-Glucosidase Inhibitory Activities of Tradescantia pallida (Rose) Hunt Leaf Extract and Fractions (팔리다자주닭개비 잎 추출물 및 분획물의 항산화 및 α-글루코시다아제 저해 활성)

  • Background: The biological activities of Tradescantia pallida grown in Korea have not been well determined, thus the aim of this study was to investigate the possibility of using it as a medicinal plant. Methods and Results: To investigate the antioxidant activity, ${\alpha}$-glucosidase inhibitory effect and antimicrobial activity of T. pallida, we performed the 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity and reducing power assay. This assay for T. pallida leaf extract showed the highest antioxidant activity for the ethyl acetate fraction ($RC_{50}=14.55{\pm}0.16{\mu}g/m{\ell}$ and Abs = 0.613 at $300{\mu}g$). Further, the ethyl acetate fraction exhibited higher ${\alpha}$-glucosidase inhibitory effect with an $IC_{50}$ value of $14.1{\pm}0.1{\mu}g/m{\ell}$ and showed antimicrobial activity against gram positive bacteria (minimum inhibitory concentration = $1,000{\mu}g/m{\ell}$). Conclusions: The ethyl acetate fraction of the crude methanol extract of T. pallida showed remarkable antioxidant activity, ${\alpha}$-glucosidase inhibitory effects and antimicrobial activity. These activities might be related to the flavonoid content in the T. pallida leaf extract.
