Comparison Analysis of Biological Activities of Three Sedum species

  • Sedum kamchaticum, Sedum middendorffianum and Sedum takesimense belong to the sebum species. Especially, Sedum takesimense is a Korean native species growing only on Ullenung-do. Few studies for the biological activities of these plants have been reported. In this study, we examined biological activity and the contents of functional components of the aerial part and the root part of Sedum kamchaticum, Sedum middendorffianum and Sedum takesimense. As the results, the roots of Sedum takesimense showed the highest total phenolic contents (TPC : $13040{\pm}50.0mg\;GAE{\cdot}100g^{-1}$), and the aerial part of Sedum takesimens showed the highest total flavonoid contents (TFC : $2722.2{\pm}107.1mg\;CAE{\cdot}100g^{-1}$). Meanwhile, Sedum middendorffianum exhibited the highest anti-oxidant activity (DPPH $RC_{50}$ value of aerial part : $50.69{\pm}0.75{\mu}g{\cdot}mL^{-1}$, DPPH $RC_{50}$ value of root part : $27.27{\pm}0.55{\mu}g{\cdot}mL^{-1}$; ABTS $RC_{50}$ value of aerial part : $11.32{\pm}0.38{\mu}g{\cdot}mL^{-1}$, ABTS $RC_{50}$ value of roots part $5.54{\pm}0.02{\mu}g{\cdot}mL^{-1}$). The root part of Sedum middendorffianum showed the potent alpha glucosidase inhibitory activity (AGI $IC_{50}$ : $60.69{\pm}0.72{\mu}g{\cdot}mL^{-1}$). In order to elucidate active principle of Sedum middendorffianum that was shown the most potent antioxidant and anti-diabetic activities, the methanol extract of Sedum middendorffianum was fractionated with various solvents according to the polarity successively. As a result, EtOAc fraction of Sedum middendorffianum showed the highest contents of functional components (TPC : $16245.8{\pm}1025.7mg\;GAE{\cdot}100g^{-1}$, TFC : $4850.4{\pm}182.9mg\;CAE{\cdot}100g^{-1}$). And it also showed excellent anti-oxidant activities (DPPH $RC_{50}$ : $14.0{\pm}0.6{\mu}g{\cdot}mL^{-1}$, ABTS $RC_{50}$ : $3.4{\pm}0.1{\mu}g{\cdot}mL^{-1}$) and anti-diabetic activity (AGI $IC_{50}$ : $79.0{\pm}2.2{\mu}g{\cdot}mL^{-1}$). Above results suggest that Sedum middendorffianum can be developed to health functional material as a plant resource with potent antioxidant activity.

Genotoxicity Study of Polysaccharide Fraction from Astragalus membranaceus's Aerial Parts

  • Radix Astragali, the root of Astragalus (A.) membranaceus, has been applied in a variety of diseases for a long time in Asian countries such as Korea and China. In addition, the aerial parts such as leaves and stems of A. membranaceus have received a great deal of attention. Recently, the polysaccharide fraction showing a potent immunomoduating activity was isolated from the aerial parts of A. membranaceus. Thus, the aerial parts of A. membranaceus would be worthy enough for a food material and a dietary supplement. However, they should be safe even though valuable. In our previous study, it was estimated that NOAEL for female rats are 5000 mg/kg/day of the crude polysaccharide fraction from A. membranaceus-aboveground parts. As a series of safety evaluation, genotoxicity test for the crude polysaccharide fraction was carried out in this study. In conclusion, the three genotoxicity assays provided strong overall support that the crude polysaccharide fraction lacks mutagenic and/or clastogenic potential under the GLP-based test conditions. This indicates the aerial parts of A. membranaceus would be safe enough for a food material and a dietary supplement.

On-site Demonstration of Topographic Surveying Techniques at Open-pit Mines using a Fixed-wing Unmanned Aerial Vehicle (Drone) (고정익 무인항공기(드론)를 이용한 노천광산 지형측량 기술의 현장실증)

  • This study performed an on-site demonstration of the topographic surveying technique at a large-scale open-pit limestone mine in Korea using a fixed-wing unmanned aerial vehicle (UAV, Drone, SenseFly eBee). 288 sheets of aerial photos were taken by an automatic flight for 30 minutes under conditions of 300 m altitude and 12 m/s speed. Except for 37 aerial photos in which no keypoint was detected, 251 aerial photos were utilized for data processing including correction and matching, then an orthomosaic image and digital surface model with 7 cm grid spacing could be generated. A comparison of the X, Y, Z-coordinates of 4 ground control points measured by differential global positioning system and those determined by fixed-wing UAV photogrammetry revealed that the root mean squared errors were around 15 cm. Because the fixed-wing UAV has relatively longer flight time and larger coverage area than rotary-wing UAVs, it can be effectively utilized in large-scale open-pit mines as a topographic surveying tool.

Antibacterial Activity of Some Medicinal Plants against Propionibacterium acnes (여드름 원인균 Propionibacterium acnes에 대한 생약 추출물의 항균활성 측정)

  • Acne, one of the most common skin disease, is a chronic inflammatory disease and is caused by uncertain and multiple factors. Propionibacterium acnes belongs to the human cutaneous normal flora and is a major etiologic agent of acne vulgaris. In this study, we evaluated the antibacterial effect of 19 medicinal plants. Antibacterial activity of extracts prepared from the 19 medicinal plants was investigated against bacteria related to acnes, Propionibacterium acnes. Among them, some medicinal plants inhibited the growth of the P. acnes. Minimum inhibitory concentration (MIC) of Polygonum aviculare (Herb) was 0.2 mg/ml against P. acnes. MIC of Dianthus chinensis (Aerial Part), Forsythia viridissima (Fruit), Lygodium japonicum (Spore) and Sophora flavescens (Root) were 0.4 mg/ml. Based on these results, Polygonum aviculare (Herb), Dianthus chinensis (Aerial Part), Forsythia viridissima (Fruit), Lygodium japonicum (Spore) and Sophora flavescens (Root) may be considered as a candidate for a good medicine for acne.

Quality Evaluation of Orthoimage and DSM Based on Fixed-Wing UAV Corresponding to Overlap and GCPs (중복도와 지상기준점에 따른 고정익 UAV 기반 정사영상 및 DSM의 품질 평가)

  • UAV(unmanned aerial vehicle) can quickly produce orthoimage with high-spatial resolution and DSM(digital surface model) at low cost. However, vertical and horizontal positioning accuracy of orthoimage and DSM, which are obtained by UAV, are influenced by image processing techniques, quality of aerial photo, the number and position of GCPs(ground control points) and overlap in flight plan. In this study, effects of overlap and the number of GCPs are analyzed in orthoimage and DSM. Positioning accuracy are estimated based on RMSE(root mean square error) by using dataset of nine pairs. In the experiments, Overlaps and the number of GCPs have influence on horizontal and vertical accuracy of orthoimage and DSM.

Evaluation of Gastric Motility Enhancement of the Extracts and Isolates from Traditional Medicinal Herbs (한약재 추출물 및 유래 화합물들의 위장관 운동 촉진 효능 연구)

  • To identify potential gastrointestinal prokinetic agents, water and 70% ethanol extracts and isolated compounds from 41 different traditional medicinal herbs were evaluated for the stimulation of gastrointestinal (GI) motility in vivo. Of the 41 water and 70% ethanol extracts, 12 extracts were found to enhance GI motility activity in mice by more than 10%. The 12 extracts are as follows: Atractylodes japonica (root), Crataegus pinnatifida (flower), Aucklandia lappa (root), Inula helenium (root), Cynanchum wilfordii (root), Chinese Liriope platyphylla (root), Codonopsis pilosula (root), Glehnia littoralis (root), Pinellia ternate (tuber), Agastache rugosa (aerial part), Angelica decursiva (whole plant), and Peucedanum praeruptorum (whole plant). In particular, the extracts from Atractylodes japonica (root), Cynanchum wilfordii (root) and Angelica decursiva (whole plant) have demonstrated the highest GI motility activity. In addition, 26 isolated compounds from the medicinal herbs were tested, and 8 isolated compounds were found to be active. They are ${\alpha}$-ionone, ${\beta}$-ionone, trans-caryophyllene, cedrol, methyl-3,5-di-O-E-caffeoyl-quinate, lobetyolin, oleoyllinoleoylolein and cis-jasmone. ${\beta}$-ionone from Aucklandia lappa (root) showed the most potent GI motility activity. The active traditional medicinal herbs and isolated compounds might be therapeutically advantageous in the treatment of GI motility disorders.

Assessment of Parallel Computing Performance of Agisoft Metashape for Orthomosaic Generation (정사모자이크 제작을 위한 Agisoft Metashape의 병렬처리 성능 평가)

  • In the present study, we assessed the parallel computing performance of Agisoft Metashape for orthomosaic generation, which can implement aerial triangulation, generate a three-dimensional point cloud, and make an orthomosaic based on SfM (Structure from Motion) technology. Due to the nature of SfM, most of the time is spent on Align photos, which runs as a relative orientation, and Build dense cloud, which generates a three-dimensional point cloud. Metashape can parallelize the two processes by using multi-cores of CPU (Central Processing Unit) and GPU (Graphics Processing Unit). An orthomosaic was created from large UAV (Unmanned Aerial Vehicle) images by six conditions combined by three parallel methods (CPU only, GPU only, and CPU + GPU) and two operating systems (Windows and Linux). To assess the consistency of the results of the conditions, RMSE (Root Mean Square Error) of aerial triangulation was measured using ground control points which were automatically detected on the images without human intervention. The results of orthomosaic generation from 521 UAV images of 42.2 million pixels showed that the combination of CPU and GPU showed the best performance using the present system, and Linux showed better performance than Windows in all conditions. However, the RMSE values of aerial triangulation revealed a slight difference within an error range among the combinations. Therefore, Metashape seems to leave things to be desired so that the consistency is obtained regardless of parallel methods and operating systems.

Accuracy Analysis of Medium Format CCD Camera RCD105 (중형카메라 RCD105 정확도 분석)

  • Lately, airborne digital camera and airborne laser scanner in field of airborne surveying are used to build geography information such as digital ortho photo map and DEM(Digital Elevation Model). In this study, 3D position accuracy is compared medium format CCD camera RCD105 with airborne digital camera DMC. For this, test area was decided for aerial photograph. And using 1/1,000 scale digital map, ground control points were selected for aerial triangulation and check points were selected for horizontal/vertical accuracy analysis using softcopy stereoplotter. Accuracy of RCD105 and DMC was estimated by result of aerial triangulation and result of check points measurement of using softcopy stereoplotter. In result of aerial triangulation, RMSE(Root Mean Square Error) X, Y, Z of RCD105 is 2.1, 2.2, 1.3 times larger than DMC. In result of check point measurement using softcopy stereoplotter, horizontal/ vertical RMSE of RCD105 is 2.5, 4.3 times larger than DMC. Even though accuracy of RCD105 is lower than DMC, it is maybe possible to make digital map and ortho photo using RCD105.

Analysis of Chemical Components for Aerial and Underground Parts of Wild Ginseng and Evaluation of Skin Anti-aging Efficacy (야생 산삼 지상부 및 지하부의 화학성분 분석과 피부 항노화 효능 평가)

  • Objectives : This study was intended to reveal the chemical profiles of aerial(leaf, stem) and underground(rhizome, radix) parts of wild ginseng, and to investigate their anti-aging effects on human skin cells. Methods : Wild ginseng, estimated for over 20 years, was divided into the aerial and underground parts. Total phenolic contents of each extracts were measured using a Folin-ciocalteu method. The contents of 18 amino acids, 8 minerals and 27 ginsenosides were determined by GC-FID, ICP-MS and LC-MS, respectively. The anti-aging effects, including the radical scavenging activity, the activation of mitochondrial function on human fibroblasts, and the proliferation activity on human keratinocyte progenitor cells, for the whole plant and underground part of wild ginseng were evaluated. Results : The total phenolic acids, amino acids, and minerals in the aerial part were more than twice as high as in the underground part. Compared to the cultivated ginseng root, there were various types of ginsenosides in both parts of wild ginseng, and the total amount was more than twice as high. In particular, the aerial part significantly contained ginsenoside F1, F2, C-Mc1, and C-O, and the distinctive patterns that distinguish each parts of wild ginseng from the cultivated ginseng root were derived. The whole plant and underground part of wild ginseng exhibited significant antioxidant effect(14.3-45.6%), activation of mitochondrial membrane potential(105.5-120.1%), and cell proliferation(112.1-125.4%). Conclusions : The entire plant and underground part of wild ginseng are high value-added plants and have beneficial effects on skin anti-aging properties through its abundant metabolites.

Automatic Extraction of Tree Information in Forest Areas Using Local Maxima Based on Aerial LiDAR (항공 LiDAR 기반 Local Maxima를 이용한 산림지역 수목정보 추출 자동화)

  • Currently, the National Forest Inventory (NFI) collects tree information by human, so the range and time of the survey are limited. Research is actively being conducted to extract tree information from a large area using aerial Light Detection And Ranging (LiDAR) and aerial photographs, but it does not reflect the characteristics of forest areas in Korea because it is conducted in areas with wide tree spacing or evenly spaced trees. Therefore, this study proposed a methodology for generating Digital Surface Model (DSM), Digital Elevation Model (DEM), and Canopy Height Model (CHM) images using aerial LiDAR, extracting the tree height through the local Maxima, and calculating the Diameter at Breath Height (DBH) through the DBH-tree height formula. The detection accuracy of trees extracted through the proposed methodology was 88.46%, 86.14%, and 84.31%, respectively, and the Root Mean Squared Error (RMSE) of DBH calculated based on the tree height formula was around 5cm, confirming the possibility of using the proposed methodology. It is believed that if standardized research on various types of forests is conducted in the future, the scope of automation application of the manual national forest resource survey can be expanded.
