A Comparative Study on the Statistical Methodology to Determine the Optimal Aggregation Interval for Travel Time Estimation of the Interrupted Traffic Flow (단속류 통행시간 추정을 위한 적정 집락간격 결정에 관한 통계적 방법론 비교 연구)

  • The goals of this paper are two folds: i) to evaluate whether the data collected by a license plate matching AVI equipment being operated on some segment of a national highway are suitable or not for use in travel time estimation of interrupted traffic flows; ii) to study the statistical methodologies to be used for the determination of the optimal aggregation interval for travel time estimation. In this study it was found that the AVI data are not representative because the data are collected on some selected lanes of a roadway where main traffic is thru-traffic and, thus the AVI data are different from those collected from all lanes in traffic characteristics. For the determination of the optimal aggregation interval for travel time estimation. two statistical methods. namely point estimation and interval estimation. were tested. The test shows that the point estimation method is more sensitive and gives more desirable results in determing the optimal aggregation interval than the interval estimation method. And it turned out that the optimal aggregation interval on interrupted traffic flows has been calculated as 5 minute and thus the existing aggregation interval. 5 minute is proper.

Feasibility of Coculture Method for Production of Chimeric Mice Using J1 Embryonic Stem Cells

  • The demand for the production of gene-defective mice from embryonic stem (ES) cells is increasing to clarify decisive gene function in vivo. Although blastocyst injection is widely used to generate ES cell-mediated knockout mice, coculture method has been alternatively used because of several advantages, such as low cost and simple procedure. Thus, this experiment was designed to demonstrate the feasibility of the coculture method using J1 ES cells, which are known to be efficient for blastocyst injection. Eight-cell embryos were harvested from 2.5 days post-coitum (dpc), denuded with acid tyrode's solution, and transferred onto trypsinized J1 ES cells. Aggregation was carried out following two typical methods, which are simple coculture method and aggregation in groove prepared by aggregation needle. Successfully aggregated-embryos were developed to blastocysts for 24 h and transferred into uterus of pseudo-pregnant foster mother. Chimeric offspring was judged by coat pigmentation. In this study, we could obtain chimeric mice from all the two aggregation methods, but the chimera production efficiencies in coculture using groove were three times higher at least than those in the other group. In conclusion, these observations suggest that coculture method should be available for production of knockout mice from J1 ES cells. Presently, the germ-line transmission rates of the chimeras produced from the two methods are under investigation.

  • Spatial data warehouses provide analytical information for decision supports using SOLAP (Spatial On-Line Analytical Processing) operations. Many researches have been studied to reduce analysis cost of SOLAP operations using pre-aggregation methods. These methods use the index composed of fixed size nodes for supporting the concept hierarchy. Therefore, these methods have many unused entries in sparse data area. Also, it is impossible to support the concept hierarchy in dense data area. In this paper, we propose a dynamic pre-aggregation index method based on the spatial hierarchy. The proposed method uses the level of the index for supporting the concept hierarchy. In sparse data area, if sibling nodes have a few used entries, those entries are integrated in a node and the parent entries share the node. In dense data area, if a node has many objects, the node is connected with linked list of several nodes and data is stored in linked nodes. Therefore, the proposed method saves the space of unused entries by integrating nodes. Moreover it can support the concept hierarchy because a node is not divided by linked nodes. Experimental result shows that the proposed method saves both space and aggregation search cost with the similar building cost of other methods.

  • Co(II)-tetrasulfonated phthalocyanine (CoTSP) is known to be aggregated to dimer at high concentration levels in water. A study on the aggregation of CoTSP using multivariate curve resolution analysis of the visible absorbance spectra over a concentration range of 30, 40 and 50 ${\mu}M$ in the presence of dimethyl sulfoxide (DMSO), dimethyl formamide (DMF), acetonitrile (AN) and ethanol (EtOH) in the concentration range of 0 to 3.57 M is conducted. A hard modeling-based multivariate curve resolution method was applied to determine the dissociation constants of the CoTSP aggregates at various temperatures ranging from 25, 45 and $65^{\circ}C$ and in the presence of various co-solvents. Dissociation constant for aggregation was increased and then decrease by temperature and concentration of phthalocyanine, respectively. Utilizing the vant Hoff relation, the enthalpy and entropy of the dissociation equilibriums were calculated. For the dissociation of both aggregates, the enthalpy and entropy changes were positive and negative, respectively. Molecular dynamics simulation of cosolvent effect on CoTSP aggregation was done to confirm spectroscopy results. Results of radial distribution function (RDF), root mean square deviation (RMSD) and distance curves confirmed more effect of polar solvent to decrease monomer formation.

Effects of Acori Rhizoma Extract on the in vitro Anti-platelet Activity in Human Whole Blood (창포류 추출물이 인간 전혈혈소판 응집억제에 미치는 영향)

  • Objectives : Acori Rhizoma is one of the common widely used herbal medicines with diverse bioactive effects. However, little evidence has been reported about the potential anti-platelet activity of Acori Rhizoma. The present study examined the effects on platelet aggregation by Acori Rhizoma. Methods : In this study, we tested the in vitro effect of 16 kinds of Acori Rhizoma extracts by hot water or 70% ethanol on collagen-induced platelet aggregation in human whole blood using the impedance method of aggregometry. Results : Among them, 2 kinds of 70% ethanol extract and 1 kind of hot water extract showed the significant inhibiting effect on whole blood aggregation. In particular, Acorus gramineus extracts were selected as the most effective candidate. Conclusiions : The results from this experiment provide pharmacological evidence for the traditional medicine, suggesting that Acorus gramineus could be help problems of blood circulation more than Acorus tatarinowii.

Development of Protein Delivery System using Pullulan Acetate Microspheres (PAM) (플루란 아세테이트 미립구를 이용한 단백질 전달 시스템 개발)

  • The aim of this study was to develop new protein/peptide depot system instead of poly(DL-lactic acid-coglycolic acid) (PLGA) microspheres. Pullulan was chemically modified by the addition of acetic anhydride (pullulan acetate; PA) and then investigated as new depot system for protein/peptide delivery. PA microspheres (PAM) with lysozyme as a model protein were prepared by w/o/w double emulsion method. The microspheres had a mean size of 10-50 mm with a spherical shape. The size distributions reduced with increasing the degree of acetylation. The loading efficiency of lysozyme was also increased. Lysozyme aggregation behavior in the microsphere was monitored to estimate the change of protein stability during preparation step. The ratios of protein aggregation in PAMs are lower than that of PLGA microsphere, in particular, PA 5 showed lowest as about 16%. The result indicated that the increase of acetylation suppressed the aggregation of protein. The release profiles of lysozyme from PAMs were significantly different. High acetylation effectively improved lysozyme release kinetics by reducing initial burst release and extending continuous release over a period of time. To check the effect of preservation for structural stability of lysozyme, the activity of lysozyme released from PA 5 was also observed. The activity of lysozyme was maintained almost 100% for 25 day. Therefore, PAM may become to a useful carrier for delivery of protein/peptide drugs, if it will be supported by biocompatibility and biodegradability results.

Sensor Network Routing using Data Aggregation (데이터 병합을 이용한 센서 네트워크 라우팅)

  • In this paper we investigate the benefits of a data aggregation to prolong the lifetime of wireless sensor networks. To reduce the overload of messages from source node to sink node, data aggregation technique is generally used at intermediate node in path. The DD-G(Directed Diffusion-Greedy) can diminish the consumption of node energy by establishing energy effective single path from source to destination. In this case, the nodes near sink node have some problems, i) overly concentration of energy consumption, ii) increase of message delay time. To solve these problems, we propose a new data aggregation method which consider distribution of network overload, especially at the nodes close to sink node. The result shows that it can save energy and network delay time.

Characterization of a Cell Aggregation Factor from Aspergillus sp.LAM 94-142 (Aspergillus sp. LAM 94-142가 생산하는 세포응집물질의 특성)

  • A cell aggregation factor produced by Aspergillus sp. LAM 94-142 was purified and partially characterized. The factor was purified about 15 folds from culture broth by IRA 420 and IRC 120 treatment, 1% NaCl added acetone precipitation, and Sepharose 4B column chromatography with overall yield of 48%. It was heteropolysaccharide consisted of mannose, arabinose, and glucose with a molar ratio, 31:17:2, and its molecular weight was estimated to be about 900,000 daltons by Sepharodse 4B gel filtration method. The optimum pH and temperature was 8 and 40$\circ$C, respectively. The factor was stable in pH range of 3-9 and at 100$\circ$C for 90 min. The cell aggregation activity of the factor was inhibited by the addition of Hg$^{2+}$, Fe$^{2+}$, Cu$^{2+}$, and some polypeptides such as milk casein or hemoglobin. The factor aggregated Bacillus subtilis, B. macerans, B. turingiensis, E. coli, Peudomonas aeruginosa, P. fluorescens, P. malophilia, and weakly aggregated Staphylococcus sp., Sarcina lutea, P. putida and Cryptococcus neoformnans, but it didn't aggregate various strains of Candida sp. and Saccharomyces sp.

Numerical Study of Aggregation and Breakage of Particles in Taylor Reactor (테일러 반응기 내의 입자응집과 분해에 관한 수치 연구)

  • Using the computational fluid dynamics (CFD) technique, we simulated the fluid flow in a Taylor reactor considering the aggregation and breakage of particles. We calculated the population balance equation (PBE) to determine the particle-size distribution by implementing the quadrature method-of-moment (QMOM). It was used that six moments for an initial moments, the sum of Brownian kernel and turbulent kernel for aggregation kernel, and power-law kernel for breakage kernel. We predicted the final mean particle size when the particle had various initial volume fraction values. The result showed that the mean particle size and initial growth rate increased as the initial volume fraction of the particle increased.

Structure analysis of polymeric micelles using SANS (중성자 소각 산란(Small Angle Neutron Scattering)을 이용한 모델 고분자 미셀의 구조 분석)

  • Structure analysis of model polymeric micelles, both for non-asosciative and associative cases is done by small angle neutron scattering method. Aggregation number of the hydrophobic cores and the lyotropic ordering transitions of aqueous solutions of poly(ethylene glycol)(PEG) (6 K or 10 K g/mol) end-capped with perfluoroalkyl groups $(-(CH_2)_2C_OnF_{2n+1}$ (n =6,8, or 10) are characterized. Aggregation number is mainly determined by the hydrophobe end group only, and is insensitive to polymer concentration or temperature. Also, there is no difference between non-associative micelles and associative micelles in terms of aggregation number. The model systems order into a BCC structure with increasing concentration.
