This study was carried out to get infomation on the nitric oxide production ability and its formation mechanisms of polysaccharides extracted from Ganoderma lucidum(PSG) by using murine macrophage cell line. The cultured mycelial cells of Ganoderma lucidum were extracted by alkali, and than neutralized by acid. The extract were passed through the column of DEAE cellulose for more purification. The neutral fraction was concentrated and precipitated with 95% ethanol. The precipitate was lyophilized and PSG was obtained. The immunomodulating effects of PSG on macrophage were performed by using murine macrophage cell line ATCC TIB 71 cells with PSG 0.5mg. PSG alone could not induce the production of nitrite, but it had a significant potential effect on nitrite secretion when the cells were primed and triggered with BCG and Interferon(IFN)-${\gamma}$. Also it was prominent by using calcium channel blocker(verapamil) and adenylate cyclase activator(forskolin).
본 연구는 adenylate cyclase의 촉진제인 forskolin과 cholera toxin이 생쥐난자의 핵막붕괴 및 cAMP 합성에 미치는 영향을 조사하고자 수행되었다. 체외난자 배양방법과 adenylate cyclase assay 방법을 이용한 연구의 결과는 아래와 같다. 생쥐난자를 4시간 배양한 결과 대조군의 핵막붕괴율은 93%인데 반해서 forskolin (20-40$\\mu$g/ml)이 함유된 배양액에서 배양한 난자의 핵막붕괴율은 56-36%로써, forskolin의 농도에 비례하여 생쥐난자의 핵막붕괴가 현저하게 억제되었다. Forskolin (80 $\\mu$g/ml)을 3시간 처리한 후, 난자를 forskolin이 제거된 배양액으로 옮겼을 때 난자의 핵막붕괴율이 대조군과 비슷한 정도를 나타내고 있어 forskolin에 의한 핵막붕괴 억제현상은 가역적이었다. 한편, cholera toxin (10-1,000 ng/ml)은 생쥐난자의 핵막붕괴를 억제시키지 못했다. Forskolin (10-80 $\\mu$g/ml)을 생쥐난자 추출물에 첨가할 경우 cAMP합성이 5-18배 증가되었으나, cholera toxin (10-1,000 ng/ml)은 효과가 없었다. 덧붙여, adenylate cyclase의 regulatory unit의 촉진제인 guanidylimido-diphosphate (100$\\mu$M)를 forskolin과 함께 처리하여도 forskolin만 처리한 실험군에 비하여 cAMP합성정도에 변화가 없었다. 또한, cholera toxin과 guanidylimido-diphosphate(100$\\mu$M)를 함께 처리하여도 생쥐난자의 cAMP합성은 증가되지 않았다. 이상의 결과에서 forskolin에 의한 생쥐난자의 핵막붕괴 억제 현상은 난자내의 cAMP농도가 높아짐으로써 야기된 것이라 추측되며, 난자내의 cAMP 농도의 변화가 생쥐난자 성숙에 중요한 역할을 수행할 것이라고 사료된다.
흰쥐 해마(hippocampus)에서 acetylcholine(Ach) 유리에 미치는 $A_1-adenosine$ 수용체의 역할과 post-receptor 기전에 있어서 adenylate cyclase 계의 관여여부에 관한 지견을 얻고자 하여 $[^3H]-choline$으로 평형시킨 해마 slice를 사용하여 $[^3H]-ACh$ 유리에 미치는 여러가지 약물들의 영향을 관찰하였다. $A_1-adenosine$ 수용체 흥분제인 $N^6-cyclopentyladenosine(CPA,\;0.1{\sim}10\;{\mu}M)$은 전기자극$(3Hz,\;5\;Vcm^{-1},\;2\;ms,\;rectangular\;pulses)$에 의한 $[^3H]-ACh$ 유리를 용량 의존적으로 감소시켰다. $A_1-adenosine$ 수용체 차단제인 8-cyclopentyl-1, 3-dipropylxanthine$(DPCPX,\;1{\sim}10\;{\mu}M)$은 용량 의존적으로 $[^3H]-ACh$ 유리를 증가시켰으며, 이때 기저(basal)유리 또한 증가됨을 관찰할 수 있었고, $2\;{\mu}M$ DPCPX 전처리는 CPA의 효과를 길항하여 CPA에 의한 용량반응곡선을 우측으로 이동시킴을 볼 수 있었다. G protein 억제제인 N-ethylmaleimide$(NEM,\;10\;&\;30\;{\mu}M)$는 그 자체에 의하여 자극에 의한 ACh 유리를 증가시켰으며, 기저유리 또한 증가함을 볼 수 있었다. NEM 전처리에 의하여 CPA의 효과는 완전히 소실되었다. 한편 adenylate cyclase 활성화제인 forskolin$(0.3{\sim}10\;{\mu}M)$은 기저유리에 변함없이 용량의존적인 $[^3H]-ACh$ 유리의 증가를 초래하였으며 $3\;{\mu}M$ forskolin 전처리는 대량$(10\;{\mu}M)$의 CPA의 효과를 제외하고는 CPA의 효과를 억제시킴을 관찰할 수 있었다. 이상의 실험 결과로 흰쥐 해마의 choline 작동성신경의 presynaptic $A_1-adenosine$ heteroreceptor는 ACh 유리에 중요한 역할을 하고 있으며, ACh 유리의 조절에 Gi-단백질을 통한 adenylate cyclase 계의 관여가 확실하다 하겠다.
As it has been reported that the depolarization-induced norepinephrine (NE) release is modulated by activation of presynaptic $A_1-adenosine$ heteroreceptor and various lines of evidence indicate the involvement of adenylate cyclase system in $A_1-adenosine$ post-receptor mechanism in hippocampus, it was attempted to delineate the role of adenylate cyclase system in the $A_1-receptor-mediated$ control of NE release in this study. Slices from rat hippocampus were equilibrated with $[^3H]-NE$ and the release of the labelled products was evoked by electrical stimulation.(3 Hz, $5Vcm^{-1}$, 2 ms, rectangular pulses). The influence of various agents on the evoked tritium-outflow was investigated. $N^6-Cyclopentyladenosine$ (CPA), a specific $A_1-adenosine$ receptor agonist, in concentrations Tanging from 0.1 to $10{\mu}M$ decreased the $[^3H]-NE$ release in a dose-dependent mauler without any change of basal rate of release. 8-Cyclopentyl-1,3-dipropylxanthine (DPCPX, $2{\mu}M$), a selective $A_1-receptor$ antagonist, inhibited the CPA effect. The responses to N-ethylmaleimide $(3&10{\mu}M)$, a SH-alkylating agent of G-protein, were characterized by increments of the evoked NE-release and the CPA effects were completely abolished by NEM pretreatment. Forskolin, a specific adenylate cyclase activator, in concentrations ranging from 0.1 to $30{\mu}M$ increased the evoked and basal rate of NE release in a dose-dependent manner and the CPA effects were inhibited by forskolin pretreatment. Rolipram $(1&10{\mu}M)$, a phosphodiesterase inhibitor, did not affect the evoked NE release but reduced the CPA effect. And 8-bromo-cAMP $(100&300{\mu}M)$, a membrane permeable cAMP analogue inhibited the CPA effect significantly. These results suggest that the $A_1-adenosine$ heteroreceptor plays an important role in NE-release via nucleotide-binding protein $G_i$ in the rat hippocampus and that the adenylate cyclase system might be participated in this process.
세포내 cAMP의 농도 변화가 Toxoplasma의 성장 및 증식에 미치는 영향을 검토하기 위하여 끽접 cAMP와 cAMP의 analogue를 첨가하거나 간접적으로 세포내 cAMP의 합성이나 분해를 담당한 효소들의 활성을 변화시 킴으로써 CAMP의 농도를 조절하여 그 효과를 측정하였다. HL-60세포를 숙주 세포로 사용하여 동수의 Toxoplasma를 첨가하여 배양하였으며, 처리 효과는 배양 세포계에서 Toxoplnsma에만 특이하게 표지되는 $^3H-uracil$과 Toxoplasma및 HL-60세포에 공통으로 표지되는 $^3H-thymidine$을 각각 매 12시간마다 2시간씩 배양하여 그 표지량을 측정하여 비교 분석하였다. 직접 cAMP와 cAMP의 analogue인 dbcAMP 및 br-cAMP를 첨가하였을 때, 각각 특이한 농도에서, 즉 1 mM, 0.5~5mM 및 0.1mM에서 Toxoplasma의 성장 및 증식을 향상시켰다. 이때, 성장 및 증식을 유도하는 시기 및 그 증가도에서도 차이가 있는 것으로 나타났다. 이 결과들은 세포내 cAMP의 농도를 증가시키도록 cAMP의 합성 및 분해에 관여하는 효소들의 황성을 변화 시켰을 때에도 같은 양상으로 나타났다. cAMP의 합성 효소인 adenylate cyclase의 활성제인 pNHppG, cAMP의 분해 효소인 cAMP phosphodiesterase의 활성제인 A23187과 imidazole 및 억제제인 IBMX, compound 48/80 및 theophylline을 각각 처리하였는데, 세포내 cAMP의 농도가 증가되었을 대에는 Toxoplasmn의 성장 및 증식 을 향상시켰으나, cAMP의 농도를 감소시켰을 때에는 억제하였다. 이 때 배양계에 독성을 일으키는 농도 이하에서 농도 의존성의 경향을 보였으며, 유도 시기 및 그 증가도에는 차이가 있었다. 따라서 세포 내에서 향상된 수준의 cAMP가 어떤 기전을 활성화시키며, 그 결과 세포질에서의 Toxcplasma의 성장 및 증식을 자극하게 된다는 것을 시사하였다.
가토 해마(hippocampus)에서 acetylcholine(ACh) 유리에 미치는 cAMP의 역할에 관한 지견을 얻고자하여 $[^3H]-choline$으로 평형시킨 해마 slice 및 시냅토솜(synaptosome)을 사용하여 $[^3H]-ACh$ 유리에 미치는 여러가지 약물들의 영향을 관찰하였다. Adenylate cyclase 활성화제인 forskolin$(0.1{\sim}30{\mu}M)$은 전기 및 고농도 $K^+$ 자극에 의한 $[^3H]-ACh$ 유리를 용량 의존적으로 증가시켰으며, dbcAMP 또한 ACh 유리를 강화시켰다. Muscarine성 흥분제인 oxotremorine$(0.1{\sim}30{\mu}M)$은 전기 및 $K^+$ 자극에 의한 $[^3H]-ACh$ 유리 효과를 용량 의존적으로 감소시켰으며, 이러한 효과는 forskolin 및 atropine에 의하여 차단됨을 관찰할 수 있었다. 한편 $K^+-channel$ 억제제인 glibenclamide$(1,\;10{\mu}M)$는 자체로는 $K^+$ 자극에 의한 $[^3H]-ACh$ 유리를 약간 억제시킴을 관찰할 수 있었으며, 대량의 forskolin$(10\;{\mu}M)$에 의한 $[^3H]-ACh$의 증가효과를 감약시킴을 알 수 있었다. 이상의 실험 결과로 가토 해마의 presynaptic muscarinic autoreceptor를 통한 ACh 유리의 조절에는 세포내 cAMP의 관여가 확실하다 하겠으나, 일부 $K^+-currents$의 관여를 배제할 수는 없을 것으로 사료된다.
The role of cAMP in the differentiation process of F9 cells induced by ginsenosides was examined by performing transient transfixion assay with CRE-luciferase reporter plasmid, GR thansactivation assay with GRE-luciferase activity with or without treatment of CAMP and forskolin, an activator of adenylate cyclase, and protein klnase A assay in the presence of ginsenosides. Ginsenosides had no effect on CRE-transactivation activity, whereas retinoic acid induced the activity. When cAMP or forskolin was treated with ginsenosides, GRE-luciferase activity was further augumented by them. In addition, ginsenosides induced protein kinase A activity in the presence of cAMP. These results suggest that ginsenosides activate cAMP-dependent protein kinase A which, in turn, increase GR activity in F9 cells.

Forskolin, an activator of adenylate cyclase, was used to enhance cAMP production, and the consequence of this enhancement on the spontaneous and potassium stimulated release of dopamine and acetylcholine was evaluated. Neostriatal slices were prepared from Fischer 344 rats and after a preincubation period the release of each endogenous neurotransmitter was measured from the same slice preparation. To measure acetylcholine release the slice acetylcholinesterase (AChE) activity was inhibited with physostigmine, but the release from slices with intact AChE activity was also determined (choline, instead of acetylcholine was detected in the medium). Under both conditions forskolin induced a significant dose-dependent increase in the potassium-evoked release of dopamine. In the same tissue preparations the release of neither acetylcholine (AChE inhibited) nor choline (AChE intact) was affected by forskolin. The results indicate that the CAMP second messenger system might be involved in neuronal mechanisms that enhance neostriatal dopamine release, but stimulation of this second messenger by forskolin does not further enhance neostriatal acetylcholine release.

A possible potentiation between cholecystokinin (CCK) and secretin in amylase secretion from isolated rat pancreatic acini was investigated. Combined treatment of acini with secretin and CCK at low concentrations, which are known to be physiological, resulted in enzyme secretion larger than the arithmetic sum of their separate effects. Such a potentiating effect also occurred between secretin and A23187 (Ca ionophore), between forskolin (adenylate cyclase activator) and CCK, and between forskolin and A23187. Staurosporin (protein kinase C inhibitor) and W7 (calmodulin antagonist) inhibited markedly the potentiated amylase release induced by the agonists, but KT5720 (protein kinase A inhibitor) did not affect the potentiated amylase release. Therefore, we concluded that the action of CCK in a physiological concentration is potentiated by secretin in a physiological concentration range and vice versa, and that the intracellular mechanism necessary for the potentiation is associated with $Ca^{2+}$. However, it is uncertain what mechanisms are involved in potentiation of amylase release after CAMP and $Ca^{2+}$.
