Ethanol-soluble amino acids and phenolics in healthy and blast-infected leaves of the susceptible rice cultivar Nakdong and adult-plant-resistant cultivar Dobong were quantitatively analyzed, At the 3 days after inoculation, the levels of soluble amino acids and phenolics in the infected fifth and eighth leaves of the two cultivars ere similar to those of comparable healthy controls. As blast lesions appeared on the leaves at the 5 days after inoculation, the soluble amino acids and phenolics began to increase. At the 7 days after inoculation, the levels of amino acids and phenolics were about 1.5-3 times more than those in healthy controls at the five-and eight leaf stages. The adult-plant-resistant cultivar Dobong showed higher amounts of soluble amino acids and phenolics in both healthy and infected fifth and eighth leaves than did the susceptible cultivar Nakdong, although Dobong was less infected by Pyricularia oryzae than Nakdong, The pronounced increases in amino acids and phenolics in rice leaves of the cultivar Dobong during the blast infection may play an important role in the expression of adult-plant resistance to blast.
Thirty two rice cultivars which have been cultivated or used as breeding materials in Korea were tested for screening rice cultivars resistant to leaf blast at adult-plant stages in the blast nursery hill plots. When compared on the basis of disease severities in individual leaves at different growth stages of rice plants under natural field infection, the 16 indica-japonica hybrids tested were highly resistant but the 16 japonica cultivars tested showed various degrees of resistance to leaf blast. With aging of rice plants, the quantitative levels of resistance to leaf blast increased in all the cultivars, although the levels of resistance to leaf blast varied according to rice genotypes. The leaf position of rice plants in which changed from susceptible to resistant reactions varied also with rice genotypes. The susceptible reactions of the rice cultivars to Pyricularia oryzae were distinctly changed to a resistant reaction on upper leaves of rice plants. The rice cultivars, in which the quantitative level of resistance to leaf blast was higher, were resistant on the lower leaves of rice plants. The cultivars Akibare, Palkeum, Jinheung, Olchal, Dobong and Ginga which drastically decreased blast infection at late growth stage were evaluated as adult-plant-resistant to leaf blast in the field. The adult-plant-resistant cultivars became resistant to leaf blast, both qualitatively and quantitatively, as rice plants matured.
The development of resistance to bifenazate (resistance ratio of egg=40.3 folds) was found in population of the twospotted spider mite, Tetranychus urticae, collected from rose greenhouses in Chilgok, Gyeongbuk Province in December 2000. This population was selected for 4 years with bifenazate treatment (over 150 times), and showed 248.8 folds increase in resistance as compared to susceptible (S) strain. Inheritance of bifenazate resistant strain (R) and cross resistance of this strain to 9 acaricides were investigated. There were differences of susceptibility in the bifenazate concentration-mortality relationships in F1 progenies obtained from reciprocal cross with the S and R strain $(R{\times}S,\;S{\times}R)$. Degrees of dominance were 0.48 and 0.94 in adult females and eggs of $R{\times}S$ and -0.85 and -0.17 in adult females and eggs of $S{\times}R$, respectively. Inheritance type in the F1 progeny of $R{\times}S$ was incomplete dominant, and F1 progeny of $S{\times}R$ was incomplete recessive. These results suggest that inheritance of bifenazate resistance is controlled by a complete dominance. The R strain exhibited cross resistance to acequinocyl and fenpyroximate in adult females, and amitraz, emamectin benzoate, fenpyroximate, milbemectin, pyridaben and spirodiclofen in eggs. However they showed negatively correlated cross-resistance to emamectin benzoate and milbemectin in adult females, and abamectin in eggs.
Six rice cultivars showing various types of resistance or susceptibility to Pyricularia oryzae in the field were used to study the relationship between susceptibility and contents of carbohydrates, amino acids and phenolics at different plant maturities and leaf ages under controlled environmental conditions. No consistent differences between the susceptible cultivars and adult-plant-resistant cultivars were found in terms of the contents of individual carbohydrates, total amino acids and phenolics in rice leaves throughout the plant development. Only the adult-plant-resistant cultivar Dobong had lower contents of inositol throughout the plant development than the other cultivars. The amounts of sucrose, inositol, glucose, and fructose increased in all tested cultivars at eight leaf stage than those at five leaf stage, but slightly decreased at twelve leaf stage. In contrast, total amino acids and phenolics gradually decreased during plant development. With increasing age of rice leaves, the amounts of total soluble amino acids and phenolics gradually increased in healthy fifth leaf tissues, although there were no significant differences between the cultivars. In particular, a high level of phenolics existed in old fifth leaves of the cultivar Dobong. These results suggest that increased resistance to blast of matured rice plants and old leaves may be the two different phenomina derived from the physiological changes occurring during plant development and leaf senescence, probably functioning differently in P. oryzae development.
Starch contents in healthy and powdery mildew-infected spring barley leaves at first and fifth leaf stages were measured at different intervals after inoculation. Different patterns of starch accumulation in the susceptible cultivar Peruvian and the adult plant resistant cultivar Asse were found in infected first and fifth leaves. At the early phase of infection, the amount of starch in infected first leaves was slightly changed in both cultivars. During colony development and sporulation, the decrease in starch content was .more marked in the susceptible Peruvian, whereas the amounts of starch in the adult plant resistant Asse increased during colony development and then decreased sharply before and after sporulation. In heavily infected first leaves, the susceptible cultivar Peruvian showed a drastic decrease in starch content. In the adult plant resistant cultivar Asse, the higher amount of starch retained in these infected leaves, regardless of infection intensity. The regulation of starch accumulation in mildewed barley plants at different leaf stages was discussed in relation to adult plant resistance of spring barley.
Fusarium head blight (FHB; scab) caused mainly by Fusarium graminearum is a devastating disease of wheat and barley around the world. FHB causes yield reductions and contamination of grain with trichothecene mycotoxins such as deoxynivalenol (DON) which are a major health concern for humans and animals. The objective of this research was to develop an easy seed or seedling inoculation assay, and to compare these assays with whole plant resistance of twenty-nine Korean winter wheat cultivars to FHB. The clip-dipping assay consists of cutting off the coleoptiles apex, dipping the coleoptiles apex in conidial suspension, covering in plastic bag for 3 days, and measuring the lengths of lesions 7 days after inoculation. There were significant cultivar differences after inoculation with F. graminearum in seedling relative to the controls. Correlation coefficients between the lesion lengths of clip-dipping inoculation and FHB Type II resistance from adult plants were significant (r=0.45; P<0.05). Results from two other seedling inoculation methods, spraying and pin-point inoculation, were not correlated with adult FHB resistance. Single linear correlation was not significant between seed germination assays (soaking and soak-dry) and FHB resistance (Type I and Type II), respectively. These results showed that clip-dipping inoculation method using F. graminearum may offer a real possibility of simple, rapid, and reliable for the early screening of FHB resistance in wheat.
Ethanol-soluble amino acids in healthy and powdery mildew-infected leaves of the susceptible cultivar Peruvian and the adult-plant-resistant cultivar Asse of spring barley were quantitatively analysed. At I day after inoculation, the levels of amino acids in the infected first leaves of the two cultivars were similar to those of comparable healthy controls. During sporulation, increases in amino acids were more pronounced in Peruvian than those in Asse. The changes in amino acid content in the infected first and fifth leaves were closely related to the number of colonies per leaf. The susceptible cultivar Peruvian showed higher amounts of amino acids in infected first and fifth leaves at all infection intensitives than did Asse.

To breed powdery mildew resistant varieties, 104 accessions of Cucurbita spp. germplasm were evaluated. Powdery mildew resistance and horticultural characteristics of the accessions were investigated in the field conditions. C. martinezii was selected for high resistance to powdery mildew in the field. The growth of powdery mildew pathogen and response of leaf surface tissue to the pathogen were observed after artificial inoculation to resistant C martinezii and susceptible C. moschata cv. 'Wonye No.402'. Mycelium growth was very clearly different between resistant C. martinezii and susceptible C moschata cv. 'Wonye No. 402' at 5 days after inoculation at the adult stage. Interspecific hybridization was attempted to transfer powdery mildew resistance gene(s) from C. martinezii to C. moschata. A single dominant gene action was postulated as deter mining powdery mildew resistance at the seedling stage. But for the effective selection of powdery mildew resistant individuals, it was established that adult plant resistance should be confirmed in the field conditions after the seedling test. Through the seedling and field tests, C. moschata lines resistant to powdery mildew and superior to horticultural characteristics, were selected after 2 backcrossing and 3 selfing generations. The fruit characteristics of newly developed lines were similar to those of the recurrent parent, 'Jaechenjarae'.
This study was conducted to evaluated the nature of the variety Tongil(Suweon 213-1) in resistance to the striped rice borer, Chilo suppressalis Walker, comparing with those of Rexoro(susceptible check) and TKM-6(resistant check) selected at IRRI. 1. The striped rice borer moths much more preferred the variety Tongil for oviposition than the varieties Rexoro and TKM-6. The variety Tongil and more egg masses and number of eggs than the varieties Rexoro and TKM-6, while TKM-6 having more egg masses and more number of eggs than Rexoro. This reaction was consistent throughout the test regardless of the number of tillers per hill. 2. In laboratory, preference of larvae for feeding was studied with 5cm of stem pieces of the varieties. The results showed, in contrast to the case of ovipositional preference, that the striped rice borer larvae least preferred the stems of Tongil among the test varieties, while larvae much more preferred the stems of Rexoro than those of TKM-6. 3. The larval weights at 20 days later infested on the 40 day-old plants were the lowest on Tongil among the test varieties. On the variety Rexoro the larvae had heavier body weights(43.0mg), higher pupation(64.9%) and higher adult emergence(83.3%) than those on Tongil(larval weights 30.3mg, pupation 60%, adult emergence 60.7%) and TKM-6(larval weights 35.7mg, pupation 56.3%, adult emergence 51.9%). The pupal weights, however, were not consistent among the test varieties and/or sexes in comparison with the larval weights, pupation and adult emergence above mentioned. 4. Field experiments indicated that the incidence of dead hearts at 70 days after transplanting was relatively higher on the variety Tongil(11.1%) than those on Rexoro(8.9%) and TKM-6(8.4%), and the incidence of white heads at harvest was, in contrast to the dead hearts, lower on Tongil(9.8%) than those on Rexoro(27.4%) and TKM-6(13.9%). At harvest lower larval survival observed on Tongil (49 larvae/40 hills) than those on the susceptible variety Rexoro(104 larvae/40 hills) and on the resistant variety TKM-6(70 larvae/40 hills). The average larval weights collected from three test varieties at harvest were 80.5mg from Tongil, 83.7mg from TKM-6 and 99.6mg from Rexoro. 5. Increased nitrogen fertilizer application to the variety Tongil, the striped rice borer damage was increased. Also, preference of larvae for feeding significantly increased with the increase of nitrogen fertilizer application. 6. Any specific association between the plant characters and striped rice borer resistance could not be found. The variety Tongil even having large number of tillers, short plant height, large stem, broad leaf, etc, had still high preference of moths for oviposition, low preference of larvae for feeding, low damage, and relatively high antibiosis. 7. Resistance of the variety Tongil to the striped rice borers seemed to be associated with the low feeding preference and the relatively high antibiosis, not associated directly with the ovipositional preference.

The effect of powdery mildew infection on the $^{14}CO_2$ assimilation and metabolism of $^{14}C-$assimilates was studied with spring barley cultivars, susceptible Peruvian and adult-plant resistant Asse at the four-leaf stage. No consistent differences between Peruvian and Asse were revealed in $^{14}CO_2$ assimilation and metabolism of $^{14}C-$assimilates in healthy whole plants. In the two cultivars, $^{14}CO_2$ assimilation and translocation of assimilates decreased as the number of infected leaves increased. Despite the same infection intensity, $^{14}CO_2$ assimilation was less inhibited in Asse than Peruvian. Infection reduced the fixation of $^{14}CO_2$ in noninfected fourth leaves of Peruvian more severely than that of Asse. Infection of the lower 3 leaves also inhibited the incorporation of 14 C into carbohydrates such as fructose and glucose in noninfected fourth leaves and their translocation into leaf sheathes, the inhibitions being greater in Peruvian than Asse. In the infected third leaves, there was a reduction of 14 C-activity in carbohydrates, more $^{14}C-$labeled fructose and glucose being retained in Peruvian. The stimulation of $^{14}C-$organic acid synthesis in all plant organs was more pronounced in Peruvian than Asse. Powdery mildew markedly increased the incorporation of $^{14}C$ into amino acids in infected third and noninfected fourth leaves, but reduced their translocation to the leaf sheathes. A greater rise of $^{14}C-$ activity in some amino acids in the two leaves was found in Peruvian than Asse.
