Origin of Organic Matter and Geochemical Variation of Upper Quaternary Sediments from the Ulleung Basin (울릉분지 상부 제4기 퇴적물의 유기물 기원 및 지화학적 분포)

  • Elemental, Rock-Eval pyrolysis and isotopic analysis of the core sediments from the northwestern and eastern Ulleung Basin of the East were carried out to identify their geochemical characteristics, spatial and vertical variation and origin of organic matter in Upper Quaternary sediments from the northwestern and eastern Ulleung Basin of the East Sea. TOC, m and TS did not show spatial variation between the sampling locations whereas they showed systematic vertical variation associated with MIS stages related to the sea-level change of the East Sea. It is suggested that these past changes of sea-level influenced the sedimentary depositional environments and/or diagenesis which resulted the patterns observed in this study. Based on the results of TOC/N, TS/TOC, ${\delta}^{13}C_{org}\;and\;{\delta}^{15}N_{org}$ analysis, organic matters in the study area appears to be predominantly originated from the marine algae rather than land plant and deposited under normal marine oxic condition during MIS I and MIS III period, and under euxinic/anoxic condition during MIS II period. TOC/N, ${\delta}^{13}C_{org}\;and\;{\delta}^{15}N_{org}$ have a relatively constant value irrespective of MIS stages, implying that the organic matter source does not change by the sea-level fluctuations. However, the results of Rock-Eval pyrolysis indicates that the organic matter is in immature stage and originated from land-plant (Type III), locating in the immature stage land plant (Type III). Similar differences were reported from other areas such as the Atlantic Ocean, Iberia Abyssal Plain, Mediterranean Sea, suggesting that Rock-Eval method does not exactly reflect the characteristic of immature organic matters. Accordingly, the application of Rock-Eval pyrolysis for delineating the source of immature organic matters should be approached with caution and all other geochemical proxies should be considered altogether at the same time.

Filter-Feeding Effect of a Freshwater Bivalve (Corbicula leana PRIME) on Phytoplankton (식물플랑크톤에 대한 담수산 패류 (참재첩)의 섭식효과)

  • The purpose of this study was to evaluate the filtering-feeding effect of a freshwater mussel (Corbicula leana) on the phytoplankton communities in two lakes with different trophic conditions between June and September, 2000. Manipulation experiments were conducted with two treatments (the control and mussel addition), and each established in duplicate 10 l chambers. Both ambient nutrient (TN, TP) and chlorophyll-a concentrations were significantly (p<0.01) higher in Lake Ilgam than Lake Soyang. Cyanophytes (Microcystis, Oscillatoria, Lyngbya and Dactylococcopis) consistently dominated algal community in Lake llgam, while flagellated algae (Dinobryon divergence, Mallomonas, Rhodomonas) and cyanophytes (Microcystis)dominated in Lake Soyang. The net exponential death rate ($R\;=\;day^{-1}$) of total phytoplankton in the mussel treatment ranged $1.70{\sim}7.39$ and $0.38{\sim}1.64$ in Lakes Soyang and Ilgam, respectively. Mean filtering rate standardized by mussel AFDW ($ml\;mgAFDW^{1}\;h^{-1}$) was much higher in Lake Soyang ($1.70{\sim}3.06$) than in Lake Ilgam($0.24{\sim}0.88$0.24~o.88). Estimating FR per mussel, 1 mussel filtered $1.6{\sim}7.8\;l$ per day and $1.7{\sim}3.0\;l$ per day in Lakes Soyang and Ilgam, respectively. Based on tile C-flux tobiomass ratio, Corbicula leana consumed $0.8{\sim}4.4$ fold of phytoplankton standing stock in Lake Soyang, and $0.4{\sim}1.6$ fold in Lake Ilgam per day. Mussel feeding resulted in increase of SRP concentration by $30{\sim}50%$, compared with the control. The results of this study suggest that filter-feeding activity of Corbicula leana varies depending on the phytoplankton density and community composition. The high seston consumption rate of Corsicuja Jeaua even in a eutrophic lake suggests that biomanipulation approach using filter-feeding mussels can be used far wate rquality management in small eutrophic reservoirs.

Analysis of Water Quality Improvement of Ceratophyllum demersum under Laboratory Condition - by Nutrients Removal Efficiency (실험실 조건에서 붕어마름의 수질개선 효과 분석 - 영양염류 제거 효율을 중심으로)

  • To evaluate the ability of the submerged plant, Ceratophyllum demersum's (C. demersum) to remove nutrients and to inhibit growth of cyanobacteria, a total of 6 mesocosms were conducted in a batch reactor for 9 days. From the 84 hr of the experiment, C. demersum was stabilized and showed daily cycle trends according to changes in pH and DO levels. The concentration of nutrients, $NH_3{^+}$, $NO_3{^-}$ and $PO_4{^3}$ continuously decreased until 9 days of the experiment, with the rapid decrease in nutrient concentration for the first 24 hours. High correlation coefficient ($r^2{\geq}0.96$, p<0.001) between the amount of C. demersum's biomass per unit area and the nutrients removal level were derived, and greater C. demersum's biomass per unit area showed higher removal efficiency of nutrients. However, there were differences in the C. demersum's activity level between batch reactors with higher and similar density of the C. demersum, but nonetheless water purification effect appears to have a significant influence due to attached algae and microorganisms. The growth rate of harmful cyanobacteria, Microcystis aeruginosa (M. aeruginosa) with C. demersum's density of 2,500 g $fw/m^2$ (100% of cover degree) was 0.31 /day, compared to the growth rate of 0.47 /day for the control group (0% of cover degree). In terms of number of cells, the control group had 1.7 times higher number of cells than the experimental group, proving that C. demersum has the ability to inhibit the growth of harmful cyanobacteria.

Effect of Chlorella vulgaris CHK0008 Fertilization on Enhancement of Storage and Freshness in Organic Strawberry and Leaf Vegetables (Chlorella vulgaris CHK0008 시비가 유기농 딸기와 엽채소의 저장성과 신선도 향상에 미치는 영향)

  • This study aimed to enhance storage and freshness of strawberry fruits and foliage vegetables by spray treatment with Chlorella vulgaris as a bio-fertilizer. The tested strain, C. vulgaris CHK0008, was isolated from an organically cultivated rice paddy and identified as C. vulgaris by its morphology and 18S rDNA and 23S rDNA sequence homology. We successfully cultured C. vulgaris CHK0008 in BG11 modified medium (BG11MM) and adjusted $2.15{\times}10^6cell/mL$ C. vulgaris CHK0008 to one OD value by measuring the optical density at 680 nm using a UV-vis spectrophotometer. The soluble solid content of 'Seolhyang' and 'Yukbo' strawberry fruits treated by spray application with C. vulgaris CHK0008 was enhanced by 22.2% and 11.5% respectively, compared to untreated controls. Additionally, the decay rates of treated 'Seolhyang' and 'Yukbo' strawberry fruits decreased 63.8% and 74.4% respectively, compared to untreated control. Surface color changes and chlorosis of leaves in leaf vegetables such as lettuce, kale, red ornamental kale, white ornamental kale and beet were observed in samples treated with water spray for 10 days after cold storage. However, the decay rate of leafy vegetables treated with foliar application of 25% C. vulgaris CHK0008 liquid culture was significantly decreased compared to that of the untreated control during storage at $4^{\circ}C$.

Antioxidant and Cellular Protective Activities of Ecklonia cava Extracts against Reactive Oxyegen Species (감태(Ecklonia cava) 추출물의 항산화 및 세포보호 활성)

  • In this study, we investigated the antioxidative effects of brown seaweed Ecklonia cava extract and its subfractions. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction of E. cava. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}$) of ethyl acetate fraction ($FSC_{50}=6.98{\mu}g/mL$) and aglycone fraction ($7.03{\mu}g/mL$) are similar to that of (+)-${\alpha}$-tocopherol ($8.98{\mu}g/mL$) which is a reference control. Reactive oxygen species (ROS) scavenging activity (total antioxidant capacity, $OSC_{50}$) of the aglycone fraction ($OSC_{50}=14.48{\mu}g/mL$) on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay was the strongest among all extract and fractions. However, all samples showed lower antioxidant activities than that of L-ascorbic acid ($6.88{\mu}g/mL$) known as a powerful antioxidant. The protective effect of 50% ethanol extract on the $^1O_2$-induced cellular damage of human erythrocytes was dependent on the concentration from 5 to $50{\mu}g/mL$. Both ethyl acetate fraction and aglycone fraction showed strong cellular protective activities at $10{\mu}g/mL$, where the cellular protective effects (${\tau}_{50}$) of each fraction were recorded 442.0 min and 539.9 min, respectively. Three kinds of extract/fractions of E. cava showed much greater cellular protective activities at $10{\mu}g/mL$ than that of liposoluble antioxidant (+)-${\alpha}$-tocopherol (40.6 min) which is a reference control. These results suggest E. cava extracts and its fractions can be applied as an antioxidant ingredient in a field of cosmetics.

Induction of Growth Inhibition and Apoptosis in Human Cancer Cells by a Brown Algae Extract (갈조류 추출물에 의한 인간 암세포 성장 억제 및 세포 사멸 유도)

  • In this study, we investigated the effects of Undaria pinnatifida (UP), Petalonia binghamiae (PB) and Punctaria latifolia (PL) extracts on the inhibition of proliferation and apoptosis in human gastric and breast cancer cells. AGS, MDA-MB-231 and SK-BR-3 cells were treated with 0, 50, 100, and 200 μg/ml concentrations of the extracts to determine their anti-proliferative effects, using the MTT assay. The UP, PB and PL extracts inhibited proliferation of AGS, MDA-MB-231 and SK-BR-3 cells in a dose-dependent manner, and the PL extract was found to be the most effective. DAPI staining was also performed to determine changes in the cell nucleus. Further, the AGS, MDA-MB-231 and SK-BR-3 cells were treated with 0, 50, 100, and 200 μg/ml of only the PL extract. DAPI staining showed increased chromatin condensation, which is indicative of apoptosis, in the 200 μg/ml group. The expression of the Bax, Bcl-2, and PARP proteins in AGS, MDA-MB-231 and SK-BR-3 cells treated with the PL extract was also determined by western blot analysis. The expression of Bax (a pro-apoptotic protein) and cleaved-PARP was increased, whereas the expression of Bcl-2 (an anti-apoptotic protein) was decreased compared with the control. These findings indicate that the PL extract may have potential as an alternative anticancer drug and nutraceutical.

The Effect of Ecklonia stolonifera Extracts on Blood Flow and Serum Lipid Level in Ovariectomized Rats (곰피 추출물이 난소를 절제한 흰쥐의 혈액 유동성 및 혈중 지질함량에 미치는 영향)

  • Estrogen deficiency in peri- and postmenopausal women results in variety of neurovegetative, psychic and somatic symptoms, and may contribute to severe diseases within the aged female population. Ecklonia stolonifera (ES) is an edible brown algae traditionally used in fishery towns in Far East Asia. This study was performed to investigate the effects of ES extracts on blood flow and serum lipid concentration in ovariectomized rats. Weight-matched female Sprague-Dawley strain rats were assigned to four groups. Three groups were surgically ovariectomized (OVX). The fourth group was sham operated. Rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-CON), ovariectomized rats supplemented with ES extract at 50 mg/kg bw/day (OVX-ES50) and ovariectomized rats supplemented with ES extract at 200 mg/kg bw/day (OVX-ES200). Serum total cholesterol and triglyceride contents decreased in the sham group compared to the OVX-CON group. Six weeks feeding of ES extract resulted in a significant (p<0.05) lowering of serum triglyceride and a lowering tendency of total cholesterol level. The level of Hdl-cholesterol in serum increased by supplementation of ES extracts at 50 mg/kg and 200 mg/kg bw/day (p<0.05). Blood passage time of the ES extracts-supplemented group was higher than the OVX-CON group. The ability of platelet aggregation of groups treated with ES extracts was less than that of the OVX-CON group. These results suggest that the beneficial effects of ES extract may be used to possibly improve on the metabolic syndrome of menopausal women.

Understanding of Phytoplankton Community Dynamics Through Algae Bioassay Experiment During Winter Season of Jinhae bay, Korea (생물검정실험을 통한 동계 진해만 식물플랑크톤의 군집 변동 특성 파악)

  • The distributions of phytoplankton assemblages and environmental factors in Jinhae Bay and their relationships were investigated to estimate the potential limiting nutrient for phytoplankton growth and community structure. In situ algal bioassay experiments were also conducted to assess the species-specific characteristics in phytoplankton responses under different nutrient conditions (control, N(+) and P(+) treatment). During the study periods, bacillariophyceae and cryptophyceae occupied more than 90% of total phytoplankton assemblages. Phytoplankton standing crops in the inner part of Masan Bay were higher than that of Jinhae Bay. The DIN:DIP ratio, pH and transparency showed the significant positive correlation with phytoplankton biomass. According to cluster and multidimensiolnal scaling (MDS) analysis based on phytoplankton community data from each station, the bay was divided into three groups. The first group included stations from the south-western part of Jinhae bay where cryptophyta species were dominated. The second group was distinguished from inner stations in Masan Bay. These stations showed low transpancy and high DIN:DIP ratio. The other cluster included the stations from the eastern part and central part of Jinhae Bay, which was characterized by the high DSi:DIP ratio and dominant of diatom species. Phosphorous (P) was limited in Masan Bay due to significantly increases in the phytoplankton abundances. Based on stoichiometric limitation and algal bio-assay in Jinhae Bay, nitrogen (N) was a major limiting factor for phytoplankton production. However, silicate (Si) was not considered as limiting factor, since Si/DIN and Si/P ratio and absolute concentration of nutrient did not create any potential stoichiometric limitation in the bay. This implies that high Si availability in winter season contributes favorably to the maintenances of diatom species.

The Study on the Marine Eco-toxicity and Ecological Risk of Treated Discharge Water from Ballast Water Management System Using Electrolysis (전기분해원리를 이용한 선박평형수관리장치의 배출수에 대한 해양생태독성 및 해양환경위해성에 관한 연구)

  • The International Convention for the Control and Management of Ship's Ballast Water and Sediments was adopted at 2004 and then various BWMS (ballast water management system) have been developed. In this study, WET (whole effluent toxicity) test with algae (diatom) Skeletonema costatum as primary producer, invertebrate (rotifera) Brachionus plicatilis as 1st consumer and fish (olive flounder) Paralichthys olivaceus as predator, chemical analysis and ERA (environmental risk assessment) were conducted to assess the unacceptable effect on marine ecosystem by emitting the discharge water treated with AquaStar$^{TM}$ BWMS using electrolysis as main treatment equipment for removing the marine organisms in the ship's ballast water. The most sensitive test organism on discharge water treated with AquaStar$^{TM}$ BWMS was S. costatum that gave the NOEC value of 25.00%, LOEC value of 50.00% and 72hr-$EC_{50}$ value of 69.97% from WET test result for 20 psu salinity treated discharge water. NOEC and LOEC value of B. plicatilis and P. olivaceus exposed at 20 psu salinity treated discharge water were 50.00% and 100.00%, respectively. In the chemical analysis results, total number of substances produced by AquaStar$^{TM}$ BWMS was 18 which were bromate, 7 volatile halogenated organic compounds, 7 halogenated acetic acids, 3 halogenated acetonitriles and chloropicrin. Eighteen substances did not consider as persistence and bioaccumulative chemicals. Uncertainty of toxic property of 18 substances was high. PECs of 18 substances calculated by MAMPEC model were ranged from $4.58{\times}10^{-4}$ to $4.87{\mu}g\;L^{-1}$, PNECs of them were ranged from $1.6{\times}10^{-2}$ to $3.2{\times}10^2{\mu}g\;L^{-1}$. And, the PEC/PNEC ratio of 18 substances did not exceed 1. Therefore, ERA for produced substances indicate that the discharge water treated with AquaStar$^{TM}$ BWMS does not pose unacceptable effect on marine life. And $EC_{50}$ value of S. costatum on discharge water treated by BWMS using the electrolysis had positive correlation with initial TRO concentration, concentration and kind & level of HAAs.

Effects of Myelophycus Simplex Papenfuss Methanol Extract on Adipocyte Differentiation and Adipogenesis in 3T3-L1 Preadipocytes (바위수염 메탄올 추출물이 3T3-L1 지방전구세포의 분화에 미치는 영향)

  • Myelophycus simplex Papenfuss is distributed over the northern Pacific and southern coast of Korea, and is a member of the brown algae family. The objective of this study was to investigate the effect of M. simplex methanol extract on adipocyte differentiation and adipogenesis in 3T3-L1 preadipocytes. Treatment with M. simplex methanol extract significantly suppressed terminal differentiation of 3T3-L1 preadipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet content observed by Oil Red O staining. Also, the M. simplex methanol extract significantly suppressed the triglyceride content of 3T3-L1 preadipocytes in a dose-dependent manner. Treatment with 300 and $500{\mu}g/ml$ of M. simplex methanol extract caused a 42% and 76% reduction in lipid droplet content, respectively. In order to understand the anti-adipogenic effects of M. simplex methanol extract, the changes in the expression of several adipogenic transcription factors, including peroxisome proliferator-activated receptor (PPAR) ${\gamma}$-cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding protein (C/EBP) ${\alpha}$ and ${\beta}$, were investigated using immunoblotting. M. simplex suppressed the expression of $PPAR{\gamma}$, $C/EBP{\alpha}$, and $C/EBP{\beta}$ proteins compared with control. Therefore, the results of this study suggest that M. simplex methanol extract inhibits adipocyte differentiation and thus may have applications as a potential source for an anti-obesity functional food agent.
