The effect of herbal mixture on alcohol metabolism in Sprague Dawley rats (알코올을 투여한 흰쥐의 알코올 대사에 미치는 한약재 혼합추출물의 영향)

  • Objectives : Alcohol hangover is a common phenomenon which basically occurs after heavy drinking. Moreover, heavy alcohol consumption leads to acute and chronic diseases. We investigated the effect of herbal mixture (SJ) on alcohol metabolism in serum or/and liver. Methods : 5W-old Sprague Dawley rats were used for the study. To overnight fasted rats, 0.9% saline or SJ extract was administrated per os before alcohol treatment. Then, 40% alcohol was orally administrated to all rats in 30 mins. Ethanol, acetaldehyde concentrations, alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH) activities were measured by assay kits. Aspartate aminotransferase (AST) and alanine transaminase (ALT) were measured by analyzer. Results : Ethanol and acetaldehyde concentrations lowered in SJ groups compared with CON group. Especially, acetaldehyde concentration significantly decreased in SJ groups compared with CON group. AST and ALT levels tended to increase in SJ groups compared with CON group but there was no significant difference between CON group and SJ groups. ADH activity in serum was higher in SJ groups than CON group but no significant difference in liver. ALDH activity in both serum and liver showed significantly increased in SJ groups compared with CON group. Conclusions : Treatment of SJ extract showed not only reducing concentration of alcohol and acetaldehyde but also increasing activities of ADH and ALDH. These results suggest SJ may influence in alcohol metabolism via control of metabolic enzyme activities and metabolite. Therefore, SJ, herbal mixture, might have a function of preventing hangover after drinking alcohol.

  • The cDNA of alcohol dehydrogenase (PgADH) was isolated and characterized from the leaf of Panax ginseng. The cDNA had an open reading frame of 801 bp and a deduced amino acid sequence of 266 residues. The calculated molecular mass of the mature protein is approximately 29 kDa with a predicated isoelectric point of 6.84. Homology analysis revealed that the deduced amino acid of PgADH shares a high degree of homology with the short-chain ADH proteins of other plants. Genomic DNA hybridization analysis indicated that PgADH represents a multi-gene family. The expression of PgADH under various environmental stresses was analyzed at different time points using real-time PCR. ABA, SA and especially JA (80-fold) significantly induced PgADH expression within 24 h of treatment. The positive responses of PgADH to abiotic stimuli suggest that ginseng ADH may protect against hormone-related environmental stresses.

  • A Zymomonas mobilis DNA fragment consisting of 207 nucleotides, which corresponded to the 5'-flanking region of an adhB gene encoding alcohol dehydrogenase II, was fused to the structural gene coding for a Bacillus endo-.betha.--1, 4-glucanase. The Z. mobilis DNA framgment waw identified to promote 50-fold increase in the expression of endo-.betha.1. 4 glucanase gene in Escherichia coli.

  • Allyl alcohol은 간에서 두 단계의 효소 반응을 거쳐 대사되는데, 먼저, alcohol dehydrogenase (ADH)에 의해 독성 활성체인 acrolein으로 바뀌고, 이후 계속하여 aldehyde dehydrogenase (ALDH)에 의해 acrylic acid로 무독화되어 배설된다. Ethanol 역시 간에서 대사되는데 있어 같은 효소들을 공유하므로 allyl alcohol과 경쟁적으로 반응할 것이다. 따라서, 본 실험에서는 ethanol에 의한 대사 효소 경쟁반응에 의해 allyl alcohol 의 간독성이 어떻게 변화하는지를 연구하였다. 우선 ethanol과 allyl alcohol을 동시 투여할 경우 5시간째에 allyl alcohol에 의해 증가된 ALT level을 낮춘다는 보고를 확인하고자 ethanol 2 g/kg과 allyl alcohol 40 mg/kg을 동시투여했으나 오히려 치사율이 증가했고, ethanol을 2시간 전처리한 군에서도 역시 치사율이 증가되고, 간의 glutathione 양은 allyl alcohol 단독 처리군에 비해 현저히 감소되는 양상을 보였다.

  • Despite the use of antibiotics and vaccines, Streptococcus pneumoniae remains a serious cause of morbidity and morality in human. In this study, pathogenic and immunological natures of alcohol dehydrogenase (ADH) in S. pneumoniae were elucidated. In vitro cytotoxicity test determined by lactate dedydrogenase release from A549 cells revealed that adh mutation significantly reduced cytotoxicity although in vivo intraperitoneal challenge of the adh mutant to BALB/C mouse exhibited marginal increase of survival time than the wild type. Changes of Antioxidative Enzymes and Alcohol Dehydrogenase in Young Rice Seedlings Submerged in Water (담수조건하 벼 발아 및 입모단계시 항산화효소 및 Alcohol dehyrogenase활성 변화)

  • Successful germination and establishment of seedlings in flooded paddy are critical in direct seeding cultivation of rice. In this study, we examined the relationship between antioxidant enzymes and alcohol dehydrogenase (ADH) activities and coleoptile elongation under submergence of deep water with two rice cultivars, Iksan429 and Woodrose, which show characteristic coleoptile elongation under hypoxic condition. The growth of shoot under submerged in water was faster than the root. The survival duration was longer in Iksan429 than in Woodrose under submerged in water. The alcohol dehydrogenase (ADH) activities were significantly increased under hypoxia compared to in aerated condition. The ADH activity was increased in Iksan429 more than in Woodrose under hypoxia. The superoxide dismutase (SOD) activity in Iksan429 was gradually increased up to 5 days after treatment (DAT) then decreased slowly till 14 DAT under water, whereas in Woodrose it was dramatically decreased after 5 DAT. The peroxidase (POX) activity in Iksan429 was significantly increased until 7 DAT under hypoxia, whereas it was not significantly different in Woodrose during hypoxic treatment. However, in non-treated condition, POX activity in Woodrose was increased more than Iksan429. The changes of catalase (CAT) activities showed no differences in both cultivars. We suggest that the overexpression of ADH, SOD and POX activities is responsible for the hypoxic tolerance and plays an important role in the surviving of rice seedling.

  • Alcohol dehydrogenase (ADH) and Aldehyde dehydroganase (ALDH) activities of turnip extracts with ultrasonification at $60,\;100^{\circ}C$ temperature were investigated. Ultrasonification extracts showed a increase as compared with the normal extracts. ALDH activities were high than ADH activities. So, turnip extracts was shown efficient plants in alcohol oxidation. Appraisers liked powder extracts better than liquid extracts in sensory score.

  • Objectives : The aim of this study was to investigate effects of the Sungjuchunggan-tang(Xingjiuqinggan-tang), which has been used for alcoholic diseases in Oriental medicine, on alcoholic metabolism and alcoholic liver damage. Methods : We evaluated the activities of alcohol dehydrogenase(ADH)and aldehyde dehydrogenase(ALDH), and measured the levels of AST, ALT, glucose, triglyceride, HDL-cholesterol, LDL-cholesterol and phospholipid in serum of guinea pigs. Results and Conclusions : In this experiment, Sungjuchunggan-tang, Pharbitidis Semen and Puerariae Radix significantly suppressed the activity of ADH which is a precursor enzyme of acetaldehyde. Sungjuchunggan-tang and Pharbitidis Semen significantly suppressed the activity of ALDH, which is a catabolic enzyme, and increased its level. Due to alcohol consumption, level of ALT and AST were significantly reduced. These experimental results suggest that Sungjuchunggan-tang and Pharbitidis Semen inhibit the formation of acetaldehyde in the metabolism of alcohol, and affect the recovery of weakened liver functions caused by alcohol.

고온성 알콜발효 효모의 Alcohol Dehydrogenase의 특성

  • The characteristics of alcohol dehydrogenase (ADH, EC 1.1.1.1, alcohol:NAD oxidoreductase) of thermotolerant alcohol-producing yeasts, Saccharomyces cerevisiae RA-74-2 and Kluyveromyces marxianus RA-912, were compared with that of mesophilic S. cerevisiae D, an industrial strain. Under anaerobic culture condition, both S. cerevisiae RA-74-2 and D had similar level of ADH activity at 30$\circ$C, and the activity of S. cerevisiae RA-74-2 at 37$\circ$C was the same level at 30$\circ$C. However, the level of ADH activity of S. cerevisiae D at 37$\circ$C decreased about 70% of that at 30$\circ$C. The level of enzyme activity of K. marxianus RA-912, which showed lower alcohol productivity than S. cerevisiae RA-74-2 and D, was about 43% of those strains at 30$\circ$C, and decreased somewhat at 37$\circ$C. The results showed a good correlation between the alcohol productivities and the level of ADH activities of these strains grown at 30$\circ$C and 37$\circ$C. And the higher heat stability of ADH of S. cerevisiae RA-74-2 than that of S. cerevisiae D seemed to reflect the ability of high temperature fermentation. Despite of its fermentation ability even at 45$\circ$C, however, the ADH of K. marxianus RA-912 showed lower heat stability than that of S. cerevisiae D. Both S. cerevisiae RA-74-2 and D showed similar patterns of two bands of ADH isozyme, and the low band of S. cerevisiae RA-74-2 moved slightly faster than that of S. cerevisiae D. The staining intensity of the bands of S. cerevisiae D at 37$\circ$C was weaker than those at 30$\circ$C. However, S. cerevisiae RA-74-2 showed no differences in total intensity of the bands of 30$\circ$C and 37$\circ$C. As the patterns of cellular proteins and ADH isozyme of K. marxianus RA-912 were different from S. cerevisiae RA-74-2 and D, K. marxianus might have its own characteristic ADH system.
