A Study of Organic Acid Formation and Dialysis Culture in E. coli Fermentation (대장균 배양에서의 유기산 생성과 투석배양에 관한 연구)

  • In order to elucidate the effect of acetate ion on the growth of Escherichia coli, flask and fermentor cultures were performed using M9 and LB media. Acetic acid was secreted at a higher rate under the conditions of high glucose concentration as well as of richer medium, i. e., LB broth. The pH in flask culture could not be controlled as i fermentor and pH decreased with the formation of acetic acid. The inhibition effect of acetic acid was pronounced at a lower pH, and the effective inhibitory concentrations of acetic acid were 2.0g/l for LB flask culture, 4.0g/l for M9 flask culture, and 8.0g/l for M9 fermentor culture. Dialysis flask culture was designed to slowly provide E coli cells with glucose. Solid LB agar was layered under LB liquid medium with the variation of agar concentration and solid volume, The increase in the solid portion in the total volume(agar+liquid) resulted in the increase of the final cell concentration. This can be ascribed to the fact that the larger solid phase behaves like a larger reservoir for glucose and controls the growth of E. coli with a controlled rate.

Anti-microbial Activity of Platycodon Grandiflorum Extracts Against Oral Microbes (도라지 추출물의 구강미생물에 대한 항균효과)

  • Objectives: The objective of this study was to assess the antimicrobial effect of Platycodon grandiflorum extracts against oral microorganisms. Methods: The anti-microbial activity and minimal inhibitory concentration were measured the agar dilution method. Results: Platycodon grandiflorum extracts grew in the free agar plates all of the oral microorganisms. In the bark-free Platycodon grandiflorum extracts all the oral microorganisms grew in the free agar plates. Growth was inhibited at a concentration of 0.5 mg/ml. Oral microorganisms showed an absence of growth at a concentration of 1 mg/ml. Conclusions: It was confirmed that the extracts of Platycodon grandiflorum having a higher saponin content than the bark - free Platycodon grandiflorum extract showed excellent antimicrobial effect.

Fluid Properties and Quality of Agar Solution from Cheju Seaweed, Gellidium amansii (제주산 우뭇가사리로 부터 추출한 한천의 품질 및 용액 특성)

  • To bring out the functional properties of Agar-agar extracted with 0.01N $H_2SO_4$ from Gellidium amansii produced in Che-ju Udo and its fluid properties were investigated. The yield of agar from Gellidium amansii produced was $32.7\%$, the content ratio of agarose and agaropectin was 79 to 21, gelation ability was $0.19\%$ and jelly strength was $413.8\;dyne/cm^2$. Agar-agar solution showed the movement of non-Newtonian fluid and pseudoplastic property was emerged as its concentration go higher. The yield stresses in the range of $0.5\~5\%$ agar-agar solution were $0.09\~1.21\;dyne/cm^2\;at\;80^{\circ}C,\;0.12\~5.29\;dyne/cm^2\;at\;60^{\circ}C\;and\;4.84\~58.37\;dyne/cm^2\;at\;50^{\circ}C$.

Preparation Conditions of Agar Oligosaccharides with Organic Acids (한천 올리고당 제조를 위한 유기산 처리 조건)

  • The optimum organic acid and temperature conditions were investigated for the preparation of oligosaccharides from agar. The tested organic acids were acetate, citrate, lactate, malate, and succinate and the conditions for oligosaccharides preparation were $0.3\%,\;0.5%;and\;0.7\%$ organic acid concentrations at $80\~120^{\circ}C.$ The low concentration of organic acid below $0.3\%$ decreased the degrading ratio and the high concentration up $0.5\%$ could not changed the degrading ratio. Conditions below $100^{\circ}C$ was not good for degrading agar. But $100^{\circ}C\;or\;120^{\circ}C$ was optimal temperature conditions for agarooligosaccharides according to the organic acid type and concentration. The organic acid concentration was $0.5\%$ and organic acid was the citrate or malate. The treatment time considered optimum was 120$\~$180 min. The maximal degrading ratio giving optimum conditions such as $100^{\circ}C\;and\;120^{\circ}C\;was\;35.5\%\;and\;38.7\%,$ respectively. The agarooligosaccharides prepared by autoclaving at $100^{\circ}C\;and\;120^{\circ}C$ were 2$\~$7 species oligomer.

Characteristics of OCP of Reinforced Concrete Using Socket-type Electrodes during Periodic Salt Damage Test (주기적 염해 시험에 따른 소켓 타입 전극을 활용한 철근 콘크리트의 OCP 특성)

  • It is known that buried rebars inside concrete structures are protected from corrosion due to passive layer. It is very important to delay the timing of corrosion or evaluate a detection of corrosion initiation for the purpose of cost-beneficiary service life of a structure. In this study, corrosion monitoring was performed on concrete specimens considering 3 levels of cover depth(60 mm, 45 mm, and 30 mm), W/C(water to cement) ratio(40.0%, 50.0%, and 60.0%) and chloride concentration(0.0%, 3.5%, and 7.0%). OCP(Open Circuit Potential) was measured using agar-based socket type sensors. The OCP measurement showed the consistent behavior where the potential was reduced in wet conditions and it was partially recovered in dry conditions. In the case of 30 mm of cover depth for most W/C ratio cases, the lowest OCP value was measured and rapid OCP recovery was evaluated in increasing cover depth from 30 mm to 45 mm, since cover depth was an effective protection against chloride ion ingress. As the chloride concentration increased, the effect on the cover depth tended to be more dominant than the that of W/C ratio. After additional monitoring and physical evaluation of chloride concentration after specimen dismantling, the proposed system can be improved with increasing reliability of the corrosion monitoring.

In Vitro Propagation by Shoot-tip and Node-bud Culture of Rehmannia glutinosa (정단 및 마디조직 배양을 통한 지황의 기내 증식)

  • Multiple shoots obtained in MS medium suppler with 5.0 mg/L BA though shoot-tip culture. The frequency of vitrified shoot was lower on Bacto-agar medium than on Gelrite as gelling agent. Addition of activated charcoal at concentrations of 0.1~0.3% reduced vitrification and markedly increased shoot growth, and formation and growth of roots, but significantly reduced the number of shoots formed. The ratio of fresh weight to dry weight was decreased by increasing light intensity and agar concentration. Eight-tenths times of macroelement of MS medium was observed to be effective for shoot formation. Addition of IAA effectively promoted shoot formation in both shoot tip and node-bud explants. Supplement of 5.0 mg/L BA, 0.3 mg/L IAA to MS medium was most effective in shoot proliferation on shoot tip and node-bud explants.Multiple shoots obtained in MS medium suppler with 5.0 mg/L BA though shoot-tip culture. Comparison of the E-Test with Agar Dilution Susceptibility Test by Using Bacteroides fragilis (Bacteroides fragilis의 E-test와 한천 평판 희석법에 의한 항균제 감수성상의 비교)

  • The susceptibilities of 45 clinical isolates of bacteroides frogilis to cefaclor, ciproflxacin and imipenem were determined by new method, E-test (AB Bidisk, Solna, Sweden) and were compared with those from conventional agar dilution method by using brain heart infusion, Mueller-Hinton and Wilkins Chalgren agar plates. And the susceptibility of 60 clinical isolates of Bacteroides fragilis group (B. fragilis 45 strains, B. distasonis 6 strains, B. ovatus 5 strains, B. thetaiotaomicron 4 strains) to 5 quinolones (ciprofloxacin, enoxacin, norfloxacin, ofloxacin, pefloxacin) were determined by in vitro agar dilution method. Compared with agar dilution MICs for B. fragilis 45 strains, 90.3% of E-test MICs were within ${\pm}$1 dilution of the agar dilutions, and 98.4% were within 2 dilutions. And there were little effect of different medium bases to determine MICs except Mueller-Hinton agar. On Mueller-Hinton agar, B. fragilis showed have or no growth activity. In vitro susceptibility of B. fragjlis group to quinolones, most of the test strains showed resistant patterns to quinolones except ofloxacin and there was little difference of susceptibility patterns between species of B. fragilis group.

Effect of Manganese Sulfate Concentration in Media on Production Speed of Insecticidal Crystal by Bacillus thuringiensis (배지 중 Manganese sulfate 농도가 Bacillus thuringiensis의 곤충독소 생성 시간에 미치는 영향)

  • In this study, the effect of MnSO4 on the insecticidal crystal (IC) produced by Bacillus thuringiensis for a rapid detection medium was analyzed. The strains used included one B. thuringiensis reference (KCTC 1511) and nine wild-type strains. The IC in B. thuringiensis was detected following the method published by the Ministry of Food and Drug Safety in Korea. In the nutrient agar to which 0.005% MnSO4 was added, IC was observed on two of the three plates after 48 hours of incubation and on all three plates after 120 hours. In AK agar, IC was observed on one and two of the three plates after 48 and 96 hours of incubation, respectively. These results indicated that 0.005% MnSO4 nutrient agar is more appropriate than AK agar for production of IC in B. thuringiensis. The effect of various MnSO4 concentrations on IC production was studied after 24 hours of incubation. IC was produced on 1 of the 10 plates with 0.000% MnSO4 nutrient agar, 2 of the 10 plates with 0.001% MnSO4 nutrient agar, and 3 of the 10 plates with 0.002% MnSO4 nutrient agar. IC was not observed for the other nutrient agars containing 0.003%-0.009% MnSO4. These results indicated that nutrient agar with 0.002% MnSO4 led to the most rapid production of IC by B. thuringiensis after 24 hours of incubation. However, the conditions for IC production by B. thuringiensis depended on the incubation conditions and strain activity. Therefore, further studies are needed to verify the effects of 0.002% MnSO4 on the production of IC by various Bacillus thuringiensis strains.

Antibacterial and Antioxidant Activity of Chamaecyparis obtusa Extracts (편백나무 추출액의 항균 및 항산화 활성)

  • In this study, we investigated the biological antioxidant and antibacterial activity of Chamaecyparis obtusa (C. obtuse) extracts by measuring DPPH radical scavenging and ABTS radical scavenging, and SOD-like activities. The DPPH and ABTS radical scavenging activities were increased in a dose-dependent manner, with maximum activities of 78% and 62% at an extract concentration of $50{\mu}l/ml$. The C. obtusa extracts also showed high SOD-like activity, with a maximum activity of 92.85% at a concentration of $50{\mu}l/ml$. The antibacterial activities of C. obtusa extracts were measured against six types of bacteria known to cause food poisoning and disease. Antibacterial activity was investigated against three gram-positive and three gram-negative bacteria using the paper disc agar diffusion method. The C. obtusa extracts showed antibacterial activities against B. cereus, E. coli, L. monocytogenes, S. aureus, S. typhi and V. parahaemolyticus, among which the activity against B. cereus was greatest. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of C. obtusa extracts were $30-40{\mu}l/ml$ for the 6 strains that showed an antimicrobial response by the paper disc agar diffusion method. These results suggest that C. obtusa extracts could serve as potential antibacterial agents to inhibit the growth of pathogens responsible for food poisoning and disease.

  • Characteristics of ${\beta}$-agarase production of Agarivorans sp, JA-1 isolated from north-eastern sea of Jeju marine environment was studied. Optimal cell growth was definite that the medium containing agar is 0.2%. The decreasing pattern of viscosity and agar concentration was same and they reached almost zero after 15 hours. Fed-batch culture was studied to improve agarase productivity by Agarivorans sp. JA-1 in marine broth containing 2.0 g/L agar with intermittent addition of 0.8 g agar two times.
