Leaching of copper and silver from ground mobile phone printed circuit boards using nitric acid (핸드폰 기판(基板)으로부터 구리와 은의 질산(窒酸) 침출(浸出) 연구(硏究))

  • Leaching of copper and silver from mobile phone PCBs(printed circuit boards) with nitric acid was performed to investigate the effects of nitric acid concentrations, leaching temperatures, agitation speeds, and pulp densities on the leaching behaviors of Cu and Ag. The leaching rate considerably increased with increasing acid concentration and temperature. The leaching ratios of Cu and Ag were found to be 96.4% and 96.5%, respectively, under the optimum condition; at $80^{\circ}C$ with 2mol/L $HNO_3$ and 120g/L in pulp density within 39minutes. The kinetic parameters were determined based on the shrinking core model with reaction control corresponding to small particles. The activation energies for the leaching of copper and silver were found to be 45.5kJ/mol and 60.5kJ/mol at $35{\sim}80^{\circ}C$ with 2mol/L $HNO_3$, respectively.

Optimization of Medium and Fermentation Conditions for Mass Production of Bacillus licheniformis SCD121067 by Statistical Experimental Design (Bacillus licheniformis SCD121067 균체 생산성 증가를 위한 통계적 생산배지 및 발효조건 최적화)

  • In this work, mass production of Bacillus licheniformis SCD121067 through medium optimization by statistical experimental method was studied. First, galactose, yeast extract and potassium phosphate dibasic were selected as carbon, nitrogen and phosphate sources for mass production of B. licheniformis SCD121067 by using one factor at a time method. Second, according to the result of Plackett-Burman experimental design, key factors was yeast extract and $K_2HPO$. Finally, the response surface methodology was performed to obtain the optimum concentrations of two selected variables. The optimized medium composition consisted of 20 g/L galactose, 36 g/L yeast extract, 0.41 g/L $K_2HPO4$, 0.25 g/L $Na_2CO_3$, 0.4g/L $MgSO_4$ and 0.01g/L $CaCl_2$. Dry cell weight (15.4 g/L) by optimum production medium were increased 10 times, as compared to that determined with basic production medium (1.5 g/L). Fermentation conditions were examined for the mass production of B. licheniformis. The effect of temperature, agitation speed, pH and aeration rate on the mass production of B. licheniformis were also studied in a batch fermenter which was carried out in a 2.5 L bioreactor with a working volume of 1.5 L containing optimized production medium. As a result, dry cell weight of batch culture was 30.7 g/L at $42^{\circ}C$, 300 rpm, pH 8.0 and 2 vvm.

Octadecyl-Modified Graphene as an Adsorbent for Hollow Fiber Liquid Phase Microextraction of Chlorophenols from Honey

  • Octadecyl-modified graphene (graphene-C18) was fabricated and used as adsorbent in hollow fiber liquid phase microextraction (HF-LPME) for the first time. The extraction performance of graphene-C18 reinforced HF-LPME was evaluated using chlorophenols as model analytes. The factors affecting the extraction efficiency, such as extraction time, pH of the sample solution, agitation rate, the concentration of graphene-C18 and salt addition were optimized. After the graphene-C18 reinforced HF-LPME of the chlorophenols from honey sample, the analytes were separated and determined by high-performance liquid chromatography. The linearity was observed in the range of 5.0-200.0 ng $g^{-1}$ for 2-chlorophenol and 3-chlorophenol, and 2.0-200.0 ng $g^{-1}$ for 2,3-dichlorophenol and 3,4-dichlorophenol, respectively. The limits of detection (S/N = 3) of the method were lower than 1.5 ng $g^{-1}$. The recoveries of the method were between 88% and 108%. The method is simple, sensitive and has been resoundingly applied to analysis of chlorophenols in honey samples.

Characterization of biopolymer produced by Bacillus coagulans DL-1 and optimization of its production (Bacillus coagulans DL-1이 생산하는 고분자 중합체의 특성과 생산조건의 최적화)

  • The microorganism isolated from soil was identified as Bacillus coagulans by morphological and biochemical analyses and API-50CH/B kit, which was an identification kit for Bacillus species, and named as B. coagulans DL-1. It produced an extracellular biopolymer. Maximum production of biopolymer was 5.00 $\pm$0.15 g/$\ell$ in a $7\ell$bioreactor with an aeration rate of 1.0 vvm and an agitation speed of 500 rpm when concentrations of glucose and yeast as the optimal carbon and nitrogen sources were 2.0% (w/v) and 0.25% (w/v), which were optimized with a flask scale. Gas chromatographic analysis showed that the biopolymer producded by B. coagulans DL-1 consisted of glucose and rhanmose and their molar ratios was about 9 : 1. Its average molecular weight was 2.80$\times$$10^5$ with gel permeation chromatographic (GPC) analysis.

Cultural Characteristics and Scale-up for Submerged Cultivation of Hericium erinaceum Through Air-lift and Jar Fermenter System (Air-lift 및 Jar Fermenter에 의한 Hericium erinaceum 심부배양의 배양특성 및 Scale-up)

  • For the study of Hericium erinaceum as a useful functional foods and materials, liquid cultivation under two different bioreactors(air-lift fermenter and jar fermenter) which was not studied systematically until now, was conducted as a method of mass cultivation for H. erinaceum. A batch cultivation in an air-lift fermenter and a jar fermenter was examined for enhancing the productivity because of small amounts of mycelial weight and slow growth in case of a liquid culture for H. erinaceum. We found that air lift fermenter system was more effective than jar fermenter for mycelial production of H. erinaceum, and mycelial morphology was a critical factor of the growth. By scale-up and cultivation based on morphological analysis, the conditions for mass production with 30 L and 500 L jar fermenter was 200 and 150 rpm of agitation speed at 1 vvm of aeration rate, respectively, and mycelial dry weight under these conditions was enhanced to about $13{\sim}14g/L$.

  • SMEDDS is mixture of oils, surfactants, and cosurfactants, which are emulsified in aqueous media under conditions of gentle agitation and digestive motility that would be encountered in the gastro-intestinal(GI) tract. The main purpose of this work is to prepare self-microemulsifying drug delivery system(SMEDDS) for oral bioavailability enhancement of a poorly water soluble drug, atorvastatin calcium. Solubility of atorvastatin calcium was determined in various vehicles. Pseudo-ternary phase diagrams were constructed to identity the efficient self-emulsification region and particle size distributions of the resultant micro emulsions were determined using a laser diffraction sizer. Optimized formulations for in vitro dissolution and bioavailability assessment were $Capryol^{(R)}$ 90(50%), Tetraglycol(16%), and $Cremophor^{(R)}$ EL(32%). The release rate of atorvastatin from SMEDDS was significantly higher than the conventional tablet ($Lipitor^{(R)}$), 2-fold. Our studies illustrated the potential use of SMEDDS for the delivery of hydrophobic compounds, such as atorvastatin calcium by the oral route.

Optimization of Fermentation Process for Acetic Acid Production (초산 생성을 위한 발효공정의 최적화)

  • Various conditions of acetic acid fermentation by Acetobacter aceti B20 strain were investigated and evaluated to optimize the fermentative production of acetic acid. The effects of the initial ethanol concentration on growth and acid productivity in a flask and fermentor were also studied. The growth of A. aceti B20 strain was inhibited as the concentration of ethanol increased. However, the highest total acidity and fermentation yield were 5.34% and 56.1%, respectively when the initial concentration of ethanol was 7% in the batch fermentation. Although the concentration of initial glucose influenced the growth rate of B20 strain, it did not influence the total acidity in the flask culture. When the agitation speed increased, the growth, total acidity and fermentation yield were all improved. In fed-batch fermentation, total acidities and fermentation yields were 7.14-8.76% and 39.1-53.0%, respectively, and their values mostly depended on the feeding methods.

Statistical Optimization of Biosurfactant Production from Aspergillus niger SA1 Fermentation Process and Mathematical Modeling

  • In this study, we sought to investigate the production and optimization of biosurfactants by soil fungi isolated from petroleum oil-contaminated soil in Saudi Arabia. Forty-four fungal isolates were isolated from ten petroleum oil-contaminated soil samples. All isolates were identified using the internal transcribed spacer (ITS) region, and biosurfactant screening showed that thirty-nine of the isolates were positive. Aspergillus niger SA1 was the highest biosurfactant producer, demonstrating surface tension, drop collapsing, oil displacement, and an emulsification index (E24) of 35.8 mN/m, 0.55 cm, 6.7 cm, and 70%, respectively. This isolate was therefore selected for biosurfactant optimization using the Fit Group model. The biosurfactant yield was increased 1.22 times higher than in the nonoptimized medium (8.02 g/l) under conditions of pH 6, temperature 35℃, waste frying oil (5.5 g), agitation rate of 200 rpm, and an incubation period of 7 days. Model significance and fitness analysis had an RMSE score of 0.852 and a p-value of 0.0016. The biosurfactant activities were surface tension (35.8 mN/m), drop collapsing (0.7 cm), oil displacement (4.5 cm), and E24 (65.0%). The time course of biosurfactant production was a growth-associated phase. The main outputs of the mathematical model for biomass yield were Yx/s (1.18), and µmax (0.0306) for biosurfactant yield was Yp/s (1.87) and Yp/x (2.51); for waste frying oil consumption the So was 55 g/l, and Ke was 2.56. To verify the model's accuracy, percentage errors between biomass and biosurfactant yields were determined by experimental work and calculated using model equations. The average error of biomass yield was 2.68%, and the average error percentage of biosurfactant yield was 3.39%.

Characteristics of the Oxygen Uptake Rate of Entomopathogenic Nematodes Steinernema spp. (곤충병원성 선충 Steinernema spp.의 산소 요구도 특성)

  • Recently, entomopathogenic nematodes have received a considerable attention as biologicalcontrol agents. For in vitro cultivation, storage and transportation of nematodes, oxygen supply isextremely impotant due to its limited solubility and mass transfer problem. The oxygen uptake rates(OURs) of four different Steinernema species were measured in a 5L bioreactor at varying temperatures.The OURs of the Steinernema spp. were below 0.5 x mmolO'||'&'||' . min in the range of 13-17$^{\circ}$C. TheOURs (mmo102/L - min) of S. glaseri Dongrae and S. carpocapsae Pocheon strains were 0.4 x lo-', 0.75x lo-\ulcorner at 21$^{\circ}$C, 1.5 x lo-\ulcorner, 3.2 x 10-2 at 25"C, and 2.8 x lo-', 5.8 x lo-\ulcorner at 29"C, respectively. However,the OURs were not significantly altered by the agitation speed of 50-150 rpm. The specific oxygenuptake rates (qol) of S. glaseri NC, S. glaseri Dongrae, S. glaseri Mungyeong and S. carpocapsaePocheon strains were 0.3 x 0.5 x 0.3 x and 0.2 x mmolO~/cell min at 25"C,respectively. Removal of Cs by Adsorption with IE911 (Crystalline Silicotitanate) from High-Radioactive Seawater Waste (IE911 (crystalline silicotitanate) 의한 고방사성해수폐액으로부터 Cs의 흡착 제거)

  • This study was performed on the removal of Cs, one of the main high- radioactive nuclides contained in the high-radioactive seawater waste (HSW), by adsorption with IE911 (crystalline silicotitanate type). For the effective removal of Cs and the minimization of secondary solid waste generation, adsorption of Cs by IE911 (hereafter denoted as IE911-Cs) was effective to carry out in the m/V (ratio of absorbent weight to solution volume) ratio of 2.5 g/L, and the adsorption time of 1 hour. In these conditions, Cs and Sr were adsorbed about 99% and less than 5%, respectively. IE911-Cs could be also expressed as a Langmuir isotherm and a pseudo-second order rate equation. The adsorption rate constants (k2) were decreased with increasing initial Cs concentrations and particle sizes, and increased with increasing ratios of m/V, solution temperatures and agitation speeds. The activation energy of IE911-Cs was about 79.9 kJ/mol. It was suggested that IE911-Cs was dominated by a chemical adsorption having a strong bonding form. From the negative values of Gibbs free energy and enthalpy, it was indicated that the reaction of IE911-Cs was a forward, exothermic and relatively active at lower temperatures. Additionally, the negative entropy values were seen that the adsorbed Cs was evenly distributed on the IE911.
