Screening of Extracts from Red Algae in Jeju for Potentials MarineAngiotensin - I Converting Enzyme (ACE) Inhibitory Activity

  • This study was conducted to screen in vitro angiotensin - I converting enzyme (ACE) inhibitory activities of methanol (MeOH) and aqueous extracts at 20°C and 70°C, respectively, prepared from twenty-six red algae obtained from the coast of Jeju Island in Korea. Among aqueous extracts at 20°C (20AE) from red algae Lomentaria catenata showed the strongest ACE inhibitory activity and Lithophyllum okamurae recorded the second highest activity. From MeOH extract at 20°C (20ME) Ahnfeltiopsis flabelliformis possessed the strongest ACE inhibitory activity. Remarkable activities from MeOH extracts at 70°C (70ME) were observed in Grateloupia filicina, Sinkoraena lancifolia and Grateloupia lanceolata. However, no significant activity was found in aqueous extracts at 70°C (70AE). The IC50 values, which are concentrations required to inhibit 50% activity of ACE, for ACE inhibitory activities of 20AE from Lithophyllum okamurae and L. catenata were 13.78 and 12.21 μg mL–1, respectively. The IC50 values of 20ME from A. flabelliformis and Laurencia okamurae were 13.84 and 106.15 μg mL–1. Those of the 70ME from Bonnemaisonia hamifera, Grateloupia filicina, Sinkoraena lancifolia, G. lanceolata, Gracilaria vermiculophylla and L. okamurae ranged from 25.82 to 124.69 μg mL–1.

해조류 추출색소의 기능성을 활용한 립밤 제조 (Preparation of Lip Balm Utilizing Functionalities of Colorants Extracted from Marine Algae)

  • The aim of this study is to utilize the algae extracts with functionalities such as antioxidant and antimicrobial properties for preparing lip balm. Functional properties of extracts from Dictyota coriacea and Ulva pertusa were evaluated in terms of DPPH radical scavenging activity and antimicrobial activity. Lip balms contained the algae extracts were tested its efficacy by measuring the moisture retention of lip skin after applying human subjects. According to the results of skin moisture retention measurement, lip balm was effective to increase the moisture retention of lip skin. It is considered that the utilization of colorants extracted from algae which are verified of antioxidant effect and antimicrobial activity is valuable and useful in developing the cosmetic products.

  • The antioxidant activity of methanol extracts of plants and marine algae was tested by using 1,1-diphenyl-2-picrylhydrazyl(DPPH). Five plant extracts(Prunus davidiana, Eriobotrya japonica, Artemisia iwayomogi, Stirodella tolyrrhiza and Ulmus davidiana) and two algae (Ecklonia stolonifera and Symphycoladia latiuscula) were found to be the most effective in DPPH radical scavenging activity. The methanol extract obtained from the stems of Prunus davidiana was fractionated with several solvents. The ethylacetate soluble fraction exhibiting the strongest antioxidant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Antioxidant flavonoids and flavonoid glycosides were isolated and the most active ones was identified as (+)-catechin by MS, $^1H-NMR$ and $^{13}C-NMR$. Its antioxidant activity was higher than that oil vitamin C.

해조류 추출물이 섬유아세포의 증식에 미치는 영향 (Cell proliferation effect of brown marine algae extracts on Mouse Fibroblast)

  • We examined cell regeneration efficiency of brown marine algae living in Jeju coast for search of a novel therapeutic device with cutaneous wound healing materials. The five algae were collected and compared with epidermal growth factor (EGF) as a positive control in the assays of cell proliferation and cell migration of NIH3T3 fibroblast cells. Among the 80% methanol extracts of these brown algae, the two algal extracts from Ishige foliacea and Colpomenia bullosa showed the proliferative effects of the cells similar to the effect of EGF. Besides it was found that Colpomenia bullosa extract significantly enhanced cell migration of NIH3T3 cell. In the study, therefore, we confirmed that the Colpomenia bullosa extract improved proliferation of NIH3T3 cell and a potential candidate for cultaneous wound healing.

Inhibitory Effects of Brown Algae Extracts on Histamine Production in Mackerel Muscle via Inhibition of Growth and Histidine Decarboxylase Activity of Morganella morganii

  • This study was performed to investigate the inhibitory effects of brown algae extracts on histamine production in mackerel muscle. First, antimicrobial activities of brown algae extracts against Morganella morganii were investigated using a disk diffusion method. An ethanol extract of Ecklonia cava (ECEE) exhibited strong antimicrobial activity. The minimum inhibitory concentration (MIC) of ECEE was 2 mg/ml. Furthermore, the brown algae extracts were examined for their ability to inhibit crude histidine decarboxylase (HDC) of M. morganii. The ethanol extract of Eisenia bicyclis (EBEE) and ECEE exhibited significant inhibitory activities (19.82% and 33.79%, respectively) at a concentration of 1 mg/ml. To obtain the phlorotannin dieckol, ECEE and EBEE were subjected to liquid-liquid extraction, silica gel column chromatography, and HPLC. Dieckol exhibited substantial inhibitory activity with an $IC_{50}$ value of 0.61 mg/ml, and exhibited competitive inhibition. These extracts were also tested on mackerel muscle. The viable cell counts and histamine production in mackerel muscle inoculated with M. morganii treated with ${\geq}2.5 $ MIC of ECEE (weight basis) were highly inhibited compared with the untreated sample. Furthermore, treatment of crude HD-Cinoculated mackerel muscle with 0.5% ECEE and 0.5% EBEE (weight basis), which exhibited excellent inhibitory activities against crude HDC, reduced the overall histamine production by 46.29% and 56.89%, respectively, compared with the untreated sample. Thus, these inhibitory effects of ECEE and EBEE should be helpful in enhancing the safety of mackerel by suppressing histamine production in this fish species.

Investigation of Antimicrobial Activity of Brown Algae Extracts and the Thermal and pH Effects on Their Activity

  • The antimicrobial activity of water and ethanol extracts from 30 species of algae was measured using the agar diffusion method and minimum inhibitory concentration (MIC) test. In agar diffusion method, the 95% ethanol extracts from 12 of the algae showed growth inhibition against the tested microorganisms. In particular, Ishige okamurai, Ecklonia stolonifera, Sargassum siliquastrum, Sargassum thunbergii, Colpomenia bullosa, and Ecklonia cava had strong antibacterial activities against Gram-positive bacteria at 4 mg/mL. In the results of the MIC test, S. siliquastrum showed the most antimicrobial activity, where its MIC values ranged from 0.005 to 0.0075% against Listeria monocytogenes, Clostridium perfringens, and Basillus subtilis. In the thermal stability test, for the ethanol extracts of I. okamurai, E. cava, S. siliquastrum, S. thunbergii, and C. bullosa, the extracts proved to maintain high antimicrobial activities when they were treated at $121^{\circ}C$ for 15 min. In the pH stability test, the antimicrobial activity of the S. siliquastrum ethanol extract was stable from pH 2 to 10, whereas the activity of the other species ethanol extracts were weakened under pH 10 against several microbes.

  • Recently, sea algae cultivation as carbon sink and carbon dioxide fixation have been considered. Also, various researches on bioenergy derived from sea algae and the utilization of fibers, saccharide, and lipid of sea algae have been performing. Till now, algae fibers has been used for manufacturing of paper and reinforcing of polymer composites and the extracts of sea algae are used for cosmetics, pharmaceutical materials and food such as agar. Especially, algae fiber has so similar properties to cellulose in terms of crystallinity and functional groups that it can be utilized as reinforcements of biocomposites. Biocomposites as alternatives of glass fiber reinforced polymer composites are environmentally friendly polymer composites reinforced with natural fibers and are actively applying to the automobiles and construction industries. In this paper, characteristics of algae fiber and biocomposites reinforced with algae fiber as environmentally friendly energy materials have been introduced.

제주도 연안 서식 해조류에 대한 주요 활성성분 및 항산화효능 연구 (Studies on Bioactive Substances and Antioxidant Activities of Marine Algae from Jeju Island)

  • The objective of this study was to investigate the useful components and potential antioxidant activities of 30 marine algae, collected from Jeju Island in Korea. The contents of bioacitve substance of extracts from marine algae, such total polyphenol and carbohydrate, were determined. The extract of Distromium decumbens had the highest amount of total polyphenol content (21.27%), and that of Gracilaria incurvata Okamura had the highest amount of total carbohydrate content (10.18%). The antioxidant activities of extracts obtained from algae were tested through the evaluation of DPPH radical and hydroxyl radical scavenging activity. The extracts of Distromium decumbens, Sargassum hemiphyllum (Turner) C.Agardh, Sargassum serratifolium (C. Agardh) C. Agardh and Acrosorium yendoi Yamada were found to have more than 80% DPPH radical scavenging activity and that of Dictyota okamurae (Dawson) $H{\ddot{o}}tning$, Schnetter, et Prud'homme van Reine, Myagropsis myagroides (Martens ex Turner) Fensholt, Sargassum serratifolium (C. Agardh) C. Agardh and Cladophora wrightiana Harvey showed more than 50% hydroxyl radical scavenging activity. These results suggest that algae collected from Jeju Island would be good raw materials for antioxidant.

해조류 추출물과 페놀성화합물의 in vitro 및 in vivo 간보호활성 (Hepatoprotective Effect of Extracts and Phenolic Compound from Marine Algae in Bromobenzene-treated Rats)

  • The methanol extracts of some marine algae were tested for investigating the effects on the formation of lipid peroxide and the activities of free radical generating enzyme in vitro in bromobenzene-treated rat. The extracts of Enteromorpha compressa, Capsosiphon fulvescens, Gelidium amansii, Hizikia fusiformis, Sargassum siliquastrum and Sargassum thunbergii which decreased the formation of lipid peroxide, inhibited the activity of xanthine and aldehyde oxidases by adding of each extracts. Phloroglucinol isolated from Ecklonia stolonifera reduced bromobenzene-induced hepatic lipid peroxidation. This compound administered daily over one week before intoxication with bromobenzene did not affect the activities of aminopyrine N-demethylase, aniline hydroxylase and glu tathione S-transferase. Epoxide hydrolase activity was decreased by bromobenzene, which was restored by pretreatment of phloroglucinol, The results suggest that the bromobenzene-induced hepatic lipid peroxidation by phloroglucinol is reduced by enhancing the activity of epoxide hydrolase.
