Direct Purification of Lysozyme from Hen Egg White Using High Density Mixed Mode Adsorbent

  • The high density mixed mode adsorbent known by the trade name of Mimo-AD was used to purify lysozyme directly from the hen egg white (HEW). The homogenized hen egg white was treated with the adsorbent in a stirred vessel for lysozyme adsorption, and then the adsorbent, easily separated from the HEW by sedimentation, was packed into a column. The remaining HEW and contaminant proteins were removed by washing with pH 11 distilled water in an expanded-bed state, and subsequently the elution was performed with pH 12 distilled water in a packed-bed state. By this simple and rapid adsorption, washing, and elution procedure, lysozyme was purified to>95% with an overall recovery yield of 66%. This process offers a great potential for industrial application by allowing the extraction of lysozyme while retaining the commercial value of HEW.

Adsoprtion Characteristic of Fancy Veneer Overlaid Charcoal Board Composite

  • This study was carried out to manufacture very thin natural elm veneer overlaid charcoal board for enhancing aesthetic value of charcoal board for the indoor application, and to use the advantageous properties of the charcoal as a building material for solving the sick house problem. The thin elm veneer had 26.9% opening ratio. The experiment results showed that the spreading area and the nonvolatile content of adhesive did not affect the gas adsoprtion of fancy veneer overlaid charcoal board. The natural thin elm veneer overlaid charcoal board enhanced not only the aesthetic beauty but also showed the same gas adsorption by the charcoal board.

  • Activated Rice-Hull carbon was developed to remove ammonia compounds in water matrix. Isotherm adsorption tests of ammonia were conducted using a bottle-point technique and column test. Residual ammonia after Jar-Test or passing through the column was determined by Indophenol method, and assessed the removal efficiency for ammonia of the adsorbent. As a result, the adsorption capacity for ammonia of activated racehull carbon was very larger than that of coconut shell carbon, because the rice hull carbon had the higher BET surface area of silicate. The activated racehull carbon is under the development as adsorbent to remove ammonia in drinking water and waste water.

  • A sensory element against carbaryl, as a widely used pesticide was prepared based on adsorbed acetylcholine esterase (AChE) from Torpedo california. Octadecyl was substituted on macro-porous silica, confirmed by infra-red (IR) spectroscopy and quantitatively estimated through thermo-gravimetric analysis (TGA). Immobilization of the enzyme was achieved by adsorption on this support. Activity of the immobilization product was measured as a function of the loaded enzyme concentration, and maximum binding capacity of the support was estimated to be 43.18 nmol.mg-1. The immobilized preparations were stable for more than two months at storage conditions and showed consistency in continuous operations. Possible application of the immobilized AChE for quantitative analysis of carbaryl is proposed in this study.

  • Fine dust, which is emerging as a global problem, attracted much attention recently in Korea. One of the reasons is that recent factory relocation and expansion in China affected the increase of domestic fine dust concentration due to the influence of the Yangtze river battalion and westerlies. Fine dust is a big problem not only in outdoor but also indoors. The room requires periodic ventilation and is also essential for improving indoor air quality. The fine dusts are designated as Group 1 carcinogens in WHO and the risk of fine dusts on the human body due to respiratory diseases is also increasing. In this study, the characteristics of the fine dust adsorption matrix are examined and the actual application direction is derived.

Interaction of Indigo Carmine with Cetyltrimethylammonium Bromide and Application to Determination of Cationic Surfactant in Wastewater

  • The microsurface adsorption - spectral correction (MSASC) technique has been applied to the interaction of indigo carmine (IC) with cetyltrimethylammonium bromide (CTAB). The aggregation of IC on CTAB obeys Langmuir isothermal adsorption. The results show that both the monomer complex $IC{\cdot}CTAB$ and the micellar complex $(IC{\cdot}CTAB)_{78}$ were formed. The binding constant of the monomer complex was calculated to be $K_{IC{\cdot}CTAB}$ = 2.20 ${\times}10^5L{\cdot}mol^{-1}$, and the molar absorptivity of the micellar complex was calculated to be ${\varepsilon}_{(IC{\cdot}CTAB)78}\;^{560nm}$ = 8.58 ${\times}10^5L{\cdot}mol^{-1}{\cdot}cm^{-1}$. The aggregation was applied to the determination of cationic surfactant in wastewater.

  • We have developed a high performance powder, which has a quenching efficacy not only for short-chain fatty acids and amines, but also for vinyl ketones (l-octen-3-one, cis-l,5-octadien-3-one), newly found as other key compounds in axillary malodor. By investigating various powders known to have a quenching efficacy, we finally developed a highly porous silica bead containing magnesium oxide. We found that the superior deodorant effect performed by this powder was the result of multiple effects due to both an excellent physical adsorption capability from its high porosity and a specific adsorption of vinyl ketones by magnesium in the powder. An antiperspirant formulation containing both this powder and a Morus alba extract showed good efficacy as a deodorant.

Application of Molecular Simulation Techniques to Estimation of Gas Permeability in Zeolite Membranes

  • Molecular modeling of gas permeation through zeolite membranes with/without intercrystalline region was carried out. Molecular dynamics (MD) and Monte Carlo (MC) simulations were performed to estimate the diffusion coefficient and adsorption parameters respectively, and our proposed combined method of molecular simulation techniques with a permeation theory (CMP) was used to estimate gas permeability. The calculated permeability of gases (Ar, He, Ne, $N_2$, $0_2$, $CH_4$) at 301 K for the single crystal membrane model was about one order of magnitude larger than the experiential values, although the dependence on the molecular weight of the permeating species agreed with experiments. On the other hand, the estimated permeability using the diffusivity and adsorption parameters of the intercrystalline region model was in good agreement with the experiments. The consistency between experiments and the estimated values means the importance of considering the intercrystalline region and the validity of CMP method to predict the performance of zeolite membranes.

  • There are serious issues with the application of surfactant flooding as a third recovery method, such as surfactant slug losses. In this study,the impact of the salinity gradient on the remobilization of oiltrapped in Berea sandstone was investigated by emphasizing the surfactant adsorption gradient and phase behavior to determine the optimal salinity of the chosen surfactant concentration for investigating the salinity gradient. Three salinity-gradient schemes were applied to six cores saturated with light and heavy oils. The positive salinity gradient provided the best recovery results with an in situ microemulsion formation that could be observed in the fluid collector.

  • Over the past decades, silk sericin (SS) received increasing attention in the academic and industrial fields. In nature, SS acts as a glue that holds the two strands of silk fibrils together. However, recent works suggest that SS might have a more diverse role during the silk spinning process, such as stabilizing the SF in the silk gland. On the other hand, the sericulture industry has been trying to find novel applications for SS discarded from the silk fabric manufacturing process. Recovery and refining of SS would be the first step of the recycling of SS. Using a proper solvent SS could be shaped into various forms, such as spherical beads, microparticles, fibers, and films. Moreover, the applicability of these SS materials has been investigated in various fields such as cosmetics, templates for nanomaterials, drug delivery, heavy metal adsorption, and enzyme immobilization.
