A novel method for cell counting of Microcystis colonies in water resources using a digital imaging flow cytometer and microscope

  • Microcystis sp. is one of the most common harmful cyanobacteria that release toxic substances. Counting algal cells is often used for effective control of harmful algal blooms. However, Microcystis sp. is commonly observed as a colony, so counting individual cells is challenging, as it requires significant time and labor. It is urgent to develop an accurate, simple, and rapid method for counting algal cells for regulatory purposes, estimating the status of blooms, and practicing proper management of water resources. The flow cytometer and microscope (FlowCAM), which is a dynamic imaging particle analyzer, can provide a promising alternative for rapid and simple cell counting. However, there is no accurate method for counting individual cells within a Microcystis colony. Furthermore, cell counting based on two-dimensional images may yield inaccurate results and underestimate the number of algal cells in a colony. In this study, a three-dimensional cell counting approach using a novel model algorithm was developed for counting individual cells in a Microcystis colony using a FlowCAM. The developed model algorithm showed satisfactory performance for Microcystis sp. cell counting in water samples collected from two rivers, and can be used for algal management in fresh water systems.

Chlorophyll-a의 히스토그램 분석을 통한 녹조발생 우심지역 분류 (Classification of the Algal Monitoring Points by Histogram Analysis of Chlorophyll-a)

  • 본 연구에서는 Chlorophyll-a (Chl-a)값을 빈도분석인 히스토그램(histogram)을 통해 녹조관리가 필요한 지점을 분류 하고자 하였다. 누적도수분포곡선(ogive curve)에 따라 지점별 녹조발생정도를 분석하였고, 히스토그램의 형태에 따라 관리지점을 3단계로 분류하였다. 4대강 중 한강에서 저농도의 Chl-a가 가장 많이 나타났고, 금강과 영산강 지점에서 높은 농도의 Chl-a가 빈번하게 나타났다. 히스토그램의 패턴에 따라 녹조의 관리 정도를 분류 할 수 있었는데, 한강의 경우 집중관리가 필요한 지점이 나타나지 않았고, 금강과 영산강은 대부분의 지점에서 녹조관리가 필요한 것으로 나타났다. 마지막으로 낙동강은 중류 및 하류 지점부터 녹조관리가 필요한 지점으로 분류되었다. 본 연구결과를 통해 빈도분석이 녹조발생의 우심지역 판단의 보조지표 활용 가능성 여부를 확인하였다.

대청호 유속에 따른 조류이동 영향 (The Relationship between Algae Transport and Current in the Daecheong Reservoir)

  • Water quality in the Daecheong reservoir has been deteriorated by algal bloom every year. Algal bloom is propagated from eutrophicated tributary into the main body of the reservoir during the wet season. Nutrients from diffuse sources trigger the propagation of the algal bloom. This study is aimed to analyze relationship between the water current by the simulation and algae transport from the main body in the Daecheong reservoir including tributary where algal bloom has occurred seriously every year. Water quality model CE-QUAL-W2 was applied to analyze water movement in draught season (2001) and flooding season (2003). The result of simulation corresponded with the observed water elevation level, 63~80 m and showed stratification of the Daecheong reservoir. In the draught season, as velocity and direction off low in the reservoir was estimated to affect algae transport including nutrient supply from small tributary, algal blooms occurred in the stagnate zone of middle stream of the reservoir. On the other hand, in the flooding season, it was resulted in nutrient transport from upstream of main reservoir and nutrients were delivered up to downstream by fast water velocity. In result, algal blooms occurred in stagnate zone of reservoir downstream as the current of downstream was retarded according to dam outflow control.

Prediction of short-term algal bloom using the M5P model-tree and extreme learning machine

  • In this study, we designed a data-driven model to predict chlorophyll-a using M5P model tree and extreme learning machine (ELM). The Juksan weir in the Youngsan River has high chlorophyll-a, which is the primary indicator of algal bloom every year. Short-term algal bloom prediction is important for environmental management and ecological assessment. Two models were developed and evaluated for short-term algal bloom prediction. M5P is a classification and regression-analysis-based method, and ELM is a feed-forward neural network with fast learning using the least square estimate for regression. The dataset used in this study includes water temperature, rainfall, solar radiation, total nitrogen, total phosphorus, N/P ratio, and chlorophyll-a, which were collected on a daily basis from January 2013 to December 2016. The M5P model showed that the prediction model after one day had the highest performance power and dropped off rapidly starting with predictions after three days. Comparing the performance power of the ELM model with the M5P model, it was found that the performance power of the 1-7 d chlorophyll-a prediction model was higher. Moreover, in a period of rapidly increasing algal blooms, the ELM model showed higher accuracy than the M5P model.

경기도 경안천에 설치한 메소코즘 내에서의 동물플랑크톤, 어류, 수생식물, 인공식물섬에 의한 조류저감효과 연구 (Effect of Algal Bloom Control Using the Mesocosms Installed with Zooplankton, Fishes, Aquatic Macrophytes and Artificial Marshy Land in Kyungan Stream)

  • We installed mesocosms including zooplankton, fishs, artificial marshy land and aquatic macrophytes in Kyoungan stream to study the reduction effects of algae from Sep. 16 to Oct. 28, 2011. The control tendency of phytoplankton taxa was compared by analyzing community structure and dominant species in each mesocosm. Under the condition where Bluegill was absent, Daphnia similoides showed good effect of algal control since it has large food area and has high grazing pressure. Bluegill selectively preys upon large zooplankton, Daphnia similoides, as it also preys on small zooplanktons that flow in. In condition that Bluegill was absent, Daphnia similoides preyed selectively large phytoplankton (Cryptomonas ovata). Due to the shading of light, removal of nutrients and providing refuge for small zooplanktons, aquatic macrophytes and artificial marshy land showed high level of algal control. In corrals with aquatic macrophytes and artificial marshy land, the dominancy of genus Coelastrum and Pediastrum, which are difficult for small zooplanktons to feed on, relatively increased. In conclusion, under conditions of small number of predators such as Bluegill, Daphnia similoides is thought to be useful in algal bloom; however, when lots of predators are present, using small sized zooplanktons along with artificial marshy land and aquatic macrophytes is thought to be more useful than using large zooplanktons.

Mitigation of Harmful Algal Blooms by Sophorolipid

  • A new method was proposed to control Harmful Algal Blooms (HABs) by a biosurfactant sophorolipid. The effect of sophorolipid on the growth, motility, precipitation, and recovery of algal cells was investigated for four common HAB species, Scripsiella trochoidea, Prorocentrum minimum, Cochlodinium polykrikoides, and Heterosigma akashiwo. The motility and growth of algal cells were inhibited significantly at the concentration of 20 and 5 mg/l sophorolipid, respectively, and no recovery was observed under the above concentrations. The concentration of 20 mg/l sophorolipid was considered to be an effective concentration for the mitigation of HABs. A sedimentation test suggested that the maximum precipitation occurred at the end of 1 h, and the algicidal effect of sophorolipid was observed by a microscope. Comparative study showed that sophorolipid had marked algicidal capability. Analysis on biodegradability, toxicity, and cost effectiveness further demonstrated the potential of sophorolipid in future HABs mitigation.

MBOD법에 의한 낙동강의 조류증식 제한인자 추정 (Evaluation of Algal Growth Limiting Factor in the Nakdong River by MBOD Method)

  • The increase of population and industrial activities had brought into eutrophication in the Nakdong river. A remarkable acceleration of eutrophication brought about serious problems for water supply. Therefore, for the purpose of conservation of water quality in the Nakdong river it is necessary to control nutrients. MBOD method was use to evaluate algal growth limiting factor and algal growth potential in the Nakdong river from June to August 1994. The modified biochemical oxygen demand(MBOD) depends on the amount of available inorganic nutrient and organic substrate during 5 day incubation in the dark at 2$0^{\circ}C$. The MBOD assay depends on inorganic nutrients such as P and N as well as reduced carbon and called the MBOD, the MBOD-P, and the MBOD-N, respectively. The results of bioassay by MBOD(Modified BOD) method showed that the MBOD, MBOD-P and MBOD-N value were found to be in the ranges of 3.8~96.0 mg$O_2$/l, 5.6~94.0 mg$O_2$/l and 42.0~220 mg$O_2$/l, respectively. And the the bioassay value was found to be the highest in Koryong area and the lowest in Waekwan area throughout the Nakdong river. The variations of MBOD-P and MBOD-N value showed similar tendencies to the variations of phosphorus and nitrogen value, respectively. By MBOD method, the relationships of MBOD, MBOD-P and MBOD-N value were MBOD ≒ MBOD-P 《 MBOD-N. The MBOD value was nearly equal to the MBOD-P value, and the MBOD-N value was 3 to 20 times more than the MBOD-P value, approximately. Therefore, in the Nakdong river, phosphorus was the limiting factor for algal growth during summer season. The algal growth potential as the concentration of chlorophyll-a in the summer was maximum 5 times more than standing crop as it.

Effects of Nitrogen Sources and C/N Ratios on the Lipid-Producing Potential of Chlorella sp. HQ

  • Microalgae are being researched for their potential as attractive biofuel feedstock, particularly for their lipid production. For maximizing biofuel production, it is necessary to explore the effects of environmental factors on algal lipid-producing potential. In this study, the effects of nitrogen (N) sources (NO2-N, NO3-N, urea-N, NH4-N, and N-deficiency) and carbon-to-nitrogen ratios (C/N= 0, 1.0, 3.0, and 5.0) on algal lipid-producing potential of Chlorella sp. HQ were investigated. The results showed that for Chlorella growth and lipid accumulation potential, NO2-N was the best amongst the nitrogen sources, and NO3-N and urea-N also contributed to algal growth and lipid accumulation potential, but NH4-N and N-deficiency instead caused inhibitory effects. Moreover, the results indicated that algal lipid-producing potential was related to C/N ratios. With NO2-N treatment and carbon addition (C/N = 1.0, 3.0, and 5.0), total lipid yield was enhanced by 12.96-20.37%, but triacylglycerol (TAG) yields decreased by 25.52-94.31%. As for NO3-N treatment, carbon addition led to a 17.82-57.43%/25.86-82.67% reduction of total lipid/TAG yields. When NH4-N was used as the nitrogen source, total lipid/TAG yields were increased by 46.67-113.33%/28.99-74.76% with carbon addition. The total lipid/TAG yields of urea-N treatment varied with C/N ratios. Overall, the highest TAG yield (TAG yield: 38.75 ± 5.21 mg/l; TAG content: 44.16 ± 4.35%) was achieved under NO2-N treatment without carbon addition (C/N = 0), the condition that had merit for biofuel production.

규산질다공체와 미생물응집제의 녹조제어 효과 (Effects of CellCaSi and Bioflocculant on the Control of Algal Bloom)

  • 부영양화된 연못에서 규산질다공체(CelICaSi)와 미생물응집제를 이용한 녹조제어의 효과를 조사하였다. 녹조 발생 남조류인 Microcystis aeruginosa에 대한 응집능이 우수한 S-2 균주가 생산한 미생물응집제를 선정하여 현장의 조류응집에 적용하였다. 초기의 인산염농도는 대조구에서 $131\mu{g}\ell^{-1}$를 기록하였음에 비해서, CelICaSi가 첨가된 3개의 처리구에서는 $1-14\mu{g}\ell^{-1}$를 기록하며 크게 감소하였다. 엽록소$-\alpha$ 농도는 현장에 설치한 en-closure에서 초기에 $215\mu{g}\ell^{-1}$ 이었으나 CellCaSi $(1g\ell^{-1}$, 미생물응집제 $(2ml\ell^{-1})$, 보조응집제 $(1g\ell^{-1})$, 염화제이철 $(2mg\;Fe\ell^{-1})$을 함께 처리한 처리구에서 $59\mu{g}\ell^{-1}$로 가장 많이 감소하였다. 실험용 쥐 및 금붕어를 이용한 독성실험에서 녹조제거에 이용되는 성분들과 미생물응집제에 대한 독성은 없는 것으로 조사되었다. 따라서, CellCaSi와 미생물응집제는 인 제거 및 엽록소-$\alpha$ 농도 감소에 의한 녹조제거에 효과적인 것으로 판단되었다.

대청호 조류발생에 따른 경보제 운영 및 개선 방안 고찰 (A study on the management and improvement of alert system according to algal bloom in the Daecheong Reservoir)

  • Following the industrialization and urbanization in Korea, algal bloom causes aesthetic displeasure and many other problems such as taste and odor, coloration, scum, increase in pH, filter-bed blockage. There were some cases involving human death by microcystins during summertime in foreign countries. In Korea, Harmful cyanobacteria such as Microcystis and Anabaena develop in summer in the Daecheong reservoir, one of the main water resources, with the retention time of above 200 days. To better control algal bloom, the Ministry of Environment has been running algal bloom alert system from 1998 for the Daecheong reservoir, which needs to be improved to reflect the characteristics of river-type lakes. For this reason, we try to find new measures to improve an algal bloom alert system for each water zone considering the characteristics of harmful cyanobacteria in this study.
