마늘의 주요 성분인 allicin투여 후 유도되는 사람 말초혈액의 단핵구의 유전자 발현에 미치는allicin의 효과를 규명하였다. DNA microarray를 이용하여, allicin이 chemokines, cytokine, 면역관련 유전자 및 신호전달 관련 유전자의 발현을 유도하는 것을 확인하였다. 반대로 allicin은 Th1 type의 획득면역 관련 유전자의 발현을 억제하였다. 염증세포에 있어서 allicin은 억제효과 및 자극 효과를 동시에 보여주었다. 이는 allicin이 휴지기 세포에서 먼저 증가시킨 특정 유전자의 발현을 이후에 감소시키는 결과를 보여주는 것으로 positive와 negative 효과를 발휘하는 새로운 기전을 제시하는 것이다. Allicin에 대한 광범위하고 새로운 관심을 고려해 볼 때 본 연구에서 보여주는 많은 유전자의 발현 양상은 좀 더 특정적이고 효과적인 치료법을 고안하는 데 유용할 것이다.
The major garlic component, Allicin [diallylthiosulfinate, or (R, S)-diallyldissulfid-S-oxide] is known for its medicinal effects, such as antihypertensive activity, microbicidal activity, and antitumor activity. Allicin and diallyldisulfide, which is a converted form of allicin, inhibited the cholesterol level in hepatocytes, in vivo and in vitro. The metabolism of allicin reportedly occurs in the microsomes of hepatocytes, predominantly with the contribution of cytochrome P-450. However, little is known about how allicin affects the genes involved in the activity of hepatocytes in vivo. In the present study, we used the short-term intravenous injection of allicin to examine the in vivo genetic profile of hepatocytes. Allicin up-regulate ten genes in the hepatocytes. For example, the interferon regulator 1 (IRF-I), the wingless-related MMTV (mouse mammary tumor virus) integration site 4 (wnt-4), and the fatty acid binding protein 1. However, allicin down-regulated three genes: namely, glutathione S-transferase mu6, a-2-HS glycoprotein, and the corticosteroid binding globulin of hepatocytes. The up-regulated wnt-4, IRF-1, and mannose binding lectin genes can enhance the growth factors, cytokines, transcription activators and repressors that are involved in the immune defense mechanism. These primary data, which were generated with the aid of the Atlas Plastic Mouse 5 K Microarray, help to explain the mechanism which enables allicin to act as a therapeutic agent, to enhance immunity, and to prevent cancer. The data suggest that these benefits of allicin are partly caused by the up-regulated or down-regulated gene profiles of hepatocytes. To evaluate the genetic profile in more detail, we need to use a more extensive mouse genome array.
Allicin was synthesized for the purpose of identification an dquantitation of a pharmaceutical dosage form in soft capsules. The identified transformed products fro allicin were dially disulfide , 3-vinyl-[4H]-1, 2-dithiin and 2-vinyl-[4H]-1, 3-dithiin in gas chromatrographic conditions and dially disulfide and ajoene in HPLC. Allicin is thermally unstable , it may be completely decomposed to vinyl dithiin isomers in GC conditions. For that reason, allicin was not found directly in the pharmaceutical dosage forms. In HPLC conditions, mobile phase was methanol /water containing 0.1% formic acid(65/35) and column was $\mu$-Bondapak C18. Detection wa-velength was 254nm. The retention time of allicin was 6.98min. The calibration ranger for allicin was 10 $\mu\textrm{g}$/ml to 200$\mu\textrm{g}$/ml and correlation coefficient(r) was 0.987.
본 연구에서는 마늘성분 allicin에 의한 항암 기전 및 화학적 암 예방법의 분자 생물학적 기전을 이해하기 위한 연구의 일환으로, 마늘성분 중의 하나인 allicin이 인체 대장암 세포주 HCT116의 증식에 미치는 영향을 확인하였다. allicin의 처리에 따라 세포사멸이 나타나며 생존율이 감소함을 확인하였다. 또한 oligo DNA microarray 실험을 통하여 allicin에 의해 발현되는 유전자 중 발현양의 차이를 보인 유전자중 2배 이상 up-regulation된 유전자는 7,840개, 2배 이상 down-regulation 된 유전자는 10,010개로 각각 분류 되었다. 이 중, ATF3유전자의 발현을 확인하였고, 또 다른 항암유전자인 NAG-1 유전자의 발현을 RT-PCR로 확인하였다. 그 결과, allicin처리에 의해 항암유전자인 ATF3의 발현과 NAG1 유전자의 발현이 증가함을 확인하였다. 또한 p53 HCT116 세포주를 이용하여 allicin에 의한 NAG1 단백질의 발현을 확인한 결과, allicin은 p53 유전자 의존적으로 NAG-1 단백질을 발현함을 확인하였다. 결론적으로 allicin은 세포주기나 세포사멸에 관련된 많은 유전자들의 발현 변화를 유도함으로써 암세포 성장억제 활성을 갖는 것으로 사료된다.
Relationship between pungency and allicin content of pickled garlic during aging was examined . Degree of pungency of pickled garlic during aging at 20 $^{\circ}C$ was determined by the sensory evaluation. A panel of 10 members evaluated seven samples of pickled garlic which were aged for 0, 10 , 20, 40 , 50 or 60 days by using scoring test (seven point scale). The sensory evaluation results showed that pungency of pickled garlic decreased gradually during aging, and scored at 3.07 on the 40 th day of aging. Content of allicin, which was a major pungent component of garlic homogenate, was quantitatively analyzed by HPLC. The level of allicin in homogenate of pickled garlic was found to decrease gradually, and to 5.9% on the 40 th day of aging compared with that of fresh garlic. Relationship between the pungency score results and the content of allicin demonstrated a highly positive correlation (r=0.9648).
Background: Artesunate, extracted from Artemisia annua, has been proven to have anti-cancer potential. Allicin, diallyl thiosulfinate, the main biologically active compound derived from garlic, is also of interest in cancer treatment research. This object of this report was to document synergistic effects of artesunate combined with allicin on osteosarcoma cell lines in vitro and in vivo. Methods: After treatment with artesunate and allicin at various concentrations, the viability of osteosarcoma cells was analyzed by MTT method, with assessment of invasion and motility, colony formation and apoptosis. Western Blotting was performed to determine the expression of caspase-3/9, and activity was also detected after drug treatment. Moreover, in a nude mouse model established with orthotopic xenograft tumors, tumor weight and volume were monitored after drug administration via the intraperitoneal (i.p.) route. Results: The viability of osteosarcoma cells in the combination group was significantly decreased in a concentration and time dependent manner; moreover, invasion, motility and colony formation ability were significantly suppressed and the apoptotic rate was significantly increased through caspase-3/9 expression and activity enhancement in the combination group. Furthermore, suppression of tumor growth was evident in vivo. Conclusion: Our results indicated that artesunate and allicin in combination exert synergistic effects on osteosarcoma cell proliferation and apoptosis.
Background: Inflammation is a frequent reaction following therapeutic irradiation. Since the upregulation of adhesion molecules on endothelial cell surface is known to be associated with inflammation, the expression of adhesion molecules is an important therapeutic target. Methods: Treatment of human umbilical endothelial cells (HUVECs) with ${\gamma}$-irradiation (${\gamma}IR$) induces the expression of adhesion proteins such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin. Changes in the expression of these proteins on ${\gamma}$-irradiated HUVECs which had been treated previously with allicin were measured by ELISA. Results: In the present study, we demonstrate that allicin inhibits the ${\gamma}IR$ induced expression of ICAM-1, VCAM-1, and E-selectin on HUVEC in a dose-dependent manner. Allicin was also found to inhibit the ${\gamma}IR$ induced production of nitric oxide (NO). Conclusion: These data suggest that allicin has a therapeutic potential for the treatment of various inflammatory disorders associated with increase numbers of endothelial leukocyte adhesion molecules.
Allicin, a garlic componente, is believed to provide protection against various diseases including inflammation. Since interactions of the cell adhesion molecules are known to play important roles in mediating inflammation, inhibiting adhesion protein upregulation is a possible therapeutic target. In this study, we demonstrate that TNF-${\alpha}$- and catalase-induced expression of ICAM-1 on human lung epithelial cells (A549) in a dose-dependent manner and catalase expression and activity were also increased in TNF-${\alpha}$-treated cells. Treatment of the TNF-${\alpha}$-treated cells with catalase inhibitor 3-amino-1,2,4-triazole resulted in a significant decreased the level of ICAM-1. These data suggest that induction of ICAM-1 expression by TNF-${\alpha}$ is associated with catalase. In addition, allicin was found to inhibit the TNF-${\alpha}$ induced expression of ICAM-1 on the A549 cells. This compound also inhibited the production of catalase induced by TNF-${\alpha}$, which suggests that the inhibition of ICAM-1 expression by allicin may be due to the modulated production of catalase.
It was attempted to observe the effect of garlic on the hepatic glutathione s-transferase and glutathione peroxidase activity in this study. Glutathione s-transferase (EC 2.5.1.18) are thought to play a physiological role in initiating the detoxication of potential alkylating agents, inclnding pharmacologically active compounds. Glutathione peroxidase (EC 1. 11. 1. 9) might play an important role in the protection of cellular structures against oxidative challenge. The activities of glutathione s-transferase and glutathione peroxidase in rat liver were increased by the treatment of garlic juice. Allicin fraction, heat-treated allicin fraction and garlic butanol fraction markedly inhibited glutathione s-transferase activity in vitro, whereas glutathione peroxidase activity was significantly increased in heat-treated allicin fraction and garlic butanol fraction.
밭마늘과 논마늘(영천산)의 매운맛 차이를 알아보고자, 마늘의 매운맛 생성에 관여하는 주요 인자들인 alliin과 allcin 그리고 SAC의 함량을 HPLC로 분석하고, 이들 함함성분의 생성에 관여하는 효소인 alliinase와 GTPase(${\gamma}$-glutamyl trans peptidase) 활성 및 pyruvate 생성량과 일반성분을 분석하였다. 수분함량은 논마늘이 밭마늘에 비하여 높았으며, 조회분, 조지방, 조단백, 탄수화물 함량 역시 밭마늘이 논마늘에 비하여 높았다. 마늘의 주요 매운맛 성분인 allicin 함량은 밭마늘이 $2.83{\pm}0.03$ mg/g, 논마늘이 $2.22{\pm}0.02$ mg/g을 나타내어 밭마늘이 논마늘에 비하여 유의적으로 높았다. Allicin의 전구체인 alliin의 함량은 밭마늘이 $8.97{\pm}0.09$ mg/g, 논마늘이 $8.22{\pm}0.08$ mg/g으로 나타났다. SAC 함량은 밭마늘이 $1.74{\pm}0.17$ mg/g으로, 논마늘의 $1.04{\pm}0.14$ mg/g에 비하여 유의적으로 높았다. 한편, 효소 alliinase 활성도는 밭마늘이 $285{\pm}43{\mu}mole/mg$ protein, 논마늘이 $239{\pm}34{\mu}mole/mg$ protein을 나타내어 유의적인 차이는 없었으나, GTPase 활성은 밭마늘이 86.3 ${\mu}mole/mg$ protein으로 논마늘의 78.4 ${\mu}mole/mg$ protein에 비하여 높게 나타내었다. 이상의 결과를 토대로 밭마늘이 매운 맛이 강한 것은 본 연구결과로부터 allicin 함량이 논마늘에 비하여 높은 데 기인된 것으로 파악되었다. 밭마늘은 논마늘에 비하여 alliinase 활성은 유사하였으나 alliin의 함량이 높아 allicin 생성량이 많았으며, SAC가 높고 GTPase의 활성이 높아 alliin의 함량이 높은 것으로 생각되었다. 따라서, 마늘을 기능성 식품소재로의 활용을 극대화하고자 한다면 밭마늘이 논마늘보다 더 적합한 것으로 생각된다.
