Antioxidative Effects and Anti-proliferative Effects of MeOH, BuOH and Ethyl Acetate Fractionated from Stephania delavayi Diels (일문전(Stephania delavayi Diels.) 메탄올, 부탄올, 에틸아세테이트 분획물의 항산화 및 세포증식억제 효과)

  • Stephania delavayi Diels. (S. delavayi Diels.) has been used as a drug for pain-relieving and acute gastroenteritis treatment in China. Because the major therapeutic mechanism of anti-inflammatory drug is to inhibit the cyclooxygenase (COX)-2 and because COX-2 proteins inhibit apoptosis, COX-2 inhibitor has been thought as the anticancer drug candidate. For this reason, we examined S. delavayi Diels. as an anticancer drug. S. delavayi Diels. was fractionated with methanol and then partitioned with ethyl acetate, n-butanol and water. The antioxidant activity was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and reducing power. DPPH radical scavenging activities of the crude fractions at the concentration of $1,000{\mu}g/mL$ were 75.23% (n-butanol), 68.11% (methanol), 63.58% (ethyl acetate), and 50.13% (water). The reducing power increased according to the concentration in dose-dependent manner. Also, when the antiproliferation effects of each fraction against human breast cancer cell-lines MDA-MB-231 and MCF-7 were examined, methanol extract, n-butanol fraction and ethyl acetate fraction exhibited cell proliferative inhibition effects in both cell-lines whereas water fraction did not. Among the crude fractions, the n-butanol fraction exhibited the most potent anti-proliferation effect. In conclusion, fractions from S. delavayi Diels. are promising anticancer drug candidates.

The Paleovegetation at Dongdo of Is. Dokdo, Korea (한국 독도 동도의 고식생)

  • Dokdo, small island located in the eastern end of the Korean Peninsula, has been an uninhabited island for long time due to long distance from the land. Moreover, the steep slope of volcanic tuff at Dokdo is well drained for high permeability and Dokdo has few plain areas, swamps and very thin soil layers. In this study, pollen analysis at Dokdo was attempted on the profile of organic sandy soil for the first time in Korea owing to the marine climate environment with high humidity and precipitation including snowfalls in spite of unprofitable condition geologically or geomorphologically. While many historical, political researches have been accumulated for territorial problem of Dokdo with Japan, natural scientific researches with field work are not sufficient, and few paleoecological researches have been done. As a result of pollen analysis, the ratio of the NAP(Non-Arboreal Pollen) and spore was higher than AP(Arboreal Pollen), and the vegetation change existed by showing dramatic decrease of AP at the upper layer. AP was composed of almost Pinus, and a little Alnus, Quercus, Betula, Carpinus, Picea and Ulmus. NAP was composed of Gramineae, Compositae, Chenopodiaceae Cyperaceae, Caryophyllaceae, Liliaceae, Umbelliferae, Artemisia, Lobelia, Rumex, Polygonum. Increase of the NAP such as Gramineae, Artemisia and Chenopodiaceae, they would mostly have been transported from the other areas because agricultural activity is impossible at Dokdo. While one reason of Pinus thunbergii decrease could be regarded as environmental change, the other reason would be lumbering of pine trees by human activity which is adapted to Dokdo environment.

Effects of a Health Diary Program on Fall-Related Outcomes in Low-Income Elderly Women with Osteoarthritis (건강다이어리 프로그램이 저소득 관절염 여성노인의 낙상관련 심리적 변수에 미치는 효과)

  • Purpose: This study was done to identify the effects of a health diary program on fall related outcomes the low-income elderly women. Methods: The study was carried out with a nonequivalent control group with pretest-posttest design. The study was composed of two groups, each made up of 24 subjects: experimental group and control group. The subjects were low-income women aged over 65 years with osteoarthritis and both the experimental and control groups were made up of subjects with the same age profiles. The independent variable was the health diary program, and the dependent variables were fall related outcomes (fear of falling, fall-efficacy, knowledge of fall) difficulty of performing activity, and mood state. The health diary program was performed for 50 minutes each session and twice a week for 8 weeks. Data were collected before the health diary program 10 weeks after the beginning of the program. Results: The experimental group showed significant differences in knowledge of fall, fear of falling, and mood state compared to the control group. However there was no significant differences in difficulty of performing activity and fall efficacy. Conclusion: The results of this study may be used as part of an education to prevent falls for low-income elderly women with osteoarthritis.

  • Synthesis and Biological activity test are described for the (E)-11-tetradecen-1-yl acetate and (Z)-11-tetradecen-1-yl acetate, the sex pheromone of the Asiatic leafroller moth (Archippus breviplicanus Walsingham). 10-Bromodecan-1-ol THP ether was prepared from 10-bromodecan-1-ol. In liquid ammonia with THF and HMPA as cosolvents, sodium acetylide could be alkylated with 10-bromodecan-1-al THP ether to give 11-dodecyn-1-ol THP ether. 11-Dodecyn-1-ol THP ether was then treated with n-Buli in THF to give the lithium acetylide, which was alkylated with bromoethane to afford 11-tetradecyn-1-ol THP ether. 11-Tetradecyn-1-ol THP ether was then reduced over $Pd/BaSO_4$ and with Na in liquid $NH_3$ to give (Z)-11-tetradecen-1-ol THP ether and (E)-11-tetradecen-1-ol THP ether, respectively. (Z)-and (E)-11-Tetradecen-1-ol THP ether thus obtained were deprotected by refluxing in the presence of PPTS and ethanol. (Z)-and (E)-11-tetradecen-1-ol were acetylated with acetic anhydride to afford the final products, (Z)-11-tetradecen-1-yl acetate (1) and (E)-11-tetradecen-1-yl acetate (2), respectively. The synthetic pheromone thus obtained was attractive to the males of the Asiatic leafroller moth in the field.

Changes of physiochemical properties and biological activity during the fermentation of Doenjnag with bitter melon (Momordica charantia L.) (여주 함유 된장의 발효 중 이화학적 특성 및 생리활성의 변화)

  • This study evaluated the changes of physiochemical properties, phytochemical compounds (isoflavones and phenolic acids), and biological activity during the fermentation of Doenjang without and with bitter melon powder (BMP). The pH decreased from 6.41-5.83 to 5.81-5.24, during the fermentation of Doenjang, while the acidity increased from 0.42-0.65% to 1.28-1.48%. The viable cell numbers of Bacillus and Yeast, salinity, and total amino acid contents increased at the end fermentation (60 day). Also, the fermented Doenjang (FD) with 10% BMP showed the highest ${\gamma}$-aminobutyric acid (GABA, 129.87 mg/100 g) contents, among all the Doenjang samples. The FD exhibited significantly higher inhibitory activities than unfermented Doenjang (UFD) on radicals and ${\alpha}$-glucosidase. The phytochemical compounds including isoflavone-aglycones and phenolic acids increased, whereas isoflavoneglycosides decreased in the BM following fermentative processing. Moreover, the total phenolic, isoflavone-aglycone, and phenolic acid contents were markedly increased, leading to a general increase in antioxidant and ${\alpha}$-glucosidase inhibition activities after fermentation. These results suggest that BMP may be used to prepare a new type of fermented Doenjang with improved antioxidant and antidiabetic activities.

  • A hyperthermophilic bacterium Thernotoga maritima produced thermostable ${\beta}-glucosidase$. The gene encoding ${\beta}-glucosidase$ from T. maritima MSB8 was cloned and expressed in Escherichia coli. The en-zyme (BgIB) hydrolyzed ${\beta}-glucosidase$ linkages between glucose and alkyl, aryl of saccharide groups such as salicin, arbutin, and $_pNPG$. The insert DNA contained ORF with 2,166 bp encodes a 721 amino acids (calculated molecular mass of 80,964 and pl of 4.93). The amino a.id sequence of BglB showed the similarity to family 3 glycosyl hydrolases. The molecular weight of the enzyme was estimated to be approximately 81kDa by MUG-nondenaturing PAGE (4-methylumbelliferyl 13-D-glucoside-nondenaturing polyacrylamide gel electophoresis) and SDS-PACE. The ${\beta}-glucosidase$ exhibited maximal activity at pH 7.0 and $80^{\circ}C$. By exchanging two possible residues (Glu-232 and Asp-242) to Ala by site-directed mutagenesis method, it was found that these were essential for enzymatic activity.

  • Selective action of root-treated oxyfluorfen [(2-chloro-4-thrifluoromethylphenyl)-3'-ethoxy-4'-nitrophenyl ether] and chlomethoxynil [2, 4-dichlorophenyl-3'-methoxy-4'-nitrophenyl ether] were investigated. Oxyfluorfen showed greater activity to all plant species than chlomethoxynil. $^{14}C$-oxyfluorfen was little metabolized in roots of the plant species and more slowly absorbed than $^{14}C$-chlomethoxynil. These results suggest that herbicidal activity of oxyfluorfen at the site of action is higher than chlomethoxynil. In the tested plants, rice, barnyardgrass, sorghum, and corn were absorbed less of the oxyfluorfen and chlomethoxynil than the broad leaf plant species. However, no clear relationship was observed between a degree of tolerance and absorption and metabolism of both herbicides by the plant species.

Chemical Compositions and Antioxidant Activities of Leaves and Stalks from Different Sweet Potato Cultivars (고구마 품종별 잎과 잎자루의 성분특성과 항산화활성)

  • This study was conducted to evaluate the vegetative value of sweet potato leaves and stalks by investigating their chemical composition and antioxidant activity. The major minerals were found to be Ca and K. The crude protein content of the leaves ranged from 15.57% in Geonpungmi to 20.34% in Yeonhwangmi, while that of stalks was between 3.71% in Yulmi and 5.97% in Sinchunmi. The total phenolics and flavonoids content of the leaves were about 3.0~4.0 times higher than those of stalks. The levels of polyphenols and flavonoids were high in Geonpungmi leaf and in Sinchunmi stalk. ABTS radical activities were high in Geonpungmi leaf (58.75 mg AA eq/g) and in Daeyumi stalk (14.95 mg AA eq/g). DPPH radical scavenging was high in Geonpungmi and Jeungmi leaves ($IC_{50}$, 0.11 mg/mL) and in Sinchunmi stalk ($IC_{50}$, 1.09 mg/mL).

Fungicidal and insecticidal activities of various grain extracts against five insect pests and six phytopathogenic fungi (다양한 잡곡 추출물의 살균.살충활성)

  • Methanol extracts from 21 grains were tested for fungicidal activities against six phytopathogenic fungi and for insecticidal activities toward five insect pests in a greenhouse. The efficacy varied with both the plant pathogen/insect pest and grain species used. Potent fungicidal activity at 5 mg/pot, were produced from extracts of Elymus sibiricus and Hordeum vulgare var. nudum against Pyricularia grisea and Erysiphe graminis and these of Sesamum indicum (W) and Triticum aestivum against Puccinia recondita and Erysiphe graminis. At 2,500 ppm, potent insecticidal activities were exhibited from the extracts of Fagopyrum esculentum against Myzus persicae and Ischaemum crassipes, and these of Oryzo sativa var. glutinosa, Panicum miliaceum, Setaria italica, Sorghum bicolor, and T. aestivum against Tetranychus urticae. All grain extracts revealed weak or no fungicidal and insecticidal effect against Phytophthora infestans, Plutella xylostella and Spodoptera litura. As a naturally occurring fungicide and insecticide, grain-derived materials described could be useful as new fungicidal and insecticidal products against phytopathogenic fungi and insect pests.

  • The roots of Brassica campestris ssp rapa were extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2O$. From the EtOAc fraction, three compounds were isolated through the repeated silica gel and octadecyl silica gel (ODS) column chromatography. From the results of spectroscopic data including NMR and MS, the chemical structures of the compounds were determined as caulilexin C (1), indoleacetonitrile (2) and arvelexin (3). The arvelexin (3) has been isolated from this plant for the first time. Compounds 1, 2 and 3 showed inhibitory activity on human Acyl CoA: cholesterol. transferase 1 (hACAT1) by $54.6{\pm}6.0%$, $69.2{\pm}4.7%$ and $68.6{\pm}3.7%$, and on human Acyl CoA: cholesterol transferase 2(hACAT2) by $4.8{\pm}13.4%$, $45.6{\pm}4.8%$ and $39.5{\pm}4.3%$, respectively, at 100 ${\mu}g/ml$.
