Activity of Chlorelaa vulgaris Associated by Escherichia coli W3110 on Removal of Total Organic Carbon in Continuous River Water Flow System

  • We investigated the association of Chlorella vulgaris and E. coli W9110 in removal of total organic carbon with the lab-scaled continuous river water flow system (CRWFS). Artificial wastewater was applied at two levels of organic carbon concentration; 1,335 $mg{\cdot}l^{-1}$ in the treatment (T)-1 and 267 $mg{\cdot}l^{-1}$ in T-2. The highest densities of C. vulgaris were $8.3{\times10^6\;cells{\cdot}ml^{-1}$ in T-1 and $6.9{\times}10^6\;cells{\cdot}ml^{-1}$ in T-2. The maximum densities of E. coli W3110 were $2.0{\times}10^8$ clony forming unit (CFU)${\cdot}ml^{-1}$ in T-1 and $3.9{\times}10^8\;CFU{\cdot}ml^{-1}$ in T-2. The densities increased during the first 11 days in T-q and 4 days in T-2, and decreased rapidly till 35th day, then increased slightly afterwards. This trend was prominent in T-2. It was inplied that wider range of nutrients was required in the growth of heterotrophic bacteria in T-2 than in T-1. The algal biomass should be increased effectively for the successful removal of organic carbon.

생물검정에 의한 남조류 Microcystis가 수질에 미치는 영향 (The Impact on Water Quality from Blue-Green Algae Microcystis Natural Phytoplankton by Algal Assay)

  • In order to understand the impact for decomposition of blue-green algae Microcystis on water quality, the algae were cultivated with collection of natural population during approximately one month, when water-bloom of Microcystis dominated at August 31, 1999 in the lower part of the Okchon Stream. The enrichment of inorganic NㆍP nutrients didn't in algal assay and the effect of Microcystis on water duality was assessed from the variation of nutrients by algal senescence. Microcystis population seemed to play a temporary role of sink for nutrients in the water body. Initial algal density of Microcystis was 2.3×10/sup 6/ cells/㎖. When Microcystis population died out under light condition, algal NㆍP nutrients between 9∼12 days affected to increase of biomass after reuse by other algal growth as soon as release to the ambient water. However, cellular nutrients under dark condition were almost moved into the water during algal cultivation. NH₄, NO₃ and SRP concentration were highly increased with 160, 17 and 79 folds, respectively relative to the early. As a result, the senescence of Microcystis population seemed to be an important biological factor in which cause more eutrophy and increase of explosive algal development by a lot of nutrients transfer to water body. There are significantly observed an effort of reduce for production of inner organic matters such a phytoplankton as well as load pollutants from watershed in side of the water quality management of reservoir.

Ultrastructures of Colletotrichum orbiculare in Cucumber Leaves Expressing Systemic Acquired Resistance Mediated by Chlorella fusca

  • Chlorella, one single-cell green algae organism that lives autotrophically by photosynthesis, can directly suppress some plant diseases. The objective of this study was to determine whether pre-spraying with Chlorella fusca suspension could induce systemic acquired resistance (SAR) in cucumber plants against anthracnose caused by Colletotrichum orbiculare. In order to illustrate SAR induced by algae, infection structures in host cells were observed under a transmission electron microscope (TEM). Cytological changes as defense responses of host mesophyll cells such as accumulation of vesicles, formation of sheath around penetration hyphae, and thickness of cell wells adjoining with intracellular hyphae were demonstrated in cucumber leaves. Similar defense responses were also found in the plant pre-treated with DL-3-aminobutyric acid, another SAR priming agent. Images showed that defense response of host cells was scarcely observed in untreated leaf tissues. These cytological observations suggest that C. fusca could induce SAR against anthracnose in cucumber plants by activating defense responses of host cells.

Effect of Different Substrate Characteristics on Abundance and Community Structure of Epilithic Diatoms in Two First-Order Streams

  • The abundance and community structure of epilithic diatoms grown on different substrata were investigated in two first-order streams located in a limestone and granite area, north of the Suzuka Mountains in Central Japan. Experiments were conducted as follows: limestone and granite without algae were submerged in their own streambed or another stream station and incubated for seven weeks, while limestone and granite with algae were transferred to another stream station and incubated at the same time. The diatom biomass was consistently high in the lime-stone station experiments compared to those at the granite station. In addition, there was more diatom biomass on granite substrata than on the limestone substrata at both stations. The present results suggested that the difference in water chemistry including the major nutrient concentrations was the limiting factor for algal growth in these two streams; however, when the water chemistry was the same in each stream, the difference in substratum characteristics became the important factor affecting the diatom abundance. The diatom community grown on the transferred substrata with and without algae became similar to those grown on the original substrata in each stream during the incubation period. It was suggested that the effect of the substrata characteristics on the diatom community structure was rather small.

Cytotoxic Activities of Red Algae Collected from Jeju Island Against Four Tumor Cell Lines

  • Methanolic and aqueous extracts of 26 red algae species collected from Jeju Island coast were prepared at a high $(70^{\circ}C)$ and a room temperature $(20^{\circ}C)$ and were examined for their cytotoxic activity against 4 tumor cell lines: U-937 (human monoblastoid leukemia cell line), HL-60 (human promyelocytic leukemia cell line), B-16 (murine melanoma cell line) and HeLa (woman cervical carcinoma cell line). $20^{\circ}C$ methanolic extract of Polysiphonia japonica showed cytotoxic activity of over 50% against U-937, HL-60 and B-16 cells. On the other hand, the $20^{\circ}C$ aqueous extract of Scinaia okamurae and $70^{\circ}C$ aqueous extract of Chondrus crispus showed cell growth inhibition activity of more than 50% against HL-60 and B-16 cells. The highest cytotoxic activity was observed in the $20^{\circ}C$ aqueous extract of Scinaia okamurae against B-16 cells (80.55%).

The Morphological and Growth Characteristics of Two Strains of Fibrocapsa japonica Isolated from New Zealand and Japan

  • The blooms caused by Fibrocapsa japonica have occurred regularly in New Zea1andean coastal waters, and no fish kills and economic impacts have been reported. However, Fibrocapsa japonica in Japan killed caged juvenile fish and has been regarded as a harmful micro algae. In this comparative study a New Zealand isolate was found to be morphologically similar to a Japanese isolate, although slightly larger on average than. Optimal temperatures for growth differed, with fastest growth rates occurring at $22-24^{\circ}C$ for the New Zealand strain and $16-22^{\circ}C$ for the Japanese strain, with a decrease in growth rate exhibited by the latter at $25^{\circ}C$. Both isolates had low salinity optima of 20-25 PSU, although they grew between 15 and 30 PSU. Growth declined significantly for both strains above 30 PSU.

Production Ecology of the Seagrass Zostera marina in Jindong Bay, Korea

  • Production dynamics of eelgrass, Zostera marina was examined in Jindong Bay on the south of the Korea peninsula. Eelgrass leaf productivities and environmental factors such as underwater photon flux density, water temperature, and nutrient availabilities in the water column and sediments were monitored from March 2002 to December 2003. While water temperature exhibited a distinct seasonal trend, underwater irradiance and nutrient availabilities exhibited high degree of fluctuation, and did not show a seasonal trend throughout the experimental periods. Eelgrass leaf elongation and production rates showed significant seasonal variations. Leaf productivity was highest in May (30.0 mg dry wt sht$^{-1}$ d$^{-1}$ or 3.7g dry wt m$^{-2}$d$^{-1}$) and lowest in November (3.2 mg dry wt sht$^{-1}$ d$^{-1}$ or 0.12 g dry wt m $^{-2}$ d$^{-1}$). Eelgrass leaf productivities did not show a strong correlation with underwater irradiance or environmental nutrient availabilities. The production rates, however, were positively correlated with water temperature during spring periods, and were correlated negatively at high water temperature exceeded 20℃ during summer months. While relative growth rates were highest in spring and lowest in high water temperature periods, plastochrone interval was longest during summer and shortest during spring. These results imply that seasonal growth dynamics of eelgrass, Z. marina was mainly controlled by water temperature.

Effects of the Loess Coating on Seed Germination and Seedling Growths of the Eelgrass, Zostera marina

  • Seagrass bed is an important component in coastal and estuarine ecosystems, providing food and habitats to a wide variety of marine organisms. Recently, seagrass coverage has declined significantly due to anthropogenic impacts such as cultural eutrophication and reclamation, and thus efforts are under way to prevent further losses and restore disturbed seagrass habitats worldwide. Seagrass transplantation techniques for habitat restoration include vegetative and seed-based methods. Seagrass seeds can be collected easily, and sowing seeds is an economically effective method for large-scale restoration. However, large numbers of seed can be lost by seed predation and physical disturbance in the planting areas. In the present study, Zostera marina seeds were coated with loess to reduce seed loss by predation and sweeping away by the water currents, and germination rates of coated seeds and seedling growth were examined to assess the feasibility of the seed-coating method for large-scale restoration. Germination rate of the coated seeds with loess was significantly higher than that of the uncoated seeds. Additionally, seedling growths were not significantly different between the coated and the uncoated seeds. These results suggest that coating of eelgrass seeds with loess enhances success of seed germintion with no harmful effects on seedling growth. Therefore, the seed coating method using loess may be an effective and applicable seedbased transplanting technique for large-scale restoration.

Calcium-related genes associated with intracellular calcification of Emiliania huxleyi (Haptophyta) CCMP 371

  • Emiliania huxleyi (a haptophyte) is the most abundant coccolithophore species that produces delicate calcite scales called coccoliths. In this study, we identified several candidate genes associated with coccolith production by comparing the transcriptomes of the calcifying (CCMP 371) and non-calcifying (CCMP 2090) strains of E. huxleyi. Among the candidates, genes highly expressed in CCMP 371 were identified. To confirm whether these genes are associated with calcification, we modulated coccolith production in CCMP 371 by culturing it at different calcium concentrations. At an ambient (10 mM) concentration of calcium in the growth medium, CCMP 371 sustained its calcifying ability. However, at a low (0.1 mM) concentration or absence of calcium, there was no calcite formation, demonstrating that calcium-limiting conditions negatively affect calcification. We also evaluated the expression patterns of the putative genes in cells grown at different calcium concentrations by quantitative reverse transcription polymerase chain reaction. In addition, we showed that the growth rate of cells cultured under calcium-limiting conditions does not differ from that under ambient conditions. Further studies are required to investigate the roles of the putative calcification-associated genes at the molecular level.

팔당호 수초재배섬의 운영 사례 분석 (Analysis of Floating Island Operation in the Paldang Lake)

  • The floating island was investigated in order to make a purification of lake water quality with the uptake of nutrients, the prevention of algae growth and the supply of habitat for aquatic plants. Although the concentrations of organics and nutrients inside facility were shown higher than outside, the quantitative analysis was not attainable. Nitrogen uptakes by Cattail, Phragmites japonica, Reed and Wild rice were shown 48.2, 98.0, 111.1 and $72.0g/m^2{\cdot}year$, respectively, and in the case of phosphorus they were 3.2, 8.3, 6.1 and $4.6g/m^2{\cdot}year$, respectively. Considering total floating areas of $2,560m^2$, the nitrogen uptake is totally estimated to $210.7kg/m^2{\cdot}year$ and $14.2kg/m^2{\cdot}year$ for the phosphorus uptake. In order to promote the spread of floating island, the selection of aquatic plants, the control of growth rate properly and the method of quantitative analysis for the purification of water quality and the application to the stream water should be thoroughly surveyed from all angles prior to the application.
