Bacterial Mutagenicity of some Hot-air Dried Shellfish and Canned Products of some Red-muscle Fish During Storage (열풍건조된 몇 가지 패류와 적색어류 통조림의 저장조건에 따른 돌연변이성 변화)

  • Bacterial mutagenicity of shellfish or canned products of red-muscle fish affected by hot-airdry processings and/or during storages was investigated by Ames preincubation method. Mussel (Mytilus edulis) and surf clam (Tapes japonica) dried under either 40$\circ$C, 50$\circ$C or 60$\circ$C until remained about 25 % water content was stored at 4$\circ$C or 25$\circ$C for 6 months. Commercially available canned product of pacific saury (Cololabis saira) and mackerel saury (Scotnberjaponicus), whose producer or date of manufacture was identical each other, was stored at 25$\circ$C for 6 months. Mutagenic substances from sample (25 g dry weight) were extracted and purified for mutagenicity assay against S. typhitnurium TA98 by the simultaneous distillation extraction (SDE) method (Lee et al. 1955). Mutagenicity of sample was expressed number of revertant per 25 g dry sample. Number of revertant from unheated mussel sample was appeared to be 135, relative to 22 from unheated surf clam sample. Mutagenicity of both shellfish was strongly affected by heat treatment, but a lesser extent by storage conditions, even at 25$\circ$C for 6 months. Revertants of the mussel sample dried at 40$\circ$C, 50$\circ$C, or 60$\circ$C was found to be 227, 779, and 883, respectively. Similar, but lower, mutagenicity was observed from the sample of surf clam dried at 40$\circ$C, 50$\circ$C, or 60$\circ$C than mussel. Mutagenicity of canned pacific saury and mackerel samples was not influenced by storage conditions. Number of revertant from pacific saury was 175 which was relatively higher than 5 from mackerel. Mutagenicity of fresh pacific saury and mackerel samples was significantly higher than that of their counterparts from canned products. These results indicate that mutagenicity of the shellfish was affected by heat treatment (40$\circ$C, 50$\circ$C, or 60$\circ$C) used for drying process, but not much by storage for 6 months at 4$\circ$C or 25$\circ$C and the mutagenicity of canned products of red-muscle fish was not influenced by storage, even at 25$\circ$C for 6 months.

  • Orostachys malacophyllus grow on the old roofing tile or on the rock of mountain and is belong to Crassulaceae family. After air drying for Orostachys malacophyllus (OM), using the mixture of lactic acid bacteria (Lactobacillus bulgaricus, Streptococcus thermophilus, Lactobacillus acidophilus) was fermented (FOM). OM and FOM extracted using water (W), ethanol (E) and methanol (M) and were measured extracts yield, pH and Brix. Extracted OM and FOM were tested by in vitro experimental models of α,α´-diphenyl-β-picrylhydrazyl (DPPH) radical scavenging activity, Fe/Cu reducing power, linoleic acid peroxidation using ferric thiocyanate and thiobarbituric acid (TBA) methods and peroxidation of rat liver homogenate. In addition, the bioactive materials (phenolic compounds, flavonoids and minerals) were measured. The highest phenolic compounds and flavonoids were OME 122.2 mg/100 g and OME 84.0 mg/100 g. OM and FOM′s major minerals were K, Ca and Mg. The highest free radical scavenging activity showed in FOMM (93.9%), OMM (93.4%), FOME (92.1%) and OME (91.9%) at 0.5% additional level. Fe reducing powers were strong in FOME and FOMM and Cu reducing powers were strong in OME and FOMM. Antioxidant activities on lipid peroxidation using rat homogenate as measured by TBARS method showed strong in FOME and on lipid peroxidation of linoleic acid as measured by ferric TBA method showed strong in OME and FOME and measured by ferric thiocyanate showed strong in FOME and FOMM.

  • Volatile flavor components of garlic and factors which influence on its flavors were reviewed. Growth, storage and processing conditions influence on the flavor intensity of garlic. To intensify garlic flavors, it is desirable that sufficient sulfate nutrition be supplied to the soil of growing garlic and that the suggested proportions of mineral composition and water content be considered. And to maintain the flavor intensity of post harvested garlic, flavor losses taken place during over inter storage mainly due to respiration, sprout and decay, have to be minimized. Among the various storage methods, combination method of post harvest hot-air drying and low temperature ($0^{\circ}C$), low humidity (RH 70-75%) is useful. The flavor of processed garlic is very much decreased as compared with that of fresh, and the decreasing rate of flavors depends on processing method. The synthetic garlic flavors were obtained by three types based on intermediate thiosulfinate, S-alk(en) yl-$\small{L}$-cyteine sulfoxlde-alliinase fission products and $\small{L}$-5-alk (en)yl thiomethylhydantoin ${\pm}$ S-oxides. These synthetic garlic flavors may be promised to be applied to food additives.

Studies on the Processing and Management Forms of Filatures (우리나라 제사공장의 공정 관리실태에 관한 조사연구)

  • The processing management forms of our country's filature factories in 1969 are summarized as follows. (1) About 80% of total cocoon collection is made within 5 days involving peak day, and 10% of cocoon collection is finished until 3 days before and after the peak day, (2) About 92% of alive cocoons transported on unpaved road, and about 40% of the cocoons purchased by all factories are loaded on trucks from common selling station which is far beyond 40km, therefore a new packing system of alive cocoons to drop the damage of cocoon qualities, should be taken. (3) 22% of all factories in our. country have only low-temperature cocoon drying machine. Therefore the installment of hot-air cocoon drying machine is required urgently. (4) In view of cocoon qualities in our country, the grouping method of cocoon for reeling. taken by about 50% of the factories at percent, which classify cocoons for reeling as high group (1,2,3,4 grades) and low group(5,6 grades), will have to be replaced by the method tat classify them high group (1,2 grades) middle group (3,4 grades), low group (5,6 grades). (5) The .ratio of cocoon assorting stood about 10% in multi-ends reeling, about 15% in automatic reeling, conclusively, the ratio of cocoon assorting for automatic reeling was higher tan that for multi-ends reeling. One person's ability for a day in cocoon assorting reaches to about 80-100kg. (6) Cocoon cooking condition requires the increase of the cooking time, the pressure and temperature used to be prolonged as much as the qualities of cocoons are material cocoon ior automatic and double cocoon machines are treated uncompletely. (7) Automatic silk reeling is being performed at 1-2$^{\circ}C$ lower in reeling water temperature and operated at about twice velocity. (8) The temperature and humidity of rereeling room stood at 25$^{\circ}C$, 67.2% R.H and 32.3$^{\circ}C$, 51.9% R.H of rereeling machine are showed, Average rereeling velocity is 233m/min and large reefs charged for one person are 7.5 reels and form of skein used in all factories is double skein. (9) About 73% of water sources for filature used under-earth water. About 48% of all filature factories in our country have not yet water purifying equipments. Installation of the equipment for these factories seems to be urgent, (10) Denier .balance, sizing reel, seriplane, are being used in most factories as self-inspection apparatus. (11) More than 90% of the factories use the vacum tank in rereeling process and about 20% of them use it in cocoon cooing process (12) Only 21% of the factories use chemicals in filature process. About all them use "Seracol 100" in cocoon cooking process and "Seracol 500" in rereeling process, (13) Above survey results explain each all factories show large difference in the processing management. Therefore, it is believed that intercommunication through seminar or technical exchange will contribute to the production evaluation of cocoon in our filature industry.

Study of Sulfur Dioxide Contents in Various Fresh Vegetables During the Drying Process (건조에 따른 채소류 중의 이산화황함량 분석)

  • This study was planned to monitor the change in the content of sulfur dioxide during the drying process of fresh vegetables. The analysis of sulfur dioxide was conducted by the Optimized Monier-Williams Method based on the Korea Food Code. The samples were kinds of vegetables which consisted of naturally-originated sulfur compounds (green onion, onion, cabbage, garlic, radish leaves, radish). Fresh vegetables (n = 182) and dried vegetables (n = 41) purchased from different local areas were investigated for the content of sulfur dioxide. The fresh vegetables were dried at 50~$60^{\circ}C$ using hot-air dryer. The moisture contents of dried samples were adjusted to keep 10 percents. The contents of sulfur dioxide in self-dried vegetables were 104.6 mg/kg in green onion, 75.4 mg/kg in onion, 129.1 mg/kg in cabbage, 197.6 mg/kg in garlic, 23.0 mg/kg in radish leaves and 52.5 mg/kg in radish, respectively. The increase of sulfur dioxide content according to the moisture content reduction was different from the expected. It means that the contents of sulfur dioxide can be altered by other factors except moisture contents. This results can be utilized as materials for the safe management of sulfites of dried vegetables.

Postharvest Treatment of Sweet Persimmon and Preparation of Its Dehydrated Product (수확 후 단감 전처리 기술 확립과 고품질 건조과 개발)

  • To maintain the quality of sweet persimmon during storage, the samples were treated with a combination of 50 ppm $ClO_2$ and 0.1% fumaric acid after harvest, packaged with low density polyethylene film, and stored at $1{\pm}1^{\circ}C$ for 35 days. The combined treatment reduced the populations of total aerobic bacteria, yeast and molds by 1.82 and 2.07 log CFU/g, respectively, compared to the control. During storage, hardness of all samples decreased, but total soluble solids and weight loss were not significantly different among treatments. In addition, high-quality dehydrated sweet persimmon was prepared using red algae extract as a dehydrating agent. The rehydration ratio and vitamin C content of red algae extract-treated sample were greater than those of hot-air dried sample. These results suggest that the combined treatment of $ClO_2$ and fumaric acid can be useful for maintaining microbiological safety of sweet persimmon during storage and dehydration of sweet persimmon slices using red algae extract is an efficient drying method for the preparation of high-quality dehydrated sweet persimmon.

Physical Properties of Lightweight and Normal Weight Concretes due to Water-Cement Ratio Changes (물-시멘트비 변화에 따른 경량콘크리트와 일반콘크리트의 물리적 성질)

  • By using the artificial lightweight aggregate for the natural aggregate depletes and destruction of environment and the application of lightweight concrete in structure, the lightweight concrete is manufactured. The fundamental characteristics by the waterbinder ratio was evaluated. It is suggested the method to control of pre-absorbed water of the lightweight aggregate. Lightweight concrete with pre-absorbed aggregate has similar characteristics compared to normal weight concrete regardless of water-binder ratio. According to the water-binder ratio, the drying condition, and the rebar, the unit mass of the lightweight concrete showed the reduction of 14.6${\sim}$21.0% as the range of 1,668${\sim}$1,998 $kg/m^3$ in comparison to the normal weight concrete. The lightweight aggregate pre-absorbed water showed the deferent evaporation quantity according to the water-binder ratio. As the water-binder ratio is lower, the oven dry vapour water is larger, therefore the internal curing water is increasing. In the same water-binder, comparing the normal concrete the lightweight concrete shows lower compressive strength which is due to the different strength of an aggregate. In the air dry curing, the normal weight concrete has a lower strength improvement effect in w/c 0.3 than the ratio 0.4 and 0.5. However, the strength improvement effect has increasing as the water-binder ratio was low in the light concrete.

  • The preservative effect of chitosan film packing on quality of lightly-salted and dried horse mackerel was studied. In preparation of chitosan film, blue crab shell chitosan was dissolved in dilute acetic acid$(1.0\%,\;v/v)$, filtered, and spreaded on plastic plate and dried at $50\pm2^{\circ}C$. The chitosan film thus obtained was neutralized with 1.0N NaOH for 2 hrs and dried at room temperature after washing several times with distilled water. The lightly-salted and dried horse mackerel product was prepared by drying for 4 hrs at $40\pm2^{\circ}C$ in hot air dryer after packing with the chitosan film. During storage at $5.0\pm0.5^{\circ}C$, moisture content of the product was higher than that of the reference, but contents of VBN(volatile basic nitrogen) , amino nitrogen, and TMA of the product on dry basis were lower than those of the reference. Viable cell count, TBA value, and peroxide value of the product were also lower than those of the reference. Judging from the result of sensory evaluation, the chitosan film packing in the storage of lightly-salted and dried horse mackerel was remarkably elongated shelf-life of the product. From the results of chemical and sensory evaluation, it was concluded that chitosan film packing was an effective method for retaining the quality of lightly-salted and dried horse mackerel.

A Study on Corrosion Resistance of the Reinforement in Concrete Using Blast-Furnace Slag Powder (고로슬래그미분말을 사용한 콘크리트의 염화물이온에 의한 철근부식 저항성 연구)

  • This paper represents the permeability of chloride ions and the corrosion performance in the concrete blended with granulate blast furnace slag exposed to chloride environment. An ordinary cement (type I ) and sulfate resisting cement(type V) were used for the experiment. The two cements were combined with $0\%$, $25 \%$, $40\%$, and $55\%$ of the granulated blast furnace slag. The accelerated permeability tests of chloride ions were performed in accordance with ASTM C1202, and the accelerated corrosion tests of steel were carried out by using the method of immersion/drying cycles. After water curing 28 days, 56 days and 91 days, these tests were conducted until 30 cycles. In every cycle, test specimens were wetted in $3\%$ NaCl solution for three days and dried again in $60^{\circ}C$ air for four days. As an experimental results, the diffusion coefficient of chloride ions of the ordinary cement Concrete Combined granulated blast furnace slag was much lower than that of non granulated blast furnace slag concrete. Moreover, the diffusion coefficient of chloride ions of sulfate resisting cement concrete was higher than that of ordinary cement concrete. On the basis of the results of accelerated corrosion tests, corrosion resistance of the concrete mixed with granulated blast furnace slag shows good to corrosion resistance, however, the concrete with sulfate resisting cement shows bad to corrosion resistance.

Changes in Quality Characteristics of Chopped Garlic with Various Storage Method (저장방법에 따른 다진 마늘의 품질특성 변화)

  • This study was carried out to investigate quality characteristics of chopped garlic stored at various temperature. Chopped garlic was stored at room temp.($25^{\circ}C$), refrigerator temp.($5^{\circ}C$) and frozen temp. ($-18^{\circ}C$) for 1, 2, 4, 8, 24, 48, 72 hours(room temp.) and 1, 3, 5, 7, 10, 15, 20, 30 days(refrigerator temp. and frozen temp.), respectively. Brown color, Hunter color value, vitamine C contents and flavor patterns by electronic noes of chopped garlic were measured. Fresh chopped garlic was used as control and garlic powder by hot air drying was used for comparisons. Brown color of chopped garlic increased with increasing storage time, and the intensity of that was reduced as following order such as chopped garlic stored at room temp., refrigerator temp. and frozen temp. Brown color of garlic powder was most dark amomg all the samples. The results of Hunter color value was consistent with then of brown color in chopped garlic, whereas those of garlic powder showed relatively low browning. Above results might be caused by the difference of measurement as only surface color was measured in Hunter color value unlike brown color. Vitamine C contents of chopped garlic decreased with increasing storage time, and in particular those stored at room temp. decreased markedly as retention of those stored at room temp. was 60% of control after 1 day storage. Vitamine C retention of chopped garlic stored at refrigerator temp. was 78% of control after 10 day storage and that stored at frozen temp. was 86% of control after 30 day storage. At room temp., flavor pattern by electronic nose showed a difference between chopped garlic stored for 1hour and control, and showed occurrence of off odor after 1 day storage. Flavor pattern of garlic powder by electronic nose showed a mild flavor. Flavor patterns of chopped garlic stored at refrigerator temp. were similiar until 10 day storage, whereas those showed occurrence of volitile components after 15 day storage. Flavor patterns of chopped garlic stored at frozen temp. were similiar after 30 day storage, and especially those after 3 day storage were almost the same as control. Therefore it is desirable to store chopped garlic within 1 day at room temp. and within 10 days at refrigerator temp. And it is most desirable to store chopped garlic at frozen temp. as retention of quality characteristics was superior than other storage temp.
