In Vivo Antipyretic, Analgesic, and Anti-inflammatory Activities of the Brown Alga Ecklonia cava Extracts in Mice

  • Dichloromethane, ethanol, and boiling water extracts of the brown alga Ecklonia cava were examined in vivo for their antipyretic, analgesic, and anti-inflammatory activities in mice. These activities were evaluated by yeast-induced pyrexia, tail-flick test, and phorbol myristate acetate-induced inflammation (edema, erythema, and blood flow). Ethanol extract of E. cava (0.4 mg/ear) inhibited the inflammatory symptoms of mouse ear edema, erythema, and blood flow by 82.6%, 69.0%, and 65.4%, respectively. This extract also demonstrated potent analgesic activity. No acute toxicity was observed after p.o. administration of each extract (5 g/kg bw). These in vivo data are in agreement with the claims of the health care industry and indigenous medicine that E. cava is an effective remedy for inflammation-related symptoms.

Identification of Carotenoids from Green Alga Haematococcus pluvialis by HPLC and LC-MS (APCI) and Their Antioxidant Properties

  • Haematococcus pluvial is, a green alga, accumulates astaxanthin (3,3'-dihydroxy-$\beta$,$\beta$'-carotene-4,4'-dione) upto 2-3% on a dry weight basis. In the present study, identification of carotenoids from Haematococcus cyst cell extract by HPLC and LC-MS (APCI) and their antioxidant properties were evaluated in in vitro model systems. The extract exhibited 89% and 78% antioxidant activities in the $\beta$-carotene linoleate model and the hydroxyl radical scavenging model, at 9 ppm of total carotenoid, respectively. The extract also showed 80%, 85%, and 79% antioxidant activities against lipid peroxidation in the kidney, brain, and liver of rats. Low-density lipoprotein oxidation induced by $Cu^{2+}$ ions was also protected (45%, 64%, and 75%) by the extract in a dose-dependent manner with different carotenoid levels. Thiobarbituric acid reactive substances concentration in the blood, liver, and kidney of rats were also significantly (p<0.005) decreased in H. pluvialis-treated rats. The potent antioxidant activity is attributable to various carotenoids present in the extract.

  • The addition of seaweed extracts was found to regulate the viability of cultures of the crustose coralline alga Lithophyllum yessoense. The viability was quantitated using a triphenyltetrazolium chloride assay, and the methanol-soluble extracts from 18 prevalent seaweed species were tested. Extracts from Codium fragile and Enteromorpha linza inhibited viability, and a Hizikia fusiform is extract slightly increased viability. The methanol extract of C. fragile, which had the strongest inhibitory activity, decreased viability to 72 or $52\%$ that of the control following addition of 0.2 or 2 mg/mL of extract to the culture, respectively. The main active compound in the C. fragile was lipid. This information is a preliminary result related to the exploration of seaweed restoration in the algal whitening area.

  • A monoacyldigalactosyl glycerol was isolated from the $CH_2Cl_2$ soluble fraction of the MeOH extract from the green alga Enteromorpha prolifera. The structure was established as $1-O-palmitoyl-3-O-[{\alpha}-D-galactopyranosyl(1{\rightarrow}6)-{\beta}-D-galactopyranosyl]-sn-glycerol$ (1) by chemical and spectroscopic methods.

  • Ecklonia stolonifera Okamura is a member of the family Laminariaceae, belonging to the order Laminariales. Previously we reported that the methanolic extract of the brown alga E stolonifera exerts antioxidative activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical.   • In the course of search for various bioactive compounds from marine algae, we found strong antioxidant activity of the methanolic extract of the brown alga G. elliptica. Chromatographic purification [ODS flash, gel-filtration on Sephadex LH-20, HPLC] of the BuOH layer of the methanolic extract afforded two known polyphenolic compounds, 6,6'-bieckol (1) and dieckol (2). Compound 1 showed acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) inhibitory activities, free radical scavenging activity on DPPH (1,1-diphenyl-2-picryl-hydazyl) with IC$\_$50/ values of 91.2, 45.6 and 15.5 $\mu\textrm{g}$/$m\ell$, respectively. Cytotoxic Activity of Biosynthesized Gold Nanoparticles with an Extract of the Red Seaweed Corallina officinalis on the MCF-7 Human Breast Cancer Cell Line

  • Background: Nano-biotechnology is recognized as offering revolutionary changes in the field of cancer therapy and biologically synthesized gold nanoparticles are known to have a wide range of medical applications. Materials and Methods: Gold nanoparticles (GNPs) were biosynthesized with an aqueous extract of the red alga Corallina officinalis, used as a reducing and stabilizing agent. GNPs were characterized using UV-Vis spectroscopy, transmission electron microscopy (TEM), energy dispersive analysis (EDX) and Fourier transform infra-red (FT-IR) spectroscopy and tested for cytotoxic activity against human breast cancer (MCF-7) cells cultured in Dulbecco's modified Eagle medium supplemented with 10% fetal bovine serum, considering their cytotoxicty and effects on cellular DNA. Results: The biosynthesized GNPs were $14.6{\pm}1nm$ in diameter. FT-IR analysis showed that the hydroxyl functional group from polyphenols and carbonyl group from proteins could assist in formation and stabilization. The GNPs showed potent cytotoxic activity against MCF-7 cells, causing necrosis at high concentrations while lower concentrations were without effect as indicated by DNA fragmentation assay. Conclusions: The antitumor activity of the biosynthesized GNPs from the red alga Corallina officinalis against human breast cancer cells may be due to the cytotoxic effects of the gold nanoparticles and the polyphenolcontent of the algal extract.

Acetylcholinesterase 억제 및 신경세포 보호 활성을 갖는 다시마목 해조 추출물 NX42의 마우스 학습능력 향상 효과 (Improvement of Learning Behavior of Mice by an Antiacetylcholinesterase and Neuroprotective Agent NX42, a Laminariales-Alga Extract)

  • 다당류 및 플로로탄닌 등을 주성분으로 하는 갈조추출물 NX42가 인지능력 향상에 미치는 영향을 평가하기 위한 in vitro 및 동물실험을 수행하였다. 그 결과 NX42는 acetylcholinesterase에 대하여 온화하지만 용량의존적인 억제효과($IC_{50}=600-700\;{\mu}g/mL$)를 나타내었다. NX42로부터 추출된 플로로탄닌 분획은 현저히 높은 용량 의존적 억제 효과($IC_{50}=54\;{\mu}g/mL$)를 나타내었다. 반면, 플로로탄닌이 제거된 분획과 푸코이단은 억제효과가 없었다. NX42 및 플로로탄닌 분획은 과산화수소에 의해 유발된 산화스트레스 조건 하에서의 SK-N-SH 세포의 파괴를 유의성 있게 억제한 반면, 플로로탄닌이 제거된 분획과 푸코이단은 보호효과를 나타내지 않았다. 스트레스 조건 하에 있는 마우스의 학습능력에 미치는 효과를 평가한 결과, NX42를 섭취한 마우스의 경우에는 섭취하지 않은 경우에 비하여 유의성 있게 개선된 학습능력을 나타내었으며, 이는 in vitro 실험 결과를 바탕으로 볼 때, NX42에 함유된 플로로탄닌의 acetylcholinesterase 억제 활성 및 신경보호활성에 의한 것으로 해석된다.

Effects of Nutrient Levels on Cell Growth and Secondary Carotenoids Formation in the Freshwater Green Alga, Chlorococcum sp.

  • The freshwater green alga Chlorococcum sp. grew on NH_4^{+},{\;}NO_3^{-}$, urea, yeast extract, and peptone as the nitrogen source showing similar pattens of growth and secondary carotenoid (SC) production. However, the most suitable nitrogen source for the induction fo SC was urea. The dffects of nutrient levels (urea, phosphate, sulfate, ferrous iron, and salt) on growth and SC production were stydied by varying the concentration of each nutrient in batch cultures. High biomass production was achieved in cultures containing 20-28 mM urea, 4.8-10 mM phosphate, 1.6 mM sulfate, 70 mM phosphate, 1.6 mM sulfate, 170 mM NACl, and $50{\;}\mu\textrm{M}$ iron. The optimum concentrations of nutrients for biomass and for the SC accumulation in biomass were evaluated and the two media for achieving high biomass production and SC production were thus developed. The extent to which each parameter to stimulate the formation of SC in the alga were varied and the potentially improned SC prodution by manipulating the nutrient levels in the modified media were descussed.
