Enterobacter sp.의 다당 생산에 미치는 통기·교반의 단계적 증가 효과 (Stepwise Increasing Effects of Agitation and Aeration on Exopolysaccharide Production by Enterobacter sp.)

  • The effects of agitation and aeration for exopolysaccharide(EPS) production through batch cultivation of an Enterobacter sp. isolated from the composter were investigated. During the EPS fermentation under conditions of constant agitation speed from 200 to 900 rpm and constant aeration rate of 0.5-2.5 vvm, the low yields of EPS(4.8-5.2g/L) was observed as the viscosity increase of culture broth. With the stepwise increases in agitation speed and aeration rate, the EPS production and the viscosity of EPS were increased 1.3~1.4 times and 2.3~3.6 times higher than those of the fixed conditions, respectively. Therefore, these stepwise increases were considered as the key operating parameters for enhancing EPS production. The max. EPS(6.8g/L) and viscosity(14,000cp) were obtained when the agitation speed was increased from 300 to 900 rpm for 54hrs at 1.5 vvm.

반응표면분석에 의한 단감식초 제조조건의 모니터링 (Monitoring for the Fermentation Conditions of Sweet Persimmon Vinegar using Response Surface Methodology)

  • To utilize deteriorated sweet persimmon effectively, response surface methodology(RSM) was used to determine the optimal vinegar fermentation conditions and monitored by a divided two stage fermentation. The optimum conditions for maximum alcohol content were obtained when the first stage (alcohol fermentation) was carried out with an initial sugar concentration of 18.5$^{\circ}$Brix, agitation rate of 140.8 rpm, fermentation time of 127.6 hr. When sugar concentration was 14$^{\circ}$Brix maximum alcohol content(7.1%) was predicted at fermentation conditions of 160 rpm in agitation rate, 140hr in fermentation time. The optimum conditions for maximum acidity were obtaiend when second stage(vinegar fermentation)was carried out 249.5 rpm in agitation rate, 148.8 hr in fermentation time. Predicted values at the optimum conditions were similar to experimental values.

Effect of Aeration-Agitation on Coenzyme Q10 Production Using Rhodobacter sphaeroides

  • With the aim of increasing the $CoQ_{10}$ production in mass culture, the effect of aeration-agitation on the $CoQ_{10}$ production using Rhodobactor sphaeroides was investigated in a l-L bioreactor. The maximum $CoQ_{10}$ production was 1.69 mg/g of dry cell weight under conditions of 50 Lux, $30^{\circ}C$, 300 rpm, and 5-vvm aeration. The $CoQ_{10}$ production was improved to produce 2.91 mg/g of dry cell weight under reduced conditions of agitation speed (200 rpm) and aeration rate (0.2 vvm). When R. sphaeroides was cultivated under more reduced DO levels during the exponential phase of the cell, the $CoQ_{10}$ production yield of 3.88-mg/g dry cell weight was the maximum obtained. Therefore, poorer conditions of aeration-agitation resulted in higher production of $CoQ_{10}$, and thus DO content was a crucial factor in the production of $CoQ_{10}$. Accordingly, it was necessary to control the DO concentration in order to enhance the $CoQ_{10}$ biosynthesis within a large-scale production.

교반속도 최적화를 통한 Kluyveromyces marxianus 36907-FMEL1로 부터 고온에서의 자일리톨 생산성 증진 (Xylitol Production by Kluyveromyces marxianus 36907-FMEL1 at High Temperature was Considerably Increased through the Optimization of Agitation Conditions)

  • 최근 모균주인 K. marxianus ATCC 36907 보다 xylose reductase의 활성이 증대된 돌연변이 K. marxianus 36907-FMEL1 균주를 개발하였다. 본 연구에서는 효율적인 xylitol의 생산을 위해 발효기를 이용하여 교반속도에 따른 효과를 확인하였다. $30^{\circ}C$ 조건에서 발효를 진행한 결과 K. marxianus 36907-FMEL1 균주는 400 rpm의 교반속도에서 가장 높은 수율 (0.57 g/g)과 생산성($0.64g{\cdot}l^{-1}{\cdot}h^{-1}$)을 확인할 수 있었다. 흥미롭게도 온도 조건을 $40^{\circ}C$로 증가하여 발효를 진행한 결과 agitation 조건 최적화를 통해 수율과 생산성이 각각 21% (0.64 g/g)와 58% ($0.90g{\cdot}l^{-1}{\cdot}h^{-1}$) 증가하였다.

  • 透析液 및 溶媒液의 攪拌이 透膜面에 미치는 流體力學的인 壓力分布狀態를 究明하였고, 攪拌으로 招來되는 透膜面兩面의 壓力不均衡을 除去할 目的으로 攪拌裝置의 開發을 試圖하였다. 다음 攪拌速度가 透析係數에 미치는 影響을 究明하여 가장 正確한 透析係數를 求할 수 있는 攪拌速度를 確定시켰다.

  • Aeration was found to affect the biological denitrification by Ochrobactrum authropi SY509. Although cell growth was vigorous under 1 vvm of aeration and an agitation speed of 400 rpm in a 3-L jar fermentor, almost no nitrate was removed. Yet under low agitation speeds (100, 200, and 300 rpm), denitrification occurred when the dissolved oxygen was exhausted shortly af-ter the inoculation of the microorganism. Ochrobactrum authropi SY509 was found to express highly active denitrifying enzymes under anaerobic conditions. The microorganism also synthesized denitrifying enzymes under aerobic conditions (1 vvm and 400 rpm), yet their activity was only 60% of the maximum level under anaerobic conditions and the nitrate removal efficiency was merely 15%. However, although the activities of the denitrifying enzymes were inhibited in the presence of oxygen, they were fully recovered when the conditions were switched to anaerobic conditions.

Optimization of submerged culture conditions for roridin E production from the poisonous mushroom Podostroma cornu-damae

  • Roridin E, produced by Podostroma cornu-damae, is a mycotoxin with anticancer activity. To increase the content of roridin E, submerged culture conditions were optimized using response surface methodology. Three factors, namely, medium initial pH, incubation time and agitation speed were optimized using a Box-Behnken design. The optimum submerged culture conditions to increase the content of roridin E included a medium with an initial pH of 4.0, an incubation time of 12.90 days, and an agitation speed of 63.03 rpm. The roridin E content in the submerged culture, under the aforementioned conditions, was 40.26 mg/L. The findings of this study can help lower the current price of roridin E and promote its related research.

딸기식초 제조를 위한 초산발효 조건 최적화 (Optimization of the Acetic Acid Fermentation Condition for Preparation of Strawberry Vinegar)

  • 딸기식초 제조를 위해 1차 단계로 Saccharomyce kluyeri DJ97을 이용하여 딸기과즙을 14$^{\circ}$Brix로 보당한 후 28$^{\circ}C$에서 50시간동안 발효시켜 딸기 알콜발효액을 얻은 후 사용하였다. 2차 단계에서, 초산발효를 시킨 결과 산도는 초기산도가 1.48%, 교반속도가 195.76 rpm, 발효시간이 7.34 day일 때 가장 높은 4.60%를 나타내었다. 색차계 a값은 초기산도가 1.78%, 교반속도가 117.63 rpm. 발효시간이 7.35 day일 때 가장 높은 11.82를 나타내었다. 딸기식초 제조를 위해서는 Acetobacter sp. PA97를 이용하여 초기산도 1.5% 및 교반속도 196 rpm에서 176시간 발효함으로서 산도 4% 이상의 식초를 얻었다.

  • Micro-structures fabricated by X-ray lithography are largely affected by doses, development conditions and other factors. For these reasons, PMMA development rates and its surface profiles under various development conditions were obseued. Development rates were measured in the rage from 1 to 6 $kJ/cm^3$ using the 9C1 white beamline of Pohang light source(PLS). In this experiment, we observed that development rates of stacked PMMA sheet using Si filter were relatively higher than that of not using Si filter. Furthermore, development rates in condition with the acoustic agitation(1 MHz, 3.67 $W/cm^3$) were twice than that in dipping condition with $35^{\circ}C$ developer considering the PMMA sheets-substrate bond strength

고율 조류 안정화지에서 교반속도가 영양염류 제거에 미치는 영향에 관한 유체동역학적 연구 (A Study on Fluid Dynamics for Effect of Agitation Velocity on Nutrients Removal in High Rate Algae Stabilization Pond)

  • HRP(high rate pond) which had kept the manufactured clay of 3cm-thickness as benthic clay in reactor and the 6 flat-blade turbine as impeller for agitation was named HRASP(high rate algae stabilization pond). And the experiment for treatment of artificial synthesis wastewater containing COD :300mg/$\ell$, NH$_3$-N : 300mg/$\ell$, T-P : 9mg/$\ell$ as nutrients was been performed successfully. This reactor was been operated under conditions : 24hrs.-irradiation and water temperature, $25^{\circ}C$ and pH 7 and agitation velocity, 15, 30, 45rpm and the effect of agitation velocity on algal bioaccumulation of nutrients was been studied with view point of fluid dynamics. The next followings could be obtained as results. 1. The agitation with a turbine impeller blade in HRASP makes clay particle indicate superior suspension effect by means of forming of excellent curl/shear flow in reactor. 2. The excessive suspension of clay particle which is created at 45rpm as rotation velocity of impeller blade of turbine disturbs the light penetration and algal photosynthesis reaction. 3. Efficiencies for removal of nutrients come out as COD : 93.9%~94.3%, ($NH_3-N + NO_3-N$) : 81.9%~99.0%, T-P : 46.8%~53.6%. 4. Kuo values of $K_1$for algal growth come out seperately as 15rpm : $1.876{\times}10^{-2}, 30rpm : 4.618{\times}10^{-3}$. 5. Kuo values of $K_2$for removal of N, P come out seperately as 15rpm : $8.403{\times}10^{-1}$ and $1.397{\times}10^{-1}$, 30rpm : $4.823{\times}10^{-1} and 2.052{\times}10^{-1}$. 6. It can be guessed easily that the excessive agitation can inhibit the algal and bacterial symbiotic reaction if it is considered that micro organism\` sense to preservation of life is relied on natural function of metabolism. Therefore the studies for this matter should be followed continuously.
