  • 토양으로 부터 diaminododecane 자화균 DAD2-2주를 분리하여 검토한 결과 Corynebacterium속으로 동정되었으며 DAD2-2주의 alkane 유도체에 대한 생육특성조사에서 putrescine dihydrochloride, dodecanethiol, dodecane. laurylamine등은 탄소원으로 이용될 수 있었으나 thioanisole, decanedithiol, dicyanooctane, 1aurylcyanide, dichlorodecane등은 이용되지 못하였다. Emulgen 25ppm을 DAD배지에 첨가하였을 경우 DAD2-2주에 의한 diaminododecane 자화가 현저하게 촉진되었으며 diaminododecane자화시 생성되는 중간생성물은 ethyl $\alpha$-ketoglutarate로 동정되었다. 그러나 diaminododecane을 탄소원 뿐만 아니라 질소원으로도 이용하였을 경우에 있어서는 ethyl $\alpha$-ketoglutarate와는 다른 생성물을 생성하였으며 diaminododecane 대신 glucose, putrescinc, n-dodecane을 생육기질로 이용하였을 때에도 ethyl $\alpha$-ketoglutarate와는 다른 생성물을 생성하였다. 또한 resting cell을 이용하여 alkane 유도체를 co-oxidation시키는 과정에서도 ethyl $\alpha$-ketoglutarate와는 상이한 생성물이 생성되었다.

Seasonal Changes in Voluntary Intake and Digestibility by Sheep Grazing Introduced Leymus chinensis Pasture

  • A study was conducted to investigate the seasonal changes in nutrient composition of pasture, voluntary intake and digestibility of sheep grazing an introduced Leymus chinensis pasture located in western Jilin Province, China. The whole-plant of L. chinensis and the samples simulating ingestion by sheep (simulating sample) were collected in spring (May, 2004), summer (July, 2004), autumn (September, 2004) and the end of winter (April, 2005). The contents of gross energy (GE), organic matter (OM), crude protein (CP), neutral detergent fibre (NDF), acid detergent fibre (ADF) and alkanes were determined. Voluntary intake and apparent digestibility of the nutrients in each season were also examined using 6 mature Chinese Northeast Merino ewes (differed among the seasons) grazing on a paddock of 1 ha size. The apparent digestibilities of GE, dry matter (DM), OM, CP, NDF and ADF of L. chinensis were significantly greater (p<0.05) in spring and summer than in autumn and winter. Metabolizable energy (ME) content was 12.13, 11.62, 8.66 and 10.40 MJ/kg DM for L. chinensis in spring, summer, autumn and winter respectively, and the corresponding voluntary feed intakes were 91, 119, 59 and 58 g/d per kg metabolic weight ($LW^{0.75}$). The ME contents and DM intakes in autumn and winter were significantly lower than in spring and summer (p<0.05). The intake of L. chinensis pasture was sufficient to provide ME requirements for maintenance by the dry ewes in all the seasons, but inadequate for maintenance protein requirement in winter. IQF 굴(Crassostrea gigas) 복합엑스분을 이용한 굴 소스의 가열향기 성분 (Volatile Flavor Constituents of Cooked Oyster Sauce Prepared from Individually Quick-frozen Oyster Crassostrea gigas Extract)

  • The pacific oyster Crassostrea gigas has a desirable taste and flavor that differs from those of other fish and shellfish. In order to develop a high value-added product from individually quick-frozen oyster extract (IQFOE), we prepared an oyster sauce from IQFOE and characterized its volatile compounds using vacuum simultaneous steam distillationsolvent extraction / gas chromatography / mass spectrometry. The moisture, crude protein, crude ash, salinity, pH and volatile basic nitrogen contents of the oyster sauce were 60.6%, 8.2%, 9.2%, 9.3%, 5.7 and 21.0 mg/100 g, respectively. Seventy-six volatile compounds were detected in the cooked odor of the oyster sauce. These volatile compounds included 14 esters, including ethyl acetate, 13 nitrogen- containing compounds, including 2,4,6-trimethyl pyridine, 13 acids, including hexadecanoic acid, 12 alcohols, including ethyl alcohol and 6-methyl heptanol, 6 alkanes, 5 aldehydes, including benzaldehyde, 5 ketones, including 1-(2-furanyl)-ethanone, 4 furans, including 2-furancarboxaldehyde and 2-furanmethanol, 3 aromatic compounds, including d-limonene, and 1 miscellaneous compound. Esters, acids and nitrogen-containing compounds, and alcohols were the most abundant compounds in the odor of the cooked oyster sauce, with some aldehydes, ketones, and furans.

용매추출과 분무건조의 통합방법을 이용한 후코이단의 냄새 저감 공정 개발 (Process Development for Deordorization of Fucoidan Using a Combined Method of Solvent Extraction and Spray Drying)

  • Volatile components of crude fucoidan and its raw material, Undaria pinnatifida sporophylls were identified by GC/MSD analysis, and the effects of a combined method of solvent extraction and drying (hot-air drying or spray drying) on deordorization of fucoidans were examined. The major components of seaweed smell (odor) in crude fucoidan and Undaria pinnatifida sporophylls were identified as alkanes (octadecane, heptadecane, tetradecane) and acids (hexadecanoic acid, oleic acid). Solvent extraction and drying were combined for the deordorization of fucoidans: hot-air drying/deordorization process (HDDP) and spray drying/deordorization process (SDDP). The deordorization effects of solvent extraction using solvents including water, ethanol, and acetone were investigated. Among the tested solvents, ethanol was selected as a deordorization solvent, because the seaweed smell and taste were mostly lowered by ethanol extraction followed by drying. In addition, HDDP and SDDP were compared, showing that SDDP gave higher sensory characteristics than HDDP. When the crude fucoidan was treated by ethanol extraction for 30 min followed by SDDP, approximately 60% of off-flavor components of crude fucoidan was removed based on GC/MSD peak area, and the total sugar and sulfate contents and APTT-anticoagulant activity significantly increased compared to crude fucoidan. In conclusion, SDDP after ethanol treatment was proposed and established as a deordorization process for fucoidan preparation.

Antimicrobial Activity and Components of Extracts from Agastache rugosa during Growth Period

  • Antimicrobial activities of volatile flavor, water and methanol extracts from Agastache rugosa were investigated. The volatile flavor extract was obtained from A. rugosa by simulataneous distillation-extraction (SDE) method. Antimicrobial activity was investigated by disc diffusion method against several microorganisms, four species of Gram positive, three species of Gram negative and tow species of yeast. The volatile flavor extracts had strong antimicrobial activity againstc. utilisand S. cerevisiae. During the growth period, a difference in antimicrobial activity among volatile flavor extracts from A. rugosa was not shown. The water extract of above 10 mg/disc showed antimicrobial activity. Methanol extracts from A. rugosa harvested in June showed antimicrobial activity against tested Gram positive and Gram negative bacteria, showed weak antimicrobial activity against the bacteria from those harvested in July and August. In particular, antimicrobial activity against V. parahaemolyticus was stronger than that against other bacteria. Water and methanol extracts did not inhibit yeast. C. utilis and S. cerevisiae. To further elucidate the effective components, volatile flavor extracts was analyzed by GC/MS. harvested in June, the components included 8 phenols (93.031%), 18 terpenes (5.230%), 12 alcohols (1.300%) 8 alkanes (0.181%), 1 ester (0.056%), 2 ketones (0.033%), 2 aldehydes (0.011%) and 1 pyrrole (0.007%). In July, the components included 6 phenols (94.366%), 19 terpenes (3.394%), 11 alcohols (2.045%), 1 ester (0.039%), 2 ketones (0.028%), 1 furan (0.005%) and 1 aldehyde (0.005%). And in August, the components included 7 phenols (95.270%), 19 terpenes (2.951%), 13 alcohols (1.399%), 1 ester (0.063%), 2 aldehydes (0.016%), 2 ketones (0.011%), 1 alkane (0.006%), 1 acid (0.005%) and 1 pyrrole (0.005%). The major component of volatile flavors was estragole, a phenolic compound.

  • 철(III) protoporphyrin과 망간(II) tetrakis(4-N-carboxyphenyl) Porphyrin을 polystyrene divinylbenzene 공중합체 수지와 반응시켜 고분자결합 금속포피린을 합성하였다. 고분자와 결합된 포피린은 공명라만분광기를 이용, 합성과정에서 포피린과 중심금속의 산화상태 등 여러가지 화학적 성질이 변화되지 않았음으로 확인하였다. 합성된 고분자결합 금속포피린은 싸이토크롬 P-450과 같은 산화촉매효소의 모형화합물로서 과산화수소에 의한 olefin의 epoxidation과 alkane의 oxidation의 촉매로서 사용이 가능하였다. 그 효율은 고분자와 결합하지 않은 동종의 포피린에 비하여 현저히 증가되었으며, 특히 포피린이 산화제에 의해 분해되지 않아 재사용이 가능하였다. 실제 효소에서와 같이 imidazole 유도체와 같은 전자주게 배위자가 포피린 중심금속에 배위한 경우 촉매효과가 더욱 향상되었다.

낙동강 수질중 유기물질과 독성 (Organic Compounds in the Nak Dong River and Its Toxicity)

  • This study aims to investigate organic compounds and its toxicity by Ames test and chromosomal aberration in the water of the Nak Dong River. Six sampling sites such as Goryung, Hagueun, Maelie, Duksan, Haedong and Myungiang were selected for these pur15oses. 200 l water samples were absorbed on XAD-2 resin columns (2.5X30cm), eluted with organic solvents mixture of acetone: cyclohexane and then dried under vacuum condition. The extracts from the XAD-2 resin was injected into GC/MS and 184 organic compounds were identified such as aldehydes, aromatic compounds, ketones, phenols, hydrocarbons, alcohols, carboxylic acids, alkanes and some unknowns. The US EPA priority pollutants such as naphthlene, bis(2-ethylhexyl)phthalate and other pollutants, 1,2-diethyl benzene, 1,2,3,4-tetrahydronaphthalene and cyclohexanol were detected in these samples. The concentration of chemical pollutants such as 1,2-diethyl benzene, nephthalene, 1,2,3,4-tetrahydronaphthalene, bis(2-ethylhexyl)phthalate and cyclohexanol were ranged into 1.228 $\mu$g/l, 298 $\mu$g/l, 30.191 $\mu$g/l, 1.147 $\mu$g/l and 2.839 $\mu$g/l, respectively. The mutagenic activity of XAD-2 extracts were tested on Salmonella typhimurium TA 98, TA 100, TA 1535 and TA 1537 and then exhibited strong mutagenic activity against S. typhimurium TA 98 and TA 100 in the presence of S$_9$. Amon them, bis(2-ethylhexyl)phtalate and 1,2-diethyl benzene showed the most strongest mutagenic activity against S. typhimurium TA 98 and TA 100 in the presence of S$_9$. On the other hands, chromosomal aberration of XAD-2 extracts in the human blood cells were not occurred by the sampling water at Goryung, Hagueun, Maelie and Duksan, Chromosomal aberration were also not occurred by the each concentration of 0.05, 0.1 amd 0.3 mg/l of each 1,2-diethyl benzol, bis(2-ethylhexyl)phthalate, naphthalene, phenol, cyclohexanol and benzothiazol test solution.

Relationship between sensory attributes and volatile compounds of polish dry-cured loin

  • Objective: The aim of this work was to determine the relationship between objective sensory descriptors and volatile flavour compound composition of Polish traditional dry-cured loin. Methods: The volatile compounds were investigated by using solid phase microextraction (SPME) and gas chromatography-mass spectrometry (GC-MS). For sensory assessment, the quantitative descriptive analysis (QDA) method was used. Results: A total of 50 volatile compounds were found and assigned to 17 chemical families. Most of the detected volatile compounds derived from smoking, lipid oxidative reactions and seasoning (46.8%, 21.7%, and 18.9%, respectively). The dominant compounds were: aromatic hydrocarbon (toluene); alkanes (hexane, heptane, and 2,2,4-trimethylpentane); aldehyde (hexanal); alcohol (2-furanmethanol); ketone (3-hydroxy-2-butanone); phenol (guaiacol); and terpenes (eucalyptol, cymene, ${\gamma}-terpinen$, and limonene). Correlation analysis showed that some compounds derived from smoking were positively correlated with the intensity of cured meat odour and flavour and negatively with the intensity of dried meat odour and flavour, while terpenes were strongly correlated with odour and flavour of added spices. Conclusion: The analysed dry-cured loins were characterized by specific and unique sensory profile. Odour and flavour of studied loins was mainly determined by volatile compounds originating from smoking, seasoning and lipid oxidation. Obtained results suggest that smoking process is a crucial stage during Polish traditional dry-cured loins production.

Seasonal Variations in Voluntary Intake and Apparent Digestibility of Forages in Goats Grazing on Introduced Leymus chinensis Pasture

  • The nutrient composition of pasture, voluntary intake and digestibility of diet ingested by goats grazing on an introduced Leymus chinensis pasture were measured across spring (May), summer (July), autumn (October) and winter (March). In each season, 12 Inner Mongolian Cashmere goats (6 wethers and 6 does with an average live weight of $22.2{\pm}1.3$ kg and $19.5{\pm}0.8$ kg, respectively) were used to graze on a 2 hectares size paddock. Diet selection was observed and the plant parts selected by grazing goats and whole plant L. chinensis were sampled simultaneously. The alkane pair $C_{32}:C_{33}$ and $C_{36}$ were used to estimate intake and digestibility, respectively. The results showed that the plant parts selected by goats had higher crude protein (CP) and lower acid detergent fiber (ADF) and neutral detergent fiber (NDF) than the whole plant, especially in the autumn and winter. The voluntary intake of dry matter (DM), CP, ADF, NDF, and metabolizable energy (ME) by goats was highest in summer (p<0.05). The goats ingested more CP, ME, and less ADF in spring than in autumn (p<0.05). The intakes of DM, CP, and ME were lowest in winter (p<0.05). There were significant differences in nutrient intake between wethers and does in each season, except for the ADF and ME intake per metabolic weight ($LW^{0.75}$). The nutrient digestibilities were higher in spring and summer, and decreased significantly during the autumn and winter (p<0.05). Goats, especially wethers, had a relative constant NDF digestibility across seasons, however, the apparent digestibility of CP in both wethers and does, decreased to negative values in winter. The grazing goats experienced relatively sufficient nutrients supply in spring and summer, and a severe deficiency of CP and ME in winter.

DBD 반응기에서 솔-젤 법으로 제조된 Pt/TiO2 촉매를 이용한 메탄의 플라즈마 전환반응 (Plasma Catalytic Methane Conversion over Sol-gel Derived Pt/TiO2 Catalyst in a Dielectric-barrier Discharge Reactor)

  • 배리어(dielectric-barrier discharge: DBD) 반응기에서 솔-젤 방법으로 제조한 $Pt/TiO_2$를 넣고 플라즈마 에너지를 사용하여 메탄전환반응 연구를 수행하였다. 제조된 촉매는 기존 금속산화물과는 다른 방법으로 환원을 하였으며, 플라즈마를 사용한 환원방법(plasma-assisted reduction: PAR)이라고 명명하였다. $Pt/TiO_2$ 촉매는 Pt 담지량 및 소성온도에 관계없이 20분 이내에 환원이 완료되었다. 3 wt% $Pt/TiO_2$ 촉매와 5 wt% $Pt/TiO_2$ 촉매를 $600^{\circ}C$에서 소성하여 PAR 방법으로 환원했을 때 메탄의 전환율은 40%로 가장 높았다. DBD 반응기에서 $Pt/TiO_2$ 촉매를 사용하였을 경우 촉매가 없을 때보다 $C_2H_6$, $C_3H_8$$C_4H_{10}$과 같은 알칸의 선택성의 매우 높게 나타났다.
