The Fabrication of Megasonic Agitated Module(MAM) for the Improved Characteristics of Wet Etching

  • The MAM(Megasonic Agitated Module) has been fabricated for improving the characteristics of wet etching. The characteristics of the MAM are investigated during the wet etching with and without megasonic agitation in this paper. The adoption of the MAM has improved the characteristics of wet etching, such as the etch rate, etch uniformity, and surface roughness. Especially, the etching uniformity on the entire wafer was less than ${\pm}1%$ in both cases of Si and glass. Generally, the initial root-mean-square roughness($R_{rms}$) of the single crystal silicon was 0.23nm. Roughnesses of 566nm and 66nm have been achieved with magnetic stirring and ultrasonic agitation, respectively, by some researchers. In this paper, the roughness of the etched Si surface is less than 60 nm. Wet etching of silicon with megasonic agitation can maintain nearly the original surface roughness during etching. The results verified that megasonic agitation is an effective way to improve etching characteristics of the etch rate, etch uniformity, and surface roughness and that the developed micromachining system is suitable for the fabrication of devices with complex structures.

Monitoring for the Fermentation Conditions of Sweet Persimmon Vinegar using Response Surface Methodology (반응표면분석에 의한 단감식초 제조조건의 모니터링)

  • To utilize deteriorated sweet persimmon effectively, response surface methodology(RSM) was used to determine the optimal vinegar fermentation conditions and monitored by a divided two stage fermentation. The optimum conditions for maximum alcohol content were obtained when the first stage (alcohol fermentation) was carried out with an initial sugar concentration of 18.5$^{\circ}$Brix, agitation rate of 140.8 rpm, fermentation time of 127.6 hr. When sugar concentration was 14$^{\circ}$Brix maximum alcohol content(7.1%) was predicted at fermentation conditions of 160 rpm in agitation rate, 140hr in fermentation time. The optimum conditions for maximum acidity were obtaiend when second stage(vinegar fermentation)was carried out 249.5 rpm in agitation rate, 148.8 hr in fermentation time. Predicted values at the optimum conditions were similar to experimental values.

The Effect of Hand-Massage on the Relaxation of Dementia Patients (치매환자에 대한 손마사지 효과 연구)

  • The agitation of dementia patient is a factor which aggravates the care-givers' burdon, and it is needed to develop intervention methods. Hall and Buckwalter hypothesized that an increase in agitation behavior occur when persons experience high levels of stress. This study was conducted to explore the efficacy of hand massage in producing relaxation and decreasing agitation behaviors in persons with dementia. Hand massage was administered once a day in afternoon for 8days to 24 patients and 4days to 23 patients. There was no difference in result between the two groups. Significant differences were found in systolic blood pressure, pulse rate, emotional state from pre to post -intervention with the use of hand massage. The change of systolic blood pressure was 26% explained by ADL, MMSE-K and institutionalized period. The most significant variable was institutionalized period. The change of diastolic blood pressure was 11% explained by ADL, and it was significant. But no decrease in agitation behavior was observed and no increase in functional capacity.

Effect of Agitation on the in vitro Permeability of Xenobiotics across Caco-2 Cell Monolayers (Caco-2 세포 단층막 투과 실험시 교반이 약물의 투과계수에 미치는 염향)

  • The unstirred water layer (UWL), which has been known to exist in the boundary of the intestinal lumen and intestinal wall, often behaves as an absorption barrier especially for lipophilic drugs. The intestinal absorption of drugs is often characterized using Caco-2 cell monolayers grown on Transwell polycarbonate membranes. The permeability $(P_{app})$ of drugs across the cell monolayer might be influenced by the agitation of the donor compartment, since the width of UWL on the surface of the cell monolayer would be reduced by the agitation. In this study, the effect of agitation of the donor compartment with 60 rpm on the permeability was measured for 12 drugs with a wide range of lipophilicity and permeability. The $P_{app}$ of mannitol, tributylmethyl ammonium, cimetidine, ranitidine, hydrocortisone, benzylpenicillin and loxoprofen was not influenced by the agitation, while the $P_{app}$ of theophylline, propranolol, YH439, phenylpropanolamine and testosterone was increased by the agitation. There was a significant correlation between the increase of $P_{app}$ by agitation and the lipophilicity for the compounds having $P_{app}>2{\times}10^{-5}$ cm/sec. No correlation was observed for the difference in $P_{app}$ by agitation and the molecular weight, or lipophilicity of the drugs. Therefore, the agitation rate of the donor compartment in the Caco-2 cell monolayer study should be carefully controlled in order to estimate $P_{app}$ reproducibly especially for lipophilic drugs.

Determination of Oxygen Transfer Coefficient in Fed-Batch Culture of Streptomyces avermitilis with Concentrated Medium Control (농축 배지 조절 유가식 배양에 의한 Streptomyces avermitilis의 산소전달계수 측정)

  • The large-scale production of antibiotics by filamentous mycelial organism requires and adequate supply of dissolved oxygen. In terms of productivity, it means that oxygen transfer is the rate-limiting step. Therefore, the oxygen transfer coefficients(K$\_$L/A) were determined in a broth involving a filamentous mycelial organism such as Streptomyces avermitilis for use in fermentations. To determine (K$\_$L/A) inn a stirred vessel, a great deal of effort is required to provide all the cells with a sufficient oxygen supply. To overcome the oxygen limitation in a batch culture, a fed-batch culture was applied to control the growth rate by an intermittent supply of nutrients. Thus, it was possible to maintain a suitable dissolved oxygen concentration at a low agitation rate. The optimal agitation speed was 350 rpm at low cell concentrations (below 7 g/L) by considering the efficiency of agitation and shear stress. The (K$\_$L/A) was found to decrease from 64.26 to 29.21h.$\^$-1/ when the biomass concentration was increased from 9.82 to 12.06 g/L. In addition, and increase in viscosity was also observed during the growth phase. By comparing the (K$\_$L/A) values for the various agitation and aeration rates, it was found that the effect of an increase in (K$\_$L/A) by aeration was reduced dramatically at high biomass concentrations. However, this effect was not observed when altering the agitation rate. This suggests that controlling the dissolved oxygen concentration by altering the agitation rate was more efficient than increase the aeration rate.

  • The effect of agitation speed and aeration rate on mTG production and cell growth by Streptoverticillum morbarense was investigated. Dissolved oxygen was controlled by on-line computer-controlled fermentation system. The agitation speed and aeration rate of 2.5 L fermentor ranged from 330 to 360 게m and 1 vvm to 4 vvm, respectively. The highest mTG production was 2.1 U/mL when dissolved oxygen level was 20%, and it was improved almost 1.1 times in comparison with that without dissolved uxygen control.

  • The extraction rate of lipids from the chopped whole fish was studied with various solvents. Factors which can influence on the extraction rate such as temperature, moisture content, agitation speed and solvent were also examined. In the early stage of extraction, it is considered that the rapid extraction was attributed to cell destruction which occurs in chopping the whole fish and in the later stage, the extraction rate was increased linearly with extraction time. The effect of agitation on the extraction rate had a great influence on the early stage of extraction. In agitation speed of 100, 200, 300 and 500 r.p.m. the slopes of extraction curve were -0.075, -0.075, -0.069 and -0.064, respectively. Extractability between hydrophilic and hydrophobic solvent showed a great difference. It is suggested that extractability difference between acetone and isopropyl alcohol is due to acetone property which can not extract phospholipids in polar lipids. Extractability of lipids was increased with increasing temperature and decreased with increasing moisture content.

  • Effect of ultrasonic agitation on Pd catalyst treatment was studied in metallization of ceramic boards by Cu electroless plating method.96% $Al_{2}$$O_{3}$ ceramic boards were used as substrate. In this study, the ultrasonic frequency of 28kHz was applied. In Pd catalyst, high density Pd nuclei of small size were formed during ultrasonic agitation. Density of Pd was more improved when using of ultrasonic then no stirring. In electroless plating, plating rate was in the range of 0.6~1.8$\mu\textrm{m}$/hr, which value increased with Rochelle Salts addition. Adhesion strength between ceramic boards and Cu layer was improved of 20% when using ultrasonic agitation at $30^{\circ}C$ ,5min.

  • The influence of temperature and agitation on the growth of Escherichia coli expressing hepatitis B core antigen (HBcAg) in stirred tank bioreactor were investigated. The highest specific growth rate for E. coli$(0.844 h^{-1})$ was achieved at a temperature of $37^{\circ}C$ and an agitation speed of 250 rpm. The activation energy for the growth of the E. coli strain W3110lQ in the stirred tank bioreactor was estimated to be 11 kcal/mol. The highest protein yield was achieved at a temperature of $44^{\circ}C$ and an agitation speed of 250 rpm. The relative protein concentration at $44^{\circ}C$ is 30 and 6% higher compared to that at 30 and $37^{\circ}C$, respectively.

Effect of Aeration-Agitation on Coenzyme Q10 Production Using Rhodobacter sphaeroides

  • With the aim of increasing the $CoQ_{10}$ production in mass culture, the effect of aeration-agitation on the $CoQ_{10}$ production using Rhodobactor sphaeroides was investigated in a l-L bioreactor. The maximum $CoQ_{10}$ production was 1.69 mg/g of dry cell weight under conditions of 50 Lux, $30^{\circ}C$, 300 rpm, and 5-vvm aeration. The $CoQ_{10}$ production was improved to produce 2.91 mg/g of dry cell weight under reduced conditions of agitation speed (200 rpm) and aeration rate (0.2 vvm). When R. sphaeroides was cultivated under more reduced DO levels during the exponential phase of the cell, the $CoQ_{10}$ production yield of 3.88-mg/g dry cell weight was the maximum obtained. Therefore, poorer conditions of aeration-agitation resulted in higher production of $CoQ_{10}$, and thus DO content was a crucial factor in the production of $CoQ_{10}$. Accordingly, it was necessary to control the DO concentration in order to enhance the $CoQ_{10}$ biosynthesis within a large-scale production.
