Variation in the microbial biomass and community structure found in sediment of heavily polluted bays and the adjacent unpolluted areas were examined using phospholipid fatty acid analysis. Total microbial biomass and microbial community structure were responding to environmental determinants, sediment grain size, depth of sediment, and pollution due to petroleum hydrocarbons. The marker fatty acids of microeukaryotes and prokaryotes - aerobic, anaerobic, and sulfate-reducing bacteria - were detected in sediments of the areas studied. Analysis of the fatty acid profiles revealed wide variations in the community structure in sediments, depending on the extent of pollution, sediment depth, and sediment grain size. The abundance of specific bacterial fatty acids points to the dominance of prokaryotic organisms, whose composition differed among the stations. Fatty acid distributions in sediments suggest the high contribution of aerobic bacteria. Sediments of polluted sites were significantly enriched with anaerobic bacteria in comparison with clean areas. The contribution of this bacterial group increased with the depth of sediments. Anaerobic bacteria were predominantly present in muddy sediments, as evidenced from the fatty acid profiles. Relatively high concentrations of marker fatty acids of sulfate-reducing bacteria were associated with organic pollution in this site. Specific fatty acids of microeukaryotes were more abundant in surface sediments than in deeper sediment layers. Among the microeukaryotes, diatoms were an important component. Significant amounts of bacterial biomass, the predominance of bacterial biomarker fatty acids with abundance of anaerobic and sulfate-reducing bacteria are indicative of a prokaryotic consortium responsive to organic pollution.
본 연구는 전통간장의 발효미생물을 규명하기 위하여 대두원료, 증자과정, 메주 배양과정 및 간장덧의 숙성과정 중의 호기성 세균수, 젖산균수, 호모균수의 정량적 변화와 성분에 대하여 조사하였다. 대두에 있던 미생물은 삶는 과정 중에 젖산균은 사멸하였으며 호기성 세균은 $10^2\;CFU/g$ 수준으로 감소하였다. 메주 배양 동안 메주 내외부의 호기성세균은 $10^9\;CFU/g$ 이상 생육하였으며, 젖산균은 외부보다 내부에서 더 많은 집락을, 효모류는 내부보다는 외부에서 월등히 많은 수를 나타내었다. 메주 배양기간 별로 제조한 간장의 미생물수는 통기구와 정치구 간에 큰 차이가 없었으나 총질소의 함량과 색도는 메주 배양기간이 길수록 더 높은 함량을 나타내었다.
We gamma-irradiated to natural casings from lamb and pork to improve sanitary quality and microbiological shelf stability of sausage stuffed in animal intestine-induced natural casing from lamb and pork. The microbial changes of irradiated casings and the microbial growth of the sausages with gamma-irradiated natural casings were observed. Population of total aerobic bacteria was decreased by gamma irradiation, dependent upon the irradiation dose. The number of microflora in lamb and pork casings was reduced by about 5.5 log cycle at 5 kGy. Population of total aerobic bacteria of sausages stuffed in non-irradiated lamb and pork casing was about 3.6 log CFU/g after just manufacture and was about 7 log after 10 day storage. However, population of total aerobic bacteria of sausages with 3 kGy-irradiated and 5 kGy-irradiated casings was 2.48 and 0.32 log in lamb, and 2.78 and 0.60 log in pork, respectively. Gamma irradiation to animal intestine- induced natural casings was effective and thus, can be used for enhancing shelf life of final products due to reduction of the number of contaminated microflora.
We investigated the contamination of animal hospital floor, beauty table, computer keyboard, exam table, operation table and forcep handle by isolations of aerobic bacteria in small animal hospitals in Gwangju. The total number of aerobic bacteria was 52 isolates and Staphylococcus spp. (38 isolates) were the predominant isolates (69.71 %) of them. The prevalent contaminated areas were floor (17 isolates), beauty table (13 isolates) and computer keyboard (9 isolates). The detection of methicillin-resistant (mecA) gene, determined by PCR, showed that 3 of the 17 coagulase-negative Staphylococcus spp. (CNS) isolates possessed the mecA gene. For evaluating the antibiotic susceptibility patterns of the isolates, disk diffusion method was used. The majority of isolates showed high susceptibility to amoxicillin (92.1 %), ceftiofur (84.2%) and polymixin B (73.7%). Also they showed the high resistant to ampicilline (66.7%), penicillin (65%) and kanamycin (56.5%). These results suggest extensive contamination of aerobic bacteria in animal hospital environment.
This study was conducted to evaluate the effects of physical characteristics. Twelve specific odorous compounds and various sources of bacteria were tested via treatment of food waste using an ultra-thermophilic aerobic composting process. Food waste was mixed with seed material and operated for 47 days. During composting, the temperature was maintained at $80-90^{\circ}C$. The variations in $O_2$, $CO_2$ and $NH_3$ production suggested typical microorganism-driven organic decomposition patterns. After composting, the concentrations of 12 specific odorous compounds other than ammonia did not exceed the allowable exhaust limits for odor. After composting, thermophiles represented 50% of all bacteria. After composting, the percentage of thermophile bacterial increased by 15%. Therefore, both stable composting operation and economic benefit can be expected when an ultra-thermophilic composting process is applied to food waste.
This study was conducted to evaluate the quality characteristics of domestic as well as imported fermented skate. Three types of fermented skate products were analyzed for proximate composition, pH, VBN, ammonia-N, free amino acids, and fatty acids. The results indicated that the domestic fermented skate contained large amounts of TMAO. Also, the domestic and imported fermented skates each contained approximately 7.1 log CFU/g and $5.8{\sim}6.5$ log CFU/g of aerobic bacteria, respectively, and 585.9 mg and $384.1{\sim}398.5$ mg of total free amino acids, respectively; all samples contained high levels of taurine, anserine, lysine, alanine, glycine, proline, and ${\beta}-alanine$. For fatty acid composition, the domestic fermented skate contained 11 different types of saturated fatty acid and 16 types of unsaturated fatty acid, whereas the imported skate contained 8 types of saturated fatty acid and $10{\sim}15$ types of unsaturated fatty acid. Overall, the results suggest that domestic fermented skate is a better source of amino acids and essential fatty acids and contains more aerobic bacteria than imported fermented skate.
The purpose of this paper is to study on the morphological and physiological nature generally observed for the identifications, and the four strains of thermal resistanting aerobic bacteria isolated from swelled cans and the different soils collected from the wide area in Korea. The results obtained in the light of the Bergey's Manual for the identification of the bacteria, have been shown that the four strains of bacteria are pertained to Bacillus subtilis. The optimum temperature, pH and the thermal resistance (2.5 min. at $121^{\circ}C$) of the bacteria have been measured.
H$_2$-producing bacteria were isolated from anaerobic granular sludge. Out of 72 colonies (36 grown under aerobic conditions and 36 under anaerobic conditions) arbitrarily chosen from the agar plate cultures of a Suspended sludge, 34 colonies (15 under aerobic conditions and 19 under anaerobic conditions) produced H2 under anaerobic conditions. Based on various biochemical tests and microscopic observations, they were classified into 13 groups and tentatively identified as follows: From aerobic isolates, Aeromonar spp. (7 strains), Pseudomonas spp. (3 strains), and Vibrio spp. (5 strains); from anaerobic isolates, Actinomyces spp. (11 Strains), Clostridium 5pp. (7 strains). and Porphyromonas sp. When glucose was used as the carbon substrate, all isolates showed a similar cell density and a H$_2$ production yield in the batch cultivations after 12 h (2.24-2.74 OD at 600 nm and 1.02-1.22 mol H$_2$/mol glucose, respectively). The major fermentation by-products were ethanol and acetate for the aerobic isolates, and ethanol, acetate and propionate for the anaerobic isolates. This study demonstrated that several H$_2$ producers in an anaerobic granular sludge exist En large proportions and their performance in terms of H$_2$ production is quite similar.
CW-4Y was identified as Stenotrophomonas sp. by morphological and physiological characteristics, and phylogenetic analysis of its 16S rDNA gene sequence. Nitrogen removal by CW-4Y was analyzed in relation to the ammonium concentration, presence of organic carbon, carbon source, and carbon-to-nitrogen ratio (C/N). Stenotrophomonas CW-4Y has heterotrophic nitrification and aerobic denitrification abilities. Stenotrophomonas CW-4Y utilized only glucose as carbon sources, and heterotrophic nitrification and aerobic denitrification were observed regardless of the type of nitrogen source. The maximum ammonium removal rate of CW-4Y was 80 $mg-N{\cdot}L^{-1}{\cdot}d^{-1}$ and its denitrification rate of 192 $mg-N{\cdot}L^{-1}{\cdot}d^{-1}$ at $NO_3{^-}-N$ (about 280 ppm) in shake culture experiments at a C/N ratio of about 15 was about 30 times higher than those of other bacteria with the same ability.
More than 8 millions of chemical have been used for human activities and lots of chemicals can not be degraded by microbial activities in this world. To show the biodegradability of a chemical, biodegradability index (B.I.) is suggested using aerobic biodegradability by $BOD_5$/COD, anaerobic biodegradability by methane potential (M.P.) and toxicity by the luminiscent bacteria. In this study, PVA (polyvinyl alcohol), HEC (hydroxy ethyl cellulose), 2,4,6-TCP (tri-chloro phenol) and 2,4-DCP (di-chloro phenol) are used for test chemicals. Though they show little toxicity, PAV and HEC have low B.I. because they are polymers having high molecular weight. That means that there are no bacteria that has enzyme to degrade polymer molecules. Also, anaerobic treatment is suggested better than aerobic treatment from B.I. 2,4,6-TCP and 2,4-DCP show high toxicity and have low B.I. Their low biodegradabilities seem to be originated from their toxicities. If B.I. is used in wastewater treatment, better treatment process can be suggested and finally it can lead our society to make more environment-friendly chemicals.
