Effects of restraint stress and its modulations by ursodeoxycholic acid(UDCA) were evaluated on some biochemical and biophysical parameters in rats. Restraint stress induced elevations in blood alkaline phosphatase (ALP). cholesterol (CHOL), aspartate transaminase (GOT), alanine transaminase (GPT), lactate dehydrogenase (LDH) levels. It was also caused adrenal hypertrophy, decrease in weight of spleen and contents of ascorbic acid in stressed rats. As a results, stress indicators such as spleen, ascorbic acid, GOT, GPT, LDH were fastly changed after imposing stress, but those such as ALP, CHOL, adrenal were induced relatively later. UDCA was tested if it has an inhibitory effect against 18-hr restraint induced stress. UDCA lowered ALP, CHOL, LDH level and also effectively elevated the ascorbic acid contents in 25 mg/kg dosage of UDCA. In organ weights. the restraint stress induced increases in spleen and adrenal were attenuated by UDCA in 50 mg/kg dosage. However. stress-induced GOT and GPT levels were unaffected by UDCA.
This study was done to investigate whether cholic acid derivatives have anti-stress activity and what is a cause of this anti-stress effect. Seven cholic acid derivatives (cholic acid, taurocholic acid, ursodeoxycholic acid, tauroursodeoxychoic acid, chenodeoxy cholic acid, dehydrocholic acid, hyodeoxycholic acid) were used, silymarin and valproic acid were used as positive controls. Stress was induced by restraint immobilization technique plus water immersion (24hrs) and adrenal weight, spleen weight, adrenal ascorbic acid, serum cholesterol, lactate dehydrogenase (LDH), alkaline phosphatase (ALP), adrenal cholesterol, glucose and corticosterone levels were measured as stress indicators. Most cholic acid derivatives markedly decreased the adrenal weight, and TUDCA and DHCA increased the spleen weight. The restraint stress induced increments in serum LDH, ALP and cholesterol were attenuated by most cholic acid derivatives. Cholic acid, taurocholic acid and tauroursodeoxycholic acid only increased the content of adrenal ascorbate. While valproic acid showed an inhibitory effect against stress, silymarin did not. Our findings suggest that most cholic acid derivatives have anti-stress effect and that their anti-stress effect is, in part, related to choleretic activity.
Systematic pharmacological studies on powdered whole part of unossified antler obtained from Cervus nippon Temminck var. mantchuricus have been carried out in mice and rats. Powdered antler, with a single oral administration, showed a very weak acute and subacute toxicity; its MLD being >5g/kg orally in mice. On daily oral treatments of the antler for 14 days, it did not cause any significant differences in body weight gain, various organ weights and serum transaminase activities compared to those of the control rats. Powdered antler, with a single oral administration, showed a remarkable analgesic activity as evaluated by writhing syndrome and tail-pressure test, a weak CNS depressant activity as well as a weak immunopotentiating action as evaluated by carbon clearance test in mice. Powdered antler, with repeated treatments, showed a moderate antifatigue effect against immobilized stress and showed significant increases in both adrenal weight and its ascorbic acid content in rats, suggesting that the mode of antifatigue effect of antler is related to adrenal and its components.

Two pure saponin components, Panax saponin C (protopanaxatriol derivative, ginsenoside Re) and Panax saponin E (protopanaxadiol derivative, ginsenoside $Rb_l$) were isolated from Panax ginseng root and their acute toxicities in mice and antistress effects in rats were investigated. Average lethal doses $(LD_{50})$ of ginsenoside Re were 130mg/kg (i.v.), more than 1,000mg/kg (i.p.) and more than 1,500mg/kg (s.c.), respectively. Average lethal dose of ginsenoside $Rb_{1}$ was 243mg/kg intravenously. Adrenal ascorbic acid and cholesterol contents were significantly decreased when normal rats were exposed to heat $(40^{\circ}C)$ for 30 min. The reduction of the adrenal ascorbic acid and cholesterol contents in rats was partially prevented when the rats received the ginseng saponins prior to exposure to heat stress and most pronounced effects were observed in rats received ginsenoside Re. However, it was found that administration of ginseng alone, without stress, did not significantly change the ascorbic acid and cholesterol contents in adrenal glands. Eosinophil counts in the blood of the rats were elevated when the rats were exposed to the heat stress, and the elevation of the eosinophil counts were prevented with the ginseng saponins under the stress, but the changes were all insignificant statistically.
Capsaicin(CAP) is a pungent ingredient of hot pepper that has been used as a spicy food additive, pre-servative, and medicine. In this study, the effect of CAP on L-ascorbic acid(AsA)level in various tissues as well as its urinary excretion. and drug-metabolizing enzyme activity in rats were investigated. Rats fed AsA-deficient diets for 17days were injected intraperitoneally with 1mg of CAP in 0.5ml of ethanol-Tween 80-saline(20 :10 : 70, v/v/v). Control rats received the equal volume of the same solution without CAP. Urine was collected for 3 day after the CAP injection, and after 5 days tissues were removed; their AsA contents were measured by high performance liquid chromatography combined with and electrochemical detector. In addition, hepatic microsomal ethoxyresorfin O-deethylase(EROD) activity as measured. Urinary AsA excretion changed significantly following CAP injection. One and two days after CAP injection, the urinary AsA increased 2-and 3-fold in the CAP injected group, compared to the control, but the contents of adrenal glands and brain were lower than those of the control Dehydroascorbic acid contents in adrenal glands of the CAP injected group were higher than that of the control These results suggested that a single large dose of CAP could temporarily cause the redistribution of AsA in tissues accompanying by its urinary excretion, by affecting probably a biological system including mixed function oxygenase system(MFOS)
In order to compare the anti-stress effect of Guibitang with that of ascorbic acid, after these medicines were administered to guinea pigs induced by heating and swimming stress, the changes of the weight of body and organ, and content of plasma catecholamines, serum total cholesterol, free cholesterol, triglyceride, protein, glucose and cortisol were measured. The results were as follows : 1. The weight of the body was increased with statistical significance in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on both heating and swimming stress. 2. The weight of spleen decreased with statistical significance in the groups administered ascorbic acid, Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress, but in case of swimming stress, the weight of spleen decreased with statistical significance in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. The weight of adrenal decreased with statistical significance in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress alone. 3. Plasma norepinephrine content decreased with statistical significance not only in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid but in the group administered ascorbic acid with Guibitang as compared with in the group administered ascorbic acid on heating stress. In case of swimming stress, norepinephrine decreased with statistical significance in the groups administered ascorbic acid, Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. 4. Plasma dopamine content decreased with statistical significance only in the group administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on both heating and swimming stress. 5. Serum total cholesterol content decreased with statistical significance in the groups administered ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress, but in case of swimming stress, it decreased with statistiscal significance only in the group administered ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. 6. Serum triglyceride content decreased with statistical significance not only in the groups administered ascorbic acid, Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid but in the group administered ascorbic acid with Guibitang as compared with in the group adminstered ascorbic acid on heating stress. In case of swimming stress, triglyceride decrease with statistical significance only in the group administered ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. 7. Serum glucose content increased with statistical significance in the groups administered Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on both heating and swimming stress, particulaly in case of swimming stress, in the group administered ascorbic acid with Guibitang, it increased with statistical significance as compared with in the group administered ascorbic acid. 8. Serum cortisol content decreased with statistical significance only in the group administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress.
Three hundred gram of Korean ginseng root was extracted with 95% ethanol on a boiling water bath for about 300 hr. Evaporation of alcohol yieled 50.2g of dark brown residue which was used by dissolving 4 mg of the residue in 1 ml of physiological saline. The ginseng group and the saline group received each day 0.5 ml per 100 g body weight of ginseng extract and physiological saline, respectively. Both the ginseng and saline group with stress were exposed to positive radial acceleration (1∼29g), cold (5$^{\circ}C$, 0$^{\circ}C$ &-10$^{\circ}C$) and heat (35$^{\circ}C$) environment, and surgical stress. After termination of the last stress, the tolerance, body weight, visceral organ weight, basal metabolism rate, rectal temperature, the number of erythrocyte and leucocyte, hemoglobin level, hematocrit ratio, total serum protein content and it's fraction and the content of adrenal ascorbic acid in the experimental animal exposed to stress were measured and at the corresponding periods, the same measurements were also carried out with the ginseng and the saline groups without stress exposure (serving as control). Results obtained were as follows. 1. Administration of ginseng does depressed the decrease of the tolerance, body weight, visceral organ weight, basal metabolism rate, the number of erythrocyte, hemoglobin value, hematocrit ratio and the A/G ratio in the mice and rats exposed to various stress. 2. The change of the rectal temperature, eosinophile counts, total serum protein content and the content of adrenal ascorbic acid of ginseng group that exposured to various stress facilitates the reaction to, and accelerates the recovery from the stress. 3. Even after hypophysectomy which served the link between the central and the peripheral portion of the stress mechanism, the adrenal ascorbic acid content of ginseng group decreased significantly more than that of the saline group 30 min. after administration of ACTH, while the value approached the normal level significantly closer in the ginseng group than in the saline group 1 and 2 hr after ACTH administration. Judging from the above results, it is concluded that administration of ginseng extract tolerated the experimental animals under the environment of stressfu1 stmuli, although the ginseng has no significant influence upon the stress mechanism in the absence of stressful stimuli. The site of action of the ginseng appears to be in the peripheral portion of the stress mechanism.
Effect of combined preparation (DWP715) containing Alaska pollack extract, maltol, ascorbic acid and nicotinamide on decreasing of blood alcohol was evaluated in human blood. Treatment of DWP715 prior to administration of 25% alcohol (100 mL) decreased alcohol concentration in blood and showed significant difference after 2 hours. The pharmacokinetic parameters such as area under the concentration-time curve (AUC), $C_{max},\;T_{max}\;and\;T_{1/2}$ were also decreased and delayed when compared with control values. Effects of DWP715 on anti-fatigue and anti-oxidation activities were also studied in the restraint stress model using various parameters (GOT, GPT, LDH values and organ weights) on mild condition and examined through the content of lipid peroxide induced by 2% $CCl_4$ in mouse livers. While GPT level, thymus and adrenal weight were not influenced by DWP715 dosing, LDH, GOT level and spleen weight used as a parameter against fatigue and stress states were recovered almost to the nomal level. Furthermore, lipid peroxidation due to $CCl_4$ was significantly inhibited by DWP715 treatment. These results suggest that DWP715 seems to metabolize the blood alcohol rapidly and to restore the damaged liver and fatigue conditions which was caused by alcohol metabolism to normal condition.
It is noted that Dioscorea quinqueloba is a medicinal herb that is widely used to treat cardiovascular disease and is assessed as useful to treat other various medical conditions. The immunopotentiating effects of the protein extract (DQP-1) from Dioscorea quinqueloba were thus formally investigated in vivo under incident of cold stress. In this case study, the spleen and thymus weight in mice was shown to have decreased after a measured exposure to cold stress, while the adrenal gland weight in the mice was shown to have increased. The systematic oral administration of DQP-1 significantly recovered the weight loss of the spleen and suppressed the adrenal gland hypertrophy during the association with cold stress. Additionally, the DQP-1 also restored the ascorbic acid level in the adrenal gland reduced after cold stress. The cold stress exposure lowered the percentage of $CD4^+$ and $CD8^+$ cells in the mouse thymus as determined by the flow cytometric analysis, as well as the levels of some serum immunological cytokines(interleukin-2, interleukin-12, and interferon-${\gamma}$) in the studied mice. The resulting identified weakened immunity caused by cold stress was also recovered by a treatment with DQP-1. The DQP-1 significantly suppressed the formation of serum enzymes of alanine aminotransferase, aspartate aminotransferase, and lactate dehydrogenase, which were systematically elevated during the cold stress episode. These results indicate that DQP-1 can improve immunity in mice that are characteristically weakened under stress.

Selective. highly stable determination of epinephrine(adrenalin) was achieved in cyclic voltammetric measurement carried out at electrochemically treated conductive boron-doped diamond electrode. Boron-doped diamond electrodes were prepared on single crystal Si wafers by microwave plasma chemical vapor deposition and $B_{2}O_{3}$ was dissolved in acetone/methanol(9:1) mixture solution so that the B/C weight ratio ca. $10^{4}ppm$. Epinephrine is a kind of catecholamines, which secreted from adrenal marrow cells. The serious problem to detection of epinephrine is the interference phenomena of electroactive constituent. including AA. In this study. electrochemical treatment of BDD was carried out to discriminate between epinephrine and AA responses. Experimental results showed that the peak potential of AA oxidation shift to the positive direction and the oxidation peak of epinephrine was unchanged. The effect of electrochemical treatment was maintained up to 40hrs.
