Molecular cloning of cDNAs for Korean garlic viruses

  • To understand the molecular structure and pathogenesis mechanism of Korean garlic viruses (GV), virus particles were isolated from field-grown garlic leaves and RNA genome was isolated from them. It was used for constructing cDNA library for GV. Several cDNA clones for GV were isolated and classified into 4 different groups on the basis of cross Southern hybridization. Northern blot analysis of GV RNA with one of these cDNA clones shows that the clone is a cDNA for GV RNA.

First Report of Bulb Canker of Garlic Caused by Embellisia allii in Korea

  • Embellisia allii causing bulb canker of white garlic(Allium sativum) has been observed on harvested or stored garlic bulbs. Identification of the fungus was determined based on the symptoms and morphological characteristics on in vitro potato dextrose agar(PDA) and malt extract agar(MEA). Colonies were effuse, blackish brown to black, velvety or powdery. Conidia were solitary, almost straight, most commonly ellipsoidal(or subcylindrical) to ovoid, mid to dark brown, smooth, with very dark 1 to 6 transverse(commonly $3{\sim}4$) and occasionally 1 or 2 oblique or longitudinal septa, and $30.4{\times}13.4{\mu}m$(av.). Conidiophores were simple or branched, straight or flexuous, up to 80 ${\mu}m$ long and $5.1{\sim}10.2{\mu}m$ thick. Chlamydospores were developed through enlargement and repeated cellular division of several adjacent hyphal cells to form a complex of thickwalled cells. The conidial measurements of garlic isolates closely matched the previous description of E. allii.

  • This study was intended to investigate the effects of solvents, particle size of a sample, sample to solvent ratio, temperature and time on the extraction of garlic oleoresin. Among eleven solvents used for oleoresin extraction from garlic, the optimal solvent was methyl alcohol. The most appropriate particle size of garlic, extracting temperature and mixing ratio of garlic to methyl alcohol were 20mesh, $25^{\circ}C$ and 1 to 3(w/w), respectively. On the basis of yield in oleoresin extraction, optimum extracting time was about 4 hours. The yield of oleoresin under the above-mentioned conditions was 21.3%. "L" and "b" as color appearance were decreased, whereas "a"was increased slightly during 60 days storage at 5$^{\circ}C$, $25^{\circ}C$ and 4$0^{\circ}C$. Changes in the pyruvate content showed close relation to pH value. During storage pyruvate content of garlic oleoresin was decreased as increasing storage temperature.

Ames Test에 의한 정유의 돌연변이원성 (Mutagenicity of the Essential Oils in Ames Test)

  • Mutagenic acivity of essential oils was tested using Salmonella typhimurium TA100 in the presence or absence of 59 fraction prepared from the mouse liver. Growth inhibitory effect of the oils on the bacteria was measured to warrant the mutagenic effect. Most oils were (round to be very strongly toxic against the bacteria at a high dose $(2,000{\mu}g/plate)$. At lower doses than this concentration, the Curcuma longa oil was found to be the most mutagenic with S9 fraction whereas it was not mutagenic without the fraction suggesting that this oil could undergo activation for the mutagenicity by cytochrome P45O. However, the mutagenicity of the Eugenia caryohpylata oil was disappeared under S9 fraction. Other oils obtained from Cinnamomum cassia, Chrysanthemum sibiricum, Paeonia moutan the flower of Artemisia princeps var. Orientalis, Allium sativum, were not mutagenic. This result suggested that antimutagenicity assay on the essential oil is necessary for the biological available substances.

  • Garlic industries in Korea faced with a increasing difficulty because of the pressure of garlic importation from China is ever increasing than before. Therefore this study was conducted to produce low-cost garlics by mechanized cultivation and the results are as follows. A. By applying machinery(stem cutter and size separator, seeder, harvester) the labor was saved over 70% compare with the conventional cultivation B. Growth characteristics and yield showed no difference in mechanized and conventional cultivation of northern-type garlic. C. When scales were sown by seeder, the sowing-depth varied between 3 to 6cm and the rate of 2-plants emergence in one hole was 14.5%.

  • The effect of garlic extract and vitamin C mixture on the various cancer cell lines in vitro and in vivo have been examined. Proliferation of human colon cancer (HT-29), human rectal cancer (HRT-18) and human hepatoma (HepG2) cells was inhibited by garlic extract and vitamin C, respectively. Based on the cytotoxic activity, mixture of garlic extract and vitamin C was demonstrated to possess a synergistic growth inhibition on HT-29, HRT-18 and HepG2 cancer cells. Mixture of garlic extract and vitamin C significantly arrested G2/M phase cells in the HepG2 cell cycle. Oral administration of mixture of garlic extract and vitamin C to sarcoma-180 tumor-bearing mice prolonged survival time compared to that of control group. These results suggested that addition of vitamin C enhances anticancer activity of garlic extract in vitro, and mixture of garlic extract and vitamin C has antitumor effect in vivo.

Detection of Allexiviruses in the Garlic Plants in Korea

  • The genomes of different allexiviruses were isolated and cloned from virus-infected garlic plants (Allium sativum), which were collected from farm fields in the southern provinces in Korea. The partial nucleotide sequences of the genomes from different allexiviruses were clearly identified in the virus-infected garlic plants. The cloned partial genomes of viruses in garlic plants showed a greater than 90% homology to previously identified allexiviruses and classified into species of GarV-A, -B, -C, -D, -E, and -X, demonstrating that species of allexivirus found in the other countries in the world are also widely distributed in the garlic plants in Korea.

마늘의 저장온도에 따른 이화학적 품질변화 (Change of Physicochemical Quality According to Its Storage Temperature in Garlic (Allium sativum L.))

  • This study was conducted to blow the effect of postharvest physiological changes on garlic quality according to its ecotypes and storage temperatures. The changes of water, total soluble solids, crude stein, md total fructans were measured and the rates of respiration and sprouting were analyzed during storage at 20$^{\circ}C$ and 30$^{\circ}C$. The decrease of water content and the increase of total soluble solids were reversely appeared during garlic storage. The crude protein content was gradually increased during storage but total fructan content was decrased. The respiration late was maximized at 60days after storage and the spouting rate was gradually increased. In the aspect of ecotypic characteristics, the water content, fructan content and sprouting rate were higher in 'Namdo' cultivar than those of southern type. The high storage temperature (30$^{\circ}C$) controlled spouting and loss of fructan, and it was effective to maintain the garlic quality.

  • 1979년(年)에 수확(收穫)한 묵은 마늘과 1980년(年)에 수확(收穫)한 햇 마늘의 분화엽(分化葉)의 엽신(葉身), 생장점(生長點)이 부착된 기부(基部)의 배양(培養)으로 부터 기관(器管)의 분화(分化)에 BA와 auxin이 미치는 영향(影響)을 검토하기 위하여 본(本) 실험(實驗)을 수행(修行)하였으며 아울러 callus를 계대배양하여 BA와 NAA의 효과(效果)를 검토하였다. 또한 햇마늘의 경우 10, 20, 30, 40일간(日間) 저온처리(低溫處理) 후(後) 엽신(葉身) 조직배양(組織培養)에서 BA와 NAA의 효과(效果)를 조사(調査)하였다. 1. 묵은 마늘의 엽신(葉身)의 배양(培養)에서 NAA 단독처리(單獨處理)에서 유식물(幼植物)의 분화(分化)는 없었고 2mg/l구(區)와 4mg/l구(區)에서 뿌리의 발생(發生)이 보였으며 유식물(幼植物)의 분화(分化)는 BA와 NAA의 복합처리(複合處理)에 의해서 이루어 졌으며 BA 2mg/l, NAA 2mg/l구(區)의 효과(效果)가 좋았다. 2. 묵은 마늘의 엽신배양(葉身培養)에서 NAA의 대신(代身) 2, 4-D를 첨가(添加)했을 때 기관(器官)의 분화(分化)는 없었고 callus의 분화(分化)가 좋았으며 이 때 callus의 상태(狀態)는 NAA의 처리구(處理區)의 callus에 비해 구상체(球狀體)를 이루고 있다. 3. 2, 4-D처리(處理)에 의해 얻어진 callus의 계대배양에서 BA와 NAA의 복합(複合處理區)에서 대체로 유식물체(幼植物體)나 뿌리의 발생이 일어났으며 특히 BA 2mg/l에 NAA 0.5 mg/l구(區)와 1mg/l 구(區)에서 효과(效果)가 좋았다. 그러나 NAA 8mg/l구(區)나 무처리구(無處理區)에서는 유식물(幼植物)의 발생(發生)이 없었다. 4. 햇 마늘의 엽조직(葉組織)의 배양(培養)에서는 묵은 마늘에서와는 마르게 무처리구(無處理區)에서 유식물(幼植物)의 발생(發生)이 많았으며 BA 2mg/l과 NAA의 혼합처리(混台處理)에서도 NAA 1mg/l구(區)에서의 유식물(幼植物)의 분화(分化)가 좋았으나, 뿌리의 발생(發生)에는 NAA 4mg/l구(區)에서 좋았다. 5. 햇 마늘 기부배양에서는 NAA의 단독처리(單獨處理)에서 2mg/l구(區)에서 발근(發根)이 있었으나 유식물체(幼植物體)의 분화(分化)는 없었고 혼합처리(混合處理)의 경우는 BA의 농도가 낮을 때는 NAA의 농도가 다소 높은 구(區)에서, BA의 농도(濃度)가 다소 높은 경우는 NAA의 농도가 낮을 때 유식물체(幼植物體)의 분화(分化)가 좋았다. 6. 저온처리(低溫處理) 후(後)의 배양에서 30일(日), 40일간(日間)의 장기간 저온처리(低溫處理)는 오히려 유식물체(幼植物體)의 분화(分化)가 저조했고 무저온처리구(無低溫處理區), 10일(日), 20일(日) 처리구(處理區) 중(中) BA 2mg/l구(區)와 NAA 0.5mg/l구(區)에서 유식물(幼植物)의 분화(分化)가 나다났다.

숙성에 의해 제조된 흑 마늘 추출물의 생리학적 활성 및 항산화 효과 (Physiological Activity and Antioxidative Effects of Aged Black Garlic (Allium sativum L.) Extract)

  • 고온에서 숙성시켜 제조된 흑 마늘의 총폴리페놀 함량은 10.0 mg/g으로 생마늘 3.7 mg/g보다 2.5배 이상 높은 값을 보였다. 생마늘과 흑 마늘 추출물의 전자공여능 실험 결과, 흑 마늘 추출물의 농도 증가에 따라 전자공여능이 급격하게 증가하는 것으로 나타났으며, 추출물의 농도가 $1,000{\mu}g/mL$일 때 생마늘은 12.9%, 흑 마늘은 101.9%로 흑 마늘이 생마늘에 비하여 항산화력이 매우 높은 것으로 나타났다. 아질산염 소거능은 추출물의 농도가 5mg/mL일 때 생마늘 추출물이 흑 마늘 추출물보다 1.7배 높게 나타났으나, 그 이상의 농도에서는 비슷하거나 흑 마늘 추출물이 유의적으로 높게 나타났다 생마늘과 흑 마늘 추출물에서 SOD 활성은 추출물의 농도가 20mg/mL일 때 각각 3.3%와 30.9%를 나타내었으며, 흑 마늘이 생마늘에 비하여 SOD 활성이 유의적으로 높게 나타났다. ACE 저해활성은 추출물의 농도가 50mg/mL 일 때 생마늘과 흑 마늘에서 각각 52.7%, 88.8%로 흑 마늘의 항고혈압활성이 높은 것으로 나타났다.
