Actinoplanes teichomyceticus ATCC 31121 was mutated with UV to obtain a superior mutant strain with increasing level of teicoplanin production. In this investigation lethal curve was obtained and the optimal condition to induce mutagenesis was determined and to isolate the desirable mutant strain. It was also confirmed that teicoplanin activities by agar diffusion method to be compared to the mother strain. One mutant strain, T991014-1 that had the highest teicoplanin productivity was finally selected for further investigation including fermentation pattern. The mutant was characterized by the various tests such as amylase activity, protease activity, antibiotic resistance, autotoxicity, and productivity.
The antimicrobial substance produced by Lactobacillus bulgaricus was inactivated by pretense, which confirmed it as a bacteriocin and referred to 'bulgaricin HJ'. The bulgaricin HJ showed the inhibitory activity against mastitis pathogens, gram-positive and gram-negative bacteria. The optimal conditions for the production of bulgaricin HJ were at the temperature of $30^{\circ}C$ and 10 h after cultivation of L. bulgaricus. Staph. and Strep. agalactiae, common bovine mastitis pathogens, were treated with bulgaricin HJ by the agar well diffusion method and showed antimirobial activities to the bovine mastitis pathogens. The activity of the bulgaricin HJ was maintained at pH 6-7 and $100^{\circ}C$ for 60 min against the mastitis pathogens. The bulgaricin HJ was determined as class IV bacteriocin by various enzyme treatments. Colony forming units analysis with indicator strains by the treatments of bulgaricin HJ indicates that the mode of bacteriocin action was bactericidal rather than bacteriostatic.
A retrospective study of 132 cases of natural Escherichia coli infection in preweaning piglets with diarrhea submitted to the Department of Veterinary Pathology in Seoul National University from 1995 to 1996 was performed to determine the biochemical characteristics and antibiotics susceptibility. Most field isolates were lysine decarboxylase-positive (99.2%), phenylalanine decarboxylase-negative (100%), and fermented sorbitol (91.7%). Antibiotic susceptibilities will be determined by agar diffusion method. A large percentage of isolates were resistant to many antibiotics in common usage. Most isolates were susceptible to colistin (98.8%), gentamycin (64.3%), amikacin (100%), and ceftiour sodium (64.2%), whereas most isolates were notably resistant to ampicillin (86.9%), neomycin (66.7%), streptomycin (84.5%), tetracycline (98.8%), penicillin (98.8%), and amoxacillin (58.3%).
This study was aimed to screen edible herbs which control the composition of intestinal microflora. With in vitro experiments, we screened the water or ethanol extracts of about 60 edible herbs and wild plants in terms of the inhibition activity on the growth of the harmful Clostridium perfringens and growth promoting activity for the beneficial Bifidobacteria. The water extracts of mugwort and small water dropwort inhibited the growth of Cl.perfringens both in agar diffusion method and broth culture. On the other hand, the water extracts of petasites, mugwort, yellow day-lily and bitter cress have shown the promotion effect on the growth of Bifidobacterium longum. In the culture test using human feces as starter, the extracts of the above selected herbs increased the population of Bifidobacteria and Lactobacillus while they reduced the numbers of Cl.perfringens and E.coli.
Yam was stepwise extracted with hexane, chloroform, ethyl acetate, butanol, and water. Antibacterial activity of each extract was investigated. 1. Methanol extract of Yam was examed against to antibacterial effects on S. mutans KCTC 5316, P. gingivalis KCTC 5352, S. aureus KCTC 1927 by means of agar diffusion method. 2. The methanol extract of Yam was showed very powerfull antibacterial activity on S. mutans, but not showed on P. gingivalis, and S. aureus. 3. The MIC of Yam hexane fraction was 0.25 mg/ml for S. mutans KCTC 5316. 4. The Yam hexane fraction strong inhibited the growth of S. mutans, in the culture medium at concentration of 0.25 mg/ml.
Escherichiae-like organisms were isolated from rectal specimens of 56 children who were either in preschool age or in elementary school. The isolated strains were subjected to tests to screen enteropathogens producing heat-labile enterotoxin and susceptibility test to various antibiotics by disc diffusion method on agar plates. Production of heat-labile enterotoxin by the strains was assyed in the sensitive and reproducible cultured adrenal tumor cell system. The assay was sterodogenesis of the cell in the presence of heat-labile enterotoxin. Among 56 strains, gave positive reaction in the test of toxin production. This meant that about 10% of the children population objected to the study harbored the toxigenic strain of enteropathogenes. Some of these toxigenic strains were resistant to the antibiotics employed in the test. This study suggested that considerable population in Korea may harbor entertoxigenic E. coli as a part of intestinal normal flora. The toxigenic strains which are resistant to antibiotics may bring issue of public health in the future.
Ethyl acetate extracts of traditional medicinal herbs were screened for their antifungal activity against Malassezia furfur through the liquid growth inhibition method with 96 well plate and the paper disk agar diffusion assay. Among the 91 kinds of herbal solvent extracts, borneolum (borneo-camphor, No. 56) was finally selected as the best antifungal effective medicinal herb. The MIC value of No. 56 against M. furfur was 0.3 mg/ml ($8.2{\times}10^6$ CFU/ml) and it was three times more efficient than midazole which is the known antifungal agent in clinical medicines.
Lactobacillus crispatus ATCC 33820 and L. gasseri ATCC 33323 were grown in MRS broth (pH 6.5) at 37$^{\circ}C$ for 24 h and the antibacterial activities of cell free culture supernatants were determined by the agar well diffusion method. The culture supernatants were inhibitory to Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Bacillus subtilis, Staphylococcus aureus, Enterococcus faecalis, Pediococcus acidilacticii, and Lactobacillus helveticus. The supernatants did not show any lysozyme activity. Addition of catalase did not affect the antibacterial activities of the supernatants. The antibacterial substances were heat stable (100$^{\circ}C$ for 60 min) and sensitive to proteases.
Dental caries is the destruction of the enamel of teeth by Streptoccus mutans. S. mutans has been isolated from human dental plaque and is associated with the initial development of enamel lesions. We have studied the antibacterial action of the fruit of Callistemon citrin us against a cariogenic bacterium, S. mutans. From the fruit of C. citrinus, piceatannol (3,3',4',5-tetrahydroxystilbene) was isolated by repeated column chromatography with $SiO_2$ and Sephadex LH-20. Its structure was elucidated by instrumental analysis using 1D-NMR, 2D-NMR and EI-MS. This compound was isolated from the fruit of C. citrin us for the first time. The anticarcinogenic activity of this compound was determined by using agar well-diffusion method and minimal inhibition concentration (MIC).
Fungal perylenequinones have photodynamic activity and are promising photosensitizers for photodynamic therapy (PDT). Here, we investigated the bactericidal and antitumor activities of phleichrome from the fungal perylenequinone family in vitro. Photodynamic bactericidal activity of phleichrome was analyzed by agar-well diffusion method under dark and illuminated conditions. The photodynamic antitumor activity of phleichrome was analyzed in MCF-7, HeLa, SW480, and HepG2 human cancer cell lines using in vitro cytotoxicity assays. Photodynamic bactericidal activities against Gram-negative and Gram-positive bacteria were species-specific. Antitumor activity against all tumor cell lines increased under the illuminated condition. Depending on the results of the analyses, Phleichrome has potential for further drug development related to its antibacterial and antitumor activities.
