PARAMYOSIN OF THE ABALONE, NOTOHALIOTIS DISCUS (전복 Paramyosin의 분리 및 그 성질)

  • The protein composition of abalone muscle was estimated with the following result: on a series of samples analyzed, water-soluble protein, $19\~22\%$, salt-soluble protein, $27\~39\%$: alkali-soluble protein, $20\~26\%$ : and stroma $20\~28\%$ : respectively. It was demonstrated by ultracentrifugal analysis that approximately $65\%$ of the salt-soluble protein is accounted for by paramyosin, $30\%$ by actomyosin, and $5\%$ by myosin, respectively. The ultracentrifugally homogenous paramyosin was prepared by BAILEY's ethanol-dried method. It showed a $S^{\circ}\;_{20,\;{\omega}$ of 3.14s, and was completely salted in with KCl beyond $0.35{\mu}$. The intrinsic viscosity at $25^{\circ}C$ was estimated at 3.1. The paramyosin is rich in several amino acids such as arginine, aspartic acid, glutamic acid, etc., and lacking of both proline and tryptophane, in rough accord with other paramyosins reported. The abalone paramyosin did not show ATPase activity over a pH range of 5 to 9,5 even in the presence of Ca++ or Mg++. So was the case with the paramyosin specimen prepared by BAILEY's wet-extraction method.

Preparation of Hydroxypropyl Methyl Cellulose with Controlled Solubility Rate by Surface Treatment Reaction (표면처리반응에 의한 용해속도조절용 Hydroxypropyl Methyl Cellulose의 제조)

  • The surface treated hydroxypropyl methylcellulose(HPMC) which could adjust the soluble time was synthesized when 40 wt % glyoxal solution and $KH_2PO_4$ were sprayed and reacted. And also, the solution dynamic at different ratios of two adding agents were identified If the surface of HPMC was treated with only glyoxal, the dispersion characteristics at different ratios of two adding agents were identified If the surface of HPMC was treated with only glyoxal, the dispersion was observed in the neutral solution and the viscosity was increased after directly dissolved as the solution become alkali condition. But the fine-powder type of HPMC which reacted with glyoxal and $KH_2PO_4$ was dispersed regardless of pH of solution and observed that it was dissolved and its viscosity increased after elapsing some time. With increasing amount of glyoxal and $KH_2PO_4$, the soluble time was delayed. The reaction condition was about 60 min at $75{\sim}85^{\circ}C$. Especially, the removal process of organic solvent after reaction was not required due to reaction under water solution without organic during glyoxal and $KH_2PO_4$ treatment. And also, the HPMC which could adjust the soluble rate in water or organic solvent by changing the degree of substitution of HPMC was synthesized.

Inorganic and Organic Solute Pattern of Costal Plants, Korea (해안 식물의 무기 및 유기용질 양상)

  • In order to elucidate the ecophysiological characteristics of coastal plants, we collected them on salt marsh and sand dune, and analyzed inorganic ($Na^+,\;K^+,\;Mg^{2+},\;Ca^{2+}$) and organic solutes (soluble carbohydrate, glycine betaine). Chenopodiaceous plant species (Atriplex gmelini, Salicornia europaea, Salsola collina, Suaeda glauca, Suaeda japonica) showed a tendency to accumulate inorganic ions such as $Na^+\;and\;Cl^-$ instead of $K^+$. However, Chenopodium serotinum which lives in ruderal habitat contained more $K^+$ and less $Na^+$ than the other Chenopodiaceous plants. Most Chenopodiaceous plant species maintained very low level of soluble $Ca^{2+}$ and relatively low concentration of carbohydrates and showed high concentration of glycine betaine which is among the most effective known compatible solutes in the leaves of plant under drought and saline conditions. On the other hand, plant species which belong to Gramineae (Ishaemum anthephoroides, Phragmites communis, Zoysia sinica) and Cyperaceae (Carex kobomugi, Carex pumila) absorbed $K^+$ selectively and excluded $Na^+\;and\;Cl^-$ effectively regardless of habitat conditions, and they accumulated more soluble carbohydrate as osmoticum than Chenopodiaceous plants. These results suggested that physiological characteristics such as high storage capacity for inorganic ions (especially alkali cations, chloride) and the accumulation of glycine betaine in chenopodiaceous plants and $K^+$-preponderance, an efficient regulation of ionic uptake (exclusion of $Na^+\;and\;Cl^-$) and the accumulation of soluble carbohydrate in monocotyledonous plants enable them to grow dry and saline habitats.

Textural Properties and Cell Wall Components of Dried Persimmon according to Varieties (품종에 따른 건시의 물성적 특성과 세포벽 성분)

  • This study is performed to investigate the relation between cell components and hardness which affect to the quality of dried persimmons. Moisture contents of dried persimmons were interrange of 30 to 36%. Enpung Junsi (EJ), Dungsi (Young dong) (DY) and Haman Susi (HS) had higher moisture contents than Dungsi (Sang Ju) (DS), Kojongsi (KS) and Hagakure (HK). Hardness was high in the varieties which has low moisture content such as DS, KS and HK. The significant differences of hardness and fracturability among varieties were appeared conspicuously but those of cohesiveness, adhesiveness and springiness were not appeared. Crude cell wall content, pectin and calcium content of cell wall were high in hard varieties. In the pectin fractions, water soluble pectin content was high in EJ, HS and DY but that of acid and alkali soluble pectin were high in DS, KS and HK. The cell wall of high hardness varieties observed thick and firm than that of low hardness varieties.

Effect of Some Treatments on the Autolysis of Baker's Yeast (Saccharomyces cerevisiae) (빵효모의 자기분해에 대한 몇가지 처리의 효과)

  • Some chemical, biochemical and physical treatments on the baker's yeasts (Saccharomyces cerevisiae) before 12-hour digestion at $50^{\circ}C$ were made to accelerate their autolysis. Dilute alkali treatment with 0.01N-NaOH solution showed a considerable increase in soluble nitrogen extraction only in the 6-hour initial autolysis. The addition of fresh yeast autolysate, 10% to the yeast slurry, could increase the autolysis rate from 65% to 83%. Microwave treatment of yeast slurry for 40 seconds also raised the autolysis rate by about 15%, while longer exposures to the microwave accompanying high temperatures repressed the autolysis process. Extrusion of the yeast cells with high pressures of 16,000 to 20,000 psi brought about significantly higher autolysis rate until the digestion time of 9 hours, but thereafter it showed a gradual drop in soluble nitrogen extraction.

Incorporation of phosphate into protein and other nitrogenous compounds in Chlorella cells (Chlorella 세포에서의 $^{32}P$-인산의 단백질 및 다른 질소화합물로의 전환)

  • In the process of the incorporation of orthophsphate into protein and other cell constituents, the role of inorganic polyphosphate and RNA-polyphosphate complex and the correlation between them were pursued by analyzing the contents of $^{32}P$ and total P in various fractions of Chlorella cells, which had been uniformly labeled with $^{32}P$ before the inoculation in a normal "cold" medium or P-free medium during the culture. The effects of ionizing radiation and various micronutritional-element deficiencies on the phosphate incorporation into, and biosynthesis of, protein and other introgenus compounds in the cells were also observed. When the uniformly $^{32}P$-labeled algae were grown in a normal "cold" medium the contents of $^{32}$ P in the fractions of protein, DNA and RNA-polyphosphate complex increased, but those in the fraction of acid-insoluble polyphosphate decreased. On the other hand, amount of $^{32}P$in the fraction of RNA was almost unchanged in spite of rapid increase of the total P. In the growing period of $^{32}P$-labeled algae in a P-free medium, amounts of $^{32}P$ in the fractions of DNA, protein and lipid increased, while those in the fractions of RNA-polyphosphate and inorganic polyphosphates decreased. When the algal cells were irradiated with about 70, 000r of gamma-rays before the inoculation in the medium, amounts of phosphate in the fractions of DNA, RNA, nucleotides and protein decreased during the culture, compared with those of the control. However, the phosphate content in the fraction of acid-insoluble polyphosphate of the irradiated cells increased than those of the control. In the growing period of the algae in a Mo-free, medium, amounts of acid-soluble total phosphate and nucleotides of the cells increased, while the amounts of residual protein and RNA decresed compared with those of the normal cells. Amounts of alkali-labile protein and phospholipid of the cells grown in a B-free medium decreased, whereas amount of phosphate in acid-soluble fraction increased compared with the control. In general, the contents of protein and RNA in each microelement deficient cells decreased more or less, compared with those in the normal cells.in the normal cells.

  • Vitamin D contents in agricultural products and foods were quantified by high performance liquid chromomatography (HPLC) with a UV/Vis detector, using external standard methods. The results were confirmed with liquid chromatography tandem mass spectrometry (LC-MS/MS). After homogenization, samples were hydrolyzed by direct alkali saponification. Thereafter, fat-soluble components were extracted with n-hexane containing 0.01% butylated hydroxytoluene (BHT). Vitamin D contents in cereals were found to be in the range of $1.882{\sim}4.856{\mu}g/100g$. Juda's ear and oak mushroom contained high amounts of vitamin D, at 363.85 and $199.42{\mu}g/100g$ of edible portion, respectively.

Conditions for the Extraction of Polyphenols from Radiata Pine (Pinus radiata) Bark for Bio-Foam Preparation

  • The use of polyphenol extracts from radiata pine (Pinus radiata) bark as raw materials for bio-foams was investigated along with the optimal NaOH extraction conditions. The targeted yield of alkaline extracts was 60%, and the targeted pH was 11 to 12. The radiata pine bark was composed of 70% of a 1% NaOH extract, which contained mainly polyphenols, such as proanthocyanidin (PA). As the particle size of the bark decreased, the yield of the 1% NaOH extracts increased from 57 to 87%. A range of NaOH concentrations, liquor ratios, and extraction times were explored to establish an economic polyphenol extraction method. More than 60% of the alkaline extract was extracted, and the pH of the extract was approximately 12 when the optimum extraction conditions were employed, i.e., a liquor to bark ratio of 5:1, a NaOH dosage of 17 to 18% based on the bark weight, and a 1 h extraction time. Following neutralization of the alkaline extract, structural analysis indicated severe structural changes in the PA during the alkaline extraction. Because the alkaline extract was barely soluble in the solvent used for the structural analyses, it is assumed that chemical modification is required to increase the solubility of the alkaline extract for the production of bio-foams.

Preparation of Fine Silk Powder and It′s Application for Surface Modification (폐견사류의 미세분말화 및 표면 가공제 적용)

  • The purification, dissolution and powdering of stained waste silk obtained from weaving and dyeing process were studied for the surface modification of textile fabric and plastic materials. The whiteness of stained waste silk could be improved through degumming and bleaching with sodium hydrosulfite. The water-soluble fibroin solution can be obtained by dissoving the degummed waste silk in a boiling solution of 50% calcium chloride for 60 minutes. The salts and heavy metals contained in fibroin solution were removed by electric dialysis, wool fiber filtration and gel filtration chromatography. The fibroin powder was prepared by using a fine grinder after the alkali treatment for weakening the silk fiber. The fine fibroin powder of particle size around 30 ㎛ was obtained with a ultra fine-mill, while it was finer below 10 ㎛ with a ball-mill. The dissolved or powdered silk was applied to the surface of fabric with addition of the binder (a urethane resin). The moisture content of polyester and nylon fabrics treated with the silk solution was improved due to hygroscopic property of silk. The fine fibroin powder mixed with the binder ws coated on the surface of synthetic film by use of the air pressed sprayer. It was revealed that the hygroscopicity as well as the softness of fibroin powder coated film was much improved. Therefore, it is thought that the fine silk fibroin powder is applicable as an coating agent for the surface modification of plastic and synthetic leather.

Green Photoresist Stripping Process with the Influence of Free Surface from Dip Withdrawal (Dip 추출에서 유체 표면의 영향을 고려한 친환경 포토레지스트 박리공정)

  • This paper describes a green stripping process to effectively strip the remaining DFR layer on a non-alkali-based ITO glass surface after an etching process. A stripper, water-soluble amine compound, is used to investigate the characteristics of stripping ability and to suggest a valid method for the green process. Increasing the composition (5-30% concentration) of the ethanol amine-based stripper was found to greatly reduce the stripping time applied in the dipping method. The composition (30%) achieved an excellent stripping effect and free-residue impurities. Additionally, it was possible to obtain the effect of stripping in a way to sustain the release before generating DFR sludge from the ITO glass surface by using dipping condition (stripping time) in the composition. An Additional stripping process (buffering) out of dipping can realize productivity improvement and cost reduction because of the higher proportion of re-use of the stripping solution used in the DFR removal step.
