Background: A model to assess the activity concentration of agricultural products and the public ingestion dose as result of a nuclear accident is necessarily required to manage the contaminated agricultural systems by the accident, or to estimate the effects of chronic exposure due to food ingestion at a Level 3 PSA. Materials and Methods: A dynamic compartment model, which is composed of three sub-modules, namely, an agricultural plant contamination assessment model, an animal product contamination assessment model, and an ingestion dose assessment model has been developed based on Korean farming characteristics such as the growth characteristics of rice and stockbreeding. Results and Discussion: The application study showed that the present model can predict well the characteristics of the activity concentration for agricultural products and ingestion dose depending on the deposition date. Conclusion: The present model is very useful to predict the radioactivity concentration of agricultural foodstuffs and public ingestion dose as consequence of a nuclear accident. Consequently, it is expected to be used effectively as a module for the ingestion dose calculation of the Korean agricultural contamination management system as well as the Level 3 PSA code, which is currently being developed.
Dynamic parameters of biochemistry including the POD (peroxidase) activity, contents of DNA and RNA, isozymes of POD and ES (esterase) in American send (Panax quinquefolium L.) seed are reported in the present paper. The dynamic changes of POD activity proved that the PAS (physiological afterripening stage) is a stage in which some substances are prepared for seed germination. The POD activity correlated with ER (embryo ratio) significantly DNA content changed little only within 0.0036∼0.013 mg/ml, which did not correlate with ER. RNA content changed from 0.1539 to 1.0313 mg/ml and correlated significantly with RE during all of the embryo afterripening. None of the POD isozyme band was obtained in ESGS (embryo slowly growth stage), but five bands in ERGS (embryo rapidly growth stage) and six bands in PAS. Four bands of ES isozymes were obtained in ESGS, but six bands in ERGS, particularly, the content of ES isozymes increased in PAS. All of these may provide some information for understanding the dormancy mechanisms of American song seed.

A novel putative toxin-antitoxin segregational stability system named KyAB system was identified in a novel native plasmid pBMB8240 from Bacillus thuringiensis strain YBT-1520, based on sequences homology with other toxin-antitoxin systems, the lethal activity of the KyB putative toxin in Escherichia coli and the stabilizing effect of the kyAB system in Bacillus thuringiensis. Secondarily, the native plasmid pBMB9741 from the same strain was resequenced and the corrected plasmid was named as pBMB7635. Based on sequence homology with the tasAB system and the lethal activity of toxin protein in Escherichia coli, a tasAB-like putative toxin-antitoxin system was identified on pBMB7635.

The purpose of this study was to characterize the coffee ground and its possibility to develop the antifungal activity. pH, EC, and Zn contents of the coffee ground from coffee shops were higher than those of commercial coffee ground, but there was no significant difference in $K_2O$, CaO, MgO, $Na_2O$, Mn contents. The antimicrobial activity of the water soluble extracts from the coffee shop and the commercial coffee ground were tested for six major plant pathogens, Rhizoctonia solani, Sclerotinia sclerotiorum, Fusarium oxysporum, Phytophthora capsici, Alternaria alternata, and Botrytis cinerea. The result showed that there was reliable antifungal activity against all of tested plant pathogenic fungi. The inhibition effects of coffee ground compost extract on the spore germination and zoospore formation were investigated. Water soluble extracts of the coffee ground compost mixture added with 10% sesame oil cake were significantly inhibited the growth of conidia germination of A. altanata and zoospore formation of P. capsici in vitro. For investigating the functional materials of coffee ground compost, it was measured the total polyphenolic compounds contents with 30 days interval during decomposing coffee ground for 90 days. The total polyphenolic content increased with decomposing periods, and it observed that the highest total polyphenolic content was $0.35{\pm}0.03mg\;GAE/g$ on the 90th day in the coffee ground compost added with 10% sesame oil cake.

Royal jelly has been widely used as a health supplement worldwide. However, royal jelly has been implicated in allergic reactions, and we developed a water-soluble royal jelly (WSRJ) without the allergy inducing protein. In this study, we aimed to identify the anti-melanogenic efficacy of WSRJ. B16F1 melanoma cells were first treated with 10 nM α-melanocyte stimulating hormone (α-MSH) and then with various doses of WSRJ. In addition, we investigated the mRNA and protein expression of melanogenesis-related genes such as tyrosinase, tyrosinase related protein-1 (TRP-1) and TRP-2 by reverse transcription-polymerase chain reaction and western blotting. WSRJ directly inhibited tyrosinase and cellular tyrosinase activity, which decreased melanin synthesis in α-MSH stimulated B16F1 melanoma cells a level comparable to that observed with arbutin. WSRJ decreased the mRNA and protein expressions of tyrosinase, TRP-1, and TRP-2, which was comparable to that observed with arbutin. WSRJ has strong anti-melanogenic activity, which invoice direct inhibition of tyrosinase enzyme activity and suppression of expression of melanogenesis related genes. Results from this study suggests that WSRJ is a potential candidate for the treatment of skin pigmentation.

A phytase with high activity at low temperatures has great potential for feed applications, especially in aquaculture. Therefore, this study used a degenerate PCR and TAIL PCR to clone a phytase gene from Erwinia carotovora var. carotovota, the cause of soft rot of vegetables in the ground or during cold storage. The full-length 2.5-kb fragment included an open reading frame of 1,302 bp and encoded a putative phytase of 45.3 kDa with a 50% amino acid identity to the Klebsiella pneumoniae phytase. The phytase contained the active site RHGXRXP and HD sequence motifs that are typical of histidine acid phosphatases. The enzyme was expressed in Escherichia coli, purified, and displayed the following characteristics: a high catalytic activity at low temperatures (retaining over 24% activity at $5^{\circ}C$) and remarkably thermal lability (losing >96% activity after incubation at $60^{\circ}C$ for 2 min). The optimal phytase activity occurred at pH 5.5 and ${\sim}49^{\circ}C$, and the enzyme activity rapidly decreased above $40^{\circ}C$. When compared with mesophilic counterparts, the phytase not only exhibited a high activity at a low temperature, but also had a low $K_m$ and high $k_{cat}$. These temperature characteristics and kinetic parameters are consistent with low-temperature-active enzymes. To our knowledge, this would appear to be the first report of a low-temperature-active phytase and its heterogeneous expression.
This research investigated classification of clay activity degree by different clay mineral components. Based on compositions of different clay and oxide minerals within 390 soil series in Korea, 7 soils were selected to analyze for CEC and specific surface area of clay minerals. As a result, soils were mainly composed with Chlorite originated from sandstone, Smectite originated from Andesite porphyry and combination of Allophane and Ferrihydrite originated from volcanic ash, if the ratio of CEC value to clay content (degree of clay activity) was greater than 0.7. If the degree of clay activity was ranged between 0.3 and 0.7, soils were composed mainly with Kaolin originated from anorthite. Soils with this ratio also was composted with combinations of Kaolin, Illite and Vermiculite originated with river deposits. When the degree of the activity was less than 0.3, soils were commonly red-yellowish color and composed with two different minerals. One type of composition was Kaolin originated from granite and granite gneiss and the soils contained Geothite and Hematite. The other type was composited mainly with Illite and Vermiculite minerals originated from granite. These soils contained Gibbsite, Geothite and Hematite. The degree of clay activity was highly related with CEC and specific surface area. The greater degree of the activity displayed greater values of clay CEC and specific surface area. It is not easy to measure actual quantity and compositions of clay minerals, while the degree of clay activity can be measured from routine soil analyses. As a conclusion, the degree of clay activity may be not just a simple but also powerful tool to estimate physical-chemical properties of soils and to evaluate the soil classification in Korean soils.

The fibrinolytic activity of aqueous extracts from Korean indigenous insects were studied. Fibrinolytic activity of aqueous extract from 5 insects (larva and adult parts of Meloimorpha japonica, Allomyrina dichotoma, Cetonia pilifera, Apis mellifera) showed 2.7-fold potent than that of plasmin used as a positive control. In addition, the fibrinolytic activity was observed through the degradation products by SDS-PAGE. The extracts efficiently hydrolyzed ${\alpha}-$, ${\beta}-$ and ${\gamma}$ chains of human fibrinogen. This study suggested that aqueous extracts from Korean indigenous insects have potential in developing a useful source of antithrombosis agent(s).

This study was performed to compare the physiological effects of puffed snack on diabetic and geriatric diseases using miscellaneous cereals and grain crops. The puffed snacks were prepared with different amounts of miscellaneous cereals and grain crops (in ratios of 10%, 20%, and 30% of brown rice). Changes in the water soluble index, water absorption index, color, antioxidant activity, total polyphenol content acidity, ${\alpha}$-glucosidase inhibition activity, and sensory evaluation were also determined. As the cereal and crop contents increased, the value of the water soluble index increased while the water absorption index decreased, with the exception of glutinous foxtail and barnyard millets. With respect to color, lightness and yellowness decreased in concert with increases in the cereal and crop contents, whereas redness increased. Furthermore, the antioxidant activity and total polyphenol content as well as ${\alpha}$-glucosidase inhibition activity increased remarkably with increasing concentrations of sorghum. There was no significant difference in the physiological activities depending on the addition of millet, barley and barnyard millets. In sensory evaluation, the puffed snacks containing sorghum, millet, and glutinous foxtail millet received higher values than other samples. Altogether, our results indicate that puffed snacks containing 10~20% sorghum could be suitable as ingredients for improving sensory and physiological activities in diabetic and geriatric diseases.
